OBJECTIVE:To study the effects of carbamylated erythropoietin(CEPO)on cardiovascular microcirculation in rats with diabetes mellitus. METHODS:Rats were randomly divided into blank control group,model group,CEPO low-dose,medi-um-dose and high-dose groups(500,1 000,2 000 u/kg)with 12 in each group. The rats in the last 4 groups were reduced diabetes mellitus model. All rats were given relevant medicine intragastrically twice a week,coronary microcirculation endothelial cells were separated after consecutive 4 weeks. Enzyme-linked immunosorbent assay was conducted to detect levels of peripheral serum prosta-cyclin (PGI2),vasoconstrictor endothelin-1 (ET-1),angiotensin Ⅱ(AngⅡ) and von Willebrand factor (vWF) of rats in each group;in vitro CCK 8 test was used to detect endothelial cell activity(OD value);real-time quantitative polymerase chain reaction was adopted to detect proliferation-related genes(Ki67,p16),poptosis-related genes(Bad,Bax),and expressions of protein vas-cular endothelial growth factor(VEGF)and AngⅠ. RESULTS:Compared with blank control group,levels of PGI2,ET-1,AngⅡand vWF in serum in model group increased;OD value deceased;Ki67,p16,Bax and VEGF expression decreased;the difference was statistically significant (P<0.05). Compared with model group,levels of PGI2,ET-1,AngⅡ and vWF in serum in CEPO low-dose,medium-dose and high-dose groups increased;OD value increased;Ki67,p16 and VEGF expression increased;expres-sions of Bad and Bax decreased;the difference was statistically significant(P<0.05). The others had no significant difference(P>0.05). CONCLUSIONS:CEPO maybe improve the coronary microcirculation function by upregulating VEGF expression in coro-nary microcirculation endothelial cells and promoting endothelial cells’regeneration.

Objective To evaluate the effectiveness and side effects of uterine arterial embolization with Pingyangmycin(a homogenous bleomycin) lipiodol emulsion(PLE) for symptomatic uterine fibroids. Methods Uterine arterial embolization with PLE was performed in 25 patients.The improvement of symptoms and uterine size changes were followed up in 3-18 months(mean 6 months) after the procedure. Results All but 2 cases were successfully treated bilaterally.Superselective angiography showed enlargement of uterine artery,accompanied by tortuous branches.The uterine size was increased.The uterus itself was significantly stained and emptied slowly. Coagulation necrosis was found in resected fibroids after embolization in 3 patients. One month after the procedure, a mean 40% reduction of uterine volume was obtained in 18 followed up cases.The clinical symptoms were relieved significantly. The main side effects were hypogastic pain(13/25),which was intense in 6 cases. Conclusion Uterine arterial embolization with PLE is a good non surgical therapy in symptomatic uterine fibroids with mild side effects.

Objective To evaluate the value of Feridex in demonstrating small focal lesions of the liver on MRI and to discuss the optimal scan time after intravenous administration of Feridex. Methods Small focal lesions in the liver were found on ultrasound or CT in 17 patients. Conventional MR plain scan and Gd-DTPA enhanced scan were performed. One to three days later, T 2 weighted imaging was repeated at 0.5 h, 3 h, and 6 h after Feridex was administered intravenously over 30-40 min at a dose of 0.05 ml/kg. The pre- and post-contrast T 2WI were compared and analyzed in terms of lesion detectability and signal change. Results Twenty-one lesions with diameter smaller than 1 cm were found on the Feridex enhanced images, and only 8 lesions were found on the conventional MR images. The small hepatic cancers, focal nodular hyperplasia (FNH), and regenerated nodules had different signal changes on the Feridex images, which made the differential diagnoses easier. The signal of the liver decreased significantly (P0.05). Conclusion Fridex enhanced MR images could not only improve the detection of small focal lesions, but also provide evidence for differential diagnoses.

Objective To study the protective effect of nalmefene hydrochloride on lung ischemia-reperfusion injury and its mechanism.Methods 40 rats were randomly divided into model group,high dose of nalmefene group,low dose nalmefene group and sham operation group equally(n =10).The lung ischemia-reperfusion model was established by occlusion of the left pulmonary hilum.The intravenous injection of nalmefene (20,10 μg·kg-1) was applied at 10 minutes before occlusion of the left pulmonary hilum in the high dose of nalmefene group and the low dose of nalmefene group,respectively.The sham operation group without occlusion of the left pulmonary hilum was not given any treatment.At 2 h after reperfusion,all rats were detected arterial blood gas value and then sacrificed.The specimens from the upper lobe of the left lung tissue were preserved to observe pulmonary lesions,detect the ratio of wet / dry weight and the expressions of β-endorphin and interleukin(IL)-17.Results Compared with the model group,the value of PCO2,the degree of pulmonary lesions,the ratio of wet / dry weight and the expressions of β-endorphin and IL-17 in lung tissue were significantly decreased (P ＜ 0.01),while the value of PO2 was significantly increased (P ＜ 0.01) in the low dose of nalmefene group.Compared with the low dose of nalmefene group,thevalue of PCO2,the degree of pulmonary lesions,the ratio of wet/dry weight and the expressions of β-endorphin and IL-17 in lung tissue were significantly decreased (P ＜ 0.01),while the value of PO2was significantly increased (P ＜ 0.01) in the high dose of nalmefene group.Conclusion Nalmefene hydrochloride may prevent lung ischemia-reperfusion injury in a dose dependent manner by reducing the production of β-endorphin and inhibiting the expression of IL-17 in lung tissue.

Objective To investigate the current situation and influencing factors of overweight and obesity in the elderly from Changsha city.Methods Using stratified sampling,2500 elderly in Changsha were included for the investigation by questionnaire and physical examination.The influencing factors were analyzed with univariate analysis and two-level models.Results Among 2500 persons,the valid questionnaires were 2254,the effective rate was 90.16％.About 39.71％ (895/2254) of Changsha's elderly were in overweight and obesity (BMI≥ 24 kg/m2).Univariate analysis showed that there were significant differences in BMI among different area,education degree,income,marital status,drinking wine,drinking tea,physical exercises and occupation (x2＝ 304.17,18.39,19.35,12.93,13.29,11.77,12.87,12.84,all P＜0.05).The results of two-level models indicated that the independent influencing factors were income,physical exercises,drinking wine,age and occupation.Conclusions It is necessary to take interventional measures against elderly obesity and to encourage the elderly taking physical exercises and keeping reasonable diets.

AIM: To investigate the changes of current and gene expression of acetylcholine sensitive K~+ channel (K_ ACh ) in chronic human atrial fibrillation (AF) and to evaluate the roles of K_ ACh expression in the occurring and maintenance of AF. METHODS: Acetylcholine sensitive K~+ currents (I_ KACh ) were recorded with the whole-cell patch clamp technique in single atrial myocyte of AF group and normal sinus rhythm group (SR group). The current densities-voltage relations were analyzed. The Kir3.4 mRNA was measured by reverse transcription-polymerase chain reaction. RESULTS: (1) Compared with SR group, acetylcholine sensitive K~+ current densities in AF group were reduced under testing potential between -80 mV and -120 mV . Objective To evaluate three different tracer-kinetic models used for dynamic contrast-enhanced MRI (DCE-MRI) data processing in the prediction of the histopathologic grade of cerebral gliomas.Methods Forty-one patients with histopathologically graded gliomas (grade Ⅱ 13,grade Ⅲ 14,grade Ⅳ 14)were imaged with DCE-MRI from March,2013 to November,2014.The values of Ktrans of gliomas were obtained by three tracer-kinetic models,which were Patlak model,Tofts model and Extended Tofts Linear model.All data were analyzed statistically by a Graphpad 6.0 statistical software.Comparison of the differences of Ktrans among different grades of gliomas was conducted using the Kruskal-Wallis test and Dunn's multiple comparisons test for the data not conform to normal distribution.Correlations of Ktrans values among those three models were analyzed using linear regression analysis,The differences of Ktrans between low grades and high grades of gliomas was conducted using the Mann-Whitney U test.Receiver operating characteristic (ROC) curve analyses were performed to determine the cut-off values for Ktrans to distinguish different low grades and high grades of gliomas.Results The Ktrans values obtained by Patlak model was 0.008(0.004-0.043) min-1for grade Ⅱ,0.021(0.005-0.088) min-1 for grade Ⅲ,and 0.035(0.017-0.061) min-1 for grade Ⅳ.The Ktrans values obtained by Tofts model was 0.085 (0.041-0.158)min 1for grade Ⅱ,0.140 (0.063-0.315) min-1for grade Ⅲ,0.229 (0.126-0.419)min 1 for grade Ⅳ.The Ktrans values obtained by Extended Tofts Linear model was 0.012 (0.004-0.092) min 1 for grade Ⅱ,0.048 (0.010-0.188) min-t for grade Ⅲ,0.094 (0.036-0.215)min 1 for grade Ⅳ.All the Ktrans values obtained by three models increased when the histological grades increased,with statistical significance between grade Ⅱ and Ⅲ (H=18.31,18.09,20.18,P＜0.05).Ktrans values among the three models had good linear correlations.The Ktrans obtained by Extended Tofts Linear model had good linear correlations with both Patlak model and Tofts model (r=0.933,0.893,P＜0.05),and the Ktrans obtained by Patlak model had less linear correlation with Tofts model (r=0.822,P＜0.05).The K values were statistically different between LGG and HGG (P＜0.01).The cut-off value of K provided good combination of sensitivity and specificity in the differentiation between LGG and HGG,and the K obtained by Extended Tofts Linear model had the best sensitivity and specificity among the three models (sensitivity 92.3％,specificity 85.7％).Conclusions K from DCE-MRI has a high performance in predicting the histopathologic grade of brain glioma,and the Extended Tofts Linear model is more suitable for the evaluation of cerebral glioma.

Objective To study ultrasonic parameters characteristics of lactation mastitis,breast cancer by using color doppler flow imaging(CDFI)and ultrasonic elastography(UE),and to explore its clinical value.Methods Retrospective analysis was conducted in the clinical data of 85 female patients (a total of 85 breast lesions).All patients were confirmed by pathology,and according to the results of pathology,they were divided into non lactation mastitis group(group NLM,a total of 28 cases),breast cancer group(group Ca,a total of 57 cases).All patients accepted CDFI and UE before treatment,and compared ultrasound parameters between the two groups.Results The proportions of class 0~I(71.43%)and RI<0.7(82.14%)in group NLM were significantly higher than those in group Ca,while the proportions of classⅡ ~Ⅲ(28.57%)and RI≥0.7 (17.86%)were significantly lower than 75.44%,84.21% in group Ca(χ2 =17.185,35.217,all P<0.05).The UE ratings(1.75 ±0.97)and the strain rate ratio (1.64 ±0.83)in NLMgroup were lower than (4.19 ±0.74),(5.03 ±1.08)in group Ca(t=-12.873,-14.623,all P<0.05).The accuracy of CDFI +UE(89.41%)was higher than the accuracy of UE(76.47%)or the accuracy of CDFI(67.06%)(χ2 =12.337,P<0.05).Conclusion CDFI,UE have a certain diagnosis ability for non lactation mastitis and breast cancer,combining both can obtain better diagnostic value,which is worthy of clinical application.
