1.The clinical significance of detecting the activites of T cells and NK cells in the peripheral blood of patients with colorectal cancer

Objective To investigate the relationship between immune condition and stages of tumor in patients with coloreetal cancer by detecting the activites of peripheral blood T cells and NK cells. Methods The activites of T cells and NK cells in peripheral blood from 90 patients with cancer of colon and 40 healthy controls were detected by flow cytometry. Results The CD3+ ratio,CD4+ rati-o,CD4+/CD8+ and NK cells were decreased in cancer of colon group. CD8+ ratio was increased com-pared with that of healthy control group. This chang became more significant following by accreseence of Dukes classification. Conclusion The cellular immune function disorder exists in cancer of colon pa-tients which a aggravation with the pathogenetic condition progress.
3.Clinical application of computer-aided electromagnetic imaging navigation in nasal endoscopic surgery

Objective To evaluate the clinical application of computer-aided electromagnetic imaging navigation in nasal endoscopic surgery.Methods Twenty-two cases of nasal endoscopic surgery with intraoperative imaging navigation were retrospectively reviewed,including 16 cases of sinusitis with or without polyp;5 cases of nasal inverting papilloma; 1 case of maxillary capillary hemangioma.All cases were operated with computer-aided electromagentic imaging navigation and nasal endoscope.Results The preoperative preparing time would take 4-10 minutes.In 22 cases,the localization accuracy between 3-D image landmarks of navigation system and actual anatomical landmarks was less than 1 mm.The optic nerve and other anatomical landmarks could be orientated accurately in intraoperative procedures.No complication occurred.Conclusions Nasal endoscope combined with computer-aided electromagnetic imaging navigation provides accurate anatomical localization of nasal cavity,sinuses and anterior skull base.It could improve the effectiveness and decrease surgical complications,especially in complicated cases.
4.Mechanism of dermal fibroblasts transdifferentiation into myofibroblasts induced by estrogen

Objective To explore the possible pathway and regulatory mechanism of dermal fibroblasts' transdifferentiation into myofibroblasts induced by estrogen. Methods Human dermal fibroblasts were divided into six groups (A: control; B: estrogen; C: estrogen + ICI-182780; D: estrogen + SB203580; E: estrogen + PD98059; F: estrogen + SP600125). The cells were collected for RNA extraction and the expression of α-SMA was detected by real time quantitative RT-PCR. Some cells were analyzed by single cell RT-PCR to detect positive expression percentage of α-SMA.Results The expression and positive rate of α-SMA in estrogen group were significantly increased (Group B vs. Group A, 7. 385±0. 246 vs 1. 367±0. 034, P<0.01) and those in ICI-182780 group and SP600125 group were significantly inhibited (Groups C and F vs. Group B, 4. 619 ±0. 164,2. 409±0. 091 vs 7. 385±0. 246, P<0. 05). Conclusions In the process of fibroblast transdifferentiation into myofibroblasts induced by estrogen, estrogen β receptor and JNK-MAPK signal transduction pathway may play an important role.
5.ANALYSIS OF PYROLYSIS GAS CHROMATOGRAPHY OF SALMONELLA TYPHI

The chromatograms of pyrolysis gas chromatography (PGC) of fifteen strains of Salmonella typhi were reported to see if there is any possible relation of the graphs to serological typing and drug-resistance spectra of the bact. In the fifteen strains there were fourteen clinical strains and one reference strain (Hg01). The results indicated that the clinical strains might be divided into group A and B (7 strains each) by PGC. The graphs drawn from group A organisms were entirely the same in shape while those of group B showed only basic similarity. According to the diversity of graphs of group B organisms, the group B organisms, may be subdivided into group B1 (4 strains), B2 (2 strains) and B3 (1 strain). The graphs within each subgroup were completely the same and those of subgroup B1 exhibited close similarity to that of the reference strain. Albeit no regular connection of chromatograms with serological types and drug-resistance could be seen, the graphs yet provided useful reference for founding a bank of chromatograms, compaed with which salmonella typhi graph could then be identified.

Objective To investigate the correlation between vertical infection of hepatitis B virus(HBV) and Hofbauer cell in placenta.Methods Placental structural change,the correlation between HBV infection and Hofbauer cell in 14 vertical transmission cases(HBsAg and HBV-DNA seropositive in both mothers and their neonates),62 non-vertical transmission cases(HBsAg and HBV-DNA seropositive in mothers,but HBsAg and HBV-DNA seronegative in their neonates) and 10 controls(HBsAg and HBV-DNA seronegative in both mothers and their neonates) were determined by inmumohistochemical techniques and photomicroscopy.Results 1.necrosis,edema,villi capillary sclerosis,intravillifibrosis,fibrinoial sendimentation,morphokarrioleukocyet,lymphacyte,granulocyte were observed in placentas both vertical transmission cases and non-vertical transmission cases.2.Placental HBsAg postive rate in vertical transmission cases was 100%,but 58% in non-vertical transmission cases.There was significant difference in two groups(P=(0.013)).The positive sign was observed in trophoblast cell,Hofbauer cell and endothelial cell.3.Total Hobauer cell and ones combined with HBV in placentas of vertical transmission cases obviously increased(P
7. Protective effects of superfine powder of Erigeron Breviscapus on chronic renal failure rats induced by adenine

Objective To observe the effects of superfine powder of Erigeron breviscapus on chronic renal failure (CRF) in rats induced by adenine. Methods SD rats were randomly divided into control group, model group, superfine powder and common powder of E. breviscapus groups, 10 rats in each group. The control group received routine feeding, while the other three groups received intragastrically (ig) with adenine (250 mg/kg) for 21 d to establish rat CRF models. Then, the rats in the control group and model group were received ig with equal volume of distilled water. Also, the rats in the superfine powder and common powder of E. breviscapus group were administered ig with E. breviscapus (3 g/kg) for 42 d. During the experiment, the general condition of each group of rats were observed and the levels of serum creatinine (Scr) and blood urea nitrogen (BUN) were determined. Expressions of transforming growth factor-β1 (TGF-β1) and plasminogen activator inhibitor-1 (PAI-1) at mRNA level were determined by qRT-PCR. Concentrations of kidney tissue inflammatory cytokines tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were determined by ELISA. Moreover, the kidney tissue was taken for histopathological observation. Results The rats in CRF model group after treatment with superfine and common powder of E. breviscapus appeared different levels of decrease of Scr, BUN, TNF-α, IL-1β, and mRNA expression of TGF-β1 and PAI-1 (P < 0.05). The pathologic changes of kidney tissue were also alleviated in different degrees. The treatment effect of superfine powder of Erigeron breviscapus was better than that of common powder group. Conclusion E. breviscapus has a good renal protective effect in rats with CRF. Furthermore, the superfine powder is more effective than the common powder in the protective effects of renal.

9.Changes of VEGF protein and mRNA in the vulva of rabbits after ultrasound irradiation

Objective To investigate ultrasound as a therapy for white lesions of the vulva.Methods Immunohistochemistry and hybridization in situ were used to study changes in VEGF protein and mRNA in the vulva skin of rabbits after ultrasound irradiation.Results After irradiation,VEGF protein and mRNA had increased markedly after 4 and 7 days,but their levels recovered by day 14.Conclusion Ultrasound irradiation causes local edema and hypoxia which enhance production of VEGF.VEGF promotes growth of local microvessels and increases the permeability of the endothelial cells of blood vessels.These changes improve nutrition of the mierovessels and nerve endings and ameliorate the microenvironment for tissue growth.One could speculate that ultrasound irradiation helps rehabilitation and regeneration of pathologic vulva skin.
10.Expression of Beclin1 in osteosarcoma and the effects of down-regulation of autophagy on the chemotherapeutic sensitivity.

To explore the expression of Beclin1 in osteosarcoma and investigate the effects of down-regulation of autophagy on the chemotherapeutic sensitivity to cisplatin (DDP), the expression of Beclin1 in 28 specimens of osteosarcoma (group A) and 19 specimens of normal bone tissues (group B) were immunohistochemically detected. The expression of Beclin1 mRNA in MG63 cells treated with different concentrations of DDP was examined with RT-PCR. After down-regulation of autophagy in MG63 cells by an autophagy inhibitor, 3-methyladenine (3-MA), the cell proliferation inhibition rate of MG63 cells treated with DDP was evaluated by using the MTT assay. The positive rates of Beclin1 were 67.85% in group A and 94.73% in group B. Its expression was lower in osteosarcoma than in normal bone tissues, with a significant difference found between them (P<0.05). RT-PCR showed that the expression of Beclin1 mRNA in the cells treated with high-dose DDP were higher than that in the non-treated cells, and no significant difference in the expression of Beclin1 mRNA was found between the cells treated with low-dose DDP and the non-treated cells. There was a positive correlation between the level of Beclin1 mRNA expression and the concentration of DDP. MTT assay showed that the proliferation inhibition rates of the cell treated with 3-MA and DDP combined were substantially increased when compared with those treated with DDP alone (P<0.01). This study demonstrated that autophagy may be implicated in the carcinogenesis of osteosarcoma, and DDP may induce autophagy in the MG63 cells. It also suggests that the down-regulated autophagy could increase chemotherapeutic sensitivity of DDP to osteosarcoma.
