Atrial fibrillation ( AF) is the most common sustained arrhythmia in clinical practice and it is associated with an in-creased thromboembolism risk , due mainly to embolism from the left atrial appendage ( LAA) .Percutaneous left atrial appendage clo-sure ( PLAAC) provides a valid alternative to oral anticoagulation ( OAC) mainly in patients who cannot tolerate this therapy due to a high bleeding risk .Recent studies showed PLAAC can substantially reduce stroke incidence .This article reviews the safety and efficacy of PLAAC preventing thromboembolism by retrospectively analyzing related studies of PLAAC .

Objective To observe the effects of PM2.5 exposure on A549 cells, and explore the mechanism of toxicity. Method The concentra?tions of PM2.5 exposure were determined utilizing cell viability assay. The morphological characteristics of exposed cell were observed with trans?mission electron microscope. Using big date from RNA?Seq and qRT?PCR,the mechanism was explored. Results The final concentration of PM2.5 exposure was 50μg/cm2;morphological detection showed that chromatin condensation after exposure which was also found on the boundary of nuclear membrane. KEGG pathway analysis and interaction network were finished ,and 8 kinds of apoptosis?related genes were found to be in?volved in the process of damage. Conclusion PM2.5 induces apoptosis in A549 cells by stimulating the changes of apoptosis?related genes ex?pressions.

Medical Journal of Chinese People's Liberation Army 2016;41(1):12-16
Objective To investigate the role of Tmubl protein in liver cell proliferation. Methods Using silenced lentivirus (LV)-Tmubl vector, over-expression LV-Tmubl vector, and corresponding empty vectors to transfect rat’s liver cell line BRL-3A to obtain stable transfected cells, which bear the expression of Tmubl protein as detected by Western blotting, cell proliferation with MTT, and cell cycle with flow cytometry. Results The Tumbl protein expression in the hepatocytes transfected by LV-Tmubl was increased significantly and that by LV-Tmubl-RNAi decreased significantly, with a statistically significant difference compared with normal hepatocytes (P<0.01). The proliferation was fastest in hepatocytes transfected by LV-Tmubl and slowest in those by LV-Tmubl-RNAi, and the difference was statistically significant (P<0.01). The G2+S phase was longer in hepatocytes transfected by over-expression LV-Tmubl vectors than in those by silenced LV-Tmubl vectors, and the difference was statistically significant (P<0.01). Conclusion Tmubl protein can promote the hepatocyte proliferation in BRL-3A cell line.
6. Research on the interaction of protein tmub1 and securin after partial hepatectomy
Medical Journal of Chinese People's Liberation Army 2014;39(11):864-867
Objective To investigate the interaction of Tmub1 and securin protein after partial hepatectomy. Methods A total of 30 adult male SD rats were randomly divided into 3 groups (10 each) according to the time after hepatectomy, i.e, 0, 24 and 48 h. Partial hepatectomy was performed in the rats in the first group, and then the primary hepatocytes were directly harvested by liver perfusion. Partial hepatectomy was also performed in the rest two groups at postoperative 24 and 48 h. Then the primary hepatocytes were also harvested respectively after liver perfusion. One part of primary hepatocytes was cultured for one night and for the observation of the colocalization of Tmub1 and securin protein with immunofluorescence. The other part of hepatocytes were used for co-immunoprecipitation and Western blotting to detect the expression of protein Tmub1/securin. Results Immunofluorescence and confocal laser scanning microscopy showed that there were almost no Tmub1 and securin proteins in the nucleus 0 h after liver resection, while a great amount of Tmub1/securin proteins were found to be co-localized in the nucleus 24 and 48 h. Co-immunoprecipitation and Western blotting showed that the expression of Tmub1/securing protein significantly enhanced in 24-48 h after operation compared with that at 0 h (P<0.01), while the difference between 24 h and 48 h showed no statistically significant difference (P>0.05). Conclusion Tmub1 hardly combines with securin protein in normal hepatocytes, but it combines with securin protein when it migrates from cytoplasm into nucleus after hepatectomy, thus inhibiting the proliferation process of hepatocytes.

Objective To investigate the correlation between vertical infection of hepatitis B virus(HBV) and Hofbauer cell in placenta. Methods Placental structural change, the correlation between HBV infection and Hofbauer cell in 14 vertical transmission cases(HBsAg and HBV-DNA seropositive in both mothers and their neonates),62 non-vertical transmission cases(HBsAg and HBV-DNA seropositive in mothers, but HBsAg and HBV-DNA seronegative in their neonates) and 10 controls(HBsAg and HBV-DNA seronegative in both mothers and their neonates) were determined by inmumohistochemical techniques and photomicroscopy. Results 1.necrosis,edema,villi capillary sclerosis, intravillifibrosis, fibrinoial sendimentation, morphokarrioleukocyet, lymphacyte, granulocyte were observed in placentas both vertical transmission cases and non-vertical transmission cases.2.Placental HBsAg postive rate in vertical transmission cases was 100%,but 58% in non-vertical transmission cases. There was significant difference in two groups(P=(0.013)).The positive sign was observed in trophoblast cell,Hofbauer cell and endothelial cell.3.Total Hobauer cell and ones combined with HBV in placentas of vertical transmission cases obviously increased(P

A PCR method was used to amplify the sequence encoding the mature peptide of?-mannanase of Bacillus subtilis. The gene was inserted into the Pichia pastoris vector pPIC9K, downstream of?-factor signal peptide sequence. The resultant recombinant plasmid pPIC9K-MAN was lineared by BglII digestion and introduced into the host Pichia pastoris GS115 by PEG method. After screen, the recombinant P. pastoris strain MAN22 was obtained and fermented in large scale 5L fermenter. The recombinant mannanase activity could reach to 1102IU/ml. The properties of the recombinant mannanase were characterized.

Objective: To observe the clinical efficacy of Tuina (Chinese therapeutic massage) plus needling the distal acupoints along the pathway of meridians for shoulder periarthritis (SP). Methods: A total of 70 patients with SP were divided into an observation group and a control group using the random number table method, with 35 cases in each group. The patients in the observation group were treated by Tuina combined with needling distal acupoints along the pathway of meridians, while those in the control group only received the same Tuina treatment as in the observation group. The visual analog scale (VAS) and shoulder joint function were scored before and after treatment, and the efficacy after treatment was evaluated. Results: After three courses of treatment, the total effective rate of the observation group was 94.3%, while the total effective rate of the control group was 80.0%, showing a statistical difference (P<0.05). After treatment, the VAS scores of the two groups were significantly lower than the baseline scores (P<0.01), and the VAS score of the observation group was lower than that of the control group (P<0.01). After treatment, the total scores of internal rotation, external rotation, back touching, ear touching and motion function in the two groups were significantly higher than those before treatment (P<0.01), and the above five scores in the observation group were significantly higher than those in the control group (P<0.01). Conclusion: Tuina combined with needling the distal acupoints along the pathway of meridians is more effective than Tuina alone in treating SP. The combined therapy can relieve the pain and improve joint function more effectively.
