1.Establishment of heterotopic cardiac transplantation model in mice

Objective To establish a mouse model of heterotopic heart transplantation and construct an animal model for the study of transplantation immunity.Methods According to the surgical procedures of Ono's pattern in mice cardiac heterotopic transplantation model,50 Kunming mice were divided into donors and recipients randomly.The donor heart aorta and the recipient ventral aorta,the donor pulmonary artery and the recipient inferior caval vein,were anastomosed by using the end-to-side suture technique respectively.Results The mouse model of heterotopic heart transplantation was established successfully with a success rate of 84% (21/25).The average time of donor operation and recipient operation was (11.0 ± 1.0) min and(60.0 ± 2.0) min respectively.Conclusions In the base of mastering the technique of operation facility,the model of mice heart heterotopic transplantation would be established successfully.
4.Reconstitution of Escherichia coli Fatty Acid Biosynthesis Reaction In vitro

Seven genes (fabD, fabG, fabH, fabA, fabZ, fabB and fabI) of E.coli fatty acid biosynthetic enzymes were cloned by PCR amplifying and appropriate expression vectors were constructed. Under induction assay expression of plasmid encoded proteins was carried out in strain BL21(DE3) and seven enzymes were purified using Ni-NTA agarose resin. In the absence of [2-14C] malonyl-CoA fatty acid synthetic reaction was reconstituted in vitro by adding seven enzymes and co-factors. And several model reactions were established for identification of special fatty acid biosynthetic enzymes. Meanwhile Clostridium acetobutylicium FabZ function was characterized by this method.
5.Relationship and conversion between persisters and antimicrobial resistant bacteria

Persisters are a sub-population of bacteria that can survive lethal concentrations of antibiotics.They contribute to recurrent or intractable chronic infections.Persisters can emerge as a result of multiple mechanisms which lead to drug tolerance.Unlike antimicrobial resistance which usually acquires genetic mutation, phenotypes of persisters are non-inheritable.Despite the distinct difference between persistence and resistance, recent studies have demonstrated that not only persistence can be observed in resistant mutants, but also persisters themselves can be regarded as an intermediate state which promotes the emergence of resistance.This review focuses on mechanisms of drug tolerance in persisters, phenomena of persistence in antimicrobial resistant bacteria and possible mechanisms of the conversion of persisters to resistant bacteria.Moreover, future research directions are also prospected is this review.
7.Risk factors for surgical site infection following ankle joint and Pilon frac-ture surgery

Objective To evaluate the risk factors for surgical site infection(SSI)following ankle joint and Pilon fracture surgery,and provide theoretical basis for the prevention of postoperative SSI.Methods Clinical data of pa-tients who underwent ankle joint and Pilon fracture surgery in a hospital between June 2005 to May 2013 were sur-veyed retrospectively,risk factors for SSI were analyzed.Results Among 356 patients with ankle joint and Pilon fracture surgery,22 developed 25 times of SSI,SSI rate was 6.18%,case infection rate was 7.02%.Univariate analysis showed that elderly patients (> 60 years old),history of diabetes,pre-operative calcaneal traction, perioperative irrational antimicrobial use,incision type,and long duration of operation (>3 h)were risk factors for SSI following ankle joint and Pilon fracture surgery (all P <0.05).Multivariate logistic regression analysis showed that the independent risk factors for SSI were incision type(OR,3.58[95%CI ,3.24-12.07]),history of diabetes (OR,2.75[95%CI ,1 .54-4.75]),duration of operation(OR,2.15[95%CI ,1 .32-3.64]),and patients age(OR, 1 .68[95%CI ,1 .25-2.37]).Conclusion Occurrence of SSI following ankle joint and Pilon fracture surgery is related to multiple factors,corresponding prevention and control measures should be taken to reduce the occurrence of SSI.
8.The Effects of Autologous Skeletal Myoblasts Transplantation on Hurted Vocal Cord of Paralysis

Objective The goal of the present experimental study is to investigate effects on the injection of a-dult rabbit autologous skeletal myoblasts into the thyroarytenoid(TA)muscle after recurrent laryngeal nerve (RLN) damage and traumatic vocal cords .Methods 20 adult New Zealand white rabbits were used for the establishment of an animal model simulating the cut -off of the left recurrent laryngeal nerve and the left vocal cord trauma scar for-mation after traumatic damage .Then from the rabbits ,the sternocleidomastoid muscle cells were cultured ,separa-ted and expanded .After 12 weeks ,the 20 animals were randomly divided into group A ,group B with 10 in each group .Group A(myoblast injection group) in the left vocal cord thyroarytenoid muscle injection of myoblasts 0 .3 ml ,the right vocal cord without any treatment served as normal controls ;Group B(saline injection control group) in the left vocal cord thyroarytenoid muscle was injected with normal saline ,the right vocal cord without any treatment as normal controls ,respectively ,were observed after 12 weeks of bilateral thyroarytenoid muscle fiber diameters and vocal cord volumes .Results After 12 weeks ,Group A's left thyroarytenoid muscle injection of bilateral thyroaryten-oid muscle fiber diameter and the vocal volume size was not significantly different (P>0 .05) ,thyroarytenoid muscle fiber diameter and the vocal cord volume on Group B's injected side was significantly smaller than Group A (P<0 .05) .Conclusion The thyroarytenoid muscle injection of autologous myoblasts can effectively treat recurrent la-ryngeal nerve (RLN) injury and traumatic vocal cords .

10.Comparison of the therapeutic effect of bed intermittent traction and sitting sustained traction on cervical spondylosis

ObjectiveTo observe the therapeutic effects of bed intermittent traction and sitting sustained traction on cervical spondylosis.Methods64 patients were randomly divided into two groups, bed intermittent traction group(n=32)and sitting sustained traction group(n=32). Patients were treated with bed intermittent traction and sitting sustained traction combined with medium frequency electrostimulation. After the treatment, the therapeutic effects were assessed and compared. ResultsThe rate of very efficiency and efficiency of two groups was 59.38%, 96.88% and 46.88%, 93.75%, there was no significant difference in two groups (P>0.05). ConclusionsThe bed intermittent traction and the sitting sustained traction are efficiency for treating the cervical spondylosis.
