AIM: To study the effects of arsenic trioxide (As_2O_3) on mouse airway fibroblast (FB) proliferation and expression of c-myc and c-sis in cultured FB. METHODS: The cultured FB was divided into seven groups. NS, dexamethasone (1.0 ?mol/L) and As_2O_3 at concentrations of 0.25, 0.5, 1.0, 2.0, 4.0 ?mol/L were instilled respectively into cultured FB. The effects of different concentration of As_2O_3 and dexamethasone on cell growth were observed at 1, 3, 5 and 7 day. The changes of cell cycle and the positive expression rate of c-myc and c-sis were examined by flow cytometry (FCM). RESULTS: Various concentrations of As_2O_3 significantly inhibited the proliferation of FB in vitro and showed a dose-dependent and time-dependent tendency. Data from FCM indicated that G_1-phase cell percentage increased and G_2/M-phase cell percentage decreased with the increase in As_2O_3 concentrations. The expression of c-myc and c-sis was significantly inhibited by As_2O_3 (2.0, 4.0 ?mol/L). CONCLUSION: As_2O_3 inhibits FB proliferation by downregulating the expression of c-myc and c-sis.

Objective To explore the prognostic value of 24 h ambulatory blood pressure monitoring (ABPM)for target organ damage (TOD) in the elderly hypertension patients. Methods Two hundred and thirty two elderly hypertension patients who were involved at least one target- organ damage experienced 24 h ABPM. All subjects were randomly divided into 3 groups according to the number of TOD and the clinical situation of cardiovascular, cerebrovascular and renal complications. 24 h ABPM recordings of each group were compared with one another. Results The number of TOD were highly correlated with 24 h average systolic pressure, nighttime average diastolic pressure, abnormal circadian rhythm and pressure burden. The protective effect of selenium against irradiation were studied by measuring the content of lipid peroxide (LPO) and activity of glutathione peroxidase (GSH-Px) in liver and spleen of BALB/C mice. The results showed that LPO contents in liver and spleen of whole-body-irradiated mice were higher than those of the control group on the third day after irradiation with 3 Gy 60Co ?-ray, while the GSH-Px activity of these tissues was significantly decreased. However, when the animals were given sodium selenite solution 0.2ml (12.5?g selenium) per day via mouth successively for 7 days before irradiation, there were remarkably reduced LPO level and increased GSH-Px activity in liver and spleen of irradiated mice. It is suggested that selenium supplementation may prevent mice from radiation injury by reducing LPO level and increasing GSH-Px activity in liver and spleen.

Objective To construct baculovirus expression system for the human plasminogen kringle 5(hK5) gene,and detect its expression in Spodoptera frugiperda 21(sf21). Methods Baculovirus Bacmid-rhK5 was prepared with bac-to-bac baculovirus expression system,and sf21 cells were infected.The expression of rhK5 in sf21 cells was determined by Western blot.The activity of purified protein was detected by vascular endothelial cell inhibition test. Results The pFastBac HTB-rhK5 was successfully constructed,and sf21 cells were transfected with the constructed vector.The output of rhK5 obtained was 90 ug/L,and the protein possessed the inhibitory activity of endothelial cell proliferation.The median effective dose(ED50) was 4 ug/mL. Conclusion The rhK5 baculovirus expression vector is highly expressed in sf21 cells,which lays a foundation for the the expression of rhK5 protein with biologic activities.

Objective It is well known that vaccines and national immunization program have helped to protect peoples,especially the children from infectious diseases such as smallpox,polio,measles and pertussis across the globe.However,adverse events may occur following immunization,which may lead to public concerns about the vaccine safety.Such concerns may threaten the success of an immunization program.Prompt and appropriate responses to adverse events can help to avoid the negative impacts to the National Immunization Program of China.
