3.The Clinical Applicaton of MRI in the Changes of the Brain of Neonatal Asphyxia

Objective To study the clinical application of MRI in neonatal asphyxia.Methods There were twenty-two cases of full term infants(≤one year old),they all had asphyxia during delivery.Transverse section on TSE T2-weighted,TSE T1-weighted and FLAIR sequences,and sagittal section on TSE T2-weighted were obtained on MRI.Results The abnormal signal intensity in white matter on MRI were found in 15 cases.The volume of white matter was decreased in three cases.Subarachnoid spaces of frontal and temporal lobe were enlarged in two cases.Two cases were normal on MRI.Conclusion We believed that the changes of the white matter of brain in neonatal asphyxia can be showed on MRI,it is of important significance for clinical diagnosis and treatment.
4.The correlation of HIF-1α,MACC1 and FOXO4 in gastric carcinoma pathology and prognosis

Objective To explore significance and impact of hypoxia inducible factor 1α,metastasis associated with colon cancer 1 and transcription factor FOXO4 in gastric carcinoma and clinical pathological characteristics and prognosis .Methods 110 samples diagnosed as gastric carcinoma and 46 samples diagnosed as pericarcinomatous tissue which was 5 cm far away from cancer were collected in first affiliated hospital of Anhui medical university from December 2006 to May 2008 .Immunohistochemical stai-ning was performed in these samples to detect the expressions of HIF-1α,MACC1 and FOXO4 .And the correlation between devel-opment of gastric carcinoma and expressions of HIF-1α,MACC1 and FOXO4 was analyzed with the help of patients′messages .Ka-plan-Meier method and Cox regression were used to analyze patients′5 years survival rate and the factors .Results The expression levels of HIF-1α(P<0 .001) and MACC1(P<0 .001) in gastric carcinoma tissues were significantly higher than pericarcinomatous tissues ,however the expression levels of FOXO4(P<0 .001) was significantly decreased in gastric carcinoma tissues .Statistical a-nalysis revealed that the expressions of HIF-1αand MACC1 were apparently increased with the lower degree of cell differentiation , the greater degree of tumor infiltration ,accompanied with lymph node metastasis and higher TNM scores ,but lowered the expres-sion of FOXO4 factor .There were significant correlations between expressions of the three factors in gastric carcinoma .The expres-sions of HIF-1α,MACC1 and FOXO4 had an apparent influence on the patients′5 years survival rate .And they were independent factors on the prognosis of gastric carcinoma .Conclusion HIF-1α,M ACC1and FOXO4 are abnormal expression in gastric cancer tissue ,and the clinical pathological features of gastric cancer and the prognosis of patients with gastric cancer are closely related .
5.EFFECTS OF MITOGEN-ACTIVATED PROTEIN KINASE ON FREE CALCIUM IN CULTURED HUMAN FIBROBLAST

To investigate the effects of basic fibroblast growth factor (bFGF) on intracellular free Ca 2+ of heating injury, and observe the relationship between mitogen activated protein kinases(MAPKs)signal pathways and Ca 2+ mobilization. Cultured human fibroblasts were heated at 45?C for 10min, then divided into four groups :①basic fibroblast growth factor (10ng/ml) treatment; ②preincubated cells with PD98059 (10?mol/L) for 30 min followed by bFGF (10ng/ml); ③preincubated cells with SB203580 (10?mol/L) for 30min followed by bFGF (10ng/ml); ④preincubated cells with PD98059 (10?mol/L) and SB203580 (10?mol/L) for 30min followed by bFGF (10ng/ml). The cells were incubated with fluorescence Ca 2+ dye fluo 3/AM at 37 C for 30min, then measured by using laser scanning confocal microscope. The results showed fluorescent intensity of fibroblast heating injury was weak, the concentration of intracellular free Ca 2+ increased after stimulation with bFGF treatment. PD98059 and SB203580 could induce calcium oscillation. A rapid decrease of fluorescent intensity was observed after cells were preincubated with PD98059 and SB203580 at the same time. bFGF induced an increase of cytoplasmic and intracellular free Ca 2+ concentrations. It is suggested that MAPKs signaling pathway has a feedback regulation for free Ca 2+ mobilization.
6.Development of gene therapy in wound healing

The basic concept of gene therapy is to introduce a therapeutic gene into a cell, whose expression can improve to healing of wound. To achieve this goal, the suitable therapeutic gene has been selected and delivered into the reparative cell, which is becoming a focal point works about gene therapy in wound healing. There have been several different therapeutic genes and gene transfer strategies that have been used in models of wound healing. This article discusses several methods that have been used to deliver genes encoding growth factor proteins, stem cells into wounds and the advantages/disadvantages of each approach. We hope a safe vectors system to deliver the effectual transgene in wound healing.
7.A cross-sectional study on the prevalence of depressive, anxiety disorder in outpatients with irritable bowel syndrome and functional dyspepsia in urban China

Objectives This study was to describe the prevalence of depressive and/or anxiety symptom and disorder in outpatients with irritable bowel syndrome (IBS) or functional dyspepsia (FD) in the comprehensive hospitals in big cities, China. Methods This was a hospital-based cross-sectional study conducted in three main cities in China in 2004. Six hundred and eight eligible subjects with IBS or FD were recruited consecutively from the outpatient departments within 3 months. Face-to-face interview was used in data collection together with the self-completed HAD scale for depressive and/or anxiety symptom screening. Subjects getting a HAD score of 9 and above were further assessed for depressive and/or anxiety disorders with HAMA scales and HAMD scales by the licensed psychologists or psychiatrists. Results The prevalence of depressive and/or anxiety symptoms was 24. 8% and 23. 6% respectively in outpatients with IBS or FD. And among detected depressive and/or anxiety symptoms, the prevalence of depressive and/or anxiety disorder was 69. 6% and 68. 7% respectively; Less than 12% of subjects had obtained a diagnosis of depressive disorders and been treated, and less than 8% of subjects had obtained a diagnosis of anxiety disorders and been treated prior to the investigation. The prevalence of anxiety symptom in female patients with IBS was significantly higher than male patients (17. 0% vs. 8. 7%; X2=4. 432,P = 0. 035); among detected depressive and/or anxiety symptoms, the prevalence of depressive disorder, anxiety disorder, depressive and anxiety disorder and depressive and/or anxiety dis order in female patients with IBS were higher than those in male patients (52. 6%vs. 25. 0% ,X2 = 4. 612, P=0. 032; 79. 5% vs. 53. 8%,X2=4. 815, P = 0. 028; 51. 4% vs. 21. 7%,X2=5. 112,P = 0. 024; 78. 6% vs. 55. 6%,X2=4. 112,P = 0. 043). Conclusions It is important for healthcare providers and health policy makers to note that the prevalence of depressive and/or anxiety symptoms and depressive and/or anxiety disorders are high and the percentages of previous diagnosis and treatment in outpatients with somatic diseases in general hospitals of urban China are rather low.
8.SIGNAL PATHWAY INVOLVED IN HEAT-STRESSED FIBROBLASTS

To explore the activities of signal transduction pathway involved in heat-stressed fibroblast in vitro. Human dermal fibroblasts were cultured in Dulbecco′s modified Eagle′s medium with 5% calf serum at 5%CO 2 in a water-saturated atmosphere. Cultured cells were heated to 45℃ for 10min. Western blotting was used to detect ERK1/2 and JNK expression, and the expression of caspase-3 protein was observed by immunoflurescence technique. The results showed that the MAPKs signal transduction pathway was activated in heat-stressed fibroblasts. The expression of JNK reached the peak at 60min, then maintained up to 180min. The expression of ERKs peaked at 30min, then lowered. The expression of caspase-3 was weak at 30min after heat-stress, and became evidently strong at 60min. The signal pathway of ERKs and JNK played important roles in the changes in biologic characteristics of fibroblasts after heat-stress.
9.Effects of sorafenib on hypoxia inducible factor-1 and vascular endothelial growth factor levels and recurrence in patients with intrahepatic cholangiocarcinoma with microvascular invasion

Objective:To investigate the effects of sorafenib on hypoxia inducible factor-1 (HIF-1) and vascular endothelial growth factor (VEGF) levels and recurrence in patients with intrahepatic cholangiocarcinoma with microvascular invasion.Methods:Ninety-two patients with intrahepatic cholangiocarcinoma who received treatment in Yiwu Central Hospital between November 2013 and November 2019 were included in this study. They were randomly assigned to undergo either conventional basic treatment (control group, n = 46) or conventional basic treatment and sorafenib treatment (study group, n = 46). Clinical efficacy, the incidence of adverse reactions and recurrence rate were compared between the two groups. Before and after treatment, HIF-1, alpha fetoprotein (AFP) and VEGF levels were also compared between the two groups. Results:After treatment, total effective rate in the study group was significantly higher than that in the control group [63.04% (29 /46) vs. 28.26% (13/46), χ2 = 11.215, P < 0.05]. After treatment, HIF-1, AFP and VEGF levels in each group were significantly lower than those before treatment (all P > 0.05). After treatment, HIF-1 [(165.23 ± 39.67) pg/mL], AFP [(109.16 ± 67.31) ng/mL] and VEGF [(297.28 ± 42.41) pg/mL] levels in the study group were significantly lower than those in the control group [(205.56 ± 40.23) pg/mL, (235.17 ± 106.41) ng/mL, (365.16 ± 40.91) pg/mL, t = 4.841, 6.788, 7.813, all P < 0.05]. There was no significant difference in the incidence of adverse reactions between the two groups ( P > 0.05). Six-month follow-up revealed that the incidence of recurrence in the study group was significantly lower than that in the control group ( χ2 = 4.792, P < 0.05). Conclusion:Sorafenib can reduce the HIF-1, AFP and VEGF levels in patients with intrahepatic cholangiocarcinoma with microvascular invasion, improve the clinical efficacy, decrease the incidence of recurrence, but cannot increase the incidence of adverse reactions.
