1.Expression of cyclooxygenase-1 in ovarian cancer and its significance

Objective To explore the expression of cyclooxygenase-1 in ovarian cancer and its significance. Methods Expression of COX-1 and CA125 was detected by immunohistochemistry in 37 cases of ovarian cancer and 31 cases of ovarian cyst. The serum level of COX-1 and CA125 was tested in 40 cases of ovarian cancer and 31 cases of ovarian cyst and 60 healthy volunteers by ELISA. Results The positive expression of COX-1 and CA125 was 78% and 57% in ovarian cancer and 3% and 7% in ovarian cyst,respectively. The expression of COX-1 and CA125 was 65% and 93% at the serum level. Conclusion Expression of COX-1 could act as an auxiliary diagnostic criterion in ovarian carcinoma.
2.Protoplast Regeneration and Mutagenesis Breeding of Streptomyces qinlingensis sp. nov.

To improve the antibiotics production of Streptomyces qinlingensis sp. nov.,protoplast regeneration combined with physical and chemical mutagenesis was used to selected high-yielding strains. The results showed that the antibacterial activities of strain R-72 from protoplast regeneration and NTG-1,H30-7 from protoplast mutagenesis against Bacillus subtilis were more than 20% higher than that of the original strain,and the heredity characters of those strains were stable in successive ten generations. The further bioassay experiments exhibited that the fungicidal and antibacterial activities of the fermentation broth from R-72,NTG-1 and H30-7 were remarkable increased comparing with that of the starting strain.
3.Outcome measurements of hearing aids in Chinese—a preliminary study

OBJECTIVE To compare the outcome measurement scores for hearing impaired people with hearing aids. Two kind of questionnaires of Chinese version--COSI and IOI-HA were developed in order to find out the more specific outcome measurements for hearing aid users in China. METHODS The COSI and IOI-HA were administered for 30 hearing aid users via telephone or by clinical interview. SPSS software was used to analyze the collecting data. RESULTS Five prevalent problems to which most of the people with hearing aids pay attention were summarized. The mean level of the satisfaction of all the participants was high. CONCLUSION The outcome measurement inventory reflects the major problems which patients concern most and it can be used to evaluate the aural intervention and rehabilitation. After further study, this method is worth extending clinical application.
4.Study on the changes of demography and behavioral characteristics of drug users in Beijing.

OBJECTIVETo understand the demography changes and behaviors in drug users.
METHODSelf-reported questionnaires was used and longitudinal investigation was conducted in one of the detoxication centers in Beijing in 1998 and 2000. Drug users were randomly chosen.
RESULTSResults showed that age of drug users tend to become younger with the numbers of drug users aged below 25, increased from 18.7% in 1998 to 28.2% in 2000. Majority of drug users remained males, but the proportion of females seemed to increase. Distribution of occupation showed that the largest increase fell among individual enterprisers, from 15.2% in 1998 to 25.9% in 2000. With educational back-ground, the proportion of lower than elementary education level, including illiterate, increased. Fifty percent of drug users were unmarried which increased from 40.8% in 1998 to 53.2% in 2000. Needle sharing was quite common, 16.5% in 1998 and 11.9% in 2000, but the decrease was not statistically significant (P > 0.05). Fifty-seven point three percent of the injecting drug users did not have constant partners to share equipments. Proportion of extra-marriage sexual practice increased from 12.5% in 1998 to 27.5% in 2000, and significant difference (Chi-square = 12.50, P < 0.001). Multiple partners in extra-married drug users was also found (mean = 2). Compared to 1998, condom use during every sexual practice increased in 2000, but 47.7% drug users still never used condom.
CONCLUSIONIn summary, as the quick increase of drug users, sharing of injecting equipment and high-risk sexual behavior, including multiple partners and unprotected sex, were quite common, with the possibility of HIV epidemic in drug users.

5.Protective effects of shark hepatic stimulator substance against acute hepatic injury induced by acetaminophen in mice.

AIMTo investigate the protective effects of shark hepatic stimulator substance (sHSS) against acute hepatic injury induced by acetaminophen (AAP) in mice.
METHODSAcute hepatic injury model of Balb/c mice was induced by a single intraperitoneal injection of AAP (200 mg.kg-1, i.p.). Serum ALT and AST activities were analyzed. The changes of microstructure and ultrastructure of hepatocyte were observed under optical and electronic microscope. The hepatocyte apoptosis was analyzed by flow cytometer and the expression level of Fas mRNA was determined by RT-PCR.
RESULTSThe activities of serum ALT and AST were significantly decreased and both necrosis and inflammatory infiltration were improved in the mice treated with sHSS 3.0 and 1.5 mg.kg-1. sHSS (3.0 mg.kg-1) prevented the ultrastructural changes of hepatocytes caused by AAP, decreased the percentage of apoptotic cells, and downregulated the expression level of Fas mRNA.
CONCLUSIONsHSS protected hepatocytes from AAP-induced injury, which might be associated with its protection of the mitochondria and inhibition of apoptosis and expression of Fas mRNA in hepatocytes.

6.The anti-injury effect of breviscapine injection on the hypoxic ischemic brain damage of neonatal rats and the expression of Bcl-2 and Bax.

OBJECTIVETo observe the protective effect of Breviscapine injection on the hypoxic ischemic brain damage of neonatal rats, and the expression of Bcl-2 and Bax.
METHODSIn this experiment 7-day-old newborn rat with hypoxic-ischemic brain damage model was used and divided into four groups: sham group, model group, control with sterile water for injection group and Breviscapine injection group. Breviscapine injection group was divided into large, medium, and small doses. Used thionin staining and immunohistochemical staining to assay the neuronal density, histological grade, and the expresssion of Bcl-2 and Bax protein in the CA1 hippocampus of each group , the number of positive cells and the integral optical density (IOD) of the immunostaining on Bcl-2, Bax protein expression in the CA1 hippocampus.
RESULTSSham group, there was no significant neuronal damage and no obvious positive cells of Bcl-2 and Bax in the CA1 hippocampus. In model group and control with sterile water for injection group, the level of Bcl-2, Bax expression peaked at 3 d after hypoxic-ischemic brain damage (HIBD) (P < 0.05 vs other groups), the value of neuronal density (ND) was decreased, and histological grade (HG) was increased compared with that in the sham group (P < 0.05). Breviscapine injection group, compared with control with sterile water for injection group, the expression of Bcl-2 protein was further increased, IOD value increased, while the expression of Bax protein was decreased, IOD value decreased, the value of ND increased, and HG decreased.
CONCLUSIONBreviscapin injection maybe reduce the delayed neuronal death, and reduce the apoptosis of neuron after severe brain injury through improving the expression of Bcl-2 protein and inhibiting expression of Bax. The study would provide a fine theoretical foundation for clinical therapy of neonatal HIBD.

7.Somatic mutations of mitochondrial DNA in thyroid papillary carcinoma.

OBJECTIVETo analyze the somatic mutations in the D-loop of mtDNA and further evaluate the possibility of mitochondrial genetic instability in thyroid papillary carcinoma.
METHODSHypervariable regions ( HVR-I and HVR-II) in the D-loop of mtDNA from the specimen of 35 thyroid papillary cancers and matched lymphocytes were amplified by PCR, and then were sequenced.
RESULTSComparing the sequences of tumors to those of matched lymphocytes and normal thyroid tissues, 5 somatic mutations in 2 patients (5.7%) were found. Two mutations were insertions of C in a poly-cytidine (nt303) microsatellite, and 3 at positions 73, 152 and 194 in HVR-II. In addition, of the 294 genetic variants detected, 292 were previously recorded polymorphisms, whereas 2 were new polymorphisms (nt324:C-->G, nt16092:T-->A).
CONCLUSIONSMutations in the D-loop of mtDNA were found in thyroid papillary cancers, this mutation rate was lower than the reported rate of alteration in tumors of epithelial origin, and further work is required to elucidate the relationship between this mutations and the development of thyroid papillary carcinoma.

8.Study for association between high risk human papillomavirus and cervical lesions in the samples from opportunistic screening

Objective To investigate the association between HPV genotypes and cervical lesion in Hybrid Capture 2 ( HC2 ) HPV test positive samples. Methods 602 cervical samples randomly obtained detected as HPV positive by the HC2 high-risk probe cocktail were determined by polymerase chain reaction (PCR)-reverse dot blot (RDB), among them 344 participated Thinprep Cytology test. Results 569 (94. 5% ) samples were successfully amplified. The most common HPV genotypes were HPV16(31.6% ),52 ( 16. 7 % ), 58 ( 15.1% ), 56 ( 8. 1% ), 39 ( 7. 9% ) ; HPV 16 or HPV 16/18 infection was significantly related to ASCUS, LSIL and HSIL; 24 samples suggested single HPV infection with the genotypes not available for HC2. Conclusion The most common HPV genotypes in random screening were HPVI6, 52 and HPV58; Our data demonstrated the pseudo-positivity of HC2 test was 4.0% ; HPV16 was a index for progression of HSIL.
9.Status survey of job satisfaction of nurses at public hospitals and recommendations

Objective To analyze the status and influencing factors of job satisfaction of nurses at public hospitals, providing references for the teambuilding strategy of nurses. Methods Questionnaire survey method was used, in October 2017, to study job satisfaction of 1,047 nurses at 13 public general hospitals, for a countermeasure study. Results Job satisfaction of the nurses was found at a medium level. Specifically, their overall job satisfaction score was 3. 68, and the satisfaction scores of the following dimensions from high to low were: on-the-job training (3. 91), hierarchical nurse use (3. 79), performance appraisal (3. 68), promotion evaluation (3. 67), and remuneration (3. 62) respectively. Contents of the five dimensions are strongly related to the overall job satisfaction (r>0. 6). Job satisfaction scores of nurses with their differences in positions, annual income, and hospital level were significantly different (P< 0. 05). The three main influencing factors for talent turnover rate are remuneration, workplace pressure and work environment. Conclusions Higher remuneration, improved hierarchical management system, and greater attention to nursing care at secondary hospitals are very important for effectively improving the job satisfaction of nurses and ensuring the health care industry.
10.Clinicopathological features of liposarcoma of the extremities: analysis of 14 cases

Objective:To investigate the clinicopathological characteristics, diagnosis and differential diagnosis of liposarcoma of the extremities.Methods:Fourteen patients with atypical lipomatous tumor/well-differentiated liposarcoma (ALT/WDL), dedifferentiated liposarcoma (DDL) and myxoid liposarcoma (ML) in Beijing Tiantan Hospital of Capital Medical University from March 2002 to December 2019 were collected. Fluorescence in situ hybridization (FISH) and immunohistochemical staining were used to detect the MDM2 gene amplification and expressions of S-100, CDK4 and MDM2 proteins in tumor tissues. The characteristics of different types of liposarcoma were comprehensively analyzed combined with clinical data and histopathological characteristics.Results:The age of 14 patients was (55.6±16.7) years old, with 10 males and 4 females. There were 2 cases of ALT/WDL, 6 cases of DDL, and 6 cases of ML. Immunohistochemistry showed that S-100 was positive in all patients and the patterns were scattered, partial and extensive. CDK4 and MDM2 were positive in both ALT/WDL and DDL, and weakly expressed in ML (4 cases). Vimentin was positive in all patients. MDM2 gene amplification was detected in 2 cases of ALT/WDL and 6 cases of DDL.Conclusion:Liposarcoma has a high incidence and obvious morphological diversity, accurate diagnosis is critical to the prognosis and treatment of the tumor.
