Objective To investigate the effects and mechanism of IL-6 on invasion and metastasis of human pancreatic cancer cells. Methods IL-6 was added into the culture media of human pancreatic cancer cells Capan-2 and SW1990. Cell growth was measured by MTT assay. Western blot and immunocytochemistry were performed to detect Phosphorylated STAT3 (P-STAT3) protein. VEGF and MMP-2 mRNA and protein expression were examined using fluorescence quantitative polymerase chain reaction (RT-PCR) and Western blot, respectively. The invasion ability of SW1990 and Capan2 cells was determined by cell invasion assay in vitro. Results 100 ng/mL IL-6 significantly promoted growth and invasion ability of Capan-2 and SW1990 cells (P<0.05). The use of IL-6 not only markedly increased the protein expression of P-STAT3, VEGF and MMP-2, but also greatly increased the mRNA expression of MMP-2 and VEGF. Conclusions STAT3 signal transducer pathway activation with IL-6 can promote the invasion ability of pancreatic cancer cells in vitro through up-regulation of MMP-2 and VEGF expression. STAT3 signal transducer may provide a novel therapeutic target for the treatment of pancreatic cancer.

Objective To explore the prevalence and associated factors of anxiety and depression symptoms in hospitalized Chinese patients with coronary artery disease(CAD). Methods From June 2007 to May 2009, 1083 hospitalized patients with confirmed coronary artery disease were recruited in this study. The ZUNG Self-rating Anxiety Scale(SAS)and the ZUNG Self-rating Depression Scale(SDS)were used for the psychological assessment. Economic status, living condition and the environment of both living and working places were evaluated by epidemiological questionnaires. Results The prevalence of pure anxiety, pure depression symptoms and the combination of anxiety and depression symptoms were 7. 9%, 28.3% and 14. 3% respectively. Incidence of anxiety and depression symptoms was significantly higher in female patients compared with in male patients(P = 0. 003, 0. 012 respectively)and in aged patients than in middle-aged patients(P = 0. 001). The elderly, less than 9 years of education and poor sleep quality increased the risk of anxiety symptom with ORs of 1.63(95% CI: 1. 21 - 2. 21), 1.54(95% CI: 1.15 -2. 07)and 1.62(95% CI: 1.34 - 1.96), respectively, while workplace noise, history of chronic disease and poor sleep quality increased the risk of depression symptom with ORs of 1.52(95% CI:1. 18 - 1.98),1.36(95%CI:1.06-1.75)and 1.27(95%CI:1.08-1.50), respectively. Female(OR=1.91, 95%CI:1. 22 -2.98), aged patient(OR = 1.84, 95% CI:1. 23 -2. 76), workplace noise(OR = 1.61, 95%CI:1. 07 -2. 42), history of chronic disease(OR = 1.84, 95% CI: 1.24 - 2. 71)and poor sleep quality (OR = 1.73, 95% CI: 1.35 -2. 21)were significantly correlated with the combined incidence of anxiety and depression symptoms. Conclusion Around half of the Chinese hospitalized CAD patients were complicated with various degrees of anxiety and/or depression symptoms. Female and aged patients were at higher risk for anxiety and depression symptoms. Sleep quality, workplace noise, years of education and history of chronic disease were independent risk factors for anxiety or depression symptoms.

Objective: To study the effect of Yangxinkang tablets on myocardial fibrosis in mice after heart failure, and to explore its mechanism.Method: The model of chronic heart failure in mice was established by thoracic aorta constriction (TAC). After successful modeling, mice were randomly divided into sham operation group, model group, 3-methyladenine(3-MA,15 mg·kg-1) autophagy inhibitor group, Yangxinkang tablets high, medium, and low dose groups (1 170,585,390 mg·kg-1).The sham operation group received equal volume of distilled water. After 30 days, cardiac ultrasound was performed to collect hemodynamic parameters. Cardiac paraffin slices were stained with Masson to observe the morphological changes and fibrosis of cardiomyocytes. Western blot was used to detect lysosome-associated membrane protein(LAMP), microtubule-associated protein light chain 3 (LC3), Beclin-1 autophagyportein, α-smooth muscle activin(α-SMA),Collagen Ⅰ,Collagen Ⅲ protein expression.Result: As compared with normal group, the left ventricular ejection fraction (LVEF) and fractional shortening(FS) were significantly decreased(P<0.05),the left ventricular end-diastolic dimension (LVDd) and left ventricular end-systolic dimension (LVDs) were significantly increased (P<0.05),the degree of myocardial fibrosis was significantly aggravated in model group (P<0.01),and the protein expression levels of α-SMA, Collagen Ⅰ, Collagen Ⅲ, LAMP, LC3, and Beclin-1 were significantly increased in model group (P<0.05).As compared with model group, the cardiac ultrasound indexes of the 3-MA group, Yangxinkang high and medium dose groups were significantly improved (P<0.05),the degree of myocardial fibrosis was significantly reduced (P<0.01),the protein expression levels of α-SMA,Collagen Ⅰ,Collagen Ⅲ,LAMP,LC3 and Beclin-1 were decreased in 3-MA group, Yangxinkang high and medium dose groups(P<0.05),while the protein expression levels between the model group and Yangxinkang low-dose group showed no significant difference. Conclusion: Yangxinkang tablets can reduce myocardial fibrosis and improve cardiac function in mice with heart failure probably by down-regulating autophagy.
