2.Synthesis and structure-activity relationship of cycloberberine as anti-cancer agent.

A series of cycloberberine derivatives were designed, synthesized and evaluated for their anti-cancer activities in vitro. Among these analogs, compounds 6c, 6e and 6g showed strong inhibition on human HepG2 cells. They afforded a potent effect against DOX-resistant MCF-7 breast cells as well. The primary mechanism showed that cell cycle was blocked at G2/M phase of HepG2 cells treated with 6g using flow cytometry assay. It significantly inhibited the activity of DNA Top I at the concentration of 0.1 mg mL-1. Our results provided a basis for the development of this kind of compounds as novel anti-cancer agents.

3.Review and Analysis on Projects Funded by NSFC in Traditional Chinese Medicine in 2017

To review the application,acceptance,evaluation and funding of the General,Youth and Regional Science Fund supported by Chinese National Natural Science Foundation in traditional Chinese medicine in 2017.There were 4 472 applications in the subject,114 of which failed in the formal examination,4 358 applications were accepted in the next assessment step;1 403 applications got more than 2 recommendations after peer assessment and 637 applications were funded in the end.The major problems in every step of Science Fund were analyzed in this paper.The author hopes all applicants should pay attention to the problems,and then improve the quality of applications in the field.
4.Zinc oxide nanoparticles inhibits Ca2+-ATPase1 expression induced by UVB irradiation in human lens epithelial cells

Objective To investigate the effect of ZnO nanoparticles on the expressions of plasma membrane calcium ATPasel (PMCA1) of human lens epithelial cell B-3 (HLEB-3) at both mRNA and protein levels in the presence and absence of ultraviolet B (UVB) irradiation.Methods HLEB-3 was cultured in RPMI 1640 medium,and the cytotoxic effect of different concentrations of ZnO (0 μg · mL-1,2.5 μg · mL-1,5.0μg · mL-1,10.0 μg · mL-1) on HLEB-3 was investigated in the presence and absence of UVB irradiation.DAPI staining was used to monitor the effect of ZnO on HLECB-3 nuclei,and cell apoptosis was evaluated using annexin V-FITC/PI staining in the presence and absence of UVB irradiation.In addition,the intracellular calcium ion (Ca2 +)levels were assayed using Fluo-3/AM staining,and the expression levels of both PMCA1 mRNA and protein within HLEB-3 were detected by real-time PCR and Western blot,respectively.Results DAPI staining showed that the ZnO-treated HLEB-3 displayed a concentration-dependent apoptosis,and UVB irradiation could further aggravate the cytotoxic effect of ZnO on HLEB-3.In addition,in the presence of UVB irradiation,concentration gradient of ZnO (2.5 μg · mL-1,5.0 μg · mL-1,10.0 μg · mL-1) increased the intracellular calcium ion levels [from (156.34 ±4.59) nmol · L-1 to (173.88 ±7.17)umol · L-1,(289.02 ± 9.09) nmol · L-1,(488.36 ± 48.16) nmol · L 1,respectively] and upregulated HLEB-3 apoptosis,with statistical difference (all P < 0.05).Moreover,the expression level of PMCA1 in the 2.5 μg · mL-1,5.0 μg · mL-1,10.0 μg · mL-1 ZnO-treated epithelial cells was accordingly 0.75,0.57 and 0.41 as much as that in the 0μg · mL-1 ZnO-treated cells in the absence of UVB irradiation (all P < 0.05),and was accordingly 0.64,0.24 and 0.09 in the present of UVB irradiation,with significant difference (all P < 0.05).Conclusion Both ZnO nanoparticle and UVB irradiation can exert cosuppression effect on HLEB-3 via calcium-mediated signaling pathway,indicating it has great potential for the treatment of posterior capsular opacification with UVB irradiation.
5.The effects of ezrin on the proliferation and the invasiveness of cells of different hepatocellular carcinoma cell lines.

OBJECTIVETo investigate the different expressions of cytoskeletal organizer ezrin and cytoskeleton protein beta- and gamma-actin in hepatocellular carcinoma (HCC) cell lines with different metastatic potentials and to explore the role of ezrin in cell growth and metastasis in HCC cell lines SF7721 and MHCC97-H.
METHODSImmunofluorescence, RT-PCR and Western blot were used to detect the gene and protein expressions of ezrin and actin in hepatocellular carcinoma cell lines with different metastatic potentials. RNA interference (RNAi) was applied to down-regulate the ezrin expression in SF7721 and MHCC97-H. Changes of the cell growth and metastasis potentials after the RNAi treatment were studied. MTT assay was used to detect cell proliferation changes and Transwell assay was applied to observe the changes of cell motility and invasiveness.
RESULTSBoth ezrin and cytoskeleton protein were demonstrated in the cytoplasma of the cells at the same time. The expression of them in cell lines with high metastatic potential, such as SF7721, MHCC-1 and MHCC97-H was obviously higher than in those with low metastatic potentials, such as SMMC-7721, Hep3B and HepG2 (chi2= 13.277, P = 0.010; chi2= 21.815). The mRNA and ezrin and cytoskeleton protein gamma-actin were over-expressed in HCC cell lines with high metastatic potentials. The expressions of beta-actin of cell lines with different metastatic potentials showed no differences. Ezrin protein was successfully down-regulated and the proliferation and the invasiveness of the cells decreased with low ezrin protein level in SF7721 and MHCC97-H.
CONCLUSIONOver-expression of ezrin and cytoskeleton protein gamma-actin are associated with the process of metastasis of hepatocellular carcinoma cells. The growth and invasiveness of SF7721 and MHCC97-H cells can be inhibited by down-regulating ezrin expression.

6.Distribution of haplotypes for four Y-sTR loci and validation in forensic science by using a double-fluorescent multiplex PCR system.

OBJECTIVE:
We focus on developing a multiplex PCR system for Y-STR loci that can be detected by double fluorescent system and assessing their usefulness in forensic mixture samples.
METHODS:
The primers of four Y-STR loci (DYS-GATA-A10, DYS531, DYS557 and DYS448) amplified by multiplex PCR technique were labeled with fluorescence, then the PCR products of these Y-STRs loci were detecting and typing by ABI PRISM310 Genetic Analyzer.
RESULTS:
When 120 unrelated individuals from the Han population in Chengdu were detected by the system, Y-GATA-A10, DYS531, DYS557 and DYS448 showed 5, 5, 8, 7 alleles, respectively. A total of 78 different haplotypes was identified and the genetic diversity reached 0.9881. To the three cases of mixture stains failed by using conventional autosomal STR analysis, our multiplex system drew conforming conclusion comparing to the suspect's Y-STRs genotypes.
CONCLUSION
Our results show that the multiplex system of four Y-STR will be very powerful for Y-STR database establishing, the paternity testing and mixture stains identifying.

7.Application of dialectical nursing of traditional Chinese medicine in patients with gout

Objective To explore the application of dialectical nursing of traditional Chinese medicine in patients with gout.Methods Totals of 68 cases with gout were randomly divided into control group( n =36)and observation group (n =34).Types such as wet- hot,heat and blood stasis block,phlegm,yin deficiency of liver and kidney of cases were divided.The observation group received the dialectical nursing and selfdesigned special care programs,many special form were used such as drawing the blood uric acid detection curve,symptom observation,daily diet records list,daily medication and external treatment record.Then,the time of symptoms better,effective,cure were observed and diet,living,grooming emotion were strictly guidanced.Control group only rceived the dialectical nursing without self-designed nursing program.Results The time of blood uric decrease to normal of observation group significantly short than that of control group(15.0 ±2.5) d vs (22.0 ±7.5) d,(t =5.1629;P <0.05).The whole effective cases number of observation group was significantly more than that of control group ( 33 vs 26 ),( x2 =4.6102 ; P < 0.05 ).Conclusions Dialectical nursing application,detailed records in every day,strict implementation of the living,diet,emotional guidance for the patients with gout have positive clinical influencing.
8.Clinical study of idiopathic dilated cardiomyopathy complicated by left ventricular aneurysm

Objective To examine the hemodynamic and electrephysiological influence of left ventricular aneurysm(LVA)formation in patients with idiopathic dilated cardiomyopathy(IDCM).Methods All hospital records were retrospectively reviewed from IDCM patients admitted to our hospital between 2003 and 2008.Patients with coronary angiography evidenced ischemic cardiomyopathy were excluded.IDCM patients with LVA(I+L)diagnosed by left ventriculography were enrolled.Twelve age-,gender-and left-ventricular-diameter-matched patients with IDCM without LVA served as control group(I-L).Results Six out of 998 patients with IDCM were confirmed to have LVA(0.60%).The LV peak-systolic pressure was higher in the I+L group than in I-L group[(130±10)mm Hg(1 mm Hg=0.133 kPa)vs.(117±9)mm Hg,P<0.05].The LV end-diastolic volume Was significandy larger in the I+L group than in I-L group[(272 ±57)ml vs.(207±60)ml,P<0.05].The LV ejection fraction was slightly lower in the I+L group than in I-L group[(27±9)%vs.(35±6)%,P=0.09].Ventricular arrhythmia occurred more frequently in I+L group than in I-L group.Conclusion LVA formation in IDCM was a rare phenomenon.IDCM patients with LVA seem to have higher LV peak-systolic pressure,larger end-diastolie volume,worse LV systolic function and more frequent ventricuLar arrhythmia than those without LVA.
9.Etiology and antimicrobial resistance of community-acquired pneumonia in adult patients in China.

BACKGROUNDAppropriate antimicrobial therapy of community-acquired pneumonia (CAP) is mainly based on the distribution of etiology and antimicrobial resistance of major pathogens. We performed a prospective observational study of adult with CAP in 36 hospitals in China.
METHODSEtiological pathogens were isolated in each of the centers, and all of the isolated pathogens were sent to Zhongshan Hospital for antimicrobial susceptibility tests using agar dilution.
RESULTSA total of 593 patients were enrolled in this study, and 242 strains of bacteria were isolated from 225 patients. Streptococcus pneumoniae (79/242, 32.6%) was the most frequently isolated pathogen, followed by Haemophilus influenzae (55/242, 22.7%) and Klebsiella pneumoniae (25/242, 10.3%). Totally 527 patients underwent serological tests for atypical pathogens; Mycoplasma pneumoniae and Chlamydia pneumoniae infections were identified in 205 (38.9%) and 60 (11.4%) patients respectively. Legionella pneumophila infections were identified in 4.0% (13/324) of patients. The non-susceptibility rate of isolated Streptococcus pneumoniae to erythromycin and penicillin was 63.2% and 19.1% respectively. Six patients died from the disease, the 30-day mortality rate was 1.1% (6/533).
CONCLUSIONSThe top three bacteria responsible for CAP in Chinese adults were Streptococcus pneumonia, Haemophilus influenza and Klebsiella pneumonia. There was also a high prevalence of atypical pathogens and mixed pathogens. The resistance rates of the major isolated pathogens were relatively low except for the high prevalence of macrolide resistance in Streptococcus pneumoniae.

10.Constructing standard allelic ladders for four short tandem repeat loci and employing them in a population study on Han Nationality of Chengdu in China.

OBJECTIVETo solve the problems in the accuracy and standardization of short tandem repeats-polymerase chain reaction (STR-PCR) typing, the authors adopted the molecular clone technology in producing the standard allelic ladders of D1S1676, D2S2735, D11S1977 and D22S444 loci and applied them in a population study on the Hans in Chengdu, China.
METHODSPCR was used to produce several different allelic fragments of these loci. PCR products were eluted from the gel and re-amplified by PCR. The purified allelic fragments were then blunt-end subcloned individually into the pGEMR-T plasmid vectors and the recombinant were transfected into competent E.coli DH5alpha TM cells. The results of sequencing confirmed that the size and the construction of the inserts were correct. The recombinant plasmids DNA with the inserts were then used as template for re-amplification to generate the four loci standard ladders.
RESULTSThe authors succeeded in producing large quantity of standard allelic ladder of these four loci, with which the genetic polymorphisms of these loci in Chengdu Han population of China were studied.
CONCLUSIONThis method is of high value for forensic DNA typing to construct standard ladders. D1S1676, D2S2735 loci are robust for forensic analysis in Chinese Han population, whereas the value of D11S1977 and D22S444 loci is limited.
