2.Spatial autocorrelation analysis of brucellosis in Zibo of Shandong Province, 2013-2015

Objective To explore the spatial distribution and spatial clustering of brucellosis in Zibo,2013-2015.Methods Spatial autocorrelation analysis was used to analyze the surveillance data of brucellosis at town level.Township as a spatial analysis unit,spatial distribution of brucellosis in small scale in Zibo City was analyzed.Results The global Moran's I indexes of brucellosis were not significantly different in 2013-2015.According to local Moran's Ⅰ statistic,the high-high regions were Xindian Street and Jiangjunlu Street in 2013.The high-high regions were Tangfang Town,Jinshan Town,Luocun Town,Zhaili Town,Xihe Town and Longquan Town in 2014.The high-high region was Wangcun Town in 2015.Condusions Our study has showed that the spatial distribution of brucellosis is local clustered in Zibo.The detection of hotspots could provide guidance for formulating regional prevention and control strategies.
3.Inhitory effect of electroacupuncture on acetylcholine M1 receptor expression in visual cortex of guinea pigs with lens-induced myopia

Background It has not been reported that if the visual cortex M receptor changed during the development of myopia and how it changed if given acupuncture treatment.Objective The aim of this study was to observe the effect of electroacupuncture stimulation on the expression of acetylcholine receptors M1 (AchRM1) in visual cortex of guinea with lens-induced myopia (LIM).Methods Forty-eight three-week-old healthy guinea pigs were randomized into the normal control group,the LIM model group and the LIM electroacupuncture group.The right eyes of the guinea pigs were selected as the experimental eyes.LIM was created by monocularly wearing of-10 D lens for 4 weeks in the right eyes in the LIM model group and LIM electroacupuncture group,and then the acupuncture at the temple and hegu point was performed for 30 minutes per day for consequent 4 weeks,in the LIM electroacupuncture group.The fellow eyes of the guinea pigs were used as the self-control eyes.The refractive power and axial length were examined with retinoscopy and A-type sonography before and 4 weeks after modeling,respectively.The animals were sacrificed by excessive anesthesia at the fourth week after acupuncture and visual vertex tissue was obtained.The expression of M1 receptor mRNA in visual vertex was detected by fluorescence quantitative PCR,and the content of M1 receptor protein in visual vertex was assyed by ELISA.The study protocal was approved by Animal Ethic Committee of Shandong University of Traditional Chinese Medicine,and the use and care complied with Statement of the Association for Research in Vision and Ophthalmology.Results At the fourth week after modeling,the mean diopters were (-3.24±0.28) D and (-3.30±0.45) D in the LIM model group and the LIM eleetroacupuncture group,which were significantly higher than (0.83 ±0.86)D in the normal control group (both at P=0.000),and there was no significant difference in the diopter between the LIM model group and the LIM electroacupuncture group (t =0.200,P =0.659).The mean axial lengths were (8.67 ±0.14) mm and (8.60±0.06) mm in the LIM model group and the LIM electroacupuncture group,which were considerably increased in comparison with (8.33±0.08)mm in the normal control group (both at P<0.05).The relative expression levels of AchRM1 mRNA in visual cortex were 0.79±0.18,1.36±0.23 and 1.13±0.13 in the normal control group,LIM model group and LIM electroacupuncture group,and the relative expression level of AchRM1 mRNA in the LIM electroacupuncture group was significantly higher than that of the normal control group and lower than that of the LIM model group (both at P<0.05).In addition,the contents of AchRM1 receptor protein in the visual cortex were 248.00±33.31,455.17±42.40 and 396.17±47.57 in the normal control group,LIM model group and the LIM electroacupuncture group,with a similar pattern among the groups (both at P<0.05).Conclusions A electroacupuncture stimmulation do not affect the myopic diopter and axial length in LIM model.The AchRM1 and AchRM1 receptor in the visual cortex up-regulate in LIM eyes,infering that electroacupuncture stimmulation can improve vision by decreasing the level of AchRM1 receptor in visual cortex in LIM eyes in guinea pigs.
4.Accuracy of cerebrospinal fluid Aβ-42 to tau level ratio in predicting postoperative cognitive dysfunction in elderly patients

Objective To evaluate the accuracy of cerebrospinal fluid (CSF) amyloid beta 42 (Aβ-42) to tau level ratio (Aβ-42/tau) in predicting postoperative cognitive dysfunction (POCD) in elderly patients.Methods Eighty ASA physical status Ⅱ or Ⅲ patients of both sexes,aged 65-85 yr,undergoing elective total hip or knee replacement under combined spinal-epidural anesthesia,were enrolled in the study.Epidural catheter was placed at T3,4 interspace,and after a catheter was successfully placed into the subarachnoid space,CSF 2 ml was obtained to measure Aβ-42 and tau levels (units:pg/ml),and the ratio between them was calculated.Neuropsychological tests were performed at 1 day before surgery and 1 week after surgery.The Z score was used to identity POCD.The cut-off value for POCD and nonPOCD patients was determined by using ROC curve.Results Thirty-two patients were diagnosed with POCD,and the incidence was 40%.The Aβ-42/tau was 2.6±0.3 and 1.7±0.4 in non-POCD and POCD groups,respectively,and the ratio was significantly lower in POCD group than in non-POCD group.The sensitivity of CSF Aβ-42/tau in predicting POCD was 91.7%,the specificity was 81.2%,and Youden index was 0.7.ROC curves revealed that the cut-off value for POCD and non-POCD patients was 2.0.Conclusion CSF Aβ-42/tau can accurately predict the occurrence of POCD in elderly patients.
5.Effect of Tuina Combined with Traditional Chinese Exercise on Nonspecific Chronic Neck Pain

Objective To explore the effect of Tuina combined with Traditional Chinese exercise (TCE) on nonspecific chronic neck pain (NCNP). Methods 80 eligible patients were recruited in our hospital from October, 2014 to October, 2015. They were randomized to observation group (n=40) and control group (n=40). The observation group received Tuina combined with TCE, and the control group re-ceived intermittent cervical traction, 5 times a week for 2 weeks. They were assessed with Visual Analog Scale (VAS) and Neck Disability Index (NDI) before, immediately after intervention, and at 1 month follow-up. Results 77 patients completed the treatment and follow-up. The scores of VAS and NDI significantly decreased after a 2-week intervention (t>9.330, P<0.001) in both groups. The VAS score were sig-nificantly lower in the observation group than in the control group immediately after intervention and at 1-month follow-up (F>8.338, P<0.01). The NDI score was significantly lower in the observation group than in the control group at 1-month follow-up (F=9.053, P=0.004). Conclusion Tuina combined with TCE could relieve pain and improve cervical function in patients with NCNP, which was superior to inter-mittent cervical traction.
6.Changes in expression of hippocampal occludin and claudin-5 after operation in aged rats

Objective To investigate the changes in the expression of hippocampal occludin and claudin-5 after operation in aged rats.Methods Eighty-one healthy male Wistar rats, aged 16-18 months, weighing 400-500 g, were randomly assigned into 3 groups (n =27 each) using a random number table: normal control group (group C), skin incision group (group I) and splenectomy group (group S).Splenectomy was performed in group S.Only skin incision and closing were performed in group I.Morris water maze test was performed before operation and on 1, 3 and 7 days after operation.The escape latency and the percentage of time spent at the target platform quadrant were recorded.Nine rats randomly selected from each group were sacrificed after the end of the test, and the hippocampi were isolated to detect the expression of occludin and claudin-5 by Western blot.Results Compared with group C, the escape latency was significantly prolonged, the percentage of time spent at the target platform quadrant was decreased, the expression of hyperphosphorylated occludin and claudin-5 was down-regulated, and the expression of hypophosphorylated occludin was up-regulated on 1 and 3 days after operation in group S (P<0.05).Conclusion The mechanism of postoperative cognitive dysfunction is related to the downregulated expression of hippocampal hyperphosphorylated occludin and claudin-5 in aged rats.
7.Impact of ethyl pyruvate on the cognitive function after splenectomy in aged rats

Objective To investigate the impact of ethyl pyruvate (EP) on cognitive function and the expression of high mobility group box 1 (HMGB1) and receptor for advanced glycation end products (RAGE) after splenectomy in aged rats.Methods Eighty-four male aged Sprague-Dawley rats, 18 months old, weighing 500-600 g, were randomly divided into 4 groups (n=21 each) by random number table method: control group (group C), surgery group (group S) ethyl pyruvate group (group E) and solution without EP group (group R).Morris water maze test was performed to evaluate cognitive function 5 days before surgery and 1, 3, 7 days after surgery.Group E was injected with EP 40 mg/kg intrapertoneally after splenectomy, group S and group C were injected with equivalent normal saline after splenectomy, group R was injected with equivalent solution without EP.Rats were killed after Morris water maze test, and the expression of HMGB1 and RAGE protein and mRNA in hippocampus were measured by Western blot and RT-PCR methods.Results Compared with group C, the escape latency and swimming distance were significantly prolonged in groups S, E and R 1 and 3 days after surgery, as well as the expression of HMGB1 and RAGE in hippocampus were significantly up-regulated (P<0.05).Compared with group S, the escape latency and swimming distance were significantly decreased and the expression of HMGB1 and RAGE were down-regulated in group E 1 and 3 days after surgery (P<0.05).Compared with the preoperative group, the escape latency and swimming distance were significantly prolonged in groups S, E and R 1 and 3 days after surgery (P<0.05).Conclusion EP may improve cognitive function in aged rats by down regulating the expression of HMGB1 and RAGE in the hippocampus.
8.Arecoline improved glucose and lipid metabolism in type 2 diabetic rats

Aim To investigate the effects of arecoline on glucose and lipid metabolism in type 2 diabetic rats and its mechanisms in glucose metabolism.Methods A type 2 diabetic rat model was established by fed with high fructose-high fat diet.The animals were randomly divided into 7 groups: control group,high fructose-high fat diet group(HF)and high fructose-high fat diet+arecoline(1 mg?kg-1,5 mg?kg-1,10 mg?kg-1,20 mg?kg-1,50 mg?kg-1)groups.The blood glucose,lipid level,hepatic function and liver histology were measured.The mRNA expression of liver glucose-6-phosphatase(G6Pase),phosphoenolpyruvate carboxykinase(PEPCK),Forkhead Box O1(FoxO1)and peroxisome proliferator-activated receptor-? coactlvator-1? (PGC-1?)were observed through RT-PCR.Results In comparison with the high fructose-high fat diet group,the fasting blood glucose and TC of the rats were significantly decreased by arecoline in a dose-dependment manner in high fructose-high fat diet+arecoline group.But hepatic function was damaged by 10 mg?kg-1,20 mg?kg-1 and 50 mg?kg-1 arecoline.The mRNA expression of hepatic G6Pase,PEPCK,FoxO1 and PGC-1? was decreased by treatment with 1 mg?kg-1 and 5 mg?kg-1 arecoline compared with the high fructose-high fat diet group.Conclusions Low dose arecoline can decrease fasting blood glucose and TC in type 2 diabettic rats,and the mechanism in glucose metabolism may be related to its effect on the inhibition of hepatic gluconeogenesis.

Objective To investigate the feasibility and accuracy of magnetic resonance microneurog- raphy of sciatic nerve fascicles in rabbil by correlation with the gross anatomy.Methods The 3D T_2-weigh- ted imaging(3D-T_2 MI),3D T_2-weighted imaging plus spectral presaturation with inversion recovety(SPIR), T_1-weighted imaging(T_1 WI)of the sciatic nerve in 10 rabbits were performed on a 1.5 Tesla magnetic reso- nance system.The radiological ananomy and imaging features of sciaticnerve fascicles were observed and the anterior-posterior diameter was measured on 3D T_2-weighted imagingThe imaging evaluation was correlated with the gross anatomy.The T_1 and T_2 relaxation time were determined by multiple echo spin echo and mix se- quecerespectively.Results The libial nerve and peroneal nerve in the main trunk of sciaticnerve in all 10 rabbits could be clearly displayed on the 3D T_2 WI,3DT_2WI plus SPIRand T_1WI.Strikingly,the 3D T_2 WI could delineate the fine branches of the sural nerve and posterior femural cutaneous nervesThe T_1 and T_2 relaxation time were 915 ms40 msrespectivelyGrosslythe anterior-posterior diameter of sciatic nerve trunk was3.17?0.21)mmwhile was(3.15?0.19)on 3D T_2 WI.There was no statistically significant difference(t=0.768,P=0.462).Conclusion With 1.5 Telsa MR system the microneurography of the sci- atie nerve could be achievable and the individual fascicles of sciatic nerve trunk could be clearly and accurately discriminated.
10.Study on Gantt charts in quality control management in nursing practice

Objective To know about the influence of quality control based on Gantt chart on work quality and stress,so as to explore effective method of management suitable for them.Methods 52 nurses from the eastern zone of a three A-level hospital in Jiujiang City were randomly selected as the experimental group,they were implemented with Gantt chart management.Another 52 nurses from the western zone of the same hospital were selected as the control group,they were executed traditional experience-based management.The scores of quality of care and nurses stress were compared between the two groups.Results For the score of the quality of care,the experimental group was higher;,for the score of the nurses stress,the experimental group was lower in the overall sides,and the differences were significant except those of the working environment and equipment,which showed no significant difference between two groups.Conclusions Gantt management method can significantly improve the quality of care,moreover,effectively alleviate the nurses'stress.
