Objective To investigate the regulation effect of bispectral index (BIS) on the depth of total intravenous anesthesia (TI-VA) by comparing with regulating the depth of anesthesia according to the changes of hemodynamics. Methods 60 ASA Ⅰ～Ⅱ patients undergoing laparoscopic cholecystectomy (LC) were randomly assigned into controlled group (group C) and trial group (group T) ,with 30 patients in each group, according to the rules of regulating the titration of propofol during TIVA. The target of group C was to keep SBP/DBP 100 ～ 140mmHg/60 ～ 89mmHg and that of group T was to keep BIS 40 ～ 60 during anesthesia. The measure indexes included SBP/DBP, HR, BIS, total dosis of Propofol, time to extubate, time to leave operating room and OAA/S (observer's assessment of alert-ness/sedation). After operation, we evaluated whether awareness during surgery happens. Results As compared with group C, there were higher SBP/DBP and BIS in group T after induction of anesthesia, during aeroperitonia, immediately after finishing operation and just before extubating endotracheal catheter, but less dosis of propofol and less time of extubation and leaving operating room in group T. The differences had significance (P <0.01). There were no significant differences in the changes of HR between two groups (P >0.05) and no awareness during surgery happened in both groups. Conclusion TIVA regulated by BIS during LC can decrease the dosis of propofol, accelerate the recovery from anesthesia and avoid extremely deep anesthesia and awareness during surgery.

Objective To define the application value of HBV DNA detection on HBsAg-negative blood donors and assess the necessity for nucleic acid detection.Methods Real-time PCR was used to detect HBV DNA on HBsAg negative blood donors.Pools of eight donor samples were used for NAT testing.Viruses were concentrated by centrifugation and the viral DNA extraction was performed using magnetic beads.If HBV DNA wag positive,serological indicators including HBsAg,anti-HBs,HBeAg, anti-Hbe,total anti-HBc was further detected.Results The HBV DNA detection limit was 25 U/ml.There were four HBV DNA positive cases among 23 225 specimens.and the detection rate was 0.17‰ The further serological examination showed anti-Hbe(+),anti-HBc(+) in the two cases and anti-HBc(+) in one case and anti-Hbs(+),anti-HBc(+)in 1 case.The viral load can range form 50 to 200 U/ml. Conclusions The results indicate that there is false negative possibility in blood screening by ELISA.It is necessary to employ anti-Hbe screening or NAT to blood donors screening.

AIM: To study the effects of Rhodiola(Rho), nitric oxid e (NO), hem oglobin (HB) on the multiple organ dysfunction syndrome(MODS) in early stage aft er burn in rabbits. METHODS:The rabbits were divided into the sh am burn group (SB), burn group (B), orally taken Rho group (R), burn and Rho therapy group (BR ). The changes of hemodynamics were monitored. The index of pulmonary permeabili ty was calculated. These data reflected separately the functions of heart, live r, lung, kidney and blood coagulation system were also determined. NO contents i n ser um and bronchoalveolar lavage fluid (BALF) were measured by Griess method. The l e vels of serum HB were measured. RESULTS:① The dysfunctions of hear t, liver and kidney achieved the criterion of MODS in group B at 48 h postburns (B 48 h ). The N O content of group B significantly increased in serum and BALF at B 48 h . ② The cardiac index (CI) and creatine phosphokinase (CK), urea nitrogen (BUN) markedly raised or decreased in group BR at 48 h postburns (BR 48 h ) than B 48 h . The NO cont ents in serum and BALF markedly raised. ③ HB contents in serum markedly raised in group B and BR at 0 h postburns (B 0 h , BR 0 h ) than group SB, B 48 h , R, BR 48 h , but NO was reverse. CONCLUTIONS: ① HB contents in serum markedly raise d at 0 h post burns, but NO was reverse. ② Rho promoted the increases of NO synthesis and the blood perfusion of organs, which might be one of mechanisms to prevent the devel opment of MODS.

Objective To analyze the effects of self-ligating bracket on enlarging the maxillary alveolar arch and to investigate the the non-extraction treatment programs.Methods All patients were used heat activated basic and precise theory for the orthodontist in order to make a plan fordeveloping nickel-titanium wire,then assessed by occlusalplane analysis method.The maxillary model andcephalometric radiographs were collected and measured before the treatment.The data of the resultswereanalyzed.Results After the treatment,the increase of the width of the dental arch was related to the /FMA,crowding degree and canines initial position.The increase of arch length was related to the ∠ANB,crowding degree,malocclusion classification,canines initial position andthe second molar eruption.Conclusion The essential factors include the crowding degree of arch,∠FMA,∠ANB,the angle of the anterior teeth,malocclusion classification,canines initial position andthe second molar eruptioncan affect the result of non-extraction quick self-ligating bracket appliance treatment on enlarging the maxillary alveolar.

Objective To analysis the human T lymphotropic virus (HTLV) infection status in Wenzhou among voluntaty blood donors.Methods Selected 72 417 voluntary blood donors of Wenzhou from from March,1,2016,to November,30,2016,to screen HTLV-Ⅰ / Ⅱ antibody by ELISA method.The positive samples were reexamined two times,two test results of samples were determined positive by ELISA.HTLV positive samples was confiemed by Western Blotting (WB).Results Screened 23 cases of anti-HTLV positive by ELISA method,then confirmed 9 cases of HTLV positive by Western Blotting (WB).HTLV infection rate of Wenzhou blood donors was 0.01％ (9/72 417).Conclusions HTLV infection was found among volunteer blood donors in Wenzhou,but the HTLV infection rate of volunteer blood donors in Wenzhou is still at a relatively low level.

Objective To detect the expression of heat shock protein 47(HSP47) in renal proximal epithelial cell lines (HK-2) and to investigate the role of HSP47 in the progress of transforming growth factor β1 (TGF-β1) induced epithelial-mesenchymal transdifferentiation (EMT) in HK-2 cells.Methods HK-2 cells were exposed to TGF-β1 (0,2.5,5,10 μg/L) for different time (0,12,24,48 h).The expression of HSP47 was examined by Western blotting.Then HK-2 cells were exposed to 10 μg/L TGF-β1,the expressions of vimentin,zona occludens-1 (ZO-1) were examined by Western blotting and real-time PCR.Furthermore,the expressions of p-Smad3 and Smad3 were examined by Western blotting.HK-2 cells were transfected with HSP47 siRNA and siRNA negative control before exposing to TGF-β1.Then the expressions of vimentin,ZO-1 were detected by Western blotting and real-time PCR,meanwhile Western blotting for HSP47,p-Smad3 and Smad3.Results Stimulating HK-2 with TGF-β1 resulted in a significant increased expression of HSP47 in time-and concentration-dependent manner (P ＜ 0.05).Meanwhile,TGF-β1 up-regulated the protein and mRNA expression of vimentin (P ＜ 0.05),and down-regulated the protein and mRNA expression of ZO-1 (P ＜ 0.05),all in time-dependent manner.Stimulating HK-2 with TGF-β1 resulted in phosphorylation of Smad3,which was peaked at 30 min,slightly decreased at 1 h,and then increased again between 24 and 48 h (P ＜ 0.05).Compared to the TGF-β1 group,inhibition of HSP4.7 expression in HK-2up-regulated the protein and mRNA expression of ZO-1,down-regulated the protein and mRNA expression of vimentin (P ＜ 0.05) and down-regulated the ratio of p-Smad3/Smad3.HSP47 siRNA negative control had no significant effect on the expressions of ZO-1,vimentin and p-Smad3/Smad3 (P ＞ 0.05).Conclusion HSP47 can promote the EMT of renal tubular epithelial cell which is possibly via the TGF-β1-Smad3 pathway.

OBJECTIVETo observe the clinical therapeutic effect of acupuncture massage for primary dysmenorrhea and its influence on prostaglandin F2alpha level in menstrual fluid.

METHODSNinety patients with primary dysmenorrhea were assigned equally to the acupuncture group, the massage group and the control group (oral administrated with Somiton tablet). The treatment course was 3 menstrual cycles. The effect in relieving menalgia was evaluated using numerical rating scale (NRS) before and after treatment and change of PGF2alpha content in menstrual fluid was tested as well.

RESULTSThe NRS score was 0.60 +/- 0.67 in the acupuncture group and 0.53 +/- 0.63 in the massage group respectively, significantly lower than that in the control group, which was 2.30 +/- 1.73 (P < 0.01). The PGF2alpha level after treatment was significantly lower in the acupuncture group (176.58 +/- 17.85 ng/L) and the massage group (171.32 +/- 17.15 ng/L) than that in the control group (220.56 +/- 54.47 ng/L), respectively (P <0.01). There showed no difference in the two aspects between the acupuncture group and the massage group (P >0.05).

CONCLUSIONAcupuncture and massage has a reliable therapeutic effect on primary dysmenorrhea, which is superior to that of Somiton tablet, and its mechanism might be exerted by regulating PGF2alpha level in menstrual fluid.

Objective: To optimize the formulation of curcumin-catanionic nanoparticles lipid carriers (Cur-CNLC) by central composite design-response surface methodology (CCD-RSM). Methods: Cur-CNLC were prepared by film dispersion-ultrasonic emulsifying method. A four factor, five-level central composite design was employed, with the solid lipid quality (X1), liquid lipid quality (X2), lecithin quality (X3), and mixed surfactant concentration (X4) as the independent variables. The dependent variables were the entrapment efficiency (Y1) and drug loading (Y2). The data were simulated using multi-linear equation and second-order polynomial equation, the possibly optimal formulation was predicted by response surface method. Results: The entrapment efficiency, drug loading, average particle size, polydispersity, and Zeta potential of the Cur-CNLCs prepared under the optimized conditions were (94.38 ± 2.67)%, (6.93 ± 0.39)%, (235.9 ± 9.6) nm, 0.272 ± 0.017, and (-28.40 ± 0.35) mV, respectively. The bias between the measured values and the predicted ones is less than 5%. Conclusion: The CCD-RSM is effective and suitable for optimizing the formulation of Cur-CNLC.

Objective To understand the present situation of happiness of the elderly living in nursing homes and discuss the related factors.Methods Using the method of questionnaire,the happiness of the elderly over sixty years old and its influencing factors in eleven nursing homes in Changchun were discussed.Results Among many influencing factors,health status,the reasons for living,marital status and self-care status affected the happiness of the elderly in nursing homes.Above all,the health status had the most significant effect on happiness.Conclusions Improvement of the health level of the elderly in nursing homes can improve the happiness of them.

Objective To investigate the effect of overexpression of cartilage oligomeric matrix protein (COMP) on BMP-2 induced cell osteogenic and chondrogenic differentiation of mesenchymal stem cells (MSCs).Methods MSCs,transfected with plasmid DNA encoding recombinant human COMP,were induced to differentiate into osteocytes and chondrocytes by BMP-2.Realtime PCR of osteogenic related markers (Col1a1,RUNX2,OPN,BGP) and chondrogenic related markers (Col2a1,SOX9,Aggrecan) were performed to evaluate the process of cell differentiation.ALP staining,Alizarin red S staining for osteogenic differentiation and alcian blue staining for chandrogenic differentiation were conducted to evaluate the tendency of cell differentiation.Results Real-time PCR assay presented the significantly higher (P＜0.05) COMP expression of MSCs when COMP gene was transfected into cells.The expression level of OPN was significantly (P＜0.05) down-regulated at all the time points in experimental group compared with that in control group.A final significant (P＜0.05) up-regulation of expression appeared in experimental group at the late stage of induction (day 7,14) compared with that in control group,even though a decrease (P＜0.05) expression of Col1a1,RUNX2 and BGP in experimental group occurred at the early stage of induction (day 3).The expression of Aggrecan and Col2a1 in experimental group was up-regulated (P＜0.05) at different time points compared with that in control group.And a significant higher (P＜0.05) expression of SOX9 in experimental group only appeared at day 7 compared with that in control group.ALP staining and Alizarin red S staining were weakened while alcian blue staining was enhanced.Conclusion COMP may inhibit BMP-2 induced osteogenic differentiation and promote BMP-2 induced chondrogenic differentiation,which may provide new insight for cartilage tissue engineering.
