1.Effects of rhPTH (1-34) on the proliferation and cell cycle of human keratinocyte HaCaT cells induced by tumor necrosis factor-α

Objective To investigate the influence of recombinant human parathyroid hormone [rhPTH (1-34)]on the proliferation of HaCaT cells induced by tumor necrosis factor-α (TNF-α) in vitro.Methods Cultured HaCaT cells were treated with various concentrations of rhPTH (1-34) for different durations after incubation with recombinant human TNF-α of 10 g/L for 24 hours.MTT assay and flow cytometry were performed to detect the proliferation and cell cycle of HaCaT cells,respectively.Results As contrast phase microscopy showed,the growth of HaCaT cells was inhibited by rhPTH (1-34) along with a decrease in the growth speed.MTT assay showed a suppressed proliferation of HaCaT cells after being treated with rhPTH (1-34) of 0.05,0.2,0.8,3.2 and 12.8 pmol/L for 36 and 48 hours (P< 0.01 or 0.05).The percentage of cells at G1 phase in HaCaT cells markedly increased (all P < 0.01 ),while that at S phase declined (all P < 0.01 )after 48-hour treatment with rhPTH(1-34) of 0.2,0.8,3.2 and 12.8 μ mol/L.Conclusions rhPTH(1-34) has an obvious inhibitive effect on the proliferation of HaCaT cells induced by TNF-α in vitro,and the effect is in a dose-dependent manner.
2.Correlation between age and general morphology of transverse section of cartilago costalis.

OBJECTIVE:
To study the correlation between age and general morphology of transverse section of cartilago costalis and its forensic significance.
METHODS:
Eighty-six corpses' cartilago costalis from the routine postmortem examination were collected and the morphological features of their transverse section were observed.
RESULTS:
With the increased age, there were regular changes in the color, structure, and material of the general morphology of transverse section of cartilago costalis. But the changes were not affected by gender.
CONCLUSION
The good correlation between general morphology of transverse section of cartilago costalis and age can be used to estimate age of the deceased rapidly.

3.1H NMR based metabonomics study on the antidepressant effect of genipin in rat hippocampus.

The purpose of this study is to explore depression metabolic markers in rat hippocampus and to investigate the anti-depressant effect of genipin and its mechanisms using nuclear magnetic resonance (NMR) metabonomics. Chronic unpredictable mild stress (CUMS) procedure was conducted to establish the depressive rat model. At the beginning of the third week, genipin low dose (25 mg x kg(-1)), middle dose (50 mg x kg(-1)), high dose (100 mg x kg(-1)), and venlafaxine (50 mg x kg(-1)) were given to the CUMS rats separately once daily for two weeks except control and model groups. Rat hippocampus was analyzed by 1H NMR based metabonomics after drug administration for 2 weeks. Significant differences in the metabolic profile of rat hippocampus of the CUMS treated group and the control group were observed with metabolic effects of CUMS including decreasing in glycine and N-acetylaspartate, increasing in inositol, glutamate, lactate, glutamine, taurine and alanine. Genipin showed ideal antidepressive effects at a dose of 50 mg x kg(-1) in rats, decrease of inositol, glutamate, lactate, alanine were observed, while glycine and N-acetylaspartate were increased. Important influence has been found on normal nervous system function of these significant changed metabolites, which suggests that the antidepressant effect of genipin may be played by enhancing the activity of neurons in hippocampus, repairing and improving the function of the neuron. The metabonomics approach is an effective tool for the investigation of the anti-depressant effect and pharmacologic mechanisms of genipin.

4.Expressions of interleukin (IL)-22 and related cytokines in the sera and peripheral blood mononuclear cells (PBMCs) of patients with psoriasis

Objective To investigate the expressions and significance of IL-22 and related cytokines (IL-23pl9 and IL-6) in sera and PBMCs of patients with psoriasis. Methods Sera and PBMCs were obtained from the venous blood samples from 58 patients with psoriasis vulgaris and 20 normal human controls. The PBMCs were subjected to culture for 5 hours followed by the collection of cells and culture supernatant. Then,quantitative real-time RT-PCR was used to examine the mRNA expressions of IL-22, IL-23pl9 and IL-6 in PBMCs, enzyme-linked immunosorbent assay (ELJSA) to detect the level of IL-22 protein in the sera and culture supernatant of PBMCs. Results In the patients with psoriasis and controls, the relative expression level in PBMCs was 4.48 ± 2.64 and 2.35 ± 0.91 respectively for IL-22 mRNA, 6.07 ± 4.09 and 2.61 ± 1.46 respectively for IL-23pl9 mRNA, 3.87 ± 1.49 and 1.48 ± 0.62 respectively for IL-6 mRNA; significant differences were observed between the two groups in all the above parameters (all P < 0.01). ELISA revealed that the level of IL-22 protein in the patients and controls was (86.23 ± 25.58) ng/L and (43.67 ± 14.82) ng/L respectively in the sera (P< 0.01), (119.11 ± 21.51) ng/L and (57.70 ± 13.17) ng/L respectively in the culture supernatant of PBMCs (P< 0.01). Conclusion There is an overexpression of IL-22 in the PBMCs and sera of patients with psoriasis, implying that IL-22 is involved in the pathogenesis of psoriasis.

6.Screening and Fingerprint Analysis of Active Site to Reduce Blood Lipid from Xin-Mai Capsule

This study was aimed to screen main active site to reduce blood lipid from Xin-Mai (XM) capsule and establish HPLC fingerprint of the site, in order to study the correlativity between active site and relevant fractions of its herbs. Solvent extraction was used to separate XM capsule into different polar fractions. Intraperitoneal injection of 75% egg-yolk emulsion was used to establish mice hyperlipidemia models. And the active site was screened. Chromatographic fingerprints of the site and relevant fractions of its herbs were configured by HPLC analysis. The
retention time of peaks was utilized as index to evaluate the correlativity. The results showed that lipid-lowering effect of ethyl acetate extract and garlic essential oil was significant (P<0.01). Fingerprint of the active site in XM capsule was established with 28 fingerprint peaks and the assignment results of 27 peaks were indicated. It was concluded that the active sites to reduce blood lipid of XM capsule were ethyl acetate extract and garlic essential oil. The established fingerprint method can effectively determine the correlativity between the active site and its relevant fractions, which contributed to pharmacodynamic material foundation and quality standard.
7.Imaging diagnosis of acetabular dysplastic coxarthrosis in adult

Objective To investigate the imaging diagnosis of acetabular dysplastic coxarthrosis. Method The imaging of the acetabular dysplasia was analyzed in 51 patients (87 hips). All but four patients were women. The age ranged from 22 to 78 years, and the mean age was 42.6 years. Pelvis radiographs were studied in all cases, and CT scan was performed in 18 cases and MRI in 10 cases. Results The fundamental signs on the plain film included shallow acetabulum, increased obliquity and insufficient coverage of the femoral head by the acetabulum. The CE angles were determined in all the cases, ranging from -10? to 30?, mean 12.9?. The charp angles ranged from 35? to 67?, mean 45.3?. The secondary osteoarthritis were revealed in 75 hips (84.2%). 54 hips (62.1%) had cyst like lesions in weight bear area of the acetabulum or femoral head. 19 hips had subluxation of femoral head. CT and MRI displayed small cystlike lesions and forward shift of the femoral head, which could not be shown on plain film, respectivly in 13 and 10 hips. Conclusion The cyst like lesion of subarticular region is a common sign in acetabular dysplastic coxarthrosis. CT or MRI can show the early osteolytic lesion and forward subluxations.
8.Strategies for Prompting the Production of Proteins in Aspergillus oryzae

Aspergillus oryzae is an important microorganism,which was widely used in the fields of food,brewing,pharmacy and fermentation industry etc and was termed as a generally regarded as safe(GRAS) organism by the United States Food and Drug Administration(FDA).The strategies for promoting the expression of homologous and heterologous proteins in Aspergillus oryzae were reviewed.Which included the usage of strong promoters,multicopies of coding genes,optimization of culture medium and overexpression of the hemoglobin domains(HBD) etc.Heterologous proteins were usually exhibited low expression in Aspergillus oryzae due to the degradation by the proteinases from host.Therefore,development of proteinase auxotrophic stains of Aspergillus oryzae as host is neccessary.In addition,fusion of the heterologous protein with a highly secreted protein of Aspergillus oryzae was an alternative way to prompt the expression of heterologous protein in Aspergillus oryzae.
9.Effect of hejie decoction on T-cell receptor V beta 7 gene expression in patients of chronic hepatitis B.

OBJECTIVETo explore the therapeutic effect of Hejie Decoction (HJD) in treating chronic hepatitis B and its relationship with T-cell receptor V beta 7 (TCRV beta 7) gene expression.
METHODSForty-five patients of chronic hepatitis B were randomly divided into two groups. The 30 patients in the treated group were treated by HJD, and the 15 patients in the control group were treated by conventional western medicine. The therapeutic effect and changes of TCRV beta 7 gene expression after treatment were observed.
RESULTSAfter 6 months treatment, the ALT level in the two groups were obviously decreased (P < 0.01). No significant difference was shown in comparison of the total effective rate between the two groups but it did show in comparison of markedly effective rate between them. TCRV beta 7 expression was detected in 5 patients of the treated group, and HBV-DNA and HBeAg in the 5 patients were all negatively converted. While in the control group, no one had TCRV beta 7 expression detected, either no one with negative conversion of HBV-DNA and HBeAg. TCRV beta 7 could not be detected in all the patients whose HBV-DNA and HBeAg hasn't negatively converted, though their liver function could be normalized.
CONCLUSIONHJD might have the effect of regulation on TCRV beta 7 expression, it possibly is the important way for HBV replication inhibition and virucidal action of HJD.

10.Willingness to influenza and pneumonia vaccination and its influence factors in community COPD patients

Objective] To investigate influence factors for willingness to influenza and pneumonia vaccination in COPD patients, thus providing a reference for relevant health education and vaccination pro-motion. [ Methods] A total of 629 COPD patients aged 60-74 were selected as subjects from 3 dis-tricts in Ningbo City of Zhejiang Province.Questionnaire involving personal information, occupation, in-come, behavior and willingness to vaccination were used for the survey through October to November in 2013 .Multivariate unconditional logistic regression was used to analyze the influence factors in vaccination willingness. [ Results] Influenza and pneumonia vaccination rate was found to be 10.49%, and there were 36.88% of the subjects who voluntarily vaccinated against influenza and pneumonia in community hospital.Factors as high income, vaccination within 5 years contributed to higher vaccination willingness;and older age, misgivings about adverse reaction of vaccination were factors affecting the willingness to vaccination. [ Conclusion ] Extensive health education for adult vaccination should be further developed.For the older, COPD patients and other special population, influenza and pneumonia vaccination should be offered free of charge by government, so as to raise the vaccination rate.
