2.Effects of puerperium pelvic floor muscle training on pelvic floor muscle strength

Objective To analyze the effects of puerperium pelvic floor muscle training on pelvic floor muscle strength and its clinical significance. Methods One hundred postpartum women were included, with full-term singleton pregnancies and with complete follow-up records from Obstetrics Department of Peking University First Hospital between March 1, 2013 and October 31, 2013. Women with vaginal birth and cesarean birth commenced pelvic floor muscle training twice a day from 24 and 72 h after delivery, respectively. According to the different training frequencies, the subjects were divided into three groups: never-training group, occasional-training group (<6 times per week) and regular-training group (≥6 times per week). All patients received pelvic floor muscle strength measurement 6-8 weeks after parturition. And the strength of type Ⅰ and type Ⅱ pelvic floor muscle fiber was divided into 0, Ⅰ, Ⅱ, Ⅲ, Ⅳ and Ⅴgrade. The abnormal strength of typeⅠand typeⅡpelvic floor muscle fiber standed for the grades lower thanⅢ. We compared the general conditions, delivery modes and abnormal ratio of type Ⅰ and type Ⅱ pelvic floor muscle fiber among the three groups, analyzed the relativity between the level of pelvic floor muscle fiber strength and pelvic floor muscle training frequency, and analyzed the influential factors of pelvic floor muscle fiber strength. Statistical analyses were performed by one-way ANOVA, rank-sum test, Pearson χ2 test, Kruskal-Wallis nonparametric test, Nemenyi test, Spearman rank correlation analysis and multivariate Logistic regression analysis. Results There was no statistical difference in age, gestation at delivery, parity, body mass index before delivery, neonatal birth weight and delivery mode among the never-training group (21 cases), occasional-training group (30 cases) and regular-training group (49 cases) (all P > 0.05). There was no statistical difference in labor time of first, second and total stage and episiotomy rate among the vaginal birth cases of the three groups. The abnormal ratio of type Ⅰ muscle fiber strength among the three groups was 100% (21/21), 77% (23/30) and 6% (3/49), respectively, while that of type Ⅱ muscle fiber strength was 100% (21/21), 53% (16/30) and 20% (10/49), respectively. And there were significant statistical differences among the three groups (F=119.16 and 77.84, both P<0.01). Spearman rank correlation analysis indicated that the level of typeⅠand typeⅡmuscle fiber strength had a significant positive correlation with pelvic floor muscle training frequency (r=0.88 and 0.79, both P<0.01). Multivariate Logistic regression analysis showed that pelvic floor muscle training was an influential factor for pelvic floor muscle strength level (95%CI:0.000-0.193, P<0.01). Conclusions Puerperium pelvic floor muscle training can help enhance the tension force of pelvic floor muscle, might benefiting the postpartum recovery of pelvic floor muscle function.
3.Inhibitory effect of insulin-like growth factor binding protein-related protein 1 on retinal angiogenesis in vitro

Background Antagonists against vascular endothelial growth factor (VECF) play key roles in treating and preventing neovascular ophthalmopathy. As a novel anti-angiogenic factor, insulin-like growth factor binding protein-related protein 1 (IGFBP-rP1) might be an antagonist against VEGF in eye. Objective This study was to explore the inhibitory effect of IGFBP-rP1, a novel anti-angiogenic factor, on VEGF-induced retinal angiogenesis in vitro. Methods The retina-choroid endothelial cell line ( RF/6A ) was cultured in DMEM containing 10% fetal bovine serum. Culture cells were divided into control group(free-serum culture group) ,10mg/L VEGF culture group and different concentrations of IGFBP-rP1 (50,100,200 mg/L) +10 mg/L VEGF group. The expression of IGFBP-rP1 in the cells was detected by immunofluorescence assay. The proliferation and migration of RF/6A cells were evaluated using MTS colorimetric assay and the chemotactic motility assay, respectively. Flow cytometry was used to detect the apoptosis of RF/6A cells. Results The immunofluorescence assay RF/6A cells showed the green fluorescence in cytoplasm and red fluorescence in nuclei after cells were exposed to any concentration of IGFBP-rP1 ,but only red fluorescence was seen in nuclei in control cells. After stimulation of 10 mg/L VEGF,the proliferation value (A490) was elevated and the numbers of cell migration were increased in comparison with control group (t = -15. 191, P = 0. 000; t = -21. 274, P = 0. 000 ) , but the cellular apoptosis rate was lower than the control group (t - 10. 228, P = 0. 000 ) . After treated with various concentrations of IGFBP-rP +10% VEGF, the proliferation and migration of RF/6A cells were significantly decreased in comparison with only 10% VEGF group (F = 534. 158,P = 0. 000;F = 2742. 323,P = 0.000,respectively) ,and the inhibitory effects were gradually enhanced with the increase of IGFBP-rP1 levels (P<0. 05). The apoptosis rate of RF/6A cells in 50,100 and 200 mg/L + 10 mg/L VEGF groups increased by ( 1. 26±0. 04)% ,( 1. 50±0. 07)% and ( 1. 93±0. 27)% respectively,showing significant differences among different groups ( F = 274. 273, P = 0. 000). Conclusion IGFBP-rP1 inhibits the proliferation and activity of retina and choroid endothelial cells induced by VEGF at a concentration-independent manner. It appears to be as a novel endogenous inhibitory factor in retinal angiogenesis.
4.A comparative study on the effect of fixation for guinea pigs eyeballs among three different fixation solution

· AIM:To compare the effect of fixation and select the optimal fixation solution and condition for PAS staining of guinea pig eyes among three different fixation solution:4% paraformaldehyde solution,4% glutaraldehyde solution and Davidson solution.· METHODS:Totally 30 healthy guinea pigs were divided into 6 groups:Group Ⅰ-Ⅱ were fixed in 4% paraformaldehyde solution and 4% glutaraldehyde solution for 24h,respectively;Group Ⅲ-Ⅴ were fixed in Davidson solution for 3,6 and 24h,respectively;and Group Ⅵ were fixed in Davidson solution for 3h and then transferred into 10% neutral formaldehyde solution for 48h.All groups were sectioned by routine section method and undergone PAS staining,and then observed by light microscope.· RESULTS:It was found that the group which was fixed by Davidson solution for 3h,remained the most complete structure for PAS staining (Group Ⅲ).While the effect of fixation for the group which was transferred into 10% neutral formaldehyde solution for preserving for 48h after fixing in Davidson solution for 3h was also acceptable for PAS staining (Group Ⅵ).The retinal cells remained clear and in order for both groups which was mentioned above.· CONCLUSION:The best fixation condition for PAS staining for eyes of guinea pigs is fixation in Davidson solution for 3h among these fixation conditions,while it is also suitable to transfer the eyes into neutral formaldehyde solution after fixing in Davidson solution for 3h for preserving for long periods,which is not severely reduced the effect of fixation for PAS staining.
5.Application of near infrared spectroscopy to identify authenticity of Polygonum multiflorum.

OBJECTIVETo establish a near-infrared qualitative analysis model to identify the authenticity of Polygonum multiflo- rum and distinguish processed products Polygoni Multiflori Radix.
METHODThe NIR spectra were peformed on over 30 batches of P. multiflorum and Polygoni Multiflori Radix samples and the adulterants Cynanchum bungei, Pteroxygonum giraldii, Polygonum cillinerve to establish the qualitative discriminant model and the conformity test model of Polygonum multifiorum , and cluster analysis was used to analyze the samples from different origins.
RESULTThe model is able to identify correctly P. multiflorum with its counterfeit, and distinguish between P. multiflorum and Polygoni multiflori Radix.
CONCLUSIONNear-infrared spectroscopy can be applied in the identification of P. multiflorum, which could be used to screen Chinese herbal medicine preliminarily.

6.Effect of two different body positions on the scalp acupuncture among infant

Objective to explore the best bady position for the scalp acupuncture among infant.Methods A total of 160 cases in out patients received scalp acupuncture were recruited and randomly divided into the experiment group ( n =80 ) and control group ( n =80 ).Experiment group adopted the type of baby cartier and control group adopted the type of parents' arms encircle when they received the scalp acupuncture.Then,the success rate of one-time puncture,the repeated puncture times among infant that fluid volume was 50 ml and the satisfaction of patients' family on the nursing skill in two groups were observed and compared.Results The success rate of one-time puncture of experiment group was 90.00% and 87.50% of the control group,and the difference was statistically significant( P >0.05).The rate of repeated puncture of experiment group was 11.25% which was significantly lower than 27.50% of the control group( x2 =0.652,P < 0.01 ),and the satisfaction of patients' family on the nursing skill of experiment group was 95.00% which was significantly higher than 86.25% of the control group( x2 = 8.721,P < 0.01 ).Conclusions The position of baby cartier is better than the position of parent' s arms encircle when infant receive the scalp acupuncture,which can be popularized in clinic.
7.Willingness to influenza and pneumonia vaccination and its influence factors in community COPD patients

Objective] To investigate influence factors for willingness to influenza and pneumonia vaccination in COPD patients, thus providing a reference for relevant health education and vaccination pro-motion. [ Methods] A total of 629 COPD patients aged 60-74 were selected as subjects from 3 dis-tricts in Ningbo City of Zhejiang Province.Questionnaire involving personal information, occupation, in-come, behavior and willingness to vaccination were used for the survey through October to November in 2013 .Multivariate unconditional logistic regression was used to analyze the influence factors in vaccination willingness. [ Results] Influenza and pneumonia vaccination rate was found to be 10.49%, and there were 36.88% of the subjects who voluntarily vaccinated against influenza and pneumonia in community hospital.Factors as high income, vaccination within 5 years contributed to higher vaccination willingness;and older age, misgivings about adverse reaction of vaccination were factors affecting the willingness to vaccination. [ Conclusion ] Extensive health education for adult vaccination should be further developed.For the older, COPD patients and other special population, influenza and pneumonia vaccination should be offered free of charge by government, so as to raise the vaccination rate.
8.Determination of luteolin and luteolin-7-beta-D-glucoside in Chrysanthemum morfolium Ramat. from different collection time by RP-HPLC.

OBJECTIVETo observe the content variation of luteolin and luteolin-7-beta-D-glucoside in Chrysanthemum morifolium Ramat. (CMR) from different collection time.
METHODSRP-HPLC was used to analyze these two components in CMR collected in 2001 and 2002.
RESULTThe content of luteolin was significantly lower than that of luteolin-7-beta-D-glucoside. Furthermore, the former showed no marked changes during collection, while the latter did not varied markedly in early collection but decreased significantly in later collection.
CONCLUSIONThe content of luteolin-7-beta-D-glucoside reflects the quality of Chrysanthemum morifolium Ramat. more viably than that of luteolin.

9.Analysis of Distribution and Drug Resistance of Bacteria in Elderly Patients with Vaginal Infection

OBJECTIVE:To provide reference for the formulation of clinical therapy plan for elderly patients with vaginal in-fection. METHODS:From Jun. 2014 to Oct. 2016,200 elderly patients with vaginal infection were selected from Wuhan Wuchang Hospital (called our hospital for short). Leucorrhea routine examination,bacterial culture,identification and drug sensitivity test were all performed for the secretions of vaginal side wall. The occurrence of vaginal infection was compared among different age groups. The distribution of bacteria,distribution of cleanliness and drug resistance were observed. RESULTS:The types of vaginal infectious diseases in 200 patients included Candida vaginitis,Trichomonas vaginitis,bacterial vaginitis,nonspecific vaginitis. The incidence of vaginal infection in 66-70 age group was significantly higher than 60-65 age group and 71-80 age group,the incidence of vaginal infection in 60-65 age group was significantly higher than 71-80 age group,with statistical significance(P<0.05). A to-tal of 205 strains of bacteria were detected,most of which were Gram-positive bacteria,except for Escherichia coli. Streptococcus was the most common pathogen,with a total of 76 strains(37.07%),followed by Staphylococci(48 strains,23.41%). The vagi-nal secretion cleanliness was grade Ⅰ,Ⅱ,Ⅲ and Ⅳ respectively in 10,70,90,30 cases. The amount of bacteria increased with the increase of vaginal secretion cleanliness,and Streptococcus and Staphylococcus infection were the most common in patients with Ⅲ to Ⅳ degree. Resistance rate of bacteria to penicillin,gentamicin and erythromycin were relatively higher while most of them to teicoplanin and linezolid were relatively lower;they were all sensitive to vancomycin (resistance rate of 0). CONCLU-SIONS:There are many types of vaginal infectious diseases in our hospital,and most of them are 66-70 years old. The majority of bacteria were Gram-positive bacteria,most of them were Streptococcus. The drugs which are sensitive to pathogen include vancomy-cin,teicoplanin and linezolid. Antibiotics should be rationally selected according to drug sensitivity test.
10.Antimicrobial resistance and resistance gene carriage by Salmonella bacteria in environmental sewage in Guangzhou

This study explored the antibiotic resistance and resistance gene carriage of 140 Salmonella strains isolated from environmental sewage in Guangzhou city between March 1,2022,and November 30,2022.The micro broth dilution method was used to select 17 antibiotics for susceptibility testing.According to whole genome sequencing results,the CARD resistance database was used to obtain corresponding resistance genes.High antimicrobial resistance rates above 80%were observed a-gainst ampicillin,tetracycline,streptomycin,chloramphenicol,and cotrimoxazole.The intermediation rate of polymyxin E and ciprofloxacin exceeded 60%.The multiple drug resistance status was severe,and the rate of multiple drug resistance was as high as 92.86%.The strains carried multiple types of resistance genes,particularly for aminoglycosides,with a carriage rate as high as 92.68%.The resistance of Salmonella in environmental wastewater in Guangzhou to one or more drugs was severe,and the overall multi-drug resistance rate gradually increased over time.The resistance spectrum was diverse,and the resistance mechanism,mediated by mobile genetic elements such as re-sistance genes,was found to be the main cause of resistance to one or more drugs.
