1.Agroinfection of sweet potato by vacuum inifltration of an infectious sweepovirus

Sweepovirus is an important monopartite begomovirus that infects plants of the genus Ipomoea worldwide. Development of artiifcial infection methods for sweepovirus using agroinoculation is a highly efficient means of studying infectivity in sweet potato. Unlike other begomoviruses, it has proven difficult to infect sweet potato plants with sweepoviruses using infectious clones. A novel sweepovirus, called Sweet potato leaf curl virus-Jiangsu (SPLCV-JS), was recently identiifed in China. In addition, the infectivity of the SPLCV-JS clone has been demonstrated in Nicotiana benthamiana. Here we describe the agroinfection of the sweet potato cultivar Xushu 22 with the SPLCV-JS infectious clone using vacuum inifltration. Yellowing symptoms were observed in newly emerged leaves. Molecular analysis confirmed successful inoculation by the detection of viral DNA. A synergistic effect of SPLCV-JS and the heterologous betasatellite DNA-βof Tomato yellow leaf curl China virus isolate Y10 (TYLCCNV-Y10) on enhanced symptom severity and viral DNA accumulation was conifrmed. The development of a routine agroinoculation system in sweet potato with SPLCV-JS using vacuum inifltration should facilitate the molecular study of sweepovirus in this host and permit the evaluation of virus resistance of sweet potato plants in breeding programs.

5.Clonidine premedication for isoflurane-induced hypotension: a rendomized double-blind clinical study

The effects of clonidine as a premedicant were studied during isoflurane-indueed hypotension(IIH). Twenty-four adult patient, ASA grade Ⅰ-Ⅱ, scheduled for elective cerebral surgery, were randomly allocated into two groups. Atropine 0.5mg and luminal sodium 100mg were given I. M. as premedication in group Ⅰ 30 mins before induction, and clonidine 5?g?kg~(-1)P. O. was combined in group Ⅱ 90 mins prior to induction. The methods of both groups were similar in induction and maintenance of anesthesia. The status of hypotension was induced and kept by inhaltion of isoflurane, with MAP being decreased by 25% or so. The arterial plasma Concentrations of adrenaline and noradrenaline were measured by electrochemistry method, at preoperation, prior to hypotension, 15 and 30 mins following IIH and 30 mins after MAP recovery, respectively. The results revealed that as compared with the values in group Ⅰ, the inspired and end-tidal concentrations of isoflurane reduced significantly during induction and maintenance of controlled hypotension (P0.05). It is suggested that clonidine premedication can increase the efficiency of isoflurane for controlled hypotension and do not affect the plasma catecholamine concentration and airway function.
6.An analysis of influence factors during isokinetic eccentric test of knee joint

Objective: To analyze some influence factors during isokinetic eccentric test. Method: Twenty normal males were tested by Biodex isokinetic test and rehabilitation system. Their peak torque(PT) and peak torque to body weight (PT/BW) were analyzed under different conditions. Result: It was found that velocity and contraction mode influence the results significantly( P<0.01). Conclusion: Many factors must be considered during isokinetic eccentric test and training.
7.The Influence of Angle and Time on Isokinetic Concentric Constriction of Knee Joint

20 normal males were examined by BIODEX isokinetic test and rehabilitation system,and data collected were used to observe the influence of angle and time on the isokinetic concentric constriction of knee joint. It was found that angle and time, especially angle factor obviously affected the testing results.

Polo-like kinase 1(PLK1)is a serine/threonine kinase which is highly conserved, and its activity is elevated in many human tumor cell lines.Lots of reports have shown that PLK1 is critical for all stages of mitosis.The newest report finds that PLK1 is required for DNA synthesis, maintenance of DNA integrity, and prevention of cell death.PLK1 physically binds to the tumor suppressor p53 in mammalian cultured cells, and inhibits its transactivation activity as well as its function in activate checkpoint protein and its pro-apoptotic function.Also PLK1 is involved in the development of tumors, and Polo-like kinase1(PLK1)regulates IFN induction by MAVS, breakdown the innate immunity. Various of inhibitors of PLK1 show the feature of high performance and low toxicity, suggesting that PLK1 may be a feasible target for cancer therapy and immunity therapy.
9.Application value of serological markers associated with iron metabolism in anemia diagnosis

Objective To explore the application value of the detection of vitamin B12 (VitB12 ) ,folic acid (FA) and four iron met-abolic indexes including serum ferritin (SF) ,transferring (TF) and total iron binding capacity (TIBC) and serum iron (SI) in the diagnosis and differential diagnosis of anemia .Methods 126 patients with anemia and 30 healthy controls were enrolled from Janu-ary to May 2013 and determined serum VitB12 ,FA ,SF ,TF ,TIBC and SI levels for conducting the comparison with each other . Meanwhile ,the patients with anemia were extracted bone marrow to carry out ferric stain and morphological test ,their detection re-sults and the results of above-mentioned serological markers determination were performed the comparison of the diagnostic accord-ance rate .Results The serum levels of various iron metabolism-related indexes had significant differences between the anemia group and the healthy control group (P<0 .05) ,and various different kinds of anemia had their comparatively specific serological characteristics .In 126 cases of anemia ,the diagnostic accordance rate of the bone marrow examination was 82 .5% ,which was sig-nificant higher than that of the serological examination(69 .8% )(χ2 =5 .600 ,P=0 .018);nevertheless ,in 23 cases anemia unable to determine the diagnosis by bone marrow examination ,the diagnostic accordance rate of the serological examination was 82 .6% (19/23);the inconsistent rate of the two examinations was 25 .4% (32/126) .Conclusion The bone marrow examination and the sero-logical examination have different emphasis points for the anemia diagnosis and have their own advantages ,which cannot replace each other .Their combination use has certain clinical value in the differential diagnosis between iron deficiency anemia (IDA) with anemia of chronic disease complicated with iron deficiency (ACD/ID) ,myelodysplastic syndromes (MDS) ,megaloblastic anemia (M A ) and mixed cellular anemia .
10.Disinfection effect of root canal with non-filling medication in root canal therapy

Objective: To observe the clinical disinfection effect of root canal with non-filling medication. Methods: 36 patients who required root canal therapy(RCT) for apical periodontitis (acute or chronic) or pulp necrosis were involved in the study.RCT was completed after two visits in both groups. Non-filling medication was used as disinfection in 18 teeth of 18 patients (experimental group) while FC in another 18 teeth of 18 patients (control group) randomly at first visit.One week later,root canal was regularly filled to complete RCT.From each case, bacteria specimens were collected and identified before sealing and filling. Results: Bacteria were detected in every specimen in both groups before sealing,which were mixture of aerobes and anaerobes.One week later,only one strain was detected in one specimen of each group. Conclusion: Non-filling medication can be used as an effective disinfection to sterilize root canal.
