1.Clinical study on auricular plaster therapy in the auxiliary treatment of insomnia caused by liver fire disturbance in a sub-healthy population

Objective:To investigate the clinical efficacy of psychotherapy combined with auricular plaster therapy in the auxiliary treatment of insomnia caused by liver fire disturbance in a sub-healthy population.Methods:Eighty sub-healthy patients with insomnia caused by liver fire disturbance who received treatment in Wenzhou Hospital of Traditional Chinese Medicine from June 1, 2017 to December 31, 2018 were included in this study. These patients were randomly assigned to receive cognitive behavioral therapy (control group, n = 40) or cognitive behavioral therapy combined with auricular plaster therapy (observation group, n = 40) for 4 weeks. Polysomnography was used to evaluate the sleep condition before and after treatment. The Pittsburgh Sleep Quality Index was used to evaluate the severity of insomnia. The World Health Organization Quality of Life Scale Brief Version (WHOQOL-BREF) was used to evaluate the quality of life of patients. The therapeutic effect was compared between the two groups. Results:After treatment, total sleep time [(421.25 ± 11.58) min vs. (456.25 ± 11.23) min], sleep efficiency [(70.32 ± 4.44)% vs. (76.58 ± 5.89)%], non-rapid eye movement stage 2 (N2) sleep [(43.25 ± 4.44)% vs.(45.62 ± 4.36)%], N3 sleep [(14.25 ± 4.52)% vs. (18.52 ± 5.55)%], rapid eye movement (R) sleep [(21.98 ± 5.21)% vs. (24.66 ± 4.87)%] in the control group were significantly shorter or lower than those in the observation group ( t = 13.723, 5.367, 9.035, 2.408, 3.772, 2.376, all P < 0.05). After treatment, sleep quality [(1.85 ± 0.54) points vs. (1.10 ± 0.42) points], sleep-onset latency [(1.52 ± 0.51) min vs. (1.08 ± 0.35) min], sleep duration [(1.30 ± 0.41) min vs. (0.75 ± 0.41) min], sleep efficiency [(1.53 ± 0.44) points vs. (1.02 ± 0.33) points], sleep disturbance [(1.20 ± 0.41) points vs. (0.78 ± 0.31) points], hypnotics [ (0.11 ± 0.02) points vs. (0.04 ± 0.01) points], daytime dysfunction [(1.65 ± 0.47) points vs. (1.03 ± 0.28) points] and total scores [(9.11 ± 1.33) points vs. (5.80 ± 1.01) points] in the control group were significantly higher than those in the observation group ( t = 6.933, 4.499, 5.999, 5.864, 5.167, 19.789, 7.167, 12 535, P < 0.05). After treatment, the scores in physical health [(13.85 ± 2.22) points vs. (17.86 ± 2.01) points], psychological domain [(15.52 ± 1.89) points vs. (16.35 ± 1.33) points], social relationships [(16.01 ± 1.58) points vs. (19.52 ± 1.85) points] and environment domain [(17.52 ± 1.90) points vs. (20.89 ± 2.22) points] in the control group were significantly lower than those in the observation group ( t = 8.468, 4.499, 9.124 and 7.294, all P < 0.05). After treatment, total effective rate in the observation group was significantly higher than that in the control group [95.0% (38/40) vs. 72.5% (29/40), χ 2 = 7.440, P < 0.05]. Conclusion:Psychotherapy combined with auricular plaster therapy can significantly increase the sleep duration and sleep efficiency and improve the quality of life of sub-healthy patients with insomnia caused by liver fire disturbance, which is worthy of clinical promotion.
2.Effects of insulinoma-glucagonoma clone 20 isoforms on cancer cells

Insulinoma-glucagonoma clone 20 can express at least 6 isoforms in human cells.These isoforms can affect cell apoptosis and proliferation through apoptosis related signaling pathway,such as TRAIL or TNF signaling pathways.Furthermore,insulinoma-glucagonoma clone 20,as a GTP-GDP exchange factor,participates in the transportation of nerve synaptophysins.
3.Nutritional risk screening in preoperative patients with colorectal carcinoma and its influence on postoperative outcomes

Objective To evaluate the preoperative nutritional status of patients with colorectal carcinoma by using the nutritional risk screening 2002 (NRS2002) score and its prediction for postoperative outcomes.Methods Retrospectively evaluated the nutritional risk of 259 colorectal cancer patients with NRS2002 score in terms of different sexes,ages,tumor location,pathology and differentiation,Dukes stages.The relationship between different NRS2002 score and postoperative complications and hospital stay were also observed.Results NRS2002 score was applicable in 243 cases,≥ 3 scores of 80 cases,< 3 scores of 163 cases.Gender,histological differentiation and before surgery NRS2002 score unrelated (P > 0.05).Ages,tumor location,and Dukes stages related NRS2002 score (P < 0.01).No case of perioperative death,hospital stay 10-101 (21.6 ± 9.7) d.The incidence of complications in ≥ 3 scores patients was significantly higher than that in < 3 scores patients [30.0% (24/80) vs.17.2 % (28/163),P < 0.05],hospital stay in ≥ 3 scores patients was significantly shorter than that in < 3 scores patients[(20.5 ± 8.2) d vs.(23.9 ± 11.9) d,P< 0.05].Multiple Logistic regression analysis showed that gender,age,tumor location,histological differentiation,Dukes stages,surgical and colorectal cancer complications unrelated (P > 0.05),NRS2002 score associated with colorectal cancer complications (P < 0.05).Pearson correlation analysis showed that hospital stay correlated with NRS2002 score (P < 0.01).Conclusions NRS2002 score is applicable in Chinese colorectal cancer patients.The youth and the elderly,the proximal part of colon,and Dukes D stage patients are more liable to develop preoperative nutritional risk than those of middle age,the distal part of colon,or other Dukes stages.≥ 3 scores predicts higher postoperative complications and longer hospital stay.Preoperative nutritional support is necessary in patients with ≥ 3 scores.
4.Study on the Clinical Feature and Influencing Factors of Pneumonia in Patients With Lung Cancer

Objective To explore the clinical feature and influencing factors of pneumonia in patients with lung cancer,in order to these patients might be timely diagnosed and treated correctly.Methods A series of 1495 cases of pneumonia in patients with lung cancer in recent 10 years were reviewed as their clinical characteristics, pathological cell types, imaging study, bacteriological monitoring, chemotherapy/radiotherapy, antibiotic treatment and prognosis.Results The infection rate of pneumonia in patients with lung cancer was 60.8%, most of them were occurred in patients treated after chemotherapy/radiotherapy, especially in patients being treated after glucocorticoid. The prognosis was poor.Conclusions The clinical situation of pneumonia in patients with lung cancer is not typical. The symptom of respiratory tract is not obvious. CT scan demonstrated obstructive pneumonia and atelectasis. Gram-negative bacteria were the mainly pathogen. The influencing factors are related to the smoking, chronic consumption, obstruction of bronchus, lower resistance after chemotherapy/radiotherapy and the basic diseases. The methods of treatment are also prelimilary discussed.
5.Study on Quality Standard for Biqing Suppository

Objective To establish a quality standard for Biqing Suppository. Methods Rhizoma Dioscoreae Septemlobae, Rhizoma Acori Tatarinowii, Radix Et Rhizoma Salviae Miltiorrhizae and Cortex Phellodendri Chinensis were qualitatively identified by TLC method. The content of berberine hydrochloride in Biqing Suppository was determined by HPLC method. The chromatographic column was Waters C18 (250 mm×4.6 mm, 5 μm);the mobile phase was acetonitrile∶water (0.2%phosphoric acid-0.02%three triethylamine)=25∶75;the flow rate was 1 mL/min;the detection wavelength was 265 nm. Results The spots of qualitative identification method were clear without interference. Berberine hydrochloride had good linear relationship in the range of 0.235-9.40 μg;the regression equation was Y=40 033 998.176 3X-85 021.2, r=0.999 7;the average recovery rate was 99.16%, RSD=3.45%. Conclusion The method is simple, flexible and reproducible, which can be used as the quality control standard for Biqing Suppository.
6.Screening of conjugated peptides of Endoglin from phage display peptide library

Objective To screen rhEndoglin-binding peptides from phage displayed 12-peptide library. Methods The rhEndoglin was used as target protein for biopanning of phage-displayed 12-peptide library. After three rounds of screening, 16 phage clones were randomly selected and identified by sandwich ELISA. The positive phage clones were sequenced, and the fuse peptides were deduced by the DNA sequence. Further we identified the affinity and speciality by competitive inhibition test. Results Six of 16 phage clones were identified as positive clones by competent ELISA which could bind to rhEndoglin. Five sequences were obtained, and the amino acid sequence in two of these five was AHKHVHHVPVRL. Conclusion The rhEndoglin-binding peptides can be obtained by screening phage random peptide library. It could play an important role in early diagnosis of ovarian cancer.
7.Determination of functional affinity of rhEndoglin conjugated eptide

Objective To assess functional affinity of rhEndoglin conjugated eptide in order to identify the affinity.Methods We measured the functional affinity of rhEndoglin conjugated eptide with non-competitive ELISA method.After coating the peptide by BSA binding with glutaral couple,affirming the best concentration,best time of peptide to coat the plate and the coating coefficient,and the proper binding time of peptide to Endoglin to reach an equilibrium,we plotted the standard curve of the binding reaction of peptide and Endoglin.Results The affinity constant of peptide and anti-hEndoglin IgG is respectively(2.956?0.749)?106mol/L and(7.403?10.76) ?108mol/L.Conclusion The peptide we got from the peptide liberary can strongly bind to Endoglin,providing a theoretical basis for its clinical use.
8.Increased carotid intima-media thickness in type 2 diabetes mellitus and risk factors.

Objective To investigate the carotid intima-media thickness(C-IMT) in type 2 diabetes mellitus(T2DM) and the relationship with risk factors. Methods From 2002-01 to 2003-03, we measured C-IMT by B-mode ultra-sonography in 67 patients with different durations of T2DM. Blood was drawn from patients for the analysis of serum creatinine and C-reactive protein(CRP). 24-hour urine collection was performed to determine the level of urine creatinine clearance. Results C-IMT in patients increased with durations and with decreasing creatinine clearance. Multiple regression analysis revealed that C-reactive protein significantly and independently affected the C-IMT. Conclusion C-IMT increases with durations of T2DM and with BMI of the patients. CRP is the risk factor for carotid atherosclerosis in T2DM.

The hemodynamic effects of deliberated hypotension induced by PGE1 or SNP on 14 dogs anesthetized by ketamine were studied separately.MAP was decreased by 30%~40% after infusing PGE,or SNP Hemodynamic variables were measured before and 15 min 30 min after hypotension and 15 min after the discontinuation of infusion.The results indicated that SVRI, PVRI, MPAP, LVSWI and RVSWI all decreased significantly during PGE1-or SNP-induced hypotension. CVP decreased and HR increased significantly during SNP-induced hypotension, but with no significant change during PGE1-induced hypotension and with SVI. CI. PCWP remained unchanged in these two groups throughout the experiments.
