1.Experimental Study of the Bionic Technology for the Construction of Small-caliber Vascular Prostheses

Objective:To study the feasibility of using the bionic technology to construct small-caliber vascular prostheses with modified SIS.Methods: The vascular endothelial cells and smooth muscle cells were separated from canine saphenous artery,the cells blended with collagen gel,which were planted respectively on the SIS films,these films were rolled into the biologic three-layer prostheses around a 3mm diameter polyethylene tube;one-layer prostheses without these cells and collagen gel served as control.These 2 types of prostheses were implanted into the defect of bilateral canine femoral by anastomosis in 15 dogs.doppler colour ultrasonic,histology detection and electron microscope examination were done postoperatively.Results: By 12 weeks postoperatively,14 biologic vascular prostheses had kept well patency,the patency rate being 93.3%,the biologic structure like blood vessels had formed,the inner surface of the vessels had been covered with full endothelial cells,a lot of smooth muscle cells had been found in the media of vascular walls in regular line;the patency rate in control group was 60.0%,the endothelial cell coverage was incomplete.Conclusions: The bionic vascular prostheses showed potent blood compatibility,which could keep long-term patency in vivo.Curative effect of repairing the small-caliber vessel defect was well satisfactory.
2.Effects of Xianfu Wenyang Tongluo Drink on ANCA Associated Antigen in Thromboangiitis Obliterans Rat Models

Objective To observe the effects of Xianfu Wenyang Tongluo Drink (XFWYTLD) on ANCA associated antigen in rats with thromboangiitis obliterans;To discuss its mechanism. Methods Totally 72 SPF male Wistar rats were randomized into sham-operation group, model group, Mailuoning Granule group and XFWYTLD low, medium, and high dose groups. Method of femoral artery injecting sodium laurate was used to duplicate models. From the second day after modeling, the rats in sham-operation group and model group were fed with distilled water, while other groups received gavage with relevant medicine. 15 days later, the activity of MPO in serum was detected through the method of ultraviolet spectrophotometry;the protein expressions of PR3 and LAMP-2 in femoral artery and the surrounding tissues were detected through the method of immunohistochemisty. Results Compared with sham-operation group, the activity of MPO in serum and the protein expressions of PR3 and LAMP-2 in rats of model group were significantly higher;Compared with model group, the activity of MPO in serum and the protein expressions of PR3 and LAMP-2 in rats of all medication administration groups decreased, among which the XFWYTLD medium dose group showed the most obvious decrease. Conclusion XFWYTLD may lower levels of ANCA associated antigen, and further inhibit humoral immune function.
3.Effects of Wuzi Yanzong Prescription on cyclophosphamide-induced DNA damage of testis cells in mice

AIM To investigate the effects of Wuzi Yanzong Prescription (Lycii Fructus,Cuscutae Semen,Rubi Fructus,Plantaginis Semen and Schisandrae chinensis Fructus) on DNA damage of testis cells in adult male mice induced by cyclophosphamide (CTX).METHODS Forty out of fifty adult male Balb/C mice were injected intraperitoneally with CTX and then were randomly and equally divided into model control group (normal saline),Wuzi Yanzong Prescription low-,medium-and high-dose groups (100,200 and 400 mg/kg),and the other ten mice served as normal control group (normal saline).All mice were anesthetized by inhalation of ether,and then were sacrificed by cervical dislocation.The sperm count,sperm motility and malformation rate of sperm were tested.The content of 8-hydroxy-deoxyguanosine (8-OHdG) in serum was measured using ELISA;the DNA damage degree of cells in testis was detected by single cell gel electrophoresis;the protein expressions of p-P53 and γ-H2AX in testis were examined by Western blot.RESULTS Compared with the model control group,Wuzi Yanzong Prescription groups showed significant increase in the sperm count,sperm motility and significantly decreased malformation rate of sperm,the level of 8-OHdG in serum,and the protein expressions of p-P53 and γ-H2AX in testis were also significantly decreased.CONCLUSION Wuzi Yanzong Prescription can significantly alleviate the DNA damage of testis cells in mice induced by CTX through down-regulating protein expressions of p-P53 and γ-H2AX.
4.Effect of acarbose on fecal bifidobacteria content in patients with type 2 diabetes mellitus

A total of 118 patients with type 2 diabetes mellitus were divided into acarbose treatment group ( A group,n =58 ) and no acarbose treatment group ( B group,n =60),and 57 healthy subjects were used as control group (C group).The quantification of fecal bifidobacteria and enterococcus faecalis in these subjects was made by realtime PCR.The results showed that fecal bifidobacteria contents in A and B groups were lower and enterococcus faecalis contents were higher compared with C group.After four weeks of intervention,fecal bifidobacteria contents in A and B groups increased ( P<0.01 ),especially in A group,while enterococcus faecalis contents decreased ( P<0.05 )compared with the baseline.Univariate correlation analysis showed that bifidobacteria content was negatively associated with lipopolysaccharides(LPS),advanced glycation index,high sensitive C reactive protein ( hs-CRP),and body mass index ( BMI ) at baseline ( P<0.05 or P<0.01 ).The enterococcus faecalis content was positively associated with levels of monocyte chemoattractant protein-1,LPS,tumor necrosis factor-α,hs-CRP,plasminogen activator inhibitor-1,BMI,and HbA1c (P <0.01 ).After four weeks of intervention,the above associations disappeared.Stepwise multivariate regression showed that basal BMI,HbA1c,and age contributed to the increase in the number of enterococcus faecalis,and BMI negatively contributed to the decrease in number of bifidobacteria.
5.Effect of Shenfu injection on nuclear factor-kappaB during myocardial ischemia/reperfusion injury in rats.

OBJECTIVETo investigate effects of Shenfu injection on the concentrations of plasma tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6), activity of Nuclear Factor kappa B (NF-kappaB) and heart tissue ultrastructure during myocardial ischemia/reperfusion (I/R) injury in rats and its potential mechanism.
METHODSMyocardial ischemia/reperfusion (I/R) was produced by ligation and release of the left anterior descending coronary artery. Ischemia lasted for 30 min and reperfusion for 60 min. Twenty-four healthy male SD rats weighing 230-280 g were randomly divided into three groups (n = 8, each): Group I (Sham-operation group); Group II (I/R group); Group III (Shenfu group), in which Shenfu injection (10 ml/kg) was intraperitoneally injected 30 min before ischemia in animals with I/R. The plasma concentrations of IL-6 and TNF-alpha were measured by ELISA, and the heart was harvested for determination of NF-kappaB levels by Ecl-western blot analysis. Electron microscopy was used to study its ultrastructure.
RESULTSAfter reperfusion, NF-kappaB binding activity in myocardial nuclei and the plasma concentrations of IL-6 and TNF-alpha were significantly increased in Group II, compared with Group I (P < 0.01), and they were markedly reduced in Group III, compared with Group II (P < 0.01). In addition, electron microscopic examination showed more serious injury of the myocardium ultrastructure in Group II, while in Group III the myocardial ultrastructure was similar to normal state.
CONCLUSIONSShenfu injection inhibits NF-kappaB activity in I/R myocardium and leads to down-regulation of proinflammatory cytokine expression, which might be one of the molecular mechanisms of Shenfu injection in cardioprotection.

6.Expression of lipopolysaccharides signal receptors CD14 and TLR4 in human dental pulp tissue and fibroblasts.

OBJECTIVETo investigate the expression of lipopolysaccharides (LPS) receptors CD14 and TLR4 in normal and inflamed human dental pulp tissue and fibroblasts and to determine the signal transduction pathway of LPS in pulpitis.
METHODSCD14 and TLR4 expression in healthy and inflammatory pulps was observed by immunohistochemistry. The rates of CD14 and TLR4 positive cells and the mean fluorescence intensity in in vitro cultured pulp fibroblasts before and after being stimulated by LPS.
RESULTSCD14 and TLR4 were not detected in healthy pulp tissues, but they were strongly positively stained in inflammatory pulps. Ratio of TLR4 positive cells and mean fluorescence intensity produced by the cells stimulated by LPS increased significantly compared with normal pulp cells. However, there were no CD14 expression in human dental pulp cells before and after LPS stimulation.
CONCLUSIONSThis study revealed a significant increase in the expression of CD14 and TLR4 in inflamed human dental pulp tissue. Human dental pulp cells could be induced by LPS to express TLR4, which may play an important role in the process of pulpitis.

7. Biomarkers for assessing motor dysfunction after stroke

Objective:
To investigate whether diffusion tensor imaging (DTI) and motor evoked potentials (MEP) can be used as biomarkers to assess the degree of motor dysfunction of stroke survivors.
Methods:
Sixty partially-paralyzed stroke survivors were given Fugl-Meyer assessments (FMAs) and MEP tests and assessed using DTI seeking any correlations among the results. The receiver operating characteristics curves (ROCs) were prepared to determine the tests′ efficacy in assessing severe motor dysfunction.
Results:
① Asymmetry in the fractional anisotropy (aFA) of the peduncles cerebra as measured by DTI was negatively correlated with the FMA scores of the upper and lower limbs on the affected side. The aFA values of the posterior limb of the internal capsule (PLIC) were negatively correlated with the FMA scores of the affected upper limb, but not with the FMA scores of the affected lower limbs. The abnormalities in central motor conduction time and motor threshold, which are MEP parameters, were negatively correlated with the FMA scores of the affected limbs. ② The ROCs showed that the aFA value of the PLIC was the best indicator for assessing severe upper limb motor dysfunction, with a cut-off value of 0.167 giving the best discrimination. MEP waveform loss could also be used. It has high sensitivity but low specificity. ③ A combination of DTI and MEP can improve specificity in assessing severe motor dysfunction in the upper limbs.
Conclusion
DTI and MEP can both be used to evaluate motor dysfunction in stroke survivors. They have high clinical value for assessing severe motor dysfunction of the upper limbs.
8.Rehabilitation exercise attitudes and compliance of patients undergoing coronary artery bypass grafting and their influencing factors

Objective:To explore the status quo of exercise rehabilitation attitudes and compliance in patients undergoing coronary artery bypass grafting (CABG), and analyze the influencing factors, so as to provide evidence for improving the compliance of rehabilitation exercise in CABG patients.Methods:From January to August 2022, convenience sampling was used to select 228 patients with CABG in three ClassⅢ Grade A hospitals in Urumqi City as the survey subject. One month after surgery, a questionnaire survey was conducted using the General Information and Disease Information Questionnaire, the Questionnaire of Rehabilitation Exercises (QRE), the Chinese version of the Tampa Scale of Kinesiophobia for the Heart (TSK Heart), and the Chinese version of the Multidimensional Fatigue Symptom Inventory-Short Form (MFSI-SF-C). This study conducted a latent category analysis of rehabilitation exercise attitudes and compliance among CABG patients, and identified the influencing factors of their latent categories through Logistic regression. A total of 220 valid questionnaires were collected, with an effective recovery rate of 96.49% (220/228) .Results:The fitting results of latent category analysis showed that the rehabilitation exercise attitudes and compliance of 220 CABG patients could be divided into 4 latent categories, which were respectively named as: excellent attitude-excellent compliance group, good attitude-single exercise group, moderate attitude-poor compliance group, poor attitude-poor compliance group. Logistic regression showed that education level, nationality, pain, exercise fear and fatigue were independent influencing factors for the latent categories of exercise rehabilitation attitudes and compliance in CABG patients ( P<0.05) . Conclusions:Postoperative exercise rehabilitation attitudes and compliance in CABG patients have 4 latent categories, and patient education level, nationality, pain, exercise fear and fatigue are all factors that affect compliance. Medical and nursing workers should formulate targeted interventions based on influencing factors to improve the compliance of exercise rehabilitation in patients with CABG.
9.Cancer Risk Assessment among Medical X-ray Workers in China

Cancer incidence(1950 ~ 1995) among 27 011 medical X-ray workers in comparison with 25 782non X-ray medical specialists between 1950 and 1980 in China was investigated. The average cumulative dose received by the X-ray workers also reconstructed by retrospective dosimetry methods. Significant cancer risk was seen among medical X-ray workers(RR = 1.2). Significantly elevated risks were found in leukemia, cancer of skin, female breast, lung, liver, bladder and esophagus, the RRs were 2. 2, 4. 1,1.3, 1.2, 1.2, 1.8 and 2. 7 respectively. The patterns of cancer risk were associated with years since beginning of X-ray work, age and calendar year of initial employment and cumulative dose suggest that the risks of leukemia, skin cancer and female breast cancer, possibly thyroid cancer were related to occupational exposure to X-rays. A significant cancer risk could be induced by prolonged exposure to low dose ionizing radiation when the cumulative dose reached a certain level.
10.Tau protein expression and possible reasons in Beagle dog model of chronic ischemic cerebral white matter dysmyelination

Objective To explore the expression level and possible reasons of Tau protein in Beagle dog model of ischemic cerebral white matter(WM) demylination.Methods Sixteen adult Beagle dogs were divided into the sham operation group(A) and observation group B,C and D according to the completely random mumber table method,4 cases in each group.Different.degrees of cerebral ischemia animal models were established by 4-vessel occlusion(4-VO) method.The bilateral ventricle edge white matter (WM) was selected.The oligodendrocyte precursors(OPCs) were labeled by NG2.The mature oligodendrocytes(Ols) were labeled by CNPase.Tau,NG2 and CNPase were detected by using the immunohistochemical method.The expression level was quantified by the mean optical value.The correlation among Tau,NG2 and CNGase was analyzed by adopting the Pearson linear correlation analysis.Results The HE staining showed obvious changes of WM demylination after chronic cerebral ischemic.The scores after LFB staining in the group A,B,C and D were(0.75 ± 0.71) points,(1.38 ± 0.06) points,(1.63 ± 0.52) points and (1.88 ± 0.64)points.Compared with the group A,the scores in the group B,C and D were much higher(P<0.05).Compared with the group A,the expression levels of Tau protein and NG2 were significantly increased(P<0.05),while the CNPase expression level was significantly decreased(P<0.05).The Pearson correlation analysis showed that Tau expression level was positively correlated with the NG2 expression level(r=0.277,P=0.006);Tau and NG2 were negatively correlated with the CNPase level(r=-0.303,-0.402,P=0.003,0.001).Conclusion The increase of Tau expression in Beagle dog model of ischemic cerebral WM demylination may be related to the differentiation dysmaturity of OPCs.
