1.The clinical effects of pemetrexed or docetaxel combined with cisplatin as the first line treatment for advanced lung adenocarcinoma

Background and purpose:At present, the advanced lung adenocarcinoma is mainly treated by chemotherapy-oriented comprehensive therapy. Pemetrexed as a multi-target antifolate chemotherapeutic drug, combined with platinum, was approved for treatment of advanced lung adenocarcinoma because of its low toxicity and high efifcacy. The purpose of this study was to compare the short-term clinical efifcacy and toxicity of pemetrexed and docetaxel combined with cisplatin for advanced lung adenocarcinoma.Methods:Fifty patients with stageⅢ-Ⅳ lung adenocarcinoma were randomly divided into two groups. Group PC received pemetrexed (500 mg/m2, d1) combined with cisplatin (75 mg/m2, d1) and group TP received docetaxel (75mg/ m2, d1) combined with cisplatin (75 mg/m2, d1) treatment. One cycle was 21 days in both two groups. The response rate (RR), disease control rate (DCR), progression-free survival (PFS) and overall survival (OS) of the two groups were compared. The peripheral blood T lymphocyte subsets and natural killer (NK) cell activity of patients in both groups were measured before and after the treatment, while compared with healthy controls (25 cases).Results:Among 50 patients, no one got complete remission (CR), 20 patients got partial remission (PR), 17 patients had stable disease (SD) and 13 patients had progressive disease (PD).It showed no signiifcant difference in RR, DCR, PFS and OS (P>0.05) between two groups. Compared with group TP, the activity of CD3+, CD4+, CD4+/CD8+ T cells and NK cells increased and CD8+ T cells reduced in group PC after treatment, but the value had no obvious change (P>0.05). The common side effects were hematological toxicity and gastrointestinal response. The incidence of side effects in group PC was lower than that in group TP, and the degree was also slighter (P<0.05).Conclusion:The clinical efifcacy of pemetrexed combined with cisplatin is equal to docetaxel combined with cisplatin in treatment of advanced lung adenocarcinoma, but the former could reduce the adverse reaction rate.
3. Metabolites of tannins in Punica granatum L. husk in rat urine

OBJECTIVE: To study the in vivo metabolism of granatum tannins by liquid chromatography-mass spectrometry and clarify their metabolic pathways. METHODS: The rat urine samples were collected after intragastric administration of tannins preparation of Punica granatum L. husk. The analysis was performed on a Venusil MP-C18 column with gradient elution system of acetonitrile and water containing 0.1% formic acid. Using electrospray ionization (ESI) in negative ion mode with scanning range from m/z 100 to 1000, the metabolites were identified by comparing the MS spectra and MS data with the literatures. RESULTS: Seven metabolites were detected in the rat urine, among which six were identified according to their second mass spectra to be U-1 (urolithin B), U-2 (urolithin A methyl ether), U-3 (urolithin A), U-4 (urolithin A glucuronide), U-5 (ellagic acid) and U-6 (urolithin C). CONCLUSION: Tannins in Punica granatum L. husk are metabolized after oral administration mainly through hydrolysis, glucuronidation gly-cosidation, decomposition and methylation reaction to generate a series of derivatives of urolithin. These results preliminarily indicate the metabolic pathway and material basis of the pharmacological activity of tannins in Punica granatum L. husk in rats.
4. Study on effect of tannins in Granati Pericarpium on renal fibrosis in rats

Objective: To study the improvement of tannins from Granati Pericarpium on renal fibrosis in rat. Methods: SD rats were randomly divided into five groups, Sham operation, model, Valsartan, high-dose (60 mg/kg), and low-dose (20 mg/kg) tannins groups. Renal fibrosis model of rats was constructed using left ureter ligation method except the rats in the Sham operation group in which the left side ureter of rats was not ligated or cut. The ig administration started at the day of operation, Urinary protein of 24 h, the concentration of Ca2+ and P3+ in urine, and the levels of Scr and BUN in serum were detected in 1-3 week after the operation. The staining of HE and Masson to the kidney tissue was carried out to investigate how tannins from Granati Pericarpium could impact the indexes of renal fibrosis in rats on the morphological basis. Results: The results of urine and blood test showed that the levels of urine protein, Scr, and BUN were deviated from the normal level. The concentration of Ca2+ was increased, while the concentration of P3+ was reduced. The staining of HE and Masson showed that the renal tissue was damaged, the left side of kidney was found renal fibrosis, tannins from Granati Pericarpium could improve the renal fibrosis in rats in some extent. Conclusion: Tannins from Granati Pericarpium have the great improvement on the renal fibrosis in rats and are able to adjust the disorders of Ca2+ and P3+ in rats.
5.Non-adherent bone marrow-derived mesenchymal stem cells differentiate into neuron-like cells in mice

Objecttve To determine whether non-adherent bone marrow-derived mesenchymal stem cells (NA-BM-MSC) can differentiate into neuron-like cells in vitro and in vivo,especially in ischemic brain of mice. Methods NA-BM-MSC of mouse were indueed with conditioned medium containing epidermal growth factor and basic-fibroblast growth factor and demonstrated by immunocytochemistry for the expression of neuron specific nuclear protein and neurofilament-200. NA-BM-MSC from β-galactosidase transgenic mice constitutively expressing β-galactosidase were transplanted into the mice model of middle cerebral artery occlusion.LacZ staining and immunobistochemistry staining were performed to detect the survival,distribution and the differentiation of the donor cells in the ischemic brain. Results NA-BM-MSC can be induced into neuron specific nuclear protein and neurofilament-200 positive cells in vitro.LacZ staining showed that NA-BM-MSC can survive in ischemic brain and express β-galactosidase.Also,numbers of the neuron specific nuclear protein positive cells in β-galactosidase-positive cells were detected in the ischemic brain with double immunohistochemistry staining. Conclusions NA-BM-BMC can be induced to differentiate into neuron-like cells in vitro,and also can differentiate into neuron-like cells in ischemic brain tissue of the mice.
6. Effects of tannins from Granati Pericarpium on endogenous substance metabolism and analysis on metabolic pathway in rats with glomeruloscerosis

Objective: A urinary metabonomics method based on liquid chromatography-mass spectrometry (HPLC-MS) was used to inspect the effect of tannins from Granati Pericarpium on endogenous metabolism of rats with glomeruloscerosis, the potential biomarkers were searched and the metabolic pathways were analyzed to provide relevant theory for treatment of glomeruloscerosis. Methods: Forty-eight SD rats were divided into control group, model group, benazepril hydrochloridec group, and three doses of pomerganate tannis groups. Glomerular sclerosis model was established by unilateral nephrectomy combined with two tail iv adriamycin; 24 h urine of rats was collected after the rats were orally given tannins from Granati Pericarpium for 8 weeks; HPLC-MS was used for testing and analyzing the urine; Principal component analysis (PCA), partial least squares-discriminant analysis (PLS-DA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were used for data treatment of each group to search the potential biomarkers and build the metabolicomics pathways. Results: The urinary metabolism of rats was clustering. Ten potential biomakers were screened by analysis and identification of important variables; The metabonomics pathways involved tryptophan, alanine, aspartate, glutamate metabolism, etc. Conclusion: The trend of endogenous metabolism of rats with glomeruloscerosis is normal after intervened by tannis in pomegranate tannins from Granati Pericarpium. It provides the basement with the further research of clarifying the mechanism of tannins from Granati Pericarpium inpacting on rats with glomeruloscerosis.

Objectives To investigate the expression of 8-Hydroxy-2'-deoxyguanine(8-oxodG)in white blood cell,plasma and urine of rhesus monkey of different age group.Methods 30 female rhesus macaques at different age(1y,5y,10y,15y,20y,25y)were selected and grouped(n=5,each).10 mL of morning urine and 5 mL of fasting venous blood were collected.The level of 8-oxodG expression in plasma,leukocyte and urine was measured by high-performance liquid chromatography-mass spectrometry(HPLC-MS) method.Results The level of 8-oxodG in leukocytes,plasma and urine was increased along with aging.The level of 8-oxodG was 1.8,1.6 and 1.4 times higher in 25 year group than in 1 year group in plasma,white blood cell and urine,respectively(P<0.05).The 8-oxodG level was more than 40 times higher in urine than in plasma.Conclusions The expression level of 8-oxodG is increased along with aging.It may be one of the experimental evidence of the aging markers.
9.The progress of the research on the relationship between stanniocalcins and cancers

Stanniocalcin (STC) was ifrst found as a calcium- and phosphate-regulating hormone produced in bony ifsh by the corpuscles of Stannius. In mammals, the homolog STC-1 displays a relative high amino acid sequence identity (nearly 50%) with ifsh STC, and STC-2 has a lower identity (nearly 35%) with STC-1 and ifsh STC. Both STC-1 and STC-2 are expressed in a variety of tissues. The functions of STC have not been understood. But some ifndings have been reported on their cellular localization, gene structure, and expression in different physiological and pathological conditions, which will be clues in elucidating the functions of STC in mammals. Moreover, STC-1 and STC-2 are expressed in many tumor cell lines, suggesting other biological functions of STC in mammals other than mineral metabolism.
10.Application of a side-to-side stapled cervical esophagogastric anastomosis after three-incision esophagectomy for cancer

Background and purpose: Three-incision esophagectomy for patients with esophageal cancer has been increasingly used, but the incidence of either postoperative anastomotic leak or stricture is higher than that in intrathoracic gastroesophageal anastomosis. The purpose of this study was to investigate the clinical effects of the side-to-side stapled cervical gastroesophageal anastomosis in preventing anastomotic leak and stricture after three-incision esophagectomy. Methods: One hundred and twenty-seven patients undergoing three-incision esophagectomy for esophageal cancer with gastric replacement were reviewed. A side-to-side stapled cervical gastroesophageal anastomosis was performed in 71 cases and manually sewn anastomosis in 56 cases. The incidence of postoperative anastomotic leak and stricture was compared between these two groups and the results were statistically analyzed using SPSS11.0 soft.Results: Anastomotic leakage was noted in seven patients (9.9%) in the stapler group and fourteen patients (25.0%) in the manually sewn group (P=0.04). After the operation two patients (2.8%) in the stapler group and nine patients (16.1%) in the manually sewn group developed a benign esophageal stricture (P=0.02).The incidence of either postoperative anastomotic leak or stricture in cases of the stapler group was significantly lower than that in the manually sewn group (P<0.05). Conclusion: Construction of the cervical esophagogastric anastomosis with a side-to-side stapled technique provides a larger luminal diameter which significantly reduces the incidence of postoperative anastomotic strictures. The surgery also greatly reduces the incidence of anastomotic leaks and strictures, so it could be used as an alternative strategy for cervical esophagogastric anastomosis after esophagectomy for esophageal cancer
