Objective To explore the expression of transforming growth factor ?1(TGF-?1)in the mucosa of guinea pigs with otitis media with effusion(OME)and the role of TGF-?1 in the development of OME.Methods 70 guinea pigs were randomly devided into 7 groups,each with 10 animals.OME was produced by injecting deactivated streptococcus pneumoniae into the typmpanic cavity of guinea pigs.The expression of TGF-?1 was examined by means of immunohistochemistry 6 h,1 d,3 d,7 d,14 d and 30 d after injection.Results TGF-?1 expression could be detected in the middle ear mucosa at 7 d after injection and reached maximum at 30 d.Conclusion TGF-?1 is expressed in the middle ear mucosa of the guinea pigs with OME induced by deactivated streptococcus pneumoniae,and this indicates that TGF-?1 may play a role in the chronic protraction of OME.
2.Effect of pure total flavonoids from citrus on hepatic SIRT1/PGC-1alpha pathway in mice with NASH.

OBJECTIVETo observe the effect of pure total flavonoids from Citrus (PTFC) on the hepatic fatty degeneration, inflammation, oxidative stress and SIRT1/PGC-1alpha expressions in mice with non-alcohol steatohepatitis (NASH), and discuss the action mechanism of PTFC on NASH.
METHODMice were given high-fat diet for 16 weeks to induce the NASH model. Since the seventh week after the model establishment, the mice were intervened with 100, 50 and 25 mg x kg(-1) x d(-1) PTFC for 10 weeks. The pathologic changes in hepatic tissues were observed with HE staining. The contents of TG, CHOL in hepatic tissue, as well as the levels of AST, ALT in serum were detected by using the biochemical process. The expression of SIRT1, PGC-1alpha and MnSOD mRNA in hepatic tissues were detected with Real-time PCR assay. SIRT1, PGC-1alpha protein and 8-OHdG expressions were determined with the immunohistochemical method. The SOD level in hepatic tissues was tested by the xanthine oxidase method. The MDA content in hepatic tissues was examined by the thiobarbituric acid method.
RESULTThe contents of TG, CHOL, NAFLD activity scores and ALT level in serum in hepatic tissues of mice in the model induced by fat-rich diet were obviously higher than that of the normal group (P < 0.010. The SIRT1, PGC-1alpha, MnSOD mRNA and protein expression in hepatic tissues were significantly lower than that of the normal group (P < 0.01). The expression of 8-OHdG and the content of MDA in hepatic tissues were obviously higher than that of the normal group (P < 0.01). After the intervention with different doses of PTFC, the NAFLD activity scores, the content of TG and the level of AST in serum were notably lower than that of the normal group (P < 0.01, P < 0.05); whereas the SIRT1, PGC-1alpha, MnSOD mRNA and protein expression were obviously higher than that of the normal group (P < 0.01, P < 0.05), with the significant decrease in the expression of 8-OHdG and the content of MDA (P < 0.01).
CONCLUSIONOxidative stress/lipid peroxidation enhancement in in NASH mice induced by high-fat diet may be related to the changes in SIRT1/PGC-1alpha signal transduction pathway. PTFC could enhance the anti-oxidant capacity in liver, relieve the damage of reactive oxygen during the fatty acid metabolic process, and prevent NASH from the occurrence and development by regulating the SIRT1/PGC-1alpha signal pathway.

3.Experimental study on intervention effect of Grifola frondosa on nonalcoholic steatohepatitis.

To study the preventive effect of Grifola frondosa on nonalcoholic steatohepatitis (NASH). The rat model of NASH was established by feeding high-fat diets for 12 weeks and intervened with 0.5 g · kg(-1) · d(-1) and 1.0 g · kg(-1) · d(-1) of C. frondosa powder suspensions. The degrees of hepatocyte fatty degeneration and inflammation were observed under the optical microscope with routine HE staining. The NAFLD activity scores (NAS) were calculated. Serum ALT, AST and hepatic TG and CHOL were tested by the biochemical method. The hepatic MDA was examined by thiobarbituric acid method. The hepatic SOD was tested by the xanthine oxidase test. The hepatic GSH-PX activity was determined by the dithio-nitrobenzoic acid method. Hepatic TNF-α and IL-6 were detected by the enzyme-linked immunosorbent assay (ELISA). The NASH model group induced by high-fat diets showed higher hepatic NAS, ser- um ALT, AST, CHOL and hepatic TG, CHOL, MDA, TNF-α, IL-6 (P < 0.01 or P < 0.05) and lower serum TG and hepatic SOD, GSH-PX (P < 0.01, P < 0.05) than the normal control group. After being intervened with different doses of G. frondosa, the NASH group revealed significantly lower hepatic NAS, serum ALT and hepatic TG, CHOL, MDA, TNF-α and IL-6 (P < 0.05) and higher hepatic SOD, GSH-PX (P < 0.05) than the model group. G. frondosa may prevent the further development of NASH by improving the disorder of lipid metabolism in rats with NASH induced by high-fat diets, relieving the level of oxidative stress and reducing the generation of inflammatory cytokines.

4.Transcranial Doppler-assisted intra-arterial thrombolysis for patients with acute ischemic stroke

Objective To investigate the clinical efficacy of transcranial Doppler(TCD)-assisted interventional intra-arterial thrombosis in patients with acute ischemic stroke.Methods This clinical trial enrolled 22 patients with acute ischemic stroke(13 with occlusion of the middle cerebral artery within 6 h of symptom onset; 9 with occlusion of the base artery within 12 h of symptom onset); occlusive arterial cannula was performed and recombinant tissue plasminogen activator(rt-PA,20 mg)was injected to perform interventional intra-arterial thrombosis; and TCD ultrasonography of low frequency(2 MHz)and low intensity(0.25 W/cm2)was performed to assist the thrombosis.Cranial CT was performed again right after the operation and 24 h after the operation to observe the recanalization rate and intracranial hemorrhage; NIHSS scores and Barthel index scores were compared before and after the operation.Results The percentage ofrecanalization was 77.27%(17/22),including 22.73%(5/22)complete recanalization and 54.55%(12/22)partial recanalization; non-symptomatic intracerebral hemorrhage occurred in 3 patients and no symptomatic intracerebral hemorrhage was noted.NIHSS and Barthel index scores after the operation were obviously higher than those before the operation,indicating that favorable outcomes were achieved after thrombosis.Conclusion TCD-assited interventional intra-arterial thrombosis with rt-PA,significantly improving the recanalization rate of occlusive artery and remarkably reducing the happening ofintracerebral hemorrhage,can improve the prognosis and enjoy good clinical efficacy and safety in treatment of acute ischemic stroke.
5.Bactericidal activity of linezolid and clindamycin against methicillin -resistant Staphylococcus aureus in vitro

Objective To determinate the antibacterial activity of linezolid and clindamycin agents of clinical methicillin -resistant Staphy-lococcus aureus ( MRSA ) in vitro.Methods The minimum inhibitory concentration ( MIC ) and minimum bactericidal concentration ( MBC ) of antibacterial agents were determined by the micro broth dilution method . Results The linezolid inhibition rate was 100%when the concentration was 2 μg · mL-1 , but clindamycin concentration was 32 μg · mL-1 . The MIC range of linezolid for MRSA was 0.25 ~2.00 μg · mL-1 , MIC50 was 1 μg · mL -1 , MIC90 was 2 μg · mL-1 , MBC50 was 8 μg· mL-1 , MBC90 was 16 μg· mL -1.The MIC range of clindamycin for MRSA was 0.25 ~32.00 μg · mL-1 , MIC50 was 16 μg · mL-1 , MIC90 was 32 μg · mL-1 , MBC50 was 32 μg · mL-1 , MBC90 was >64 μg· mL-1.Conclusion Clindamycin in the treatment of MRSA infection or suspected MRSA infection must depend on drug sensi-tivity test results.Linezolid in the treatment of MRSA infection or suspec-ted MRSA infection has higher reliability .
6.Construction of protein-protein interaction network of parkinson's disease associated proteins

Objective To construct the protein-protein interaction network of Parkinson's disease (PD) associated proteins. Methods PD-related proteins and their interaction proteins were collected by bioinformatics method from HGNC, OPHID and UniHI database. A protein-protein interaction network of PD associated proteins was designed by ProteoLens software; each protein was evaluated by its contribution to the network and the relevance scores were calculated, thus the coefficient of association of each protein in the network was noted. Results A protein-protein interaction network containing 463 PD associated proteins and 767 their interaction proteins was obtained with its index aggregation reaching 90.5% (P=0.008). The relevance scores of SNCA, PARK2, DRD2, HTRA2,NDUFV2, DJ1, DRD1, DRD3, TRAP1 and ND3 were much higher than that of the others, which indicated their important roles in the pathogenesis of PD. The relevance scores of APP, UBE2I, CLIC6 and UBB were a little higher among all the preteins, indicating that they also participated in the pathogenesis of PD. Some novel proteins, such as FLNA, FREQ, BIRC7, EPB41, EPB41L1, GIPC1,GNAZ, GRB2, KCNJ9, MAPK1, BAG5 and CYC, may also involved in the pathogenesis of PD.Conclusion A scientific and practical protein-protein interaction network of PD associated proteins is successfully constructed, which further confirms the role of some proteins in the pathogenesis of PD.Some novel proteins that might involve in PD are obtained too.
7.Effects of monosialonic acid tetrahexose ganglioside sodium and edaravone on serum Hcy and hs-CRP in patients with acute cerebral infarction

Objective To investigate the effects of monosodium tetrahexose ganglioside sodium (GM-1) combined with edaravone in the treatment of acute cerebral infarction (ACI) in patients with serum homocysteine (Hcy), high sensitivity C-reactive protein (Hs-CRP). Methods 66 patients with ACI were randomly divided into observation group and control group, 33 cases were treated with conventional neurology. The control group was treated with edaravone. The observation group was treated with edaravone and GM-1. (NIHSS) score, serum hs-CRP and Hcy levels were compared between the two groups. Results The total effective rate of the observation group was 90.91%, which was significantly higher than that of the control group (P<0.05). After 21 days of treatment, the NIHSS score, serum Hcy and hs-CRP in the observation group were significantly lower than those in the control group (P<0.05 ). Conclusion GM-1 combined with edaravone is effective in the treatment of ACI, which can promote the rehabilitation of neurological function and living activity. Its mechanism may be related to its anti-inflammatory response, reduced Hcy level and protection of brain cell injury.
8.Experimental study on processing of Paeonia lactiflora.

OBJECTIVETo provide experimental data for the quality control of processed Paeonia lactiflora, a Chinese herbal medicine.
METHODTraditional processing of P. lactiflora was simulated, content of paeoniflorin and water extracts among different preparations were assayed by HPLC; The quantitative correlations among different processing conditions were analyzed, the effects of processing parameters on the contents of paeoniflorin and water extracts were assayed a nanalysed.
RESULT AND CONCLUSIONThe controlled processing parameters were correlated with covariables which showed that processing procedures was controllable, and the heating temperature was a factor impacting the content of paeoniflorin.

9.Interaction of SerpinB5 and MAFbx in gastric cancer cell and its action site.

OBJECTIVETo explore the interaction between SerpinB5 and MAFbx in gastric cancer cell and to identify the interaction sites.
METHODSThe interaction between SerpinB5 and MAFbx was screened and validated by yeast two-hybrid screening and co-immunoprecipitation. The expression of MAFbx was analyzed after SerpinB5 expression being modified by RNA interference and pGBKT7-SerpinB5 transfection. The impact of SerpinB5 on the expression of MAFbx was studied in gastric cancer cell line SUN-16. A model of MAFbx was constructed by homology modeling. The related residues for interaction were analyzed by Autodock4.0.
RESULTSThe interaction between SerpinB5 and MAFbx was validated. The expression of MAFbx changed along with SerpinB5 expression. Amino acids including PRO261, ASN361, and LYS362 were key residue in the interaction of SerpinB5 and MAFbx.
CONCLUSIONSerpinB5 interacts with MAFbx in gastric cancer cell. Amino acids including PRO261, ASN361, and LYS362 are potential binding sites.

10.Analysis of medical reimbursement rate and influencing factors under the DIP payment method

Objective:Analyze the medical reimbursement rate and influencing factors under the DIP payment method to refine the DIP payment policy,promote the optimization of internal operations in medical institutions,and ensure reasonable compensation.Methods:Based on the 2022 DIP fund settlement data from 196 medical institutions in City A,the study used multiple linear regression to analyze the factors affecting medical reimbursement rate and conducted a heterogeneity analysis for medical institutions of different levels.Results:The medical reimbursement rate for medical institutions in City A in 2022 was 103.32%.Medical institutions with lower CMI standardized inpatient costs,lower rates of deviation cases,tertiary care institutions,lower proportion of level-four surgeries,and lower ratios of resident to employee medical insurance cases have higher medical reimbursement rate(P<0.05).Heterogeneity analysis reveals that therates of deviation cases,the proportion of primary care diseases,the ratio of resident to employee medical insurance cases,and the low-standard admission rate have different impacts on medical institutions of different levels.Conclusion:Medical insurance departments should improve policies for primary care diseases,dynamically adjust disease catalogs and payment standards,optimize funding levels and institutional coefficients,and increase penalties for violations to ensure effective use of funds.Medical institutions need to strengthen their understanding of policies,focus on refined internal management,promote standardized and rational diagnosis and treatment through performance assessment transformation,and leverage their own advantages in medical services to reasonably increase the medical reimbursement rate.
