2.Clinical significance of serum CA19-9 in the diagnosis and treatment of biliopancreatic diseases

Serum CA19-9 is one of the markers most frequently detected in the diagnosis and biliopancreatic diseases.The levels of CA19-9 is important for the diagnosis,treatment and prognosis of biliopancreatic diseases.With the development of medical technology,the comprehension of the value of serum CA19-9 is more and more thoroughly.The benign and malignant diseases of biliopancreatic systems can be differentiated by the variation of the values of serum CA19-9,which has high sensitivity in malignant tumors.Serum CA19-9 is one of the independent indicators for evaluation of resectability of tumors and the survival time and life quality of the patients.In this article,the significance of serum CA19-9 in the diagnosis,treatment and prognosis evaluation for biliopancreatic diseases were reviewed.

4.Artificial ossicular chain reconstruction:biocompatibility, properties and laryngecartilage defect repair

BACKGROUND: Defected Laryngeal cartilage has many alternatives, including autologous cartilage, al ograft cartilage and metal stents. Although these materials can achieve desired outcomes in laryngeal cartilage defect repair, certain limitations exist. OBJECTIVE: To investigate the biocompatibility and properties of artificial ossicular chain reconstruction materials, and to explore the effect of artificial ossicular chain reconstruction materials on laryngeal cartilage defect repair. METHODS: Porous hydroxyapatite otosteon was prepared by high-temperature calcination of hydroxyapatite, fol owed by cultured in bone morphogenetic protein solution extracted from fresh human bone to construct bone morphogenetic protein-hydroxyapatite artificial ossicular chain reconstruction material. And then, the biocompatibility and characteristics of the material were analyzed. Forty adult male New Zealand white rabbits were randomly divided into porous hydroxyapatite group and artificial ossicular chain reconstruction material group (n=20 per group), and underwent repair with porous hydroxyapatite material and bone morphogenetic protein-hydroxyapatite artificial ossicular chain reconstruction material respectively after modeling of laryngeal cartilage defect. RESULTS AND CONCLUSION: There was a significant difference in compressive strength of artificial ossicular chain reconstruction materials with different porosities. No symmetry sphere formed in hol ows of the outer surface of the material, with polygonal appearance and with a pore size of 100-200 μm. There were no obvious adverse reactions in both two groups after implantation, but in the artificial ossicular chain reconstruction material group, numerous fibrous connective tissues and obvious bone nodules appeared, and the degradation rate of the material was faster. These results suggest that the bone morphogenetic protein-hydroxyapatite artificial ossicular chain reconstruction material exhibits good biocompatibility and properties, which wil obtain satisfactory outcomes for laryngeal cartilage defect repair. So, the material holds a great value of clinical application.
5.Association between symptoms and signs of dry eye with Sjgren syndrome

Objective To analyze the characteristics of ocular symptoms of Sjgren syndrome and to examine the relation between signs and symptoms.Design Prospective case series.Participants 36 patients with Sjgren syndrome diagnosed by rheumatisms.Methods A self-designed questionnaire was used to investigate 4 symptoms of patients including dryness,grittiness,soreness or irritation,burning or watering.A series of clinic tests were completed as follows:break-up time,fluorescein corneal staining,lissamine green conjunctival staining,Schirmer I test.The positive rate of such symptoms was calculated.The consistency of the symptoms and signs were compared.Main Outcome Measures Break-up time,fluorescein corneal staining,lissamine green conjunctival staining,Schirmer I test.Results Patients with Sjgren syndrome were marked by ocular symptoms.There were efficient statistically significant correlations between grittiness and fluorescein corneal staining,lissamine green conjunctival staining,break-up time(P
6.Optimization of Scutellaria baicalensis extraction process by orthogonal experiment combined with pharmacodynamic index.

To optimize the Scutellaria baicalensis extraction process, the filter paper method and the bacteriostatic ratio method were adopted to determine the in vitro bacteriostatic efficacy of water extracts and 60% alcohol extracts from S. baicalensis. The quantitative analysis of multi-components by single-marker (QAMS) was used to determined the contents of four active components, baicalin, wogonoside, baicalein and wogonin. In addition, with the bacteriostatic ratio and the overall desirability of the contents of four active components as indexes, the orthogonal experiment was adopted to detect the effect of water addition, extraction frequency and extraction time. The optimal extraction process was to add 12 times of water for the first time, 10 times of water for the second time, extract for 2 time, 2 h for each time. This optimization process is stable and feasible, with a higher bacteriostatic ratio in extracts.

7.Effects of polymorphisms of leptin receptor gene 3057 G/A on serum leptin level of senile depressive patients

Objective To investigate the relation of serum leptin level in senile depressive patients with type 2 diabetes and polymorphisms of leptin receptor gene 3057 G/A.Methods 10 senile depressive patients with type 2 diabetes and 47 senile depressive patients without type 2 diabetes were selected for the study.Leptin levels were determined by radioimmunoassay.Genomitic DNA were obtained by phenol/chloroform method.Leptin receptor gene 3057 G/A polymorphisms and allele frequencies were determined with the polymerase chain reaction and restricted fragment length polymorphism assay.Results (1) The differences of leptin receptor genotypes and allele distribution in the 3 groups were not found (all P > 0.05).(2) Serum leptin level in senile depressive patients with type 2 diabetes and without type 2 diabetic patients ((41.65 ± 28.19) ng/ml,(25.56 ± 21.04) ng/ml) were significantly higher than the normal people ((10.84 ± 9.16) ng/ml) (all P < 0.05).Serum leptin level in senile depressive patients with type 2 diabetes were significantly higher than without type 2 diabetic patients(P< 0.05).(3) The serum leptin level in genotype AA senile depressive patients with type 2 diabetes were significantly higher than genotype AA patients without type 2 diabetes.The differences of the serum leptin levels in genotype AA senile depressive patients without type 2 diabetes and genotype AG/GG patients without type 2 diabetes were not found(P > 0.05).The differences of the serum leptin level in genotype AA and genotype AG/GG in normal groups were not found (P> 0.05).Conclusion Senile depressive patients have higher serum leptin level.The serum leptin level is higher,especially with type 2 diabetes.Serum leptin level is related to type 2 diabetes.Serum leptin level is not related to the polymorphisms of leptin receptor gene 3057 G/A.Type 2 diabetes are not related to polymorphisms of leptin receptor gene 3057 G/A.
8.Rapid detection of K-ras gene point mutation in pancreatic adenocarcinoma by Pyrosequencing

Objective To establish a method for detecting K-rag gene point mutation in pancreatic adenocarcinoma based on the pyrosequencing and to compare its performance with that of Sanger sequencing.Methods Genomic DNA was extracted from formalin-fixed,paraffin-embedded pancreatic tissues including 49 pancreatic adenocarcinoma,10 normal pancreas,11 chronic pancreatitis,18 benign pancreatic tumor,7 insulin carcinoma,9 ampullary carcinoma,7 bile duct carcinoma and 7 duodenum papillary adenocarcinoma tissues.K-rag gene point mutations at codon 12 were analyzed by pyrosequencing and Sanger sequencing,respectively.Results No mutant K-ras gene Wag detected in normal pancreas,chronic pancreatitis,benign pancreatic tumor,insulin carcinoma,ampullary carcinoma,bile duct carcinoma and duodenum papillary adenocarcinoma tissues by either pyrosequencing or Sanger sequencing.K-rag gene point mutation was detection rate in pancreatic adenocarcinoma tissues was 71.4%(35/49)by pyrosequencing and 61.2%(30/49)by Sanger sequencing,respectively.Conclusions Pyrosequencing is more sensitive than Sanger sequencing and is also accurate,rapid and of high throushput in detecting mutant K-ras gene.It may serve as a practical method for fast batch analysis of clinical samples.
10.Quality of life in adults with strabismus

AIM: To investigate the quality of life in adults with strabismus and evaluate the improvement in quality of life after treatment.
METHODS: In this prospective study, forty-five adults with a diagnosis of strabismus conform to the inclusion and exclusion standard were selected in our hospital from October 2013 to May 2014, as experimental group and 45 normal adults were enrolled as control group. A Chinese Adult Strabismus-20 questionnaire was used to evaluate the differences of quality of life between patients with strabismus and normal adults, and to evaluate the differences of quality of life in patients with strabismus preoperative and 6mo postoperative.
RESULTS:The scores of quality of life in adult patients with strabismus preoperative and 6mo postoperative were statistically significant lower than those of normal adults (P< 0. 01). In 6mo postoperative, the scores of quality of life statistically significant increased than preoperative ( P<0. 01).
CONCLUSION: Strabismus can decrease the quality of life in psychosocial and visual functional for adults and the surgical treatment can improve quality of life in patients with strabismus.
