1.A analysis for the value of ultrasonography and MRI in diagnosis of adrenal pheochromocytoma

Objective:To analyze differential diagnosis and imaging feature of ultrasonography and MRI for adrenal pheochromocytoma.Methods: The manifestations of the preoperative ultrasound and MRI of the 37 patient with adrenal pheochromocytoma who once were confirmed by pathology were analyzed by the retrospective analysis. The result of pathology was used as standard to analyze the accuracy rate and clinical feature of the two detection methods in the diagnosis of adrenal pheochromocytoma.Results: The results of pathological diagnosis for the 37 cases showed that 35 cases were benign and 2 cases were malignant. After the results of preoperative ultrasound was tested by using pathology, the tested results of 37 cases showed 32 cases were adrenal pheochromocytoma, 2 cases were adrenocortical adenocarcinoma and 3 cases were adrenal adenoma. As these results, the accuracy rate of preoperative ultrasound was 86.49%. And after the results of preoperative MRI was tested by using pathology, the tested results of 37 cases showed that 34 cases were adrenal pheochromocytoma, 2 cases were adrenocortical adenocarcinoma and 1 case was adrenal adenoma. And its accuracy rate was 91.89%. Therefore, if patients were diagnosed with adrenal pheochromocytoma by using preoperative imaging detection, they should be considered to receive further diagnosis so as to distinguish adrenal pheochromocytoma with adrenal adenoma, adrenocortical adenocarcinoma, metastatic tumor and lipomatous. Conclusion: Both of ultrasound and MRI can accurately achieve location, diagnosis of qualitative identification for adrenal pheochromocytoma, and the clinical feature and imaging feature which combined the two detections can increase the detectable rate and accuracy rate for adrenal pheochromocytoma.
2.The effects of repeated irradiation with focused trasound on recurrent and metastasized murine cervical carcinoma

Objective To investigate the efficacy of twice irradiating with focused ultrasound on recurrent and metastasized U14 cervical cancer implanted in the legs of mice. Methods Seventy-two mice with U14 cervical cancer cells implanted in their legs were divided into three groups randomly,with 24 rats in each group.One group received a single dose of focused ultrasound,while mice in the second group were irradiated twice and surgery resection was administered to the third group 7 or 8 days after the tumor was implanted.After 23 days post implantation of the tumor,local tumor recurrence and metastasis to the lungs and lymph nodes were evaluated. Results The inhibition rate after double irradiation was 61.70% for local recurrence and 68.18% for metastasis,significantly higher than in the other two groups. Conclusions Both single and double irradiation with focused ultrasound are effective for inhibiting local recurrence and metastasis,but double irradiation is more effective.
3.Quercetin promotes macrophage polarization and function recovery after spinal cord injury in mice

Objective To investigate the effects of quercetin with a special focus on their effect on macrophage po-larization after SCI.Methods Adult C57 mice underwent T10 spinal cord clip compression injury,then were ran-domly divided into SCI group and quercetin group.1-14 d after SCI received quercetin by intraperitoneal injection(once a day)in quercetin group,and received same normal saline in the SCI group.3,7 and 14 d after SCI, the lesion section of SCI group and Quercetin group stained by immunofluorescent:M1 macrophages pheno-type cell labeled with iNOS, and M2 macrophages phenotype cell labeled with Arg1.The mRNA expression of inflammatory factors in lesion site was detected by RT-PCR.The motor function was evaluated by Basso mouse scale(BMS)after SCI.Results Compare with SCI group,the expression of arginase-1(associated with M2 mac-rophage phenotype)significantly increased in quercetin group(P<0.05).The expression of inducible nitric oxide synthase-iNOS(M1 phenotype marker)was down-regulated as demonstrated using immunohisto-chemistry(P<0.05).Furthermore,the production of NOS2 and tumor necrosis factor alpha was significantly reduced whereas the level of interleukin 10 and TGF-β were elevated in quercetin group(P<0.05).The time course of functional recov-ery revealed a gradual recovery in the subacute phase in quercetin group, little improvement was observed in SCI group(P<0.05).Conclusions It is found that quercetin may promote the shift of M1 to M2 phenotype and amelio-rate the inflammatory microenvironment.Furthermore,the roles of quercetin in immunity modulation may enhance neuroprotective effects and partially contribute to the locomotor functional recovery after SCI.
4.Effect of ultrasound stimulation at the acupoint Guanyuan on follicular development in menopausal rats.

OBJECTIVETo observe the effect of ultrasound stimulation at the acupoint Guanyuan (CV 4) on follicular development in menopausal rats.
METHODSMenopausal female SD rats were selected by vaginal smear examinations. The rats were subjected to ultrasound stimulation at the acupoint Guanyuan with the output power of 0.1 W, working frequency of 9 MHz, and focal length of 4.5-5 mm. Electrochemiluminescence immunoassay was used to detect the serum estrogen levels of the menopausal rats. The changes in the ovarian tissue histology and the follicle number were observed.
RESULTSCompared with the control group, a 10-day ultrasound stimulation for 10 and 5 min daily at Guanyuan significantly increased the serum estrogen levels and the numbers of primary and secondary follicles (P<0.05) and reduced the number of atretic follicles in the menopausal rats (P<0.05).
CONCLUSIONUltrasound stimulation at the acupoint Guanyuan can increase the estrogen secretion function and promote the development of follicles in menopausal rats.

5.Measurement of serum free light chains and its clinical significance in 20 newly diagnosed patients of multiple myeloma.

The objective of this study was to explore the clinical significance of measuring serum free light chains (sFLC) and to compare with serum total light chains (free and binded) in multiple myeloma (MM). sFLC in 20 newly diagnosed MM patients and 20 cases of healthy people as control were measured by immuno-nephelometric assays; the serum light chains and kappa/lambda ratio were measured in all patients, while immunofixation electrophoresis (IFE) tests were carried out at the same time in 18 out of 20 patients. The results showed that the abnormality of serum free light chains and kappa/lambda ratio were found in all of the 20 newly diagnosed MM patients (p < 0.01). The measurement of sFLC showed higher sensitivity than the total serum chains (p < 0.01). It is concluded that the method testing sFLC by immuno-nephelometric assay combined with kappa/lambda ratio is valuable for MM diagnosis, and the measurement of sFLC can be used as one of indicators for MM diagnosis.

6.Relationship between Electrical Conductivity and Decomposition Rate of Rat Postmortem Skeletal Muscle.

OBJECTIVES:
To analyze the relationship among electrical conductivity (EC), total volatile basic nitrogen (TVB-N), which is an index of decomposition rate for meat production, and postmortem interval (PMI). To explore the feasibility of EC as an index of cadaveric skeletal muscle decomposition rate and lay the foundation for PMI estimation.
METHODS:
Healthy Sprague-Dawley rats were sacrificed by cervical vertebrae dislocation and kept at 28 ℃. Muscle of rear limbs was removed at different PMI, homogenized in deionized water and then skeletal extraction liquid of mass concentration 0.1 g/mL was prepared. EC and TVB-N of extraction liquid were separately determined. The correlation between EC (x₁) and TVB-N (x₂) was analyzed, and their regression function was established. The relationship between PMI (y) and these two parameters were studied, and their regression functions were separately established.
RESULTS:
The change trends of EC and TVB-N of skeletal extraction liquid at different PMI were almost the same, and there was a linear positive correlation between them. The regression equation was x₂=0.14x₁-164.91(R²=0.982). EC and TVB-N of skeletal muscle changed significantly with PMI, and the regression functions were y=19.38x₁³-370.68x₁²+2 526.03 x₁-717.06(R²=0.994), and y=2.56x₂³-48.39x₂²+330.60x₂-255.04(R²=0.997), respectively.
CONCLUSIONS
EC and TVB-N of rat postmortem skeletal muscle show similar change trends, which can be used as an index for decomposition rate of cadaveric skeletal muscle and provide a method for further study of late PMI estimation.

7.Roles of monocyte chemoattractant protein-1, RANTES and Fractalkine on promoting vulnerability of atherosclerotic plaques.

OBJECTIVETo elucidate the roles of monocyte chemotactic factors (MCP-1, RANTES and Fractalkine) on the vulnerability of atherosclerotic plaques in patients with stable (SAP) and unstable angina pectoris (UAP).
METHODSPatients with SAP (n = 50) and UAP (n = 50) underwent coronary angiography (CAG) and intravenous ultrasound (IVUS) were included in the study. Monocyte chemotaxis was assayed by the transwell chamber. Concentrations of hs-CRP, MCP-1, RANTES and Fractalkine were measured by Enzyme-linked-immunosorbent assay (ELISA). mRNA expression of MCP-1, RANTES and Fractalkine in the monocytes was detected by RT-PCR.
RESULTSIVUS evidenced soft lipid plaques in 48% UAP patients and in 16% SAP patients (P < 0.05). SAP patients had mainly fibrous and mixed plaques. Plaque burden and vascular remodeling index were significantly higher in UAP patients than in SAP patients (P < 0.01). The averaged number of migrated monocytes in the UAP patients were higher than that in patients with SAP (P < 0.01). Concentration of hs-CRP, MCP-1, RANTES and Fractalkine were significantly higher in UAP patients than those of SAP patients (P < 0.05 or P < 0.01). mRNA expression of MCP-1, RANTES and Fractalkine in patients with UAP was significantly higher than those of SAP patients (P < 0.05).
CONCLUSIONUpregulated monocyte chemotactic factors (MCP-1, RANTES and Fractalkine) might promote coronary plaque vulnerability in UAP patients.

8.The value of hs-CRP and albumin ratio in predicting the prognosis of patients with in-hospital cardiac arrest

Objective:To investigate the predicting value of high sensitivity C-reactive protein (hs-CRP) and albumin (Alb) ratio on prognosis of patients with in-hospital cardiac arrest (IHCA).Methods:A total of 107 patients with IHCA and spontaneous circulation recovery (ROSC) after cardiopulmonary resuscitation (CPR) in the Affiliated Hospital of Xuzhou Medical University during January 1, 2017 and September 30, 2020 were selected as the subjects and divided into the survival group and death group according to the survival condition on day 14 after IHCA. The correlation between ratio of high sensitivity C-reactive protein/albumin (hs-CRP/Alb) and the prognosis of patients was analyzed.Results:No statistical significant differences were found between the survival and death groups in sex, age, medical history, ECG monitoring, recovery ventilation mode, percentage of first monitoring of heart rate and pre-resuscitation Alb (all P > 0.05). However, there were significant differences in the percentage of non-cardiogenic CA and adrenaline dose > 5 mg, time of CPR, concentrations of blood lactic acid, Alb, hs-CRP, and ratio of hs-CRP/Alb (all P < 0.05). Logistic regression analysis showed that percentage of adrenaline dose > 5 mg, concentration of blood lactic acid, time of CPR, and ratio of hs-CRP/Alb were independent risk factors for predicting death. ROC curve analysis showed that hs-CRP/Alb ratio, and concentration of hs-CRP and Alb had predictive value on the death of patients with IHCA; the areas under the curves of hs-CRP/Alb ratio, hs-CRP and Alb concentration were 0.876, 0.864 and 0.745, respectively. The predictive efficiency of hs-CRP/Alb ratio was better than that of hs-CRP concentration or Alb concentration. Conclusions:hs-CRP/Alb ratio has predictive value for the prognosis of patients with IHCA and the predictive value is superior to that of hs-CRP and Alb concentration.
10.Mitofusin-1 regulates osteogenic differentiation of periodontal ligament stem cells

Objective: To detect the expression of mitofusion-1(Mfn1) in periodontal ligament stem cells (PDLSCs) isolated from healthy and periodontitis tissue and to study the effect of Mfn1 on the osteogenic differentiation of PDLSCs. Methods: PDLSCs were isolated from the healthy and periodontitis human samples(H-PDLSCs and P-PDLSCs). IL-1β was applied to mimic the inflammation microenvironment(H-PDLSCs + IL-1β). RT-PCR was used to detect the expression of Mfn1 in HPDLSCs, P-PDLSCs and H-PDLSCs + IL-1β. The expression of Mfn1 in P-PDLSCs was down-regulated by siRNA of Mfn1 (siMfn1). The osteogenic differentiation of the cells was examined by RT-PCR, alizarin red staining and cetyl pyridine chloride quantitative analysis. Results: The expression level of Mfn1 in P-PDLSCs and H-PDLSCs + IL-1β (5 μg/ml) groups was higher than that in H-PDLSCs(P< 0. 05). When the expression of Mfn1 in P-PDLSCs was down-regulated by siMfn1 the osteogenic differentiation ability of P-PDSLCs was restored(P< 0. 05). Conclusion: Inflammation may promote Mfn1 expression in PDLSCs and inhibite the osteogenic differentiation of P-PDLSCs.
