1.Molecular basis of gastric cancer targeted therapy

Gastric cancer is the second leading cause of cancer mortality worldwide,and in Asian countries like Japan,Korea and China,it remains the top killing cancer.As most cases of gastric cancer are in the advanced stage of the disease when they are first diagnosed,patients receiving conventional therapies including surgery,chemotherapy and radiotherapy have poor prognosis.An emerging modality called tumor molecular targeted therapy provides promise and insight to combat this malignancy.This article discusses the molecular basis of gastric cancer targeted therapy from the perspectives of screening and application of target molecules,modulation of tumor microenvironment and antitumor immunotherapy.
2.Effect of sulfonylurea compounds on expression of sulfonylurea receptor 2 in myocardium of diabetic Goto-Kakizaki rats

3.Clinical application and prospect of bone marrow stem cells for myocardial infarction

BACKGROUND:At present,application revascularization and drug treatment may arrest aggravation of heart function and arrhythmic development,but effectiveness is limited.Infarcted myocardial cells are still impossible to regenerate.How to make necrotic myocardial cell regeneration and diastolic myocardial function is the problem to solve.OBJECTIVE:To analyze the study progress of bone marrow stem cells in treatment of myocardial infarction.RETRIEVAL STRATEGY:A computerized online database of Pubmed was searched to identify articles published from May 2000 to July 2006 with the key words of "the marrow stem-cell,cell transplant,myocardial infarction" in English.Simultaneously,articles published between October 1997 and December 2006 were identified in China Journal Full-text Database with the same key word in Chinese.Sixty-seven articles were retrieved and data were selected primarily.Inclusive criteria:①articles related to the transplantation of bone marrow stem cells for the treatment of myocardial infarction,②in the same field,articles recently published or in authoritative journals.Exclusive criteria:①articles of repetitive study,and ②Meta analysis.LITERATURE EVALUATION:Articles mainly analyzes the transplantation of stem cells for the treatment of myocardial infarction.Of the 30 included articles,10 were reviews and the rest was clinical or basic studies.DATA SYNTHESIS:①Bone marrow stem cells are composed of bone marrow mononuclear cells,hematopoietic stem cells,bone marrow mesenchymal stem cells,endothelial stem/progenitor cells,multipotent adult progenitor cells and so on.②After the transplantation,bone marrow stem cells can differentiate into myocardiocytes,and do effectively electromechanical coupling with receptors.Bone marrow stem cells can promote the angiogenesis in ischemic region,avoid ethical dispute and immunological rejection,and these cells are easy to collect.Bone marrow stem cells are adult stem cells,and its transdifferentiation deserves further study.Many clinical studies showed that autogeneic bone marrow stem cell transplantation for the treatment of acute myocardial infarction and heart failure could be achieved.③Matrix metalloproteinase,stem cell factor and stromal cell chemokine from bone marrow stem cells play important roles in stem cell mobilization,homing and myocardial repair.Tissue injury and high-level multipotential stem cells are two key factors of the differentiation of bone marrow stem cells in myocardial microenvironment.④ Problems in bone marrow stem cell transplantation included cell type,regulatory mechanism of transdifferentiation,optimal time,manner and safety of transplantation,etc.CONCLUSION:Through establishing complete animal model,the possibility of each technique in clinical procedures is explored.Bone marrow stem cells in treatment of acute myocardial infarction may become a new method,which has different from traditional drug treatment,interventional therapy of cardiovascular disease and surgical intervention.
4.Nucleic acid aptamers and their applications in the diagnosis and treatment of tumor

Nucleic acid aptamers are ligands with high specificity and affinity for their targets, which are screened from large oligonucleotide pools by the SELEX technology. Their characteristics are superior to antibodies in many aspects, which make them important in the research of recognition of molecules. They can be used to determine the levels and inhibit the bioactivity of their targets. There're broad application prospects for aptamers in the diagnosis and treatment of tumor.
5.Chinese herbal medicine for treatment of dislipidemia

Prevalence of dislipidemia is increasing rapidly in China and there has been a growing interest in Chinese herbal medicine for the treatment of hyperlipidemia both inside and outside China. In this article, lipids regulating effects of 9 herbs or their extracts and 5 herbal formulae which have been published in English-language literature are reviewed. Although evidence from animals and humans consistently supports the therapeutic activities of these Chinese herbal medicines, few multi-center large-scale clinical trials have been conducted to confirm the efficacy and evaluate their safety.
6.A Study on Ability of Facilitating Osteogenesis of Cultured Periosteal Cells Combined with Bone Morphogenetic Protein

Objective To investigate the properties of cultured periosteal cells associated with bone morphogenetic protein(BMP) to find a new treatment for bone defects.Methods The periosteal cells were cultured,expanded and passage cultured in vitro to investigate the characteristics and functional changes of rabbit periosteal cells.Animal models of 1.0cm gaps with complete removal of the bilateral radius medium bone and periosteum were induced in 24 New Zealand rabbits,which were randomly divided into 4 groups to receive the defect repair with periosteal cells combined with BMP,periosteal cells,BMP and blank control respectively.X-ray and histological examination were made periodically after operation to evaluate the effectiveness of bone formation.Results Periosteal cell morphology can be divided into three stages: cell division phase,cell growth phase and cell functional phase.Cellular configuration had no significant difference with primary cells after anagenesis.The callus can be formed after the periosteal cells were transplanted into the bone defection.The osteogenesis ability of periosteal cells combined with BMP was better than that of the BMP,and the osteogenesis ability of the BMP was better than that of the periosteal cells.The mechanism of bone regeneration for periosteal cells was similar to intramembranous ossification,and that of BMP was similar to endochondral ossification.In the periosteal cells combined with BMP group,both intramembranous and endochondral ossification existed,and the former was obvious.Conclusion Both periosteal cells and BMP can improve the repair of bone defect in vivo.Cultured periosteal cells combined with BMP had substantial ability of synergistic osteogenesis,and it was an ideal treatment for bone defection.
7.A systematic evaluation of protective ventilation for surgery in the prone position

Objective To evaluate protective ventilation(PV) versus conventional ventilation(CV) for surgery in the prone position.Methods We searched PubMed, Embase, the Cochrane Library, WanFang Data and other Chinese databases to collect the randomized controlled trails (RCTs) on intraoperative PV in comparison with CV for surgery in the prone position.Two authors independently identified the studies, performed data extraction and assessed the risks of bias in the included studies according to the Cochrane Handbook for Systematic Reviews of Interventions.The reviewers conducted data analyses with RevMan software.Results A total of 9 RCTs involving 449 participants were included.The results showed that the incidence of postoperative pulmonary complications (RR 0.30, 95% CI 0.12~0.73, P<0.01) and peak pressure (MD-3.52, 95% CI-6.93~-0.11, P<0.05) were lower in the PV group.Intraoperative PaO2/FiO2 was higher (MD 37.28, 95% CI 22.67~51.89, P<0.001) and alveolar-arterial oxygen difference was lower (MD-45.50, 95% CI-61.35~-29.65, P<0.001) in the PV group.Conclusions Low tidal volume ventilation in combination with positive end-expiratory pressure (with or without recruitment maneuver) decreases postoperative pulmonary complications,reduces peak pressure and improves oxygenation for surgery in the prone position.
8.Association between symptoms and signs of dry eye with Sjgren syndrome

Objective To analyze the characteristics of ocular symptoms of Sjgren syndrome and to examine the relation between signs and symptoms.Design Prospective case series.Participants 36 patients with Sjgren syndrome diagnosed by rheumatisms.Methods A self-designed questionnaire was used to investigate 4 symptoms of patients including dryness,grittiness,soreness or irritation,burning or watering.A series of clinic tests were completed as follows:break-up time,fluorescein corneal staining,lissamine green conjunctival staining,Schirmer I test.The positive rate of such symptoms was calculated.The consistency of the symptoms and signs were compared.Main Outcome Measures Break-up time,fluorescein corneal staining,lissamine green conjunctival staining,Schirmer I test.Results Patients with Sjgren syndrome were marked by ocular symptoms.There were efficient statistically significant correlations between grittiness and fluorescein corneal staining,lissamine green conjunctival staining,break-up time(P
9. Advances in immunoregulatory effects and bioactive compounds of polysaccharides from Dendrobium officinale

10.Gefitineb inhibits the growth and induces the apoptosis of mouse I-10 Leydig testicular cancer cells in vitro.

OBJECTIVETo observe the inhibitory effect of gefitineb on the proliferation and its inducing effect on the apoptosis of mouse I-10 Leydig testicular cancer cells in vitro.
METHODSWe treated I-10 Leydig testicular cancer cells of mice with gefitineb at 0, 1.25, 2.5, 5, 10, 20, and 40 µmol/L. Then we determined the inhibitory effect of gefitineb on the growth of the cells by MTT, detected their early and late apoptosis by Annexin V-FITC/propidium iodide double staining and Hoechst 33258 nuclear staining, respectively, and observed the expressions of apoptosis-related proteins Bcl-2, Bax and caspase 3/9 by Western blot.
RESULTSCompared with the blank control group, gefitineb significantly inhibited the proliferation of the I-10 cells at 10 and 20 µmol/L (P < 0.05). The survival rate of the cells was (32.4 ± 2.8)% (P < 0.01) and their early and late apoptosis rates were (26.7 ± 4.2)% and (59.33 ± 10.2)% in the 40 µmol/L group, significantly different from those in the control (P < 0.05 and P <0.01). In comparison with the blank control group, gefitineb at 10, 20, and 40 µmol/L increased the expression of pro-apoptotic protein Bax by (41.9 ± 7.1), (60.1 ± 9.8), and (69.0 ± 11.3)% (all P < 0.05), decreased that of apoptosis-inhibitory protein Bcl-2 by (50.3 ± 8.9), (63.9 ± 6.9), and (88.7 ± 13.9)% (all P < 0.05), and elevated that of the cleft proteins caspase-3 by (69.0 ± 6.9)% (P < 0.05), (71.5 ± 8.1)% (P < 0.05), and (110.9 ± 14.2)% (P < 0.01) and caspase-9 by (51.8 ± 4.9), (54.7 ± 6.7), and (43.8 ± 11.8)% (all P < 0.05).
CONCLUSIONGefitineb can increase the cytotoxicity of I-10 Leydig testicular cancer cells of mice and induce their apoptosis via the mitochondria-mediated apoptosis signaling pathway.
