OBJECTIVETo investigate the effects of Newcastle disease virus (NDV) infection on the expression of survivin and cell cycle in human tongue squamous carcinoma TSCCa cells.
METHODSThe proliferation of TSCCa cells infected with NDV in vitro was evaluated by means of MTT assay, and survivin expression in the infected cells was detected using RT-PCR and Western blotting. Flow cytometry was performed to assess the changes in the cell apoptosis, cell cycle and cell proliferation index (PI) of the cells.
RESULTSNDV infection resulted in decreased survivin expression and increased apoptosis of TSCCa cells, with reduced cell percentage in G2/M and S phases and lowered PI of the cells, showing significant differences from those of the negative control cells (P<0.05).
CONCLUSIONNDV infection can inhibit survivin expression, affect the cell cycle of TSCCa cells and induce their apoptosis.

2.An experimental study of the wear behavior of dental feldspathic glass-ceramic and lithium disilicate glass-ceramic.

OBJECTIVETo investigate the tribology characteristics of two ceramic materials in vitro:feldspathic glass-ceramic (veneer porcelain) and lithium disilicate glass-ceramic (heat-pressed ceramic), and to evaluate the wear resistance of different ceramic materials from the dynamic chewing perspective.
METHODSWear tests were performed in simulated oral environment with stainless steel ball antagonists (r = 3 mm), veneer porcelain (CERAMCO 3) and heat-pressed ceramic (IPS e.max Press HT type) in the chewing simulator. The tribological tests were carried out under artificial saliva lubrication condition in room temperature with a vertical load of 10 N for 1.2×10(6) cycles (f = 1.5 Hz, uniform circular motion, revolving speed = 90 r/min, radius = 0.5 mm). The wear volumes were measured using three-dimensional profiling, and surface microscopic morphology were observed using scanning electron microscopy at time point of 200 000, 400 000, 600 000, 800 000, 1 000 000, and 1 200 000 cycles.
RESULTSIn a simulated oral environment, the wear rates of veneer porcelain were (0.001 20 ± 0.00 018) , (0.000 10 ± 0.000 03) , (0.000 50 ± 0.000 05), (0.000 10 ± 0.000 02) , (0.004 10 ± 0.000 38) , and (0.019 00 ± 0.003 53) (×10(-4) mm(3)/cycles) at 200 000, 400 000, 600 000, 800 000, 1 000 000, 1 200 000 cycles. The wear rates of heat-pressed ceramic were (0.139 50 ± 0.030 94), (0.124 40 ± 0.031 20), (0.054 80 ± 0.005 38), (0.038 80 ± 0.006 10), (0.011 10 ± 0.003 75), (0.198 90 ± 0.045 80) (×10(-4) mm(3)/cycles) at 200 000, 400 000, 600 000, 800 000, 1 000 000, 1 200 000 cycles. Three stages were observed in the wear loss process of the two materials: running-in stage, steady wear stage and severe wear stage. In running-in and steady wear stage, the shallow wear tracks of veneer porcelain were produced by the fatigue effect.While in severe wear stage, the wear tracks turned into ploughing. In running-in stage, the surface of heat-pressed ceramic was characterized by dense and shallow ploughing. In steady wear stage, the wear tracks turned into flake peeling produced by fatigue effect. At last, the whole layer was worn off by the effects of ploughing.
CONCLUSIONSIn a simulated oral environment, the wear rate and wear loss of heat-pressed ceramic are greater than that of veneer porcelain.

3.Auto-mobilized adult hematopoietic stem cells advance neovasculature in diabetic retinopathy of mice.

BACKGROUNDHematopoietic stem cells (HSCs) can be used to deliver functionally active angiostatic molecules to the retinal vasculature by targeting active astrocytes and may be useful in targeting pre-angiogenic retinal lesions. We sought to determine whether HSC mobilization can ameliorate early diabetic retinopathy in mice.
METHODSMice were devided into four groups: normal mice control group, normal mice HSC-mobilized group, diabetic mice control group and diabetic mice HSC mobilized group. Murine stem cell growth factor (murine SCF) and recombined human granulocyte colony stimulating factor (rhG-csf) were administered to the mice with diabetes and without diabetes for continuous 5 days to induce autologous HSCs mobilization, and subcutaneous injection of physiological saline was used as control. Immunohistochemical double staining was conducted with anti-mouse rat CD31 monoclonal antibody and anti-BrdU rat antibody.
RESULTSMarked HSCs clearly increased after SCF plus G-csf-mobilization. Non-mobilized diabetic mice showed more HSCs than normal mice (P=0.032), and peripheral blood significantly increased in both diabetic and normal mice (P=0.000). Diabetic mice showed more CD31 positive capillary vessels (P=0.000) and accelerated endothelial cell regeneration. Only diabetic HSC-mobilized mice expressed both BrdU and CD31 antigens in the endothelial cells of new capillaries.
CONCLUSIONAuto-mobilized adult hematopoietic stem cells advance neovasculature in diabetic retinopathy of mice.

4.Fibrosis of corpus cavernosum in animals following cavernous nerve ablation.

AIMTo investigate alterations of smooth muscle cells and collagen fibers in corpus cavernosum following cavernous neurectomy and its relation to the expression of transforming growth factor-beta1 (TGF-beta1).
METHODSTen adult male SD rats (neurectomy group) were subject to a bilateral cavernous nerve (CN) resection aseptically under an operating microscope, with 6 sham-operated rats as the control. Fifteen weeks after the operation, the penile specimens were collected and prepared for quantitative-analyzing of ratio of smooth muscle to collagen fibers in corpus cavernosum with confocal microscopy, and for detecting the expression of TGF-beta1 by RT-PCR and western-blot.
RESULTSSmooth muscle cells that show red color after fluorescent-labeling with tetramethylrhodamine isothiocyanate-phalloidin and collagen fibers that produce green autofluorescence after paraformaldehyde fixation were clearly identified under the confocal microscope. Quantification of fluorescent intensity showed that the ratio of smooth muscle to collagen fibers in corpus cavernosum in neurectomy group was 0.265 +/- 0.125, which was significantly lower than that in sham-operated group (0.760 +/- 0.196, P<0.01). RT-PCR and western-blot analyses revealed a significantly higher expression of TGF-beta1 in the penile tissues of the neurectomy animals than that in sham-operated group.
CONCLUSIONBilateral ablation of CN can lead to fibrosis of corpus cavernosum, which may be related to an increased expression of TGF-beta1 induced by hypoxia in cavernous tissue after denervation.

6.A novel reassortant H3N8 influenza virus isolated from drinking water for duck in a domestic duck farm in Poyang Lake area.

OBJECTIVETo conduct a full genome sequence analysis for genetic characterization of an H3N8 influenza virus isolated from drinking water of a domestic duck farm in Poyang Lake area in 2011.
METHODSThe virus was cultivated by specific pathogen free (SPF) chicken embryo eggs and was subtyped into hemagglutinin (HA) and neuraminidase (NA) by real-time PCR method. Eight gene segments were sequenced and phylogenetic analysis was conducted.
RESULTSThe NA gene of this virus belongs to North American lineage; other seven genes belong to Eurasian lineage. Compared with the viruses containing NA gene, the PB2 and PB1 gene came from different clades. And this indicates that the virus was a novel reassortant genotype. The HA receptor binding preference was avian-like and the cleavage site sequence showed a low pathogenic feature. There was no drug resistance mutation of M2 protein. The mutations of Asn30Asp, and Thr215Ala of the M1 protein implied the potential of pathogenicity increase in mice.
CONCLUSIONThe finding of novel genotype of H3N8 virus in drinking water in this duck farm near Poyang Lake highlighted the importance of strengthening the surveillance of avian influenza in this region, which could contribute to pinpointing the influenza ecological relations among avian, swine, and human.

OBJECTIVE: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). METHODS: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. RESULTS: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=-4.39, p<0.001), were older (t=-4.69, p<0.001), reported more lifetime depressive episodes (z=-3.24, p=0.001), were more likely to experience seasonal depressive episodes (chi2=6.896, p=0.009) and depressive episodes following stressful life events (chi2=59.350, p<0.001), and were more likely to have a family history of psychiatric disorders (chi2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. CONCLUSION: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.

OBJECTIVE:
To investigate the current status of antibiotic use for very and extremely low birth weight (VLBW/ELBW) infants in neonatal intensive care units (NICUs) of Hunan Province.
METHODS:
The use of antibiotics was investigated in multiple level 3 NICUs of Hunan Province for VLBW and ELBW infants born between January, 2017 and December, 2017.
RESULTS:
The clinical data of 1 442 VLBW/ELBW infants were collected from 24 NICUs in 2017. The median antibiotic use duration was 17 days (range: 0-86 days), accounting for 53.0% of the total length of hospital stay. The highest duration of antibiotic use was up to 91.4% of the total length of hospital stay, with the lowest at 14.6%. In 16 out of 24 NICUs, the antibiotic use duration was accounted for more than 50.0% of the hospitalization days. There were 113 cases with positive bacterial culture grown in blood or cerebrospinal fluid, making the positive rate of overall bacterial culture as 7.84%. The positive rate of bacterial culture in different NICUs was significantly different from 0% to 14.9%. The common isolated bacterial pathogens Klebsiella pneumoniae was 29 cases (25.7%); Escherichia coli 12 cases (10.6%); Staphylococcus aureus 3 cases (2.7%). The most commonly used antibiotics were third-generation of cephalosporins, accounting for 41.00% of the total antibiotics, followed by penicillins, accounting for 32.10%, and followed by carbapenems, accounting for 13.15%. The proportion of antibiotic use time was negatively correlated with birth weight Z-score and the change in weight Z-score between birth and hospital discharge (r=-0.095, -0.151 respectively, P<0.01), positively correlated with death/withdrawal of care (r=0.196, P<0.01).
CONCLUSIONS
Antibiotics used for VLBW/ELBW infants in NICUs of Hunan Province are obviously prolonged in many NICUs. The proportion of routine use of third-generation of cephalosporins and carbapenems antibiotics is high among the NICUs.

A new type of L. Bulgaricus microcapsule was prepared to improve stability and resistance of L.Bulgaricus probiotics in harsh environments. An optimal method of preparation of L. Bulgaricus microcapsule is as follow. L. Bulgaricus was mixed with 3% alginate solution. The concentration of bacterial suspension was 1×109 cfu·mL-1. The mixture was microencapsulated by extrusion into 2% CaCl2 solution with a dispensing equipment. After 30 min solidification, the gel beads were lyophilized to obtain L.Bulgaricus microcapsules. The microencapsulation technology was aimed to improve the stability and survival rate of L. Bulgaricus. The microcapsule was spherical with uniform particle size and intact structure. The tolerance of acid, high temperature, high humidity and the long-term stability of freeze-dried powder and microcapsule were evaluated. The results indicated that microencapsulation technic could greatly improve stability and resistance of L. Bulgaricus probiotics in harsh environments.
