1.Research progress in brain tumor stem cell theory

Pubmed Database was undertaken to identify relevant articles of brain tumor stem cells published from January 2000 to January 2008. Books on stem cells and brain tumor stem cells were retrieved. The data were selected primarily, 39 articles related to brain tumor stem cells were selected. Of the 39 inclusive articles, 8 were on review of the brain tumor stem cells theory, 31 were about the research and experiment of brain tumor stem cells. Data synthesis shows that the CD133+ cells isolated from brain tumor have the stem cell-like properties of endless cell proliferation, uncontrolled self-renewal and multi-directional differentiation, but brain tumor stem cells have more vigorous capacity to increase than normal neural stem cells. The brain tumor stem cells express the same gene product as the normal neural stem cells, but the nuclear type of the brain tumor stem cells is abnormal. Brain tumor stem cells maybe the products of gene mutation from the normal neural stem cells. Brain tumor stem cells are the bad seeds of brain cancer, and play a key role in the progress of tumorigenesis, growth, invasion, metastasis, drug resistance and recurrence. Thus, studies on brain cancer stem cells have crucial effects on analyzing the mechanism of occurrence and development, as well as radical cure of brain tumor.
2.Stem cells transplantation for cerebral ischemia

Cerebral ischemia represents one of the leading causes of death and disability in modern time, but only few options exist for the treatment of ischemia-related disease. Neural cell replacement can reconstruct the nerve conduction circuit unit and lead to functional recovery partly in patients. The mechanism lies in: the stem cells transplanted into the lesion can differentiate into functional glial cells or neurons, and they can substitute the functions of dead neurons; to activate the endogenous neural stem cells; to excrete cytokines and improve the local environments, such as inflammatory reactions, tissue necrosis and glial scar formation. Transplantation of bone marrow mesenchymal stem cells, umbilical cord blood, embryonic stem cells and adipose mesenchymal cells have been used for stem cells transplantation in treatment of cerebral ischemia in animal models. So far, the underlying mechanisms of the cell replacement therapy for stroke lesions have remained unknown, and this therapy faces predominantly two major problems: one is how cell grafts survive in an ischemic environment; the other is how cell grafts substitute or rescue neural cells with many different phenotypes.
3.Morphological changes of rat pancreatic tissue induced by ligation of thoracic duct

Objective To observe the morphological changes of pancreatic tissue of thoracic duct ligated rats in fine and ultrastructural levels, and to determine whether lymph block animal model can affect pancreatic islet amyloid polypeptide(PIAP)deposit in rat pancreas. Methods At the 6th month after the operation, some pancreatic tissue sections of 16-month-old experimental rats were embedded in paraffin wax and stained with HE and Congo red;immunohistochemical staining was performed on some frozen sections, which were then observed with light microscope;transmission electron microscope (TEM) specimen preparation and observation were performed on other samples. Results HE and Congo red stained sections showed that the pancreatic glandular lobule space was widened, with significant connective tissue hyperplasia, and fat accumulation when the islet was stained indistinctly or vermeil and tissue space was broadened. The sections with immunohistochemical staining displayed the pancreatic islet as well as the tissues around it were stained into dark brown being positive with PIAP antigen. TEM observation showed the pancreatic glandular interlobule space was widened, while blood vessels and enlarged lymphatic vessels were visible;within widened pancreatic islet interstitial space, a great quantity of lipid droplets and some collagen fibril structures could be seen.Conclusion The ligation of thoracic duct can contribute to pancreatic lymph draining block, lymphagiectasis, connective tissue space and interstitial space widening, fat accumulation, and PIAP deposit in rat pancreas. These structural changes may affect the function of pancreatic islets.
4.High expression of angiopoietin-2 and endostatin in endometriosis

Objective To explore the effect of Angiopoietin-2(ANG-2) and Endostatin (ENS) in patho genesis and development of endometriosis. Methods Twenty-five endometriosis cases and 24 controls were enrolled. ANG-2 and ENS in endometrium were detected by Immunohistochemistry. The concentrations of ANG-2 and ENS in peri-toneal fluid were measured with enzyme linked immunoabsorbant assay. Results In the EMs group, ANG-2 positive rate in eutopic endometrium was significantly higher than that of ectopic endometrium. The ANG-2 positive rate in ectopic endometrium was significantly higher than that of control group; in EMs group, the ENS positive rate in ec-topic endometrium was significantly higher than that in eutopic endometrium. The ENS positive rate in eutopic endo-metrium was significantly higher than that in control group. There was no significant difference of ANG-2 or ENS between stage Ⅰ~Ⅱand stage Ⅲ~Ⅳ. In peritoneal fluid, ANG-2 and ENS concentration and ratio of ANG-2/ ENS in endometriosis were significantly higher than that in controls. Conclusion The effect of angiopoietin-2 and endostatin in angiogenesis of endometriosis are important play a role in pathogenesis and development of the endo-metriosis.

6.A rapid and quantitative fluorescent microsphere immunochromatographic strip test for detection of antibodies to porcine reproductive and respiratory syndrome virus

A fluorescent microsphere-based immunochromatographic strip test (FICT) was developed for the rapid, sensitive, and quantitative detection of porcine reproductive and respiratory syndrome virus (PRRSV) antibodies at the pen-side. The assay was based on the formation of a sandwich immune-complex (anti-pig IgG-PRRSV antibodies-NSP7/N), which was validated by a comparison with IDEXX-ELISA using 3325 clinical specimens. The diagnostic specificity, sensitivity, and accuracy of FICT were 97.28, 93.41, and 94.95%, respectively. FICT showed a good correlation with the virus neutralization assay. Overall, a promising pen-side diagnostic tool was developed for the rapid and quantitative detection of PRRSV antibodies within 15 min.
7.Expression of programmed death-1 and programmed death ligand-1 in the peripheral T-lymphocytes from patients with chronic periodontitis

Objective To investigate the expression of programmed death-1(PD-1) and programmed death ligand-1 (PD-L1) in peripheral T-lymphocytes from patients with chronic periodontitis and its significance and to clarify its role in the development of chronic periodontitis.Methods A total of 73 subjects were included in the study and divided into three groups,chronic periodontitis (30 cases),chronic gingivitis(25 cases) and 18 healthy controls.The peripheral blood was collected and PD-1/PD-L1 expression in the surface of CD4+ T lymphocytes and CD8+ T lymphocytes was examined by flow cytometry.Blood samples from 16 chronic periodontitis patients were collected at week 0 and 6 after initial therapy for 6 weeks and PD-1 and PD-L1 expression in the surface of CD4+ and CD8+ T lymphocytes was also determined by flow cytometry.The data were statistically analyzed.Results The percentage of PD-1 expression in CD4+ and CD8+ T lymphocytes of chronic periodontitis group [(16.7 ± 5.5) %,(20.8 ± 5.1) %] and chronic gingivitis group [(14.2 ± 6.1) %,(14.5 ± 4.3) %] were higher than that of healthy controls [(9.5 ± 2.1) %,(8.1 ± 1.9) %] (P < 0.05).The percentage of PD-L1 expression in CD4+ and CD8+ T lymphocytes of chronic periodontitis group [(24.2 ± 7.1) %,(15.3 ± 6.8) %] and chronic gingivitis group [(12.4 ±6.0) %,(11.2 ± 5.5) %] were higher than that of healthy controls [(4.7 ± 1.2) %,(3.2 ± 2.3) %] (P < 0.05).The percentage of PD-1/PD-L1 expression in CD4+ T lymphocytes and CD8+ T lymphocytes of the chronic periodontitis group were significantly decreased after initial therapy(P < 0.05).Conclusions The expression of PD-1 and PD-L1 in peripheral CD4+ T lymphocytes and CD8+ T lymphocyte of chronic periodontitis patients was up-regulated and was associated with periodontal condition.The initial therapy reduced the expression of PD-1 and PD-L1.
8.Numerical simulation of hemodynamics in internal carotid artery stenosis

Objective To investigate the hemodynamic characteristics of carotid artery models with different degrees of artificial arterial stenosis based on hemodynamic numerical simulation,and to analyze causes of internal carotid artery stenosis and the diseases due to the stenosis from the perspective of hemodynamics.Methods Two-dimensional CT data of healthy individuals were collected from Affiliated Hospital of Inner Mongolia Minzu University,and the three-dimensional model of carotid artery was reconstructed from the two-dimensional CT data using medical modeling software MIMICS20.0.Subsequently,the obtained carotid artery model was imported to 3-Matic Medical software and interfered with various degrees of artificial internal carotid artery stenosis at the same location to obtain the models with 25%,50%and 75%internal carotid artery stenosis.After format conversion,boundary condition setting and mesh generation,the models were imported to computational fluid dynamics software FLUENT14.5 for the hemodynamic numerical simulation and analysis of the carotid artery and two-phase blood flow.Results The comparison of the blood streamlines and velocity vector maps of 3 different degrees of internal carotid artery stenosis at the same location revealed that with the increase of stenosis severity,the phenomenon of blood vortex flow at the carotid sinus decreases rapidly,but obvious vortex flow appeared above and below the internal carotid artery stenosis,and the blood flow was selectively deflected.During the internal carotid artery stenosis from 25%to 50%and then to 75%,the pressure on the wall of internal carotid artery was always low,and the pressure on the wall of internal carotid artery was gradually increased,forming a high pressure area.There was high shear stress area at the stenosis,and the low shear stress ranges above and below the stenosis were obviously larger.Conclusion The technology of using computer software to obtain models with artificial stenosis is convenient and efficient.When the internal carotid artery stenosis increases from 25%to 50%and then to 75%,the blood flow of the carotid artery changes obviously,obvious vortex flow appears in the internal carotid artery,and the mechanical properties of the wall of the carotid artery also change.
