1.Expression of minichromosome maintenance protein 4 in pancreatic ductal adenocarcinoma and its correlation with prognosis

Objective:To detect the expression of microchromosome maintenance protein 4 (MCM4) in pancreatic ductal adenocarcinoma, and to explore the clinical prognosis of MCM4 and pancreatic ductal adenocarcinoma. To explore the possibility of MCM4 as a potential biomarker for pancreatic ductal adenocarcinoma.Methods:The mRNA level of MCM4 in pancreatic ductal adenocarcinoma and normal tissues adjacent to the cancer was analyzed by bioinformatics methods, and the relationship between its expression and the survival rate of patients with pancreatic ductal adenocarcinoma was analyzed. The clinicopathological data of 76 patients with pancreatic ductal adenocarcinoma who underwent surgical treatment were retrospectively analyzed. Immunohistochemical method was used to detect the expression level of MCM4 protein in pancreatic ductal adenocarcinoma tissue and normal tissues adjacent to the cancer, and to analyze its relationship with clinicopathological characteristics of patients with pancreatic ductal adenocarcinoma.Results:The results of bioinformatics analysis showed that MCM4 mRNA was significantly highly expressed in pancreatic ductal adenocarcinoma tissue, and was significantly correlated with the overall survival rate and disease-free survival rate of patients (all P<0.05). Immunohistochemical results showed that the expression of MCM4 in pancreatic ductal adenocarcinoma tissue was significantly higher than that in normal tissues adjacent to the cancer. The high expression of MCM4 in pancreatic ductal adenocarcinoma tissue was related to tumor size ( P=0.038), but not related to age, gender, and tumor grade (all P>0.05). Conclusions:MCM4 is highly expressed in pancreatic ductal adenocarcinoma tissues and indicates a poor prognosis, and its expression level is related to the tumor stage of pancreatic ductal adenocarcinoma. MCM4 may serve as a new potential biomarker for pancreatic ductal adenocarcinoma.
2.The Growth Regulation and Postreceptor Signal Transduction of Somatostatin on Cell Line of Human Colon Cancer

Objective:Our purpose was to investigate the growth regulation and postreceptor signal transduction of somatostatin (SS) on human colon cancer cell line. Methods:Low differentiated clone A cells of human carcinoma were treated with different concentrations of SS and the growth status was observed. Intracellular 1,4,5-trisphosphate (IP3) and cyclic adenosine monophosphate (cAMP) were extracted, then the concentrations of them were measured by liquid scintillation technique and gamma scintillation counter.Intracellular free calcium concentration was detected by loading Fura-2 and fluorescental technique. Protein kinase C (PKC) was extracted from cytosol and membrane, then the activity of both parts was determined with TaKai method. Results:The clone A cell growth was inhibited greatly by different concentrations of SS (10-10 to 10-5 mol/L) and it is related to the doses of SS.Somatortatin could largely inhibit the production of intracellular IP3, [Ca2+]i, and cAMP, and decrease the activity of PKC. Conclusion:The growth of Clone A cell can be inhibited by SS. The inhibition may be mediated by phosphate inositol pathway, so intracellular IP3, [Ca2+]i decreased, or inhibited the cell growth by inhibiting the activity of PKC.On the other hand, the cell proliferation may be inhibited by adenosine cyclase pathway, that is decreasing intracellular cAMP ,inhibiting cAMP-depending protein kinase.

The activation of p21 kinase 2 (PAK2) has a certain value in cancer research, since its participation in a series of intracellular biological activities. There is a relatively little research on its relationship with the development of tumors. PAK2 can be activated by a variety of upstream signals, especially small G protein Rho family of Rac and Cdc42, and it participates in a variety of important signaling pathways and cell function regulation. Abnormal expression of PAK 2 has been reported in various tumors, which involves in cytoskeletal remodeling, cell motility, apoptosis, signal transduction, gene transcription, translation and angiogenesis. Therefore, PAK2 plays an important role in tumor occurrence and development. The study on PAK2 and the targeting therapy for anti-PAK2 provides a new sight on the treatment and prevention of tumor.
4.A perturbation solution of pulsatile Casson flow in the parallel-plate flow chamber.

The parallel-plate flow chamber (PPFC) with rectangular shape, of which the height is far smaller than its own length and width, is one of the main apparatus for in vitro studies of the mechanical behavior of the cells. Considering that the Reynolds numbers of flows in the usually used PPFCs are small, a perturbation solution of laminar pulsatile Casson fluid in the PPFC is presented using Reynolds number as perturbation parameter. Furthermore, the velocities and shear stresses in the PPFC are given. The numerical results demonstrate that under the same pressure gradient the shear stresses are almost identical between Casson fluid and Newtonian fluid, whereas under the same flux the shear stresses are obviously different between Casson fluid and Newtonian fluid. The results in this paper provide a theoretical way to determine the shear stresses in the parallel-plate flow chamber under pulsatile Casson fluid.

5.3D-TV health assessment system by the multi-modal physiological signals.

In order to meet the requirements of the multi-physiological signal measurement of the 3D-TV health assessment, try to find the suitable biological acquisition chips and design the hardware system which can detect different physiological signals in real time. The systems mainly uses ARM11/S3C6410 microcontroller to control the EEG/EOG acquisition chip RHA2116 and the ECG acquisition chip ADS1298, and then the microcontroller transfer the data collected by the chips to the PC software by the USB port which can display and save the experimental data in real time, then use the Matlab software for further processing of the data, finally make a final health assessment. In the meantime, for the different varieties in the different brain regions of watching 3D-TV, developed the special brain electrode placement and the experimental data processing methods, then effectively disposed the multi-signal data in the multilevel.

7.Effect of quality control circle on improving the quality of pre-hospital care in patients with ST-segment elevation myocardial infarction

Objective To improve the quality of pre-hospital care in patients with ST-segment elevation myocardial infarction (STEMI) by quality control circle (QCC). Methods From August 2016 to February 2017, the QCC activity on the theme of "improving the quality of pre-hospital care for patients with acute STEMI"was carried out in Zhengzhou Central Hospital. The factors influencing the pre-hospital treatment of 79 patients with acute STEMI were investigated. In addition, the reasons were analyzed, so as to formulate and implement the corresponding countermeasures. The rate of bypass and the change of site disposal time in patients with acute STEMI were observed. Results After the QCC activity, the bypass rate of the pre-hospital patients with STEMI increased from 58.2% to 91.7%, and the site disposal time was shortened from 42.1 min to 29.3 min, and the differences were statistically significant (P<0.05). Conclusions QCC activity has an obvious effect on improving the quality of pre-hospital care in patients with STEMI, which significantly increases the bypass rate and greatly shortens the site disposal time. At the same time, it improves the comprehensive quality of QCC activity team members.

Objective.To study the changes of microcirculation in patients with diabetic retinopathy(DR).Methods.Examination were performed in 153 cases of type Ⅱ diabetes mellitus, among them,72 cases were male,81 cases were female,mean age 57.0±10.0 years, mean disease course 8.2±7.5 years.All cases were examined fundi by ophthalmologist,urinary albumin excretion rate (UAE)in 24 hours was measured by radioimmunoassay.Moreover,we examined the blood glucose,blood pressure,blood viscosity and observed the changes of naifold microcirculation.Results.It was found that there were more evident disturbance of microcirculation,markedly slowed velocity of blood flow(P<0.05),significantly increased aggregation of blood cells(P<0.05)and exudation around the loop(P<0.05)in the group with DR,compared with the group without DR.Conclusion.It was more evident disturbance of nailfold microcirculation in patients with diabetic retinopathy.

Objective To explore the repair of the open compound wounds in lower extremities caused by multiple factors. Methotis Transplantation of cutaneous.musculo-cutaneous or greater omentum flaps were applied to 155 patients of open compound lower extremity wounds. Results The wound healing rate following first operation was 50% and that following two operations was 14.8%.While the wounds were healed in 7.7% of patients after three operations. Conclusion Transplantations of cutaneous,musculo-cutaneous or greater omentum flaps ale effective to repair and reconstruct the open compound lower extremity wounds.
