1.Two fusion peptides promote healing of scalded mice

Objective:To study the therapeutic effect of 2 artificially synthesized fusion peptides on scalded mice. Methods: Two new fusion peptides, M-T508 and M-T?1, were synthesized based on amino acid sequences selected according to the characteristics of scald (including 3 stages: inflammation, infection and repair by cell proliferation). The 2 peptides were applied for second degree scald burn in mice and the healing of the scald burn was observed. The expression ofⅠcollagen, vascular endothelial growth factor (VEGF), proliferating cell nuclear antigen (PCNA), and cytokeratin (CK) was detected by immunohistochemical staining. Serum TNF-? concentration was determined by ELISA in the animals and the blood vessel permeability was analyzed by Evans blue staining. Results: The healing periods of scald burns in the 2 peptide groups were 1 week earlier than that in the control group. Immunohistochemical results showed that the expression of collagenⅠ, VEGF, PCNA and CK was all positive in the 2 peptide groups 10 days after scald and negative in the control group. Examination of blood vessel permeability and serum TNF-? concentration in the M-T?1 and M-T508 groups showed that the 2 fusion peptides had anti-inflammation and anti-infiltration effects in scalded mouse. Conclusion: The synthesized fusion peptide M-T?1 and M-T508 can promote the healing of scald burns in mice.
2.Progress of clinical research of neoadjuvant chemotherapy for breast cancer

Breast cancer is a systemic disease, only through multidisciplinary treatment including surgery, radiotherapy, chemotherapy, endocrine, can we enhance its effectiveness. In recent years, neoadjuvant chemotherapy for breast cancer has aroused great interest in tumor academia. We hope that this therapy can not only improve breast conservation rate, but also extend the overall survival through a series of clinical studies. This review presents the research progress of neoadjuvant chemotherapy for breast cancer.
3.Selection of peptides bind to PreS1 antigen from a phage-displayed peptide library

Objective:To screen for peptides that specifically bind to PreS1 antigen from a phage-displayed peptide library.Methods: The PreS1 antigen was used as the target molecule to screen the binding peptide from the Ph.D.-7 peptide library with phage display technique,and the positive clones were identified by ELISA.Results: After three rounds of biopanning,the binding peptides were screened from the peptide library by ELISA and competitive inhibiting ELISA.Sequencing result showed that the binding peptides had high affinity and specificity.Conclusion: A peptide binding PreS1 antigen has been successfully obtained by screening the phage display library,which paves a way for the diagnosis and treatment of hepatitis B infection.
4.Possibility of no recurrent laryngeal nerve injury in thyroid surgery

OBJECTIVE To explore the possibility of no recurrent laryngeal nerve injury in thyroid surgery. METHODS A total of 659 consecutive patients with thyroid disease undergoing thyroidectomy byotolaryngologists from March 2001 to March 2005 were retrospectively analyzed. The operative mode and incidence of complications, particularly postoperative RLN palsy, hypoparathyroidism and postoperative recurrence were evaluated. Routine dissection and identification of the RLN was performed during all operative procedures and parathyroid with the blood supply was preserved. RESULTS Unilateral total thyroidectomy with contralateral partial lobectomy was performed in 376 cases, unilateral total thyroidectomy with isthmectomy in 87, bilateral subtotal thyroidectomy with the remnant left at the upper pole in 76, total thyroidectomy in 73. The operations on 47 patients with substernal goiter have been successfully performed via cervical collar incision. None of our patients incurred unilateral or bilateral vocal cord paralysis and permanent hypocalcaemia. Of these patients, the incidence of temporary postoperative hypocalcemia was 1.67 %(11/659). Postoperative hemorrhage requiring reoperation occurred in 4 cases (0.60 %) and 5 patients developed wound haematomas (0.76 %).Postoperative hypothyroidism was found in 3 patients(0.45 %).The incidence of postoperative recurrence was 0.15 %(1/659). No patients had incision infection. CONCLUSION With knowledge of the anatomy of the RLN and routinely complete identification the RLN in performing capsular dissection high on the surface of the thyroid gland, RLN injury may be avoided in thyroid surgery. 5.Isolation and Identification of Canine Parvovirus Serotype 2a and Its VP2 Protein Expression in Transgenic Tobacco

A strain of canine parvovirus (CPV) was isolated from feces of an ill puppy in an animal hospital in Wuhan, China. It was designated as CPV/WH02/06. This isolate was identified as serotype CPV-2a by the hemagglutination test, CPV Ag detection strip, electron microscopy, and PCR. The vp2 gene was cloned and sequenced and assigned GenBank accession number EU377537. A 1242 bp segment of the 5' region of the vp2 gene was cloned and inserted into the binary vector pBI121 and used for Agrobacterium-mediated tobacco transformation. Transgenic tobacco plants were selected on MS medium supplemented with 100 μg/mL kanamycin and 100 μg/mL timentin. Integration of the vp2 gene into the tobacco genome was confirmed by PCR using T1 progeny plants, and the expression of the VP2 protein was confirmed by Western blotting.
6.Screening for simulation peptide specifically binding to the first and the second extra-cellular domain of CCR5 and its therapeutic effect on mice with autoimmune encephalomyelitis

Objective:To screen for simulation peptide binding specifically to the first and the second extra-cellular domain of CC chemokine receptor 5(CCR5),and to observe their therapeutic effect on mice with experimental autoimmune encephalomyelitis(EAE).Methods:Phage display peptide library was used to screen for peptide sequence binding specifically to CCR5;ELISA was used to identify its binding activity and analyze its DNA sequence.The simulation peptide was synthesized and was injected into abdominal cavity of the EAE mice.Spinal cord tissues were obtained and the pathologic changes were studied by H-E staining in EAE control group and simulation peptide group.Results:Twenty phage clones were randomly chosen for identification and ELISA showed that there were eighteen clones had a strong binding activity with CCR5.The positive clones were sequenced and four peptides of high frequency were obtained:STFTTTL,TPIPQLL,SLPLPKP,and QTSSAAL.Mean clinical score of mice in the EAE model group was 3 and that of the simulation peptide group was 1.H-E staining found that the spinal cord tissues in EAE model group had great number of inflammatory cells and evident demyelination changes;the simulation peptide group had less inflammatory cells and no demyelination changes.The four short peptides had an effect of suppressing and delaying the development of EAE,with the average inhibition rate being 43%(P

Objective To study the role of Lugol staining for detecting early esophageal cancer and precancerous lesions.Methods 2% Lugol solution was adopted to stain esophageal mucosa in 45 cases with esophageal suspicious lesions,observed and multiple biopsies were taken for pathological study.Results Thirty-nine cases were light or non-stained among the 45 cases with Lugol staining.Which revealed 8 esophageal cancers(5 early cancer,3 advanced cancer),5 Barrett esophagus,,11 mild-moderate dysplasias.The rate of detecting early esophageal cancer and precancerous lesions with Lugol staining was 46.7%.Conclusion Lugol staining is helpful for diagnosis of early esophageal cancer and precancerous lesions,and it is simple in operating.The role of Lugol staining in detecting early esophageal cancer and precancerous lesions has important clinical value.
9.Endoscopic ultrasonography with miniprobe in diagnosis of upper gastrointestinal tract mesenchymal tumor

Objective To investigate the diagnostic value of EUS with miniprobe on upper gastrointestinal tract mesenchymal tumor(GIMT).Methods The EUS features of 38 patients with GIMT who underwent EUS with miniprobe were studied retrospectively,and the results were compared with the postoperative pathological findings.Results Among 38 GIMT cases detected by EUS,there were 25 cases of gastrointestinal stromal tumor,11 cases of leiomyoma,and 2 cases of leiomyosarcoma.Postoperative histopathological and immunohistochemical examinations confirmed 28 cases of stromal tumor.In which 6 cases were high-risk GIST,8 cases were leiomyoma,1 case was leiomyosarcoma,and 1 case was neurofibroma.The accuracy of diagnosis with EUS was 89%.Conclusion EUS is an accurate method in diagnosis of submucosal tumors,which can make better differentiation diagnosis between GIMT and other submucosal tumors.
10.The diagnosis and management of penetrating abdominal injury with large vessels damage in 18 cases

Objective To investigate the experience of diagnosis and treatment of abdominal penetrating injury with large vessels damage.Methods The clinical data of 18 patients with abdominal large vessels injuries in our hospital from Jan 2003 to oct 2014 were analyzed retrospectively.Results 18 cases of large vessels injuries,accounting for 16.21% during the same period of all penetrating wounds (18/111),emergency laparotomy was performed.There were 8 cases of arterial injury,4 cases of concurrent arteriovenous injury,6 cases of venous injury,Arterial and venous broken walls were repaired,splenic artery and venous ligation and splenectomy were performed.Vascular repair success rate was 100%,Intraoperative blood loss was 500-6 000 ml,average (3 750 ±670) ml.Operating time varied from 2.5 to 7 h,average (3.9 ±0.8) h.All patients were cured.12 cases were followed-up for 2-36 months,mean follow-up time was (15 ± 7) months,ultrasonography showed normal blood flow without complications.Conclusions Emergency surgical exploration and limited fluid resuscitation is the most effective therapy in abdominal penetrating injury involving large vessels.
