Objective:The study is to investigate whether macrophage migration inhibitory factor(MIF)-173 single nucleotide polymorphism(SNP) correlates with inflammatory bowel disease(IBD) in Chinese Han population in Zhejiang Province.Methods:MIF-173 SNP alleles were typed using tetra- primer amplification refractory mutation system(ARMS) and restriction fragment length polymorphisms(RFLP)-PCR in 142 healthy subjects and 98 patients with inflammatory bowel disease(IBD),respectively.At the same time,the PCR products were cloned and sequenced.Results:We detected three kinds of genotypes at the MIF-173 locus,no deviation from Hardy-Weinberg equilibrium were observed.The result was the same from tetra-primer ARMS and RFLP-PCR.Statistical analysis showed,the frequency of MIF-173 CC genotype was significantly higher in patients with ulcerative colitis(UC) 15.5% than in healthy individuals 5.6%(P

4.A comparative study of narrow-band imaging (NBI) with routine endoscopy and NBI with magnifying endoscopy for the diagnosis of colorectal neoplasia

Objective To compare the diagnostic efficacies of narrow-band imaging (NBI) in distinguishing neoplastic from non-neoplastic colorectal lesions with routine endoscopy and with magnifying endoscopy. Methods Patients with colorectal lesions detected by NBI from September 2008 to February 2010 were enrolled in the study. These lesions were classified by pit pattern and capillary pattern, which was then assessed by reference to histopathology. Results A total of 100 patients with colorectal lesions were enrolled, and the lesions were observed by NBI with ordinary endoscopy (n =64) and NBI with magnifying endoscopy (n =36), respectively, and 7 cases (5 in NBI with ordinary endoscopy and 2 in NBI with magnifying endoscopy) which did not meet the diagnostic criteria were excluded. The overall diagnostic accuracy of NBI endoscopy in distinguishing neoplastic from non-neoplastic colorectal lesions was 91.4% ( 85/93 ), in which NBI with ordinary endoscopy and magnifying endoscopy was 89. 8% (53/59) and 94. 1% (32/34),respectively, with both significantly higher than that of conventional colonoscopy reported in the literature (79. 1% ) (P < 0. 05 ). However, no significant difference was detected between 2 methods ( P > 0. 05 ).Conclusion Similar with NBI magnifying endoscopy, NBI endoscopy without high magnification may also be useful to distinguish neoplastic from non-neoplastic colorectal lesions.
5.Rapamycin reduces SH-SY5 Y cell damage induced by oxygen-glucose deprivation

AIM:To observe the effect of rapamycin (Rapa) on human neuroblastoma SH-SY5Y cell injury induced by oxygen-glucose deprivation (OGD), and to explore the role of autophagy in this process .METHODS: The SH-SY5Y cells were randomly divided into 4 groups:normal control group:the cells were cultured without OGD treatment;Rapa group:the cells were pretreated with Rapa for 1 h;OGD group:the culture medium was replaced by glucose-free me-dium and the cells were transferred to a humidified incubation chamber flushed by a gas mixture of 1%O2 , 94%N2 and 5% CO2 for 12 h; Rapa+OGD group: the cultured cells were treated with Rapa for 1 h, and then were given the same treatments as those in OGD group .The cell viability was assessed by MTT assay .The degree of the cell damage was evalu-ated by determining the leakage of lactate dehydrogenase ( LDH) .The enzyme activity of caspase-3 was detected .TUNEL staining were used to detect the variation of cell apoptosis .The protein levels of apoptosis-related proteins Bax and Bcl-2, autophagy-related protein beclin-1 and autophagy marker protein LC 3B were determined by Western blot .RESULTS:Compared with OGD group, the viability of the SH-SY5Y cells was significantly increased, and the activity of caspase-3 was significantly reduced in Rapa +OGD group (P<0.05).The SH-SY5Y cell injury was apparent after OGD with a great in-crease in the apoptotic rate (P<0.05).Compared with OGD group, the apoptotic rate significantly decreased in Rapa +OGD group (P<0.05).Compared with control group, the protein level of Bcl-2 was significantly decreased (P<0.05) and the protein level of Bax was significantly increased in OGD group .Compared with OGD group , the levels of Bcl-2, be-clin-1 and LC3B-Ⅱwere significantly increased and the protein level of Bax was significantly increased in Rapa +OGD group (P<0.05).CONCLUSION: Rapamycin has a protective effect on in vitro cultured SH-SY5Y cells injured by OGD.The mechanism may be related to the promotion of autophagy .
6.Protective effect of antioxidant N-acetylcysteine on the retinal nerve tissue of early diabetic rats

ABSTRACT:Objective To explore the protective effect of the antioxidant N‐acetylcysteine (NAC) on the retinal nerve tissue of early diabetic rats .Methods We randomly divided 60 healthy adult Sprague‐Dawley (SD) rats weighing between 180 g and 220 g into 2 groups:normal control (CON , n=20) and diabetic (DM , n=40) .By intraperitoneal injection of streptozotocin (60 mg/kg) ,the model of diabetic rats was established .The rats were considered diabetic only when they had hyperglycemia (set at ≥16 .7 mmol/L) (32) .The CON group was injected with the same amount of citric acid and sodium citrate buffer solution .After successful model establishment ,the diabetic rats were randomly divided into 1‐month diabetes group and 2‐month diabetes group ,with 16 rats in each group .The left eye of each experimental diabetic rat was set for diabetes control group (D) while the right eye was set as NAC treatment group (NAC) .At 2 weeks of diabetes ,4μL (1 .6μg/μL) of NAC was injected into the vitreous chamber of NAC group and 4μL (0 .01 mmol/L) of PBS was injected into the vitreous chamber of the other diabetic rats .The thickness changes of outer nuclear layer retina was observed by HE ,ultrastructural changes of retinal ganglion cells were observed under the transmission electron microscope ,and the number of retinal ganglion cells was detected by immunofluorescence method .Results At different time points ,retina outer nuclear layer in NAC group was thicker than in D group (P<0 .01) .However ,the NAC group and the CON group did not differ (P>0 .05) .Under the transmission electron microscope ,NAC group had more retinal ganglion cell organelles ,higher electron density of the cytoplasm ,and milder mitochondria swelling than D group .The NAC group did not differ from CON group in the ultrastructure of retinal ganglion cells . NAC group had an increased number of retinal ganglion cells at different time points compared with the D group (P<0 .01) ,but the NAC and CON groups did not differ in the number of retinal ganglion cells (P> 0 .05) .Conclusion The antioxidant N‐acetylcysteine has a protective effect on the retinal nerve tissue of early diabetic rats .
7.Death Characteristics of Digestive System Cancer in Xianyou County, Fujian Province, 2014-2018

Objective To analyze the cancer mortality and epidemiological characteristics of digestive system cancer in Xianyou County, Fujian Province, and to provide a scientific evidence for formulating preventive measures for digestive system cancer. Methods Excel and SPSS software were used to collect the monitoring data of digestive system cancer death in Xianyou County from 2014 to 2018, and then analyzed the mortality, the standardized rate (according to the Chinese 2000 population census) and the cause of death. Results From 2014 to 2018, the deaths of malignant tumors in Xianyou County accounted for 35.00% of all deaths. Among them, the four common digestive system malignant tumors (gastric cancer, esophageal cancer, liver cancer and colorectal cancer) accounted for 62.44% of malignant tumors. Gastric cancer ranked first and accounted for 27.36%. Average annual rate of crude mortality and standardized mortality of malignant tumor in the digestive system were 108.93/105 and 89.34/105, respectively, and the ratio of male to female was 2.27 to 1. Significant differences were found between town and village, as the terrain gets lower, the death rate goes down gradually. Compared with the data of 2011-2012, the standardized death rate of gastric cancer, esophageal cancer and liver cancer were reduced by 29.71%, 23.92% and 12.06%, respectively, while the rate of colorectal cancer was increased by 28.80% and the average annual growth rate was 7.83%. Conclusion Although the death rates of gastric cancer, esophageal cancer and liver cancer have decreased significantly, the malignant tumor of the digestive system is still the main disease that causes the death of residents in Xianyou County, Putian City. Therefore, it is necessary to strengthen the education of health in order to achieve the early discovery, diagnosis and treatment.
8. Design of self-evaluation scale on basic ability in graduates of preventive medicine major and its reliability and validity test

Objective:
To design the self-rating scale on basic ability in graduates of preventive medicine major, and test its reliability and validity, providing a reliable tool for evaluating the basic ability of graduates.
Methods:
On the basis of constructing the Evaluation Index System on Basic Ability in Graduates of Preventive Medicine Major, the three-level indexes were converted to specific questions. Self-evaluation scale including 58 items, 22 sub-dimensions and 8 main dimensions were finally designed after pre-survey and expert argumentation. A total of 205 graduates of preventive medicine major from a medical college and 9 experts in the field of public health were selected to completed the questionnaire. Scale's reliability was tested by applying internal consistency reliability and composite reliability; it's validity was tested by applying content validity and confirmatory factor analysis.
Results:
Scale's α coefficient of internal consistency reliability was 0.976; α coefficient and combination reliability of eight main dimensions were both higher than 0.8; the content validity index S-CVI of the scale was 0.94, the content validity index I-CVI of the item level was 0.78 to 1.00; the average variance extraction amount (AVE) of the eight main dimensions was higher than 0.50. 9.Evaluation of carotid neovascularization in Takayasu arteritis with superb micro-vascular imaging

Objective:To investigate the value of superb micro-vascular imaging (SMI) in evaluating carotid neovascularization in patients with Takayasu arteritis (TA).Methods:SMI was used to analyze the wall thickness and neovascularization in a total of 38 common carotid arteries in 23 patients with TA diagnosed in Shengjing Hospital of China Medical University, and the results were compared with contrast-enhanced ultrasound (CEUS).Results:SMI could clearly show the thickening wall of the common carotid artery in TA patients, and could show very tiny blood flow channels in the thickening wall. Neovascularization was detected in a total of 34 common carotid arteries by SMI, 23 of which showed 1 point of score with scattered low-velocity blood flow and the rest of which showed 2 points of score with diffused spots or strips with low-velocity blood flow. Correspondingly, neovascularization was detected in a total of 34 common carotid artery by CEUS, 22 of which showed 1 point of score and the rest of which showed 2 points of score. The scores of the two groups were consistent ( Kappa=0.641, P<0.01). Conclusions:SMI can be used as one of the important screening methods for neovascularization assessment in thickened carotid wall in patients with TA, which shows potential application value in the diagnosis, treatment and follow-up for TA patients.
10.Construction of the evaluation index system for the core competence of preventive medicine graduates to respond to sudden biological public health events

Objective:To construct a set of evaluation index system for the core ability of preventive medicine graduates to respond to sudden biological public health events.Methods:Research methods such as in-depth interviews and group discussions were used to determine the core competence structure, the Delphi method was used to conduct two rounds of expert consultation, and the index system and its weights were finally determined based on the consultation results of 21 experts on the system. Excel and SPSS 21.0 were used for data collation and statistical analysis.Results:The positive coefficient of expert consultation was greater than 80%, the authority coefficient was greater than 0.7, and the average was 0.906. The final index system was composed of 4 first-level indicators, 10 second-level indicators, and 39 third-level indicators.Conclusion:The evaluation index system can provide a reference for exploring effective ways to improve the core competence of graduates.
