1.Evaluation performance of calibration apparatus for dosimeters used in X-ray mammography

Objective To fulfill the requirements for uncertainty of the calibration apparatus for dosimeters used in X-ray mammography through setting standard radiation quality at the SSDL and developing calibration procedures.Methods According to IEC 61267-2005 and IAEA TRS No 457 to recommend RQR-M and RQA-M series standard radiation quality,the calibration apparatus was evaluated for long term stability of the radiation field over 8 years from 2006 to 2014,including 10 response quantities,such as field homogeneity,change rate of mean air kerma and scatter radiation contributions and so on.In addition,the reference dose instrument was traced back to the PSDL of PTB in Germany by post during 2008 and 2012.Results The field homogeneity (φ 40 mm) relative error was ± 1.4%.The long term stability of the calibration apparatus was less than ± 2% (limits of variation).The scatter radiation contributions at their points of test were below 0.12%.The calibration factors traced to PTB were 0.999-1.000.As a result of the calibration apparatus,the expanded uncertainty was ± 3% (k =2,95% confidence interval).Conclusions The calibration apparatus may meet the requirements of IEC 61267-2005 and IAEA TRS No 457 and has obtained the license of metrology from national regulatory authority.The laboratory now performs very well to calibrate dosimeters used in X-ray mammography.

3.Comparison of the energy response for several dosimeters used in mammography

Objective To evaluate whether eight kinds of dosimeters satisfy the clinical mammography dose measurement through the comparison of energy response.Methods According to GB/T 19629-2005,the energy responses of eight available mammography dosimeters were determined for ten standard radiation qualities in the range 25-40 kVp at the SSDL using a refrence-class dosimeter.Results Except 2000S,the energy response factors of all the other dosimeters were consistent within ±5%.Conclusions The energy response of ionizing chamber type is better than other types of dosimeters.
4.Analysis in short-term clinical benefit of percutaneous kyphoplasty for treating osteolytic tumor of thoracic lumbar vertebrae

ObjectiveTo evaluate the relief of pain and daily activities in patients with osteolytic tumor of thoracic lumbar vertebrae by percutaneous kyphoplasty (PKP).MethodsA retrospective study was conducted to review 35 cases of osteolytic tumor of vertebrae treated with PKP.All patients had a refractories back pain while CT and MRI showed osteolytic changes in their vertebrae. The operation involved percutaneously inserting inflatable vertebral body and creating a cavity filled with bone cement.The visual analogue scales (VAS),WHO standards for pain relief and owestry disability index (ODI) were recorded after the procedures.The change of height in abnormal vertebrae and status of postoperative leakage of PMMA were also recorded.These data were followed up after 1 day,1 month and 6 months and analyzed via t student test.ResultsThe total 35 operations were done successfully.All patients got a conspicuous pain relief at 24 h after operation,and no spinal injury or compression was found.The X-ray showed that the vertebrates treated were filled well by cerment.There were significant differences at pain levels[(7.52±0.81)vs(4.31±0.32)],locomotor activity scales (ODI) [(69.45±0.87) vs (54.45±0.85)] and the height of vertebral bodies [(2.0±0.5) mm vs (2.7±0.4) mm] before and after operation (t value was -7.34,-5.32 and -3.36,respectively,P < 0.05).At 6 months,follow up studies showed that clinic therapeutic effects were stabled for odynolysis and locomotor activity scales.The rates of pain relief in 24 h,1 month and 6 months were 80.0 % (28/35),85.7 % (30/35),82.8 % (29/35),respectively.ConclusionPKP is a single safe and effective way to treat osteolytic tumor of thoracic lumbar vertebrae.It can simply,quickly,and effectively relief the pain caused by osteolytic spinal tumor,recover height of the abnormal vertebra,and improve patient's life quality safely with less complications.
5.Study on the quantity value transfer of enzymes from the reference measurement system

Objective Using the frozen human-pooled serum samples,with values assigned by the enzyme reference procedure in laboratory network to explore the way of quantity value transfer from enzyme reference measurement system to routine ones.Methods The study is about quantity value transfer.The frozen human-pooled serum sample was assigned by the enzyme reference laboratory network including six domestic reference procedure laboratories,namely Beijing Chaoyang Hospital affiliate of Capital University of Medical Sciences,Beijing Aerospace general Hospital,Beijing Shijitan Hospital affiliate of Capital University of Medical Sciences,Peking Union Medical College Hospital,Beijing Leadman Biochemistry Co.,Ltd.and Sichuan Maker Biotechnology Co.,Ltd.,which used the reference measurement procedures of creatine kinase (CK) and lactate dehydrogenase (LDH) from the International Federation of Clinical Chemistry and Laboratory Medicine (IFCC).First,the assigned serum was used to calibrate or verify trueness of routine measurement systems from the 49 laboratories of tertiary hospitals in Beijing in Feb,2013 by Beijing Centerfor Clinical Laboratories.Second,the assigned serum and two levels of patient samples were tested in duplicate by laboratory 's measurement systems after calibration with manufacturer's calibrator and with assigned serum.During this procedure,all laboratories data were collected.Harmonization of results was observed after calibration with the assigned serum according to the statistics.Results After calibration,the interlaboratory coefficients of variation (CVs) of two patient samples were decreased from 4.67% and 5.96% to 1.86% and 1.47% for CK and from 5.72% and 5.04% to 3.67% and 3.62% for LDH.For peer group of A and B instrument,after calibration,both CVs of the assigned serum and two patient samples were less than 2% for CK,and less than 4% for LDH ; For C instrument group,which was not calibrated with the assigned serum,the biases were-4.75% for CK and-13.1% for LDH,while the biggest CVs were 3.64% for CK and 7.57% for LDH.For peer group of reagents,after calibration,the CVs were decreased from 1.60%-4.44% to 0.23%-1.62% for CK and from 0.93%-6.07% to 1.06%-2.72% for LDH.Conclusions It is one of the feasible ways for quantity value transfer by using the frozen human-pooled serum samples,which were assigned by the enzyme reference laboratory network for calibration or verification.It can effectively solve the problem of trueness and harmonization of enzyme testing results.In addition,for those measurement systems which are not applicable for this calibration.
6.Expression and purification of murine ?-1,3-galactosyltransferase catalytic domain

Objective:To express and purificate catalytic domain of murine?-1,3-galactosyltransferase and provide the feasible method in the tumor cell surface production ?-gal epitopes..Methods:This research established expression system in pET-15b to express catalytic domain of murine ?-1,3-galactosyltransferase,then identified its activity by HPLC with anion exchange column.Results:(1)Constructed successfully recombinant ?-1,3-galactosyltransferase catalytic domain with His-tag.(2)?-1,3-galactosyltransferase with His-tag in a soluble form was expressed and purificated effeciently.(3)Its activity by HPLC with anion exchange column showed.Conclusion:This research shows ?-1,3-galactosyltransferase in a soluble form has been expressed successfully.
7.The mechanism of nuclear factor-kappa B in thioactamide induced acute hepatic injury

Objective To investigate the mechanism of nuclear factor-kappa B(NF-κB)in thioactamide(TAA)induced acute hepatic injury.Methods Seventy-eight Wistar rats were randomly divided into normal group(n=18),TAA model group(n=30)and pyrrolidine dithiocarbamate (PDTC)pretreated group(n=30).The rats in PDTC group were received 100 rag/kg of PDTC 1 h before induction of the model.Every 10 rats in TAA group and PDTC pretreated group and 6 rats in normal group were sacrificed at 6,24,48 hour after induction of the model tO mesure levels of endotoxin,tumor necrosis factor(TNF)-α and interleukin(IL)-6.The expression of the intercellular adhesion molecular(ICAM)-1 in hepatic tissue was tested using RT-PCR and the NF-κB activity was 48 hour were higher in TAA group[(87.11±8.23)%.(78.55±6.82)%and(74.27±6.26)%,respectively]than those in normal group[(4.64±1.82)%,(4.55±1.67)%and(4.91±2.12)%,all comparison with normal group(P<0.01).The serum concentration of endotoxin and TNF-α in PDTC pretreated group were higher than those in normal group(P<0.0 1).but lower than those in TAA with ticrmal group(P<0.01).The serum concentration of IL-6 in PDTC pretreated group was higher at 24 and 48 hour than those in normal group(P<0.01),but lower than thoes in TAA group(P<group(P<0.01).The expression of ICAM-1 in PDTC pretreated group was higher than that in immunohistochemical examination showed that liver necrosis was folund in TAA group and PDTC pretreated group.Conclusion The Nuclear factor-κB may aggravate the injury of liver by promoting expressions of TNF-α,IL-6 and ICAM-1.
8.Quality control testing and evaluation for digital mammography

Objective To carry out the quality control testing and evaluation for three digital mammography systems.Methods The performance of three digital mammography systems was assessed by applying methods recommended in the European guidelines for quality assurance in breast cancer screening and diagnosis and Chinese specification for testing of quality control in X-ray mammography.The performance of X-ray generator of three digital mammography systems were tested and evaluated.CDMAM 3.4 phantom with four different thickness(30,40,50,60 mm) were exposured in DR,PCM,and CR system,respectively.The average glandular dose (AGD) value was measured and image quality figure (IQF) analysis was performed in each thickness.Results The X-ray machine performance of DR and CR was in accordance with existing standard,however the standard was inappropriate to evaluate part of X-ray machine performance of PCM system.The AGDs for system DR were 1.20,1.42,1.75 and 2.20 mGy for 30,40,50 and 60 mm PMMA thickness,respectively.The respective AGDs for system PCM and CR were 0.82,1.19,1.33,1.70 mGy and 0.59,0.88,1.47,2.19 mGy.For the same phantom thickness sequence,the IQFs were 21.36,21.57,27.25 and 30.58 for system DR,28.02,29.10,35.90,and 41.24 for system PCM,whereas they were 39.78,39.30,43.85 and 48.08 for system CR.Conclusions The AGDs of all three systems were in accordance with the values recommended in European guideline.The AGD and IQF could provide an effective way for performance assessment and constancy checks for digital mammography systems.
9.Energy dependence of three passive solid detectors on low energy X-ray

Objective To assess the feasibility of ESD measurements in the mammography by the comparative study of energy dependence of the three passive solid detectors on low energy X-ray.Methods Two thermoluminescent TL) detectors ( LiF(Mg,Ti) and LiF( Mg,Cu, P) and one optically stimulated luminescence (OSL) detector( Al2O3: C ) were irradiated for the same air kerma by ten kinds of beam quality between 25-40 kV in a Mo/Mo low energy X-ray generator,respectively. Results The energy dependences of TL and OSL were 25% and 11% between 25-40 kV, respectively. Conclusions considerable potential for OSL detector can be used in routine quality control and ESD measurements in the mammography.
10.Research of angular dependence of mammographic dosimeters

Objective To study the angular dependence of four different mammographic dosimeters to monoenergetic X-ray beams and observe the variation tendency in angular dependence to different energy X-rays and to discuss the evaluation of average glandular dose(AGD)in digital breast tomosynthesis(DBT)system.Methods One ionization chamber and three semiconductor dosimeters were exposed by four kinds of beam quality.The signal of each dosimeter was tested over an angular range of 0° to 90°.The dosimeters were exposed for three times at each selected angular position.The measurement at each angle was normalized to the appropriate measurement at perpendicular incidence.Results The responses from all the dosimeters had a maximum at 0° and decreased with increasing angles.When the radiation incidence angle was 10°,the angular dependence were 0.99,0.99,0.96 and 0.96 for dosimeters A,B,C and D,respectively.And the angular dependence decreased to 0.96,1.00,0.78 and 0.52 when the incidence angle was up to 25°.For semiconductor dosimeters,no signal was recorded when incidence angle was beyond 45°.For ionization chamber,the magnitude of the angular dependence generally decreased with increasing energy.Conclusions All of the dosimeters will underestimate the dose if used in DBT.In order to minimize the error due to the rotation angle of X-ray tube,correction factor of mammography dosimeter should be used for the measurement of AGD in DBT systems.
