Objective Using the frozen human-pooled serum samples,with values assigned by the enzyme reference procedure in laboratory network to explore the way of quantity value transfer from enzyme reference measurement system to routine ones.Methods The study is about quantity value transfer.The frozen human-pooled serum sample was assigned by the enzyme reference laboratory network including six domestic reference procedure laboratories,namely Beijing Chaoyang Hospital affiliate of Capital University of Medical Sciences,Beijing Aerospace general Hospital,Beijing Shijitan Hospital affiliate of Capital University of Medical Sciences,Peking Union Medical College Hospital,Beijing Leadman Biochemistry Co.,Ltd.and Sichuan Maker Biotechnology Co.,Ltd.,which used the reference measurement procedures of creatine kinase (CK) and lactate dehydrogenase (LDH) from the International Federation of Clinical Chemistry and Laboratory Medicine (IFCC).First,the assigned serum was used to calibrate or verify trueness of routine measurement systems from the 49 laboratories of tertiary hospitals in Beijing in Feb,2013 by Beijing Centerfor Clinical Laboratories.Second,the assigned serum and two levels of patient samples were tested in duplicate by laboratory 's measurement systems after calibration with manufacturer's calibrator and with assigned serum.During this procedure,all laboratories data were collected.Harmonization of results was observed after calibration with the assigned serum according to the statistics.Results After calibration,the interlaboratory coefficients of variation (CVs) of two patient samples were decreased from 4.67％ and 5.96％ to 1.86％ and 1.47％ for CK and from 5.72％ and 5.04％ to 3.67％ and 3.62％ for LDH.For peer group of A and B instrument,after calibration,both CVs of the assigned serum and two patient samples were less than 2％ for CK,and less than 4％ for LDH ; For C instrument group,which was not calibrated with the assigned serum,the biases were-4.75％ for CK and-13.1％ for LDH,while the biggest CVs were 3.64％ for CK and 7.57％ for LDH.For peer group of reagents,after calibration,the CVs were decreased from 1.60％-4.44％ to 0.23％-1.62％ for CK and from 0.93％-6.07％ to 1.06％-2.72％ for LDH.Conclusions It is one of the feasible ways for quantity value transfer by using the frozen human-pooled serum samples,which were assigned by the enzyme reference laboratory network for calibration or verification.It can effectively solve the problem of trueness and harmonization of enzyme testing results.In addition,for those measurement systems which are not applicable for this calibration.

Objective To reproduce the inflammatory bowel disease(IBD) models induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS),and to investigate the effect of Changkangyin(CKY) on it.Methods Total 60 rats were divided into 6 groups by random:alcohol control group,model group,salicylazosulfapyridine(SASP)group,low,middle and high dosages CKY groups.IBD model was induced by TNBS.The changes of macroscopic morphous and pathohistology of colon were observed after intervened with drug for 21 d.Results The body weight in model group was obviously lower than that in alcohol control group(P0.05).Conclusion Colitis can be induced by TNBS in rats.The change of it is the same as human's.It is a ideal model to study pathogenesis of IBD and investigate the effect of medicine.CKY has a batter therapeutic effect on IBD model.

Objective To clone different lengths cell division autoantigen1(CDA1) promoters(CDA1P) and measure their activities in order to provide basis for fumctional study of CDA1P.Methods Different lengths CDA1P were amplified by PCR using mouse liver genomic as template and cloned into BglⅡ digested pGL3-basic plasmid to produce recombinant plasmid pGL3-basicmCDA1P,then they were transfected into Lewis lung cell line(LLC) and RAW 264.7 cells differently,these cells were collected after transfected for 48 h,finally luciferase activities of pGL3-basic-mCDA1P were measured.Results ①Different lengths CDA1P were cloned and characterized by restriction enzymes.②Different lengths CDA1P were ligated into pGL3basic vector,and were proved with PCR and DNA sequencing .③Activities of pGL3-basic-mCDA1A were different in the two kinds of Leuwis and mononuclear macrophage cells(RAW264.7 cells).Conclusion The different activities of CDA1P may be related with different cellular types.

Objective To investigate the efficacy of transplantation of induced bone marrow mesenchymal stem cells(MSCs)via various route for treatment of chronic liver injury.Methods MSCs were isolated and expanded by density gradient centrifugation combined with adherent culture.MSCs were induced to differentiate into hepatocyte-like cells in vitro.The markers of hepatocyte lineage were examined by RT-PCR and immunocytochemistry.The changes of ultramicrostructure of MSCs were observed by transmission electron microscope.One hundred and fourteen Wistar rats were treated with mixture of 40%CCl4 and peanut oil to establish chronic liver injury model and another 6 rats were served as control.Twelve rats in model group were died within 8 weeks,and the rest rats were divided into portal vein,spleen and tail vein transplantation groups.The rats were transplanted with DAPI-labelled MSCs which were induced for 14 days.The distribution of the DAPI-labelled cells were observed by fluorescence microscopy.The liver function and pathology were examined.Results AFP mRNA expression were detected at 7th and 14th day after induction,expressions of ALB mRNA,CK18 and hepatocyte antigen were found at 14th and 2lth day.The ultrastructure examination revealed that MSCs were enlarged with a lot of endoplasmic reticulum,mitochondrion,lysosome and glycogen granule.DAPI labelled cells were found in the spleen and liver transplantion groups.The pathological changes of liver were alleviated in all treated groups,especially in intraspleenic transplantion group(P＜0.05).Conclusions Induced bone marrow MSCs can ameliorate liver function of chronic hepatic injury after transplantation.The intraspleenic transplantation is better than others.

Objective To compare the biochemical and immunological parameters and histopathological characteristics in the patients with autoimmune hepatitis (AIH),biliary cirrhosis (PBC)and AIH/PBC overlap syndrome,and to provide basis for working out the reference standard of clinical diagnosis and reasonable treatment.Methods 135 cases of autoimmune liver disease patients were selected,including 49 cases of AIH patients,43 cases of PBC patients, and 43 cases of AIH/PBC overlap syndrome patients. The biochemical, immunological parameters and histopathological changes of the patients were detected. The patients with AIH/PBC overlap syndrome were treated with different therapy methods including ursodesoxycholic acid (UDCA ) treatment,prednisone treatment and combination treatment with UDCA and prednisone,and the effectiveness of different treatment programs were evaluated.Results The activities of GGT and ALP and IgM level of the patients in the AIH/PBC group were significantly higher than those in AIH group,there were significant differences (P<0.05);the activities of ALT and AST and IgG level of the patients in AIH/PBC group was significantly higher than those in PBC group,there were significant differences (P<0.05).The detection rates of AMA and AMA M2 of the patients in AIH/PBC group were higher than those in AIH group, there were significant differences (P<0.05);the detection rates of ANA and SMA of the patients in AIH/PBC group were higher than those in PBC group,there were significant differences (P<0.05).There were high incidence of piecemeal necrosis (100.0%), liver cell rosette-like changes (83.72%)and bile duct lesions (69.77%)of the patients in AIH/PBC group.The effective percentage in combination therapy group was 85.7% which was significantly higher than those in various drug alone groups (P< 0.05 ). Conclusion The changes of biochemical and immunological indicators and pathological features of the patients with AIH/PBC overlap syndrome are in combination with the particular indicators of AIH and PBC which would provide the diagnostic basis for AIH/PBC overlap syndrome. The combined therapy for AIH/PBC overlap syndrome is effective and should be popularized.

Objective To investigate the significance of fibrin monomer(FM ) ,D‐Dimer(D‐D) and other blood coagulation indica‐tors detection in the early diagnosis of disseminated intravascular coagulation(DIC) in obstetrics .Methods 90 healthy women with‐out pregnancy(control group) ,270 normal pregnant women including early ,mid‐,late pregnancy(90 cases for each) ,36 early DIC puerperas(early DIC group) were enrolled in the study .FM ,D‐D ,prothrombin time (PT ) ,activated partial coagulation activity (APTT) ,fibrinogen(FIB) levels were detected for those people .Results The levels of D‐D and FM in early DIC group were signif‐icantly higher than those in control group (P<0 .05) ,and PT ,APTT in late pregnant group were significantly shorter than those in early ,mid‐pregnant group and control group ,while FIB was significantly higher ,the difference was statistically significant(P<0 .05) .Conclusion PT ,APTT and FIB can reflect the high blood coagulation state in the late pregnancy ,but can not be used in the diagnosis of early DIC .D‐D and FM have important value for the early diagnosis of DIC in obstetrics .

Objective To investigate the routine methods of α-amylase (AMY) test in scrum which meets the requirements of ISO 15189.Methods Fifty human serum samples with different concentrations of AMY (40- 750 U/L) were collected from March to December in 2008,to form the patients' frozen serum group.Four AMY measurement systems including Roche,Wako,MINDRAY and MAKER were used.The frozen standard materials with concentrations of ( 70.1 ± 3.7 ) U/L and ( 418.3 ± 22.1 ) U/L and the patients' frozen serum group were measured simultaneously by using IFCC reference method and 4 AMY measurement systcms based on 7170A automatic biochemistry analyzer.Thc linear regression analysis was made between the measurement results of each system and IFCC reference method.The equivalence,agreement and trueness were also evaluated by using the file EP9-A2 method.Bland-Altman GraphicalAnalysis and the improved Bland-Altman Graphical-Analysis of MVS1.80 software.Results Judging by the standards of IFCC reference method,the measurement results of 4 measurement systems were obviously different. ( 1 ) When measuring standard materials the results were 66.4,70.6,69.4 and 49.2 U/L respectively and 394.0,456.4,406.7,302.4 U/L respectively.The measurement results of MINDRAY were in agreement with that of IFCC reference method.( 2 ) When mcasuring the patients' scrum group by 4 measurement systems and IFCC reference method,the slopes of the linear regression equations were 0.934,1.070,0.930 and 0.731.respectively.And the intercepts were 0.886,6.249,5.388 and 3.574,respectively.According to the EP9-A2 method,the measurement results of Roche Wako,MINDRAY were equivalent to that of IFCC reference method.According to Bland-Altman Graphical-Analysis, the measurement results of Roche and MINDRAY were in agreement with that of IFCC reference method.The average biases of each measurement system were - 6.11％ ( Average bias ± 2s were 2.81％ and -9.40％ ),1.99％ ( Average bias ± 2s were 10.35％ and - 6.36％ ),- 2.70％ (Average bias ± 2s were 2.37％ and -7.77％ ) and -34.72％ ( Average hias ±2s were -24.20％ and -45.24％ ),respectively.According to the improved Bland-Altman Graphical-Analysis, the measurement results of MINDRAY are correct.The average biases of each measurement system were - 5.92％ (Average bias ± 2s were -2.81％ and -9.03％),2.10％ (Average bias ±2s were 10.74％ and -6.53％),-2.64％ ( Average bias ± 2s were2.24％ and -7.51％ ) and - 29.51％ ( Average bias ± 2s were 21.82％ and - 37.21％ ),respectively.Conclusions ( 1 ) The measurement results of different measurement systems do not necessarily have crreet results though they have claimed to have traceability.(2) The trueness of measurement results using the same system may not come to the same conclusion when evaluated by different methods.So laboratories should select and establish a procedure to evaluate trueness of routine methods and adopt those meeting the trueness requirements of ISO 15189.

Objective To summarize the experiences of the diagnosis and treatment of the intraspinal tumors. Methods One hundredand twenty-three cases of the intraspinal tumors were analyzed retrospectively. Results The intraspinal tumors in those patients included meningiomas/neurofibroma in 32 cases (19.5%), neurinoma in 32 cases (26.0%), ependymoma in 8 cases (6.5%), astrocytoma in 13 cases (10.6%), lipoma in 10 cases (8.1%), metastatic tumors and hemangioma each in 5 cases (4.1%), arachnoid cyst in 9 cases (7.3%), and others in 17 cases (13.8%). The extramedullary subdural, epidural and intramedullary tumors accounted for 56.9%, 21.1% and 22.8%. According to the locations, 30.1% tumors located at cervical region, 37.4% at thoracic region, 19.5% at lumbar region and 13.0% at sacral segments. The total removal rates of tumors were 69.1% (the total removal rates of extramedullary tumors were 70.1%, and the removal rates of intramedullary tumors were 60.7%).The improvement rates of different degree of movement, sense and sphincter function were 75.2%, 73.8% and 77.8%. Conclusion Most of the intraspinal tumors are benign, and the surgical outcome is satisfied. The keys of therapeutic efficacy were MRI and microsurgical techniques application. The spinal stability reconstruction is in the extreme need if spinal stability is breached in surgery.
