1.Adverse Drug Reactions Induced by Anti-infectives:Analysis of 657 Cases

OBJECTIVE:To investigate the characteristics and regularities of adverse drug reactions(ADR) induced by anti-infective drugs in our hospital.METHODS:657 ADR cases induced by anti-infective drugs collected by the ADR monitoring center in our hospital in 2006 were classified and analyzed.RESULTS:13 categories(49 kinds) of anti-infective drugs were involved in the total 657 ADR cases.Of which,those patients aged above 60 years accounted for 30.14%.The ADR were chiefly induced by quinolones,followed by cephalosporins and penicillins etc,which chiefly manifested as lesion of skin and its accessories(53.12%),followed by the lesions of digestive system and nervous system etc.CONCLUSION:Multiple factors contributed to the ADR,to which great importance should be attached so as to reduce the incidence of ADR induced by anti-infectives.
2.Significance and expression of zinc finger protein 139 and matrix metalloproteinase 7 in endometrial adenocarcinoma

Objective To observe the expression of ZNF139 and MMP-7 in endometrial adenocarcinoma tissues and discuss the correlation with clinical pathological significances.Methods In 71 patients diagnosed as endometrial adenocarcinoma,71 cancer tissues(adenocarcinoma group) and 71 adjacent normal tiuues(adjacent cancer group) were collected in operations.Immunohistochemical methods was used to detect the expression of ZNF139 and MMP-7 in the two groups,the relationship between the two genes and communication with clinicopathological indexes were discussed.Western blot and qRT-PCR were used to detected the expression of ZNF139 and MMP-7 at protein and mRNA level.Results The positive rates of ZNF139 was 66.20% in adenocarcinoma group,higher than in adjacent cancer group(21.42%,P=0.016);the positive rates of MMP-7 was 74.65% in adenocarcinoma group,higher than7 than in adjacent cancer group(26.76%,P=0.012).The expression of ZNF139 and MMP-7 was correlated with lymph node metastasis,muscle invasion and FIGO stage(P=0.028,0.031;P=0.004,0.016;P=0.008,0.011).Spearman rank correlation analysis showed there was positive correlation between ZNF139 and MMP-7 in adenocarcinoma tissues(r=0.716,P=0.039).Western blot showed that ZNF139 and MMP-7 expression in adenocarcinoma group were higher than in adjacent cancer group(t=6.92,7.34;P<0.01);qRT-PCR showed that ZNF139 mRNA and MMP-7 mRNA expression in adenocarcinoma group were higher than in adjacent cancer group(t=4.27,4.06;P<0.05).Conclusion ZNF139 and MMP-7 are correlated with malignant pathological characteristics in endometrial adenocarcinoma,they may modulate the malignant behavior together.
3.Mechanism and effect of everolimus on endometrial cancer cell RL95-2 proliferation and apoptosis

Objective To observe the effect of everolimus on proliferation,cycle and apoptosis of endometrial cancer cells and discuss the mechanism.Methods Proliferation of RL95-2 interfered with everolimus in different concentration and time was observed by MTT assay.Cell cycle and apoptosis affected by everolimus was detected by flow cytometry.PI3K-Akt-mTOR signaling pathway was detected by Western blot.Results (1) MTT assay showed that,cell viability in everolimus group was time-dependent decreased,compared with control group,there were significant differences in 48 h,72 h and 96 h ((64.36±4.78)%,(48.18±5.93) %,(42.72±6.91) %;F inner grouP =3.278,P< 0.05;F between groups =12.327,P< 0.01;F across groups=7.729,P<0.05).Cell viability in everolimus group was dose-dependent decreased(F=3.264,P<0.05).Compared with control group,RL95-2 cells in G1 phase and G2 phase was (69.28±2.61)% and (4.75±0.84) %,decreased in everolimus group,and in S phase (27.31±0.69) % was increased (t=5.743,P<0.05;t =4.528,P<0.05;t=6.209,P<0.05).The apoptosis in everolimus group was 29.78%,higher than in control group (47.29%),the difference was significant (t =19.381,P<0.01).(2) Western blot showed that both mTOR and p-Akt in everolimus group were dose-dependent decreased (F=3.589,P<0.05;F =5.292,P<0.05),Akt was dose-dependent increased (F =4.294,P<0.05).Conclusion Everolimus decrease the cell viability and promote apoptosis via target inhibiting mTOR expression.
4.Expression of Axin in Peutz-Jeghers syndrome

Objective To study Axin expression in colorectal adenocarcinoma, colorectal adenoma, Peutz-Jeghers syndrome (PJS) polyp, and normal intestinal mucosa at mRNA and protein levels, and to explore the role of Axin in the pathogenesis of PJS. Methods The experiment samples, which were from the exsected normal mucosa, PJS polyps, adenomatous polyps and colorectal adenocarcinoma between Jan, 2001 and Mar, 2008 in Nanfang Hospital, each includes 32 sapamples. Immunohistochemistry was adopted to detect the Axin protein expression in each sample distinctively. Results transcriptase -polymerse chain reaction (RT-PCR) was used to detected and compare the Axin mRNA expression in 16 samples of each group. Results Of the tissues in each group, the positive Axin protein expression was located in the endochylema. 5.Effects of Self-Efficacy Training on Hemiplegics after Stroke

Objective To explore the effect of comprehensive rehabilitation combined with self- efficacy training on hemiplegics after stroke. Methods 60 ischemic stroke patients following hemiplegia were divided into three groups: control group 1 (n=20), control group 2 (n=20), and experiment group (n=20). All the patients received routine rehabilitation, the control group 2 received tasks analysis approach,and the experiment group received tasks analysis approach and self-efficacy training. All the patients were evaluated by Fugl-Meyer Assessment (FMA), Berg Balance Scale (BBS), Self- rating Depression Scale (SDS), Self- rating Anxiety Scale (SAS), modified Barthel Index (MBI) and General Self-efficacy Scale (GSES) before and 8 weeks after treatment. Results The scores of FMA, BBS, SDS, SAS, MBI and GSES improved significantly in the experiment group than in the control group 1 and the control group 2 after treatment (P<0.05). Conclusion Comprehensive rehabilitation combined with self-efficacy training can improve the ability of mood and motion, and the activities of daily living of ischemic stroke patient with hemiplegia.
6.Experiment of the ultrasound radiation force enhancement in adhesion of targeted microbubbles in vivo

Objective To investigate the effects of ultrasound radiation force(USRF) on adhering of targeted microbubbles in vivo.Methods A rat model of cremaster muscle Inflammation was developed,and microbubble(MB) or targeted microbubble of intercellular adhesion molecule-1 (MBICAM) was injected through the caudal vein.Immediately after intravenous MB or MBICAM injection,the cremaster was insonated for 5 minutes and the control group was in sham ultrasound exposure.All mice were divided into four groups randomly:① MB + false USRF; ② MB + USRF; ③ MBICAM + false USRF; ④ MBICAM + USRF.Fluorescence microscope was performed in twenty mice to evaluate microbubbles adhesion and video recordings were made with a high-resolution camera.Results The adhesion number of MBICAM was significantly greater in USRF group than that of MBICAM in false USRF group [(43.4 ± 2.1)/view vs (14.8± 1.8)/view,P =0.000].The adhesion number of MB in USRF group was greater than in false USRF group [(6.2 ± 1.3)/view vs (4.6 ± 0.9)/view].But there is no statistical significance (P =0.129).The adhesion number of MBICAM in both USRF group and false USRF group was significantly greater than the adhesion number of MB.Conclusions The ultrasound radiation force can significantly improve the adhesion of targeted microbubbles in vivo.
7.Value of transoral carotid ultrasonography in detecting the change of distal extracranial internal carotid artery with stenosis and occlusion in its initial segment

Objective To investigate the value of transoral carotid ultrasonography (TOCU) in detecting the change of distal extracranial internal carotid artery(ICA) with stenosis and occlusion in its initial segmen.Methods One hundred and ten patients with stenosis at least 50% or occlusion in the initial segment of ICA diagnosed by high-frequency linear probe were enrolled as case group,while thirty two with healthy carotid arteries as control group.Both of the case and control groups were undergone high-frequency linear probe to examine CCA and the initial segment of ICA,and TOCU to observe their lumen transparency,color Doppler flow display and change of Doppler flow spectrum,even measuring the internal diameter,peak systolic velocity (PSV),end-diastolic velocity(EDV) and PSVICA/PSVCCA ratio,respectively.Digital subtraction angiography (DSA)were performed within two weeks,then compared with ultrasonography.Results 92.7% patients were performed TOCU examination in case group and 93.8% in control group.The internal diameter and PSV of the distal extracranial ICA became gradually smaller from control group to near occlusion group,and there were statistical significance between groups ( P <0.05),however,near occlusion group had no statistical significance compared with occlusion group( P >0.05).There were 80% arteries having thrombus echo in occlusion group,and the others in this group having no thrombus had gloomy colour flow,even got low-velocity artery spectrum.Doppler flow spectrum of distal extracranial ICA showed characteristics of slow wave or single peak in near occlusion group,and spike shape or slow wave in 76.7 % arteries of 70% ~near-occlusion stenosis group,but spike shape in 30.8% arteries of 50%~69% stenosis group,and the others were normal.Conclusions TOCU is useful for detecting the changing regularity of distal extracranial ICA with stenosis or occlusion in its initial segment and has the value of clinical application in association with high-frequency linear probe.
8.Ultrasound imaging of acute renal allograft rejection with microbubbles targeted to intercellular adhesion molecule-1

ObjectiveTo assess the feasibility of evaluation of renal allograft acute rejection in rat withcontrast-enhancedultrasound( CEUS )andtargetedmicrobubbles.MethodsPhospholipid microbubbles targeted to intercellular adhesion molecule -1 (ICAM-1)(MBI) and control microbubbles (MB) were created by conjugating monoclonal antibody against ICAM-1 or isotype control antibody to the lipid capsule via “avidin-biotin” bridging.Ten SD rats with acute renal allograft rejection were injected intravenous of MBI and MB in random order with a 30-min interval.After 3 min of intravenous injection of microbubbles,targeted CEUS imaging was performed in all rats.And then the video intensity (VI) was determined.ResultsIn MBI group,a significant ultrasonic enhancement was observed,but it was not very obvious in MB group.Increment in VI value of transplant kidney in MBI group was great and it amounted to (27.0 ± 7.4)U,however,increment in VI value of in MB group was minor and it was merely (10.2 ± 2.4) U,Difference was evident in transplant kidney between of the two groups (P < 0.05).Conclusions Molecular imaging of ICAM-1 with targeted CEUS can evaluate renal allograft acute rejection effectively.
9.Changes of platelet function after clopidogrel discontinuation in patients with percutaneous coronary interventions

Objective To investigate the changes of platelet aggregation rate and platelet surface CD 41+CD62P+ expression af-ter clopidogrel discontinuation in the patients with percutaneous coronary interventions (PCI)) .Methods The platelet aggregation rates and platelet surface CD41+CD62P+ in 52 PCI patients with oral clopidogrel for near 12 months and discontinuation soon were measured before clopidogrel therapy(T0 ) ,in 1 week after clopidogrel therapy(T1 ) ,1 week before clopidogrel discontinuation(T2 ) , 1 week(T3 ) and 1 month after clopidogrel discontinuation (T4 ) .Results Compared with T0 ,the platelet aggregation rate and the expression of platelet CD41+CD62P+ at T1 were significantly decreased ,the difference showing statistical significance (P<0 .05) , which at T2 maintained the lower levels ;which at T3 were increased ,which at T4 were recovered to those at T0 .The platelet aggre-gation rates at various time points were (44 .20 ± 18 .36)% ,(25 .38 ± 12 .10)% ,(23 .74 ± 8 .15)% ,(51 .79 ± 10 .55)% and(45 .97 ± 16 .42)% respectively ,and the positive rates of CD41+ CD62P+ were(12 .96 ± 11 .48)% ,(3 .93 ± 3 .33)% ,(4 .72 ± 3 .14)% , (13 .90 ± 10 .38)% and(10 .84 ± 8 .13)% ,respectively .Conclusion In the patients treated with 12-month clopidogrel after PCI ,the platelet aggregation rate and the CD41+CD62P+ positive rate are mildly increased at 1 week after clopidogrel discontinuation and gradually returned to the level before discontinuation at 1 month after clopidogrel discontinuation .
10.Research and preparation for nanoemulsion of Fructus Perillae oil

Objective To study the preparation of Fructus perrillae oil nanoemulsion and its quality evaluation. Methods The formulations of Fructus Perrillae oil nanoemulsion were optimized by drawing the pseudoternary phase diagrams and investigating the form, viscosity, particle size distribution, stability, conductance, and refractiratio. Results The optimal formulation was composed of wheat oil and cremophor RH40, electron microscopy presented Fructus Perrillae oil nanoemulsion as small spherical drops, viscosity was (5.49?0.02)/mm2?s-1, conductance was 316/ms-1, refractiratio was (1.124?0.002 50), and mean diameter of particle was (50.3?1.1) nm.The average recovery determined by GC was 98.9%, its mean RSD was 0.88%. Conclusion The solubility and stability of Fructus Perrillae oil are promoted by nanoemulsion with better produncibility. It is a favorable medical matrix.
