1.Determination of Three Sulfonamides Residues in Animal Derived Food by UPLC-MS/MS

Objective:To establish a determination method for sulfadoxine, sulfathiazole and sulfadiazine in animal derived prod-ucts by ultra-performance liquid chromatography-tandem mass spectrometry ( UPLC-MS/MS) . Methods:Trichloroacetic acid and ace-tonitrile were used in the extraction and removal of proteins. A Waters Acquity UPLC BEH C18 (100 mm × 2. 1 mm,1. 7 μm) column was used as the separation column. The mobile phase was 0. 1% formic acid in water-methanol with gradient elution. The three sulfona-mides residues were detected with multiple reaction monitoring( MRM) mode in multi-period. Results:Under the conditions,the linear range of sulfadoxine, sulfathiazole and sulfadiazine was 1. 0-200. 0 ng·ml-1 ,the linear correlation coefficient was above 0. 99,the re-covery was more than 70%,the detection limit was 0. 5 ng·ml-1 , and the quantification limit was 1. 0 ng·ml-1 . Conclusion: The sample preparation method is simple and fast, and the method can be used to analyze sulfadoxine, sulfathiazole and sulfadiazine in ani-mal derived food efficiently and sensitively.

4.Expression of RECK in pancreatic carcinoma and its relationship with prognosis

Objective To study the expression of reversion-inducing-cysteine-rich protein with Kazal motifs (RECK) in human pancreatic carcinoma tissues and pancreatic carcinoma cell lines; the effects of recombinant lentiviruses carrying RECK gene(LV-RECK) therapy on human pancreatic carcinoma xenograft in nude mice; and to find out the relationship between the expression of RECK and the prognosis of pancreatic carcinoma.Methods Immunohistochemical method was used to detect the expression of RECK in the resected specimens of pancreatic carcinoma and their corresponding normal pancreatic tissues in 42 patients.Western blotting was used to examine the expression of RECK in human pancreatic carcinoma cell lines (PANC-1,MIAPaCa-2,AsPC-1).Statistical analyses were performed to determine the relationship between RECK expression and the clinicopathological characteristics and prognosis in pancreatic carcinoma.Subcutaneous xenograft tumor models of human pancreatic carcinoma were established in nude mice.These nude mice were then divided into the experimental group,the negative control group and the blank control group randomly.The three groups of nude mice were intratumorally injected with LV-RECK,LV-EGFP and normal saline (NS) respectively.The antitumor effect was studied.Immunohistochemical method was used to detect the expression of RECK and microvessel density (MVD).Terminal deoxynucleotidyl transferase mediated dUTP-DIG nick end labeling (TUNEL) was used to detect the apoptosis of tumor cells.Survival analysis was performed.Results All three pancreatic carcinoma cell lines did not express RECK.The overall positive rate of RECK expression was 45.2 % (19/42) in pancreatic carcinoma,and 88.1 % (37/42) in normal pancreatic tissue.The expression level of RECK was significantly lower in the tumor tissues than in the normal tissues (P<0.01).The expression of RECK was significantly associated with TNM stage,lymph node metastasis and local infiltration of pancreatic carcinoma (P<0.05).Kaplan-Meier survival analysis revealed that the survival time was significantly longer in the RECK positive patient group than in the RECK negative patient group.Univariate Cox regression analysis revealed that RECK expression,TNM stage,lymph node metastasis and local infiltration were significantly related with prognosis for pancreatic carcinoma (P<0.05).Multivariate Cox regression analysis revealed that only RECK expression remained as an independent significant factor in predicting the prognosis of pancreatic carcinoma (P < 0.001).When compared with the negative control and the blank control groups,the volume of subcutaneous xenograft tumor in the experimental group was significantly decreased (P<0.05).RECK protein in the experimental group was re-expressed.MVD of the experimental group was significantly less than those of the control groups (P<0.05).Apoptotic index (AI)of the experimental group was significantly higher than those of the control groups (P<0.05).The survival time of nude mice in the experimental group was significantly longer than those in the control groups (P<0.05).Conclusions RECK expression was closely related to invasion,metastasis and prognosis of pancreatic carcinoma and it was an independent prognostic marker.RECK gene over-expression inhibited neovascularization of pancreatic carcinoma,induced apoptosis of tumor cells,inhibited the growth of tumor xenograft and improved the prognosis of tumor-bearing mice.These results suggest a possible new treatment for pancreatic carcinoma.
5.The diagnosis and treatment of rectal carcinoid: a report of 33 cases

Objective To study the clinical and pathological features of rectal carcinoid and factors influencing the prognosis.Methods Data of 33 rectal carcinoid patients were analyzed retrospectively. 29 patients were followed up postoperatively. Primary tumors were classified by size (2*!cm) and muscularis invasion respectively.Results The main symptoms were changes in bowel habits(17/33,52%) and hematochezia (14/33, 42%), or rectal mass (14/33,42%), asymptomatic patients were detected during a survey.Only 4(12%) patients had tumors larger than 2 cm and tumors invading muscularis.The 10 year survival rate was 84% for all patients( n =29), and 94% for those with tumors 2*!cm( n =3) died in 5 years( P = 1 0?10 -9 ) and only one out of the 4 with muscularis invasion lived longer than 5 years( P =1 2?10 -5 ).Conclusions Most rectum carcinoid are at early stage suitable for local excision.
6.Application of Color Doppler Flow Imaging in Portal Vein Chemotherapy by Ultrasound Guided Percutaneous Transhepatic Portal Injection

To evaluate the role of color Doppler flow imaging in the portal vein chemotherapy by ultrasound guided percutaneous transhepatic portal injection, the blood supply of liver tumor and portal hemodynamics were studied in 15 patients with primary hepatic carcinoma and one patient with secondary hepatic carcinoma. The continuous wave in the lesion and forward continuous wave in the portal vein seen by pulsed Doppler in nine cases, were indications for this therapeutic method. The distribution of the drugs in the liver could be observed by color Doppler flow imaging when injection was being done. The therapeutic efficacy of portal vein chemotherapy in liver tumor may be evaluated by color Doppler folw imaging and puncture biopsy.
7.Effect of zein/chitosan composite membrane on the osteogenic differentiation of bone marrow mesenchymal stem cells

BACKGROUND:Some scholars have prepared zein/chitosan composite membrane based on blending methods, and preliminary evaluation ofitsphysical and chemical properties showsthat chitosan partly improvesthe mechanical properties and hydrophilic properties of zein. Therefore, zein/chitosan composite membrane presumably has good cytocompati bility, which is beneficial to osteogenic differentiation of bone marrow mesenchymal stem cels.
OBJECTIVE:To explore the effect of zein/chitosan composite membrane on the differentiation of bone marrow mesenchymal stem cels into osteoblasts and its feasibility asabone tissue-engineered material.
METHODS:With 60% acetic acid as solvent, zein/chitosan composite membrane was prepared by blending and casting method. The structure and physicochemical properties of the composite membrane were investigated by Fourier transform infrared spectroscopy, tensile testing, water absorption testing and scanning electron microscopy. And the cytocompatibility of the membrane was evaluated byin vitrocel cufture. Besides,bone marrow mesenchymal stem celsfrom Sprague-Dawley ratwere isolatedvia adherence screening method, andthe effects of thecompositemembrane on theosteogenic differentiation ofthese celswere observedby scanning electron microscopy, fluorescent labeling and alkaline phosphatase assay.
RESULTS AND CONCLUSION:The tensile strength, water absorption and hydrophilicity of the films were improved with the chitosan increased; chitosan could promote cel proliferation indicating the good cytocompatibility of the composite films. Moreover, osteogenic induction occurredin bone marrow mesenchymal stem cels cultured on the compositem embrane, and with an increase of chitosan, the induction was promoted. In conclusion, zein/chitosan composite membrane can be applied widely in the field of bone tissue engineering.
8.Lead exposure during different periods of pregnancy in placental apoptosis in rats

Objective To investigate the effects of lead exposure on rat placental apoptosis during different periods of pregnancy. Methods All Wistar rats were randomly divided into four groups with 27 for each group with the sex ratio of 2 : 1 (female : male).The groups with lead exposure consumed water with 0. 025% lead acetate during the entire, early or late period of pregnancy. Controls were given distilled water without lead.Blood lead levels were determined by atomic absorption spectrophotometry at the end of pregnancy. Placental apoptosis were assessed by both Hoechst staining and terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling (TUNEL) assay. Data were assessed by one-way analysis of variance, and differences between groups were compared by q-test. ResultsBlood lead levels at the end of pregnancy of the groups with lead exposure (entire, early or late pregnancy) and the control group were (1. 74±0. 19) μmol/L,(1.27±0.26) μmol/L, (0.60±0. 11) μmol/L and (0.04±0.01) μmol/L respectively(F= 12. 10,P<0.01).In the groups with lead exposure, Hoechst staining showed hyperchromatic nuclei in placental trophoblast apoptotic cells and compact fluorescent particles in some nucleus; TUNEL assay showed brown-staining apoptotic cells nuclei with some nuclei particles staining brown. Two assays showed the same results: the apoptotic index of the groups with lead exposure were higher than that of the control group; the apoptosis index of the group with lead exposure during entire pregnancy was higher than that of the group with lead exposure during early and late pregnancy (P < 0. 05).ConclusionsLead exposure during pregnancy could elevate the blood levels of lead and the degree of placental apoptosis.
9.A new terpenoid from Ganoderma lucidum.

A new terpenoid, lucidone D (1), has been isolated from Ganoderma lucidum. Its structure was determined to be 7beta, 15alpha-dihydroxy-4, 4, 14alpha-trimethyl-3, 11, 20-trioxo-5alpha-pregn-8-en on the basis of 1D and 2D-NMR spectral analysis.
10.Simple evaluation on combined wound with seawater immersion

BACKGROUND: To establish experimental animal model for rapid evaluation of combined wound with seawater immersion so as to benefit wound healing and recovery.OBJECTIVE: To explore the plan of simple evaluation of combined wound with seawater immersion.DESIGN: Group division and controlled experiment was designed.SETTING: Second Department of Orthopaedics, the 113 Hospital of Chinese PLA; Research Institute of Surgery, Daping Hospital, Third Military Medical University of Chinese PLA MATERIALS: Totally 32 health hybridized adult dogs were employed,mass weighted varied from 12 to 15 kg, of either sex.METHODS: The experiment was performed partially in the specific sea area of the Fifth and Sixth Room , Research Institute of Surgery of Daping Hospital of the Third Military Medical University of Chinese PLA from October 2001 to June 2002. The dogs were randomized into burn-blast combined wound group and projectile-burn combined wound group and each group was subdivided into immersion group (10 dogs) and the control (6 dogs). Burn-blast combined wound group: The model of burn and mild blast injury was prepared. In the control, the dogs were put in a room at 25 ℃ directly after injury; But in immersion group, the dogs were immersed immediately in simulated seawater with holder. Projectile-burn combined wound group: The model of burn and mild projectile injury was prepared. The managements were same as the previous after injury in the control and immersion group. The change of physiologic indexes was observed in 28 hours after injury. The indexes related to death were picked up for correlative analysis and establishing wound scoring scale, inducing body temperature, heart rate, mean arterial pressure (MAP), chloride concentration, PaO2 and causative factor, 0-5 score were designed, the higher the score was, the higher mortality would be.MAIN OUTCOME MEASURES: ① Analysis of dead cases in each group. ② Wound scoring. ③ Verification of wound in scoring scale.RESULTS: 32 dogs all entered result analysis. ① Analysis of dead cases and relationship: Totally, 12 dogs were died. The peak of death was in 10-12hours, no matter the immersion happened or not. 5 indexes were associated significantly with death, named body temperature, heart rate, MAP, PaO2 and chloride concentration (P < 0.05). ② Comparison of wound scoring, predictive mortality and actual mortality in each group: The results in burn-blast control were (5.89±3.25) score, 0 and 17% successively. Those in projectileburn control were (6.78±2.02), 25% and 33% successively. Those in burn blast immersion group were (9.50±4.20) scores, 50% and 30% and those in projectile-burn immersion group were (11.13±3.57) scores, 50% and 60%.CONCLUSION: ① The established scoring scale is apt to obtain common physiological indexes so as to evaluate the wound rapidly, which is applica ble for either seawater immersed cases or non-immersed cases, indicating extensively applications. ② The correction of evaluation is acceptable with verification. ③ The scale is based on animal experimental data, for which,it is still different from the one of real wound evaluation.
