1.Protective effects of Zhenbao pill on acute spinal cord injury

Objective To explore the protective effect of Zhenbao pill on acute spinal cord injury of rat and its possible mechanisms.Methods 64 Wistar rats were randomly divided into a injury group and a drug group, with 32 rats in each group, A T10 acute incomplete spinal cord injury model was produced by using modified Allen technique in the two groups,Zhenbao pill mixed suspension of 0.6 g/kg was given by gavage method daily in the drug group 30 min after moding, the same amount of saline was given in the injury group. Behavior detection were conducted in the first day, third day, and the seventh day after modeling to each 8 rats of the two groups. Spinal cord specimens was got centered about the damage points after rats were executed. histopathological changes were observed under HE staining, the MDA content were determined by thiobarbituric acid spectrophotometer and the apoptotic cells were marked by TUNEL method.Results After 1, 3, 7 d, compared with the injury group, the MDA level in the rat spinal cord tissue in the drug group (15.12 ± 1.27 nmol/mgvs.19.71 ± 1.29 nmol/mg, 20.42 ± 1.33 nmol/mgvs.24.65 ±1.36 nmol/mg, 10.46±1.49 nmol/mg vs.15.46 ± 1.44 nmol/mg) decreased(P<0.05); the apoptosis rate of drug groups (19.61% ± 1.49%vs.26.61% ±1.52%, 21.49% ± 1.37% vs.32.37% ± 1.24%, 13.04% ± 1.30%vs.18.35% ± 1.27%)decreased (P<0.05); the BBB score of drug groups (6.52 ± 1.27vs. 2.63 ± 1.27), (12.68 ± 1.32 vs.6.17 ± 1.34), (15.47 ± 1.27 vs.11.57 ± 1.29) was higher than the injury group (P<0.05). Conclusion Zhenbao pill can significantly improve the histopathological examination,reduce the MDA content in the injured spinal cord specimen of the rat and the rate of neuronal apoptosis, which may promote recovery of neurologic function in the rat suffered from ASCI.
2.Growth and decline of Oncomelania hupensis status in Duchang County at North shore of Poyang Lake,2005-2012

Objective To understand the dynamic of Oncomelania hupensis snail status in Duchang County at north shore of Poyang Lake so as to provide the evidence for formulating the strategy of schistosomiasis control. Methods The data about the snail status in Duchang County from 2005 to 2012 were collected and the change trend of the snail status was analyzed. Re?sults From 2005 to 2012 all the average density of living snails the occurrence rate of frames with living snails the density of infected snails and the area with infected snails showed a trend of sharp decline and stayed at a low level. Compared to 2005 the above 4 indexes in 2008 decreased by 70.06% 90.33% 79.48% and 37.88% respectively and those in 2012 decreased by 99.35% 98.98% 99.33% and 93.84% respectively. The infection rates of snails fluctuated between 0.06%and 0.53%which showed a trend of first decrease and then increase. The snail area maintained at the level of 2 052.5 hm2 since 2007. Con?clusions Most of the indexes of the snail status in Duchang County have decreased to the lowest level in history since 2012 but the snail area is stable and the infection rate of snails shows a fluctuation trend which suggests that the risks of schistosomia?sis transmission still exist and the snail control still should be strengthened.
3.The differential diagnosis effect of CT total tumor perfusion parameters on benign and malignant occupying lesions of the lung

Objective To investigate the effect of CT total tumor perfusion parameters in the identification of benign and malignant lung lesions. Methods 80 patients with pulmonary space-occupying lesions who were treated in our hospital from December 2015 to May 2017 were enrolled. After pathological histological diagnosis, 52 cases of malignant lesions and 28 cases of benign lesions were confirmed. And they all underwent CT whole tumor perfusion scan and perfusion imaging analysis. And the blood flow, blood volume, average transit time and surface permeability were obtained.Some cases of histopathological specimens were treated by monoclonal CD34 staining method, and the microvascularrich areas in the specimens were selected for blood vessel counting. The morphology of blood vessels was observed and microvessel density was obtained. Results The levels of BF, BV, MTT and PS in benign lesions were significantly lower than those in patients with malignant lesions. The best diagnostic threshold, sensitivity and specificity of BF in lung occupying lesions was> 49.12 mL/100 mL, 80.00% and 52.40%. The best diagnostic threshold, sensitivity and specificity of BV was 6.68 m L/100 mL, 70.00% and 81.00%. The best diagnostic threshold, sensitivity and specificity of MTT was3.82 m L/100 mL, 97.50 % and 52.40%. The best diagnostic threshold, sensitivity and specificity of PS was 7.25 mL/100 mL, 92.50% and 90.50%. The malignant lesion microvessel density count in the malignant lesions was significantly higher than that of benign lesions, and the difference was statistically significant (P<0.05). Conclusion CT total tumor perfusion parameters have a great auxiliary effect on the diagnosis of benign and malignant lung occupying lesions. Among them, PS and BV have the strongest sensitivity and are worthy of clinical promotion.
4.Effect of pingxian granules on protein expression of apoptosis regulatory genes in hippocampus of pentylenetetrazol-induced epileptic model rats.

OBJECTIVETo observe the effect of Pingxian granules on the protein expression of apoptosis regulatory genes Bcl-2 and Bax in the hippocampus of epileptic model rats and study the molecular biological mechanism of the anti-epileptic effect of Pingxian granules.
METHODTotally 60 45-days-old Wistar rats were selected and then randomly assigned into 5 groups: the normal control group, the model group, the positive control group, the Pingxian high dose group and the Pingxian low dose group, with 12 in each group. Except the normal control group, all the groups were intraperitoneally injected with 35 mg x kg(-1) pentylenetetrazol to establish the models of epilepsy. The Pingxian high dose (1.66 g x mL(-1)) and low dose (0.42 g x mL(-1)) groups were intragastrically infused with Pingxian granules 2 mL x d(-1). The positive control group received 3.6 g x L(-1) phenobarbital suspension by gastric perfusion. The normal group and the model group were drenched with distilled water, 2 mL x d(-1), for 5 weeks. Bcl-2 and Bax protein positive cells were labeled with immunohistochemical SABC at 3, 5 w.
RESULT(1) Rats in the model group appeared the epileptic behavior at the 1st week, and became serious with the kindle frequency; grade VIepileptic behavior appeared at the 4th week. The attack frequency and grade of the Pingxian group were less and lower, the highest grade were only IV, and there were no significant differences in the attack grade and frequency. (2) With the increase in kindle frequency, the model group showed a notable decrease in the Bcl-2 expression compared with the normal control group at the 3rd and 5th weekend (P < 0.01), but a significant increase in Bax protein expression (P <0. 01). The number of the Bcl-2 protein expression in Pingxian groups and the positive control group were obviously more than the model group (P < 0.01); and the number of the Bax protein expression in Pingxian groups and the positive control group were obviously less than the model group (P < 0.01).
CONCLUSIONPingxian granules may decrease neuronal cell apoptosis by improving the protein expression of apoptosis regulatory gene Bcl-2 and inhibiting the protein expression of apoptosis regulatory gene Bax with a view of anti-epilepsy.

5.Effects of self-assembled IKVAV peptide nanofibers on olfactory ensheathing cells.

We observed the effects of IKVAV self-assembling peptide nanofiber scaffold (SAPNS) on olfactory ensheathing cells (OECs). The IKVAV molecules were triggered to self-assemble to interconnected nanofibers hydrogel by adjusting pH of solution and adding of DMEM/F12 culture medium. Atomic Force Microscopy (AFM) showed that self-assembly hydrogel was consisted of the interconnected nanofibers, which varied from three nm to five nm in diameter and hundreds nanometer in length. The primary OECs were isolated from rat olfactory bulb and purified by differential adhesion twice. At days 12, the purity of OECs was 85% according to immunostaining of P75 NGFR antibody. OECs were cultured with IKVAV peptide. The adhesion, viability and proliferation of OECs were observed with inverted microscope, Calcein-AM/PI staining and Cell Counting Kit-8. OECs cultured on IKVAV SAPNS grew well and the viable cell count was 95%. IKVAV SAPNS can promote the adhesion of OECs and did no hinder the proliferation of OECs. IKVAV SAPNS nanofiber gel has good biocompatibility and bioactivity for OECs. It can serve as a good nerve tissue engineering scaffold.

6.Research advances in the degradation of hepatic lipid droplets through the autophagy pathway

Autophagy is a highly conserved cellular degradation pathway that degrades lipid droplets through a process called “lipophagy”. Lipophagy can selectively recognize lipid substances and degrade them, promoting β oxidation and thereby maintaining the balance of intracellular lipid metabolism. The liver regulates lipid droplet metabolism through lipophagy signaling pathways or key molecules, thereby alleviating hepatic steatosis and improving nonalcoholic fatty liver disease (NAFLD). This article reviews the latest advances in the degradation of hepatic lipid droplets through the three autophagic pathways of macroautophagy, molecular chaperone-mediated autophagy, and microautophagy. The major signaling pathways of AMPK/mTOR-ULK1, ATGL-SIRT1, FGF21-JMJD3, and Akt are involved in the regulation of the lipophagy process and help to maintain the homeostasis of lipid metabolism in the liver, so as to provide new ideas for clinical prevention and treatment of NAFLD.
