1.Study on the role of active oxygen in the pathogenesis of respiratory distress syndrome in dogs

Twenty-five mongrel dogs were randomized into 3 groups;the animals of group Ⅰ were traumatized with bone marrow extract,those of group Ⅱ were similarily traumatized and then treated with anisodamine,and those of group Ⅲ received saline only and served as control.The specimens of arterial and venous blood were collected and bronchoalveolar lavage fluid (BALF) and lung homogenate were obtained after the animal was killed.The activity of superoxide dismutase (SOD) and the concentration of lipid peroxides (LPO) were determined.Meanwhile,the clinical manifestations,blood gas analysis.chest radiography,and pathological examinations were performed or observed.It was found there were following findings:(1)In group I,SOD activity and LPO level in the lung homogenate and BALF were markedly elevated immediately after injury,which suggests that there had been a rapid production of active oxygen.As a result.various degrees of lung damages were pricipitated.(2)In group Ⅱ,an increase of SOD activity and LPO level in the blood,lung homogenate and BALF:The elevation was more marked than that in group Ⅲ but less marked than that in group Ⅰ.which indicates that there was a relatively of less amount of active oxygen production by the interference anisodamine.(3)In group Ⅱ,no significant changes of SOD activity and LPO livel were found.Our findings suggest that active oxygen is likely to play a very important role in the pathogenesis of acute lung damages in respiratory distress syndrome.
2.A study of osteoporosis in patients with type-2 diabetes

Objective To investigate osteoporosis and its main associated factors in patients with type 2 diabetes (DM). Methods Bone mineral density(BMD)of lumbar vertebrae 2~4 and proximal femur in 40 type 2 diabetic patients were measured with dual energy X-ray absorptiometry and compared with control group(30~35 years old). Serum Ca, P, ALP and connective peptide in type 2 diabetes mellitus patients were measured. Results The area under the connective peptide releasing curre in osteoporosis group was lower than that of non-osteoporosis in type 2 diabetes. Conclu.sion BMD is correlated with the pancreatic islet function. The patients with pancreatic islet failure develop osteoporosis easily.
3.MULTIPLE BIOMARKER ASSAY IN PATIENTS WITH LUNG CANCER

The levels of CEA, Lys, Fr and ?2-MG of serum and brochoalveolar lavage fluid (BALF) were determined in 99 patients with inflammatory lung diseases and 23 healthy subjects for controls. Their levels in BALF were compared between diseased and normal sides of lungs. The value of diagnosis of individual formulas was suggested. The combined ditermination of multiple biomarkers in serum and BALF can help to raise the positivity and spcificity of diagnosis of lung cancer.
4.Expression of HLA-DQA1 gene polymorphism in the lung cancer patients with pneumoconiosis

Objective To detect the difference of the expression of HLA-DQA1 genes polymorphism in the lung cancer patients with pneumoconiosis and the non-occupational lung cancer patients.Methods Multiple polymerase chain reaction-ligase detection reaction classification method (PCR-LDR) was used to detect 6 alleles in HLA-DQA1 and the expression of HLA-DQA1 gene between the 21 cases of pneumoconiosis complicated with lung cancer and 40 cases of non-occupational lung cancer respectively.The correlation between them with pneumoconiosis complicated with lung cancer was analysed.Results HLA-DQA1 0301 gene frequencies were 19% in pneumoconiosis complicated with lung cancer group and 2.5% in non-occupational lung cancer group respectively,with a statistically significant difference (x2 =10.022,P =0.002).There was no statistically significant difference in the other allele between the two groups.Conclusion HLA-DQA1 gene polymorphism may be related to the susceptibility of the pneumoconiosis complicated with lung cancer.DQA1 0301 is one of the important genes of pneumoconiosis complicated with lung cancer,which maybe the risk factor for the susceptibility of the pneumoconiosis complicated with lung cancer.
5.Clinical observation of dynamic blood sugar monitoring combined with insulin pump in the treatment of newly diagnosed type 2 diabetes mellitus in the elderly

Objective To evaluate islet cells functional recovery and clinical effects of dynamic blood sugar monitoring combined with insulin pump in newly diagnosed type 2 diabetes mellitus in the elderly.Methods 300 cases with newly diagnosed type 2 diabetes mellitus were randomly divided into treatment group (n=150) and control group (n=150) accompanying with three months follow-up.Body mass indexes (BMI),levels of hemoglobin A1c (HbA1c),fasting plasma glucose (FPG),2-h postprandial blood glucose,fasting insulin,2-h postprandial insulin,fasting C-peptide,2-h postprandial C peptide,and the percentage of patients who took oral hypoglycemic drugs alone without insulin could well control the blood glucose were compared between the two groups before versus after treatment.Results The levels of BMI,HbA1c,FPG,2-h postprandial blood glucose,fasting insulin,2-h postprandial insulin,fasting C peptide,2-h postprandial C peptide in the two groups were significantly decreased after treatment as compared with the pretreatment (P < 0.05).The improvement of blood sugar spectrum was better in treatment group than in control group(P<0.05) and the percentage of patients who took oral hypoglycemic drugs alone without insulin could well control the blood glucose was significantly higher in treatment group than in control group(21% vs.3%,x2 =22.475,P<0.05).Conclusions The clinical effects of dynamic blood sugar monitoring combined with insulin pump on newly diagnosed type 2 diabetes mellitus in the elderly is definite and can improve the recovery of islet cells function.It is worthy of clinical promotion.
6.Isolation and identification of marine symbiotic and epiphyte microorganisms with antimicrobial activity

125 strains of the symbiotic and epiphyte microorganisms were isolated from marine organisms (sea cucumber, sea urchin,anemone, sea actinia, Ulra, Sargassum, Undaria). Among them,21 strains of bacteria,8 strains of actinomycetes and 2 strains of fungi showed antagonistic activity on bacterial or fungal growth. In the 21 strains of bacteria, 7 strains belong to Bacillus sp., 11 strains Vibrio sp., and 3 strains Pseudomonas sp.. In the 8 strains of actinomycetes, 5 strains belong to Streptomyces sp., 3 strains Micromonospora sp., 2 strains fungi Penicillum sp..

Objective To observe the inhibitory effect of caspase\|3 mRNA antisense oligodeoxynucleotides (ASODN) on the apoptosis of HL\|60 cells, and to screen the effective ASOND. Methods By means of the lipofectimine\|mediating transfection, caspase\|3 mRNA ASODNs of four different sequences were introduced into HL\|60 cells followed by ? irradiation.The gel electrophoresis was performed for the DNA ladder, Hoechst 33258\|propidium iodide fluorecent staining for the percentage of apoptotic cells, and FCM for quantitative analysis of apoptosis. Results When HL\|60 cells were transfected with caspase\|3 mRNA ASODNs targeting 5'noncoding region (sites -62 to -46) and initiative coding region (sites -1 to 16) at the final concentration of ≥3?? mol/L, the DNA ladder was not detected with the gel electrophoresis, nor was the apoptotic peak found with FCM. Fluorescent staining analysis showed that the percentage of apoptotic cells was significantly lower than that in the control groups including non\|transfection group and mismatched oligodeoxynuleotide group ( P

OBJECTIVE To investigate the clinical characteristics and prevention of urinary system infections in type 2 diabetic patients.METHODS An analysis of pathogens and clinical characteristics of 120 hospitalized type 2 diabetic patients complicating urinary system infections was performed.RESULTS In type 2 diabetic patients complicating urinary system infections,the most common pathogen was Escherichia coli,the next were Staphylococcus epidermidis,Klebsiella pneumoniae,etc.There were no significant difference of pathogens between symptom group and non-symptom group.But they were obviously more among senile patients aged over 60 in non-symptom group than that in symptom group(P

Objective To explore the effects of polyclonal antibody to lipopolysaccharide binding protein (pLBPab) on IL 10 and IL 18 mRNA expressions of alveolar macrophages in lipopolysaccharide LPS induced acute lung injury in rats. Methods A total of 40 male Wistar rats were randomly divided into 5 groups: normal control (A), LPS treated group (B), group of pLBPab preconditioning at 30 min before injection of LPS (C), group of treatment with LPS and pLBPab (D), and group of pLBPab preconditioning at 30 min after injection of LPS (E). mRNA expressions of IL 10 and IL 18 in the alveolar macrophages in each group were measured by semi quantitative reverse transcription polymerase chain reaction (RT PCR). Results The IL 10 and IL 18 mRNA expressions were highly increased respectively in the alveolar macrophage (AM?) stimulated with single LPS, but they were significantly decreased in the AM? after stimulation with LPS + pLBPab, particularly stimulation with pLBPab 30 min before LPS injection. Conclusion pLBPab can decrease the mRNA expressions such as IL 10 and IL 18 by alveolar macrophages in acute lung injury in rats induced by LPS, and LBP antibody could be used to cure some diseases such as SIRS, acute lung injury, ARDS, and septic syndrome.
10.Effects of Processing on Contents of Amino Acid and Part of Inorganic Elements in Arisaema Amurense Maxim.

Objective: To investigate the effects of different processing methods(steeping, boiling) and different adjuvants(alum, ginger, bile) on contents of amino acid and part of inorganic elements in Arisaema amurense Maxim.. Methods: The amino acid was determined by HPLC, and the inorganic elements were determined by AAS. Results: The total content of amino acid was the highest in raw Arisaema amurense Maxim. and the lowest in Arisaema amurense Maxim. processed with bile, wherease it was similar to other processed samples. The inorganic element content changed after it was processed. Conclusion: The long rinse is the key factor of losing amino acid in Arisaema amurense Maxim. after being processed. The content of inorganic element in adjuvants is the key factor of their content changes in Arisaema amurense Maxim. after it is processed.
