1.Effect of lipopolysaccharide on apoptosis of human umbilical vein endothelial cells

AIM: To explore the mechanism of lipopolysaccharide (LPS) on vascular endothelial cell(VEC) damage. METHODS: By using cytometry techniques, we studied the effects of LPS on apoptosis of human umbilical vein endothelial cells (HUVECs) in vitro . RESULTS: LPS was able to induce apoptosis of HUVECs in a time-dose-dependent fashion.CONCLUSION:Apoptosis might play a role in LPS-induced damage of vascular endothelial cells.
2.Changes of Beta-adrenergic Receptors and cAMP-PDE Activity of Asthmatic Lung Tissue in Guinea Pigs

The changes of the number of beta-adrenergic receptors, the activity of cAMP-PDE and the level of cAMP were observed in the guinea pigs with allergic asthma with radioligand assay, radioenzyme assay and radioimu-noassay respectively. It was found that there were a 25% decrease of the number of beta-adrenergic receptors on the lung membrane, a 36% decrease of the level of cAMP in the lung tissue, and a 35% increase of cAMP-PDE activity in the asthmatic animals as compared with those of the control. A close positive correlation was observed between the number of beta-adrenergic receptors and cAMP level,but a negative correlation between cAMP-PDE activity and cAMP level in the asthmatic group.

Objective To observe the inhibitory effect of caspase\|3 mRNA antisense oligodeoxynucleotides (ASODN) on the apoptosis of HL\|60 cells, and to screen the effective ASOND. Methods By means of the lipofectimine\|mediating transfection, caspase\|3 mRNA ASODNs of four different sequences were introduced into HL\|60 cells followed by ? irradiation.The gel electrophoresis was performed for the DNA ladder, Hoechst 33258\|propidium iodide fluorecent staining for the percentage of apoptotic cells, and FCM for quantitative analysis of apoptosis. Results When HL\|60 cells were transfected with caspase\|3 mRNA ASODNs targeting 5'noncoding region (sites -62 to -46) and initiative coding region (sites -1 to 16) at the final concentration of ≥3?? mol/L, the DNA ladder was not detected with the gel electrophoresis, nor was the apoptotic peak found with FCM. 4.Exploration of estradiol (E2) on the expression of aromatase of renal tissue in systemic lupus erythematosus (SLE) model mice

Objective:To investigate the expression of aromatase of renal tissue in systemic lupus erythematosus(SLE)model mice.Methods:BALB/c mice were induced SLE with homologuous splenic cell activated with ConA after being ovariectomized,and in the same time administered different doses of benzestrofol.E2 in peripheral blood and renal tissue was detected by ELISA and the expression of mRNA of aromatase in renal tissue was detected by RT-PCR in the 4th,6th,8th and 10th weeks.Results:The level of E2 of peripheral blood and renal tissue of SLE model mice became higher as benzestrofol exogenously administered heightened.Compared with control mice,the level of E2 in SLE model mice increased,and the expression of mRNA of aromatase of renal tissue increased with E2 increment.Conclusion:E2 promotes development of SLE through regulating expression of aromatase mRNA.
5.Meta-analysis of Pitavastatin Comparison of Atorvastatin in the Treatment of Primary Hyperlipidemia in Chinese Adults

OBJECTIVE:To systematically evaluate therapeutic efficacy and safety of pitavastatin comparison of atorvastatin in the treatment of primary hypedipemia in Chinese adults,and to provide evidence-based reference for clinic.METHODS:Retrieved from The Cochrane Library,PubMed,Chinese Journal Full-text Database,Wanfang database,and manually search Google Scholar,Baidu academic search engine,randomized controlled trials (RCTs) about pitavastatin (trial group) vs.atorvastatin (control group) in the treatment of primary hyperlipemia in Chinese adults were collected.After literature screening,data extraction,quality evaluation of included studies with modified Jadad scale,Meta-analysis of the levels of total cholesterol (TC),low density lipoprotein cholesterol (LDL-C),triglyceride (TG) and high-density lipoprotein cholesterol (HDL-C),response rate and the incidence of ADR was conducted by using Rev Man 5.3 statistical software.RESULTS:A total of 5 RCTs were included,involving 456 patients.Results of Meta-analysis showed that the decrease of TC level [MD=0.09,95%CI(0.01,0.16),P=0.03] in trial group was more better than control group,while the increase of HDL-C level [MD=0.08,95% CI (0.01,0.14),P=0.03] and the decrease of the TG level [MD=-0.13,95% CI (-0.20,-0.06),P=0.000 4] in trial group were worse than control group,with statistical significance.There was no statistical difference in the decrease of LDL-C[MD=-0.01,95% CI (-0.13,0.10),P=0.84],response rate [OR=0.75,95%CI (0.15,3.66),P=0.72] or the incidence of ADR [OR=0.68,95 % CI (0.44,1.05),P=0.08] between 2 groups.CONCLUSIONS:Pitavastatin has better therapeutic efficacy in decreasing TC,but its therapeutic efficacy in decreasing LDL-C is similar to that of atorvastatin;its therapeutic efficacy in decreasing TG and increasing HDL-C is worse than that of atorvastatin.The safety of them is equivalent.
