1.Three-dimensional finite element analysis of biomechanical properties of proximal femoral structure under the normal standing position

BACKGROUND:At present,there are many established femoral finite element studies,but previous studies focused on femoral biomechanics of the outside are few.OBJECTIVE:To establish three-dimensional finite element model of structure of the proximal femur,and to analyze its biomechanics in normal standing position to provide reasonable evidence for clinical application.DESIGN,TIME AND SETTING:A three-dimensional finite element model analysis was performed in General Hospital of Tianjin Medical University from December 2008 to April 2009.PARTICIPANTS:One male volunteer aged 30 years was enrolled at General Hospital of Tianjin Medical University.Femur lesion or injury was excluded by means of X-ray.METHODS:The primitive femur CT data were adopted according to Dicom 3.0 standard in Mimics so as to obtain the primitive three-dimensional femur model which was established via threshold defining,region enlarging and morphological performing.The primitive model was remeshed and combined with ANSYS software to create a final finite-element model.The model obtained by using the Mimics was loaded with 200 N vertical loading to check out the difference of the stress distribution.MAIN OUTCOME MEASURES:Using 200 N as the external load in the vertical direction,the Vonmises stress distribution of the femur was detected.RESULTS:The applications of Mimics and Ansys software could create a finite element model of proximal femur.It was found that the correspondent relationship between stress and structure in proximal femur was verified.CONCLUSION:Mimics can build more three-dimensional finite element models in line with the proximal femoral mechanical structure and mechanical properties.The mechanical properties of the femur are more credible,and the results of the analysis support the clinical application of the guidance.
2.Analysis of amino acid and fatty acid of Fructus Broussonetiae

To analyze amino acid and fatty acid of Fructus Broussonetiae . The amino acids of Fructus Broussonetiae were analyzed with automatic amino acid instrument and fatty acids were analyzed with GC MS Computer system. More than 14 amino acids were analyzed and the fatty acid from Fructus Broussonetiae contains 17 compounds, of which 8,11 octadecadienoic acid and Hexadecanoic acid are main components.The abundance of amino acids and unsaturated fatty acid may contribute to the tonic function of Fructus Broussonetiae .
3.Effects of various seed sources and different storage conditions on seed germination rate of Valeriana officinalis

Objective To investigate the effects of seed sources and storage conditions on the seed germination rate of Valeriana officinalis. Methods The seeds of V. officinalis under different storage conditions were used as materials for germination test, and the germination rate was calculated and compared according to the results. Results The seed germination rate of valerians is varied with different sources. The seed germination rate of V. officinalis is higher than that of V. officinalis var. latifolia, and the seed germination rate of cultivated valeriana is much higher than that of the wild plant. The storage periods, storage temperature, and germination conditions also affect the germination rate. Conclusion To maintain the seed germination rate, the seed of V. officinalis should be stored in conditions with ventilation and at suitable low temperature. Lower temperature helps to prolong the seed life with relatively high germination rate. Valeriana seed should also be propagated at available temperature conditions in proper seasons.

5.Effect of Liriope platyphylla total saponin on learning, memory and metabolites in aging mice induced by D-galactose

OBJECTIVE: To investigate the effects of Liriope platyphylla total saponin (LPTS) on learning, memory, neuromediators and metabolites in aging mice induced by D-galactose. METHODS: Ninety Kunming mice were randomly divided into nine groups: normal saline (NS)-treated group, untreated group, high- (100 mg/kg), medium- (50 mg/kg) and low-dose (10 mg/kg) LPTS-treated groups, Shuxuening-treated group, jiaogulanosidi-treated group, flunarizine-treated group and vitamin E-treated group. The Kunming mice in the NS-treated group were administered with NS by intraperitoneal injection, while the aging mice in the other eight groups were administered with D-galactose by intraperitoneal injection. At the same time, the aging mice in different groups were fed with corresponding drugs for 42 days, and the aging items of the mice in different groups were measured, respectively. RESULTS: LPTS could improve the memory of aging mice induced by D-galactose, promote its body weight, and increase the thymus and spleen indexes of the aging mice. LPTS could decrease the levels of MDA and lipofuscin, inhibit MAO activity and increase SOD activity and GSH-Px level. CONCLUSION: LPTS may improve the ability of learning and memory and delay aging.
6.Correlation between the expression of TPSAB1 and the proliferation of CD4+T cells in primary biliary cirrhosis

This study performed to investigate the differences of plasma gene expression profiles between primary biliary cirrhosis(PBC)patients and healthy controls,and the effect of tryptase Alpha/Beta 1(TPSAB1)on the proliferation of naive CD4+T cells derived from peripheral blood mononuclear cell(PBMC)in patients with PBC.The plasma samples of three PBC patients and three healthy controls were analyzed by microarray;Immune cell isolation kits were used to isolate natural killer(NK)cells,B cells,naive CD4+T and naive CD8+T cells from PBMC;The expression level of TPSAB1 in NK cells,B cells,naive CD4+T cells and naive CD8+T cells were analyzed by qRT-PCR;The expression of TPSAB1 in CD4+T cells was analyzed by Western blotting after interference by short hairpin RNA(shRNA)targeting TPSAB1.WST-1 proliferation kit and 5-Ethynyl-2'-deoxyuridine(EdU)staining assay were used to detect the proliferation of naive CD4+T cells after CD3/CD28 stimulation in vitro.The apoptosis of CD4+T cells was measured by flow cytometry.Enzyme linked immunosorbent assay(ELISA)was used to determine interferon-γ(IFN-γ),interleukin-4(IL-22),and IL-17 secretion in TPSAB1 knock-down CD4+T cells.Data showed that the expression level of TPSAB1 in NK cells and naive CD4+T cells from PBC patients were significantly higher than those of healthy control group.Naive CD4+T cells from PBC patients showed enhanced proliferative capacity after CD3/CD28 stimulation compared with controls.However,there was no difference in apoptosis between the two groups.The levels of IL-22,IL-17 and IFN-γ were significantly increased in PBC patients.Compared with the control group,knockdown of TPSAB1 inhibited the proliferation ability of naive CD4+T cells under CD3/CD28 stimulation.At the same time,the levels of IL-22,IL-17 and IFN-γ were decreased.In conclusion,inhibition of TPSAB1 protein expression can block the proliferation of naive CD4+T cells,suggesting that TPSAB1 may be a new target for the treatment of PBC.
7.A new method to reconstruct the spatial structure of human proximal femur and establishment of the finite element model.

In current study of femoral head necrosis and femoral neck fractures, more attentions has been paid to relationship between the femoral head trabecular bone within the spatial structure and its biomechanics. In this connection, PMMA (polymethyl methacrylate), special square iron, dental base acrylic resin liquid and powder were used to embed and fix human dry femur. Then, M618 Lie Axle Rectangle Desk Plane Grinding Machine was applied to grind the femur specimen, and then air blower clean, two-dimensional cross section image was obtained by using scanner. With Mimics software reconstruction, a three-dimensional model of spatial structure of trabecular bone was obtained, and the trabecular bone three-dimensional parameters were calculated. The authors obtained clear three-dimensional model of trabecular bone, reconstructed the real anatomic morphology of proximal femur. This is a good method to research into the interior structure of femur and to provide the foundation for the three-dimensional finite element analysis.

8.Prediction of characteristic chromatogram for Abri Herba based on network pharmacology and molecular docking

Objective To predict the potential Q-markers of Abri Herba based on network pharmacology and molecular docking and establish a quality control characteristic.Methods The network relationship of"Abri Herba-component-target-pathway"was constructed by using a variety of databases and the method of network pharmacology.The potential Q-markers of Abri Herba were predicted and then the characteristic Chromatogram of Abri Herba was established by high performance liquid chromatography Results Through the network pharmacological prediction,it was found that the components of abrine hypaphorine,schaftoside in Abri Herba were closely associated with the main targets,such as AKT1,STAT3,HIF1A,GRB2,MMP9,which could act on HIF-1,PI3K-Akt,JAK-STAT and other signaling pathways and have good pharmacological activities to be potential Q-markers of Abri Herba.Then HPLC was used to establish the characteristic according to retention time.Conclusion Through network pharmacology and molecular dock-prediction combined with HPLC detection,the characteristic chromatogram was established with the components of abrine hypaphorine,schaftoside as Q-markers,which could control the quality of Abri Herba by combining the components and pharmacological activities.
9.Study on quality standard and characteristic chromatogram for Hirtula Roses

Objective To establish the quality standards of Hirtula Roses for the quality control of production, supervision, circulation and application. Methods The moisture content, total ash, ethanol extract content and characteristic chromatogram of Rosa roxburghii Tratt. were determined according to the related determination method in Ⅳ-Part of Chinese Pharmacopeia 2020. Results There should be 6 characteristic peaks in the characteristic chromatogram of Rosa roxburghii Tratt., which should correspond to the retention time of 5 characteristic peaks in the reference chromatogram of the standard herbs, and another peak should correspond to the retention time of the reference chromatogram of isoquercitrin. The moisture content of Rosa roxburghii Tratt. should be less than 14.0%; the total ash content should be less than 5.0%; the ethanol extract content should be more than 20.0%. Conclusion According to the experimental results, the quality standard of Rosa roxburghii Tratt. is conformed to the national requirements of quality standards for traditional Chinese medicine, which could provide a reference for the quality control of Hirtula Roses.
10.Quality evaluation of Abri Mollis Herba

Objective To establish a quality evaluation method for Abri Mollis Herba based on its morphological characteristics, microscopic features, and the determination of principal component contents. Methods The morphological characteristics of Abri Mollis Herba were identified by morphological authentication methods. Microscopic techniques were employed to observe the microscopic features of both the powdered form and cross-sectional tissue of Abri Mollis Herba. Additionally, a high-performance liquid chromatography (HPLC) method was developed to establish the quantify the main components, abrine and soyasaponin Bb, in Abri Mollis Herba. Results The morphological characteristics of Abri Mollis Herba were defined by numerous long pubescence on both the upper and lower surfaces of the leaflets, with indistinct veins and vein islands. The microscopic features mainly included non-glandular hairs, prismatic crystals, and crystal-sheathed fibers in the powdered form. In the root cross-section, xylem bundles, rays, vessels, and stone cells were visible. The stem cross-section displayed rays, vessels, and a hollow pith, while the leaf cross-section revealed collateral vascular bundles, vessels, and prismatic crystals. Conclusion The quality of Abri Mollis Herba could be effectively evaluated by the combination of morphological identification, microscopic authentication, and the quantification of main components abrine and soyasaponin Bb .
