2.Study of renal cortical perfusion in rabbits by microbubbles-enhanced ultrasound cavitation

ObjectiveTo explore the change of renal cortical perfusion in rabbits by microbubbles-enhanced ultrasound cavitation.MethodsTotally 20 New Zealand rabbits were randomly divided into four groups, 5 rabbits in each group. SonoVue was injected via the ear vein at a dose of 0.1 ml/kg. The renal perfusion imaging 30 s modeling was made. Under the mode of contrast enhancement, the rabbit kidneys irradiated with different mechanical indexs (MI) were 0.6 (MI 0.6 group) , 0.75 (MI 0.75 group) , 1.1 (MI 1.1 group) and 1.3 (MI 1.3 group) for 60 s, 15 min respectively. After the irradiation and conventional ultrasound contrast was made again, the renal perfusion imaging was mad. The time-intensity curves (TIC) were analyzed obtained from the following parameters: the slope rate of ascending curve (A) , the slope rate of descending curve (α) , area under the curve (AUC) , time to peak (TTP) , and derived peak intensity (DPI) . One-way ANOVA and LSD-t were used to analyze all experimental data.ResultsCompared with the MI 0.6 group, the MI 1.1 group and the MI1.3 group, the A, α were less than MI 0.6 group, the AUC, DPI, TTP were greater than MI 0.6 group, and the differences were statistically signiifcant (MI 1.1 groupvs MI 0.6 group:t=3.13, 5.31, 4.25, 4.53, 5.72, allP<0.05; MI 1.3 groupvs MI 0.6 group:t=3.67, 6.23, 4.47, 5.48, 6.86, allP<0.05). Compared with the MI 0.75 group, the MI 1.1 group and the MI 1.3 group, the α were less than MI 0.75 group, the AUC, DPI, TTP were greater than MI 0.75 group, and the differences were statistically signiifcant (MI 1.1 groupvs MI 0.75 group:t=6.02, 4.36, 4.85, 5.83, allP<0.05; MI 1.3 groupvs MI 0.75 group:t=6.19, 4.51, 5.73, 6.97, allP<0.05); the MI 1.1 group and the MI 1.3 group, the A were less than MI 0.75 group, and the difference were not statistically signiifcant. The MI 1.1 group and the MI 1.3 group, the MI 0.6 and the MI 0.75 group, and the difference were not statistically signiifcant.ConclusionsTo the change of hemodynamic of the rabbits renal cortical perfusion in the microbubbles-enhanced ultrasound cavitation contrast agent can be observed. Along with the mechanical index increased renal cortical perfusion flow stasis and the perfusion on of slowed in rabbit, may cause renal damage, the TIC curve displayed further changes.
3.Clinical Observation of Warm Acupuncture at Biguan (ST 31 ) in Treating Post-apoplectic Restless Legs Syndrome

Objective:To investigate the efficacy of warm acupuncture at Biguan(ST 31) in treating post-apoplectic restless legs syndrome. Methods:Fifty patients were randomly divided into treatment and control groups by a waiting control method in order of hospitalization. The treatment group received deep acupuncture at point Biguan(ST 31) plus moxibustion and the control group took L-dopa orally. The curative effects were compared after 28 days' treatment. Results: The total efficacy rate was 76.00% in the treatment group and 26.00% in the control group. There was a significant difference. Conclusion:Warm acupuncture at Biguan(ST 31) is effective in treating post-apoplectic restless legs syndrome.
4.Percutaneous interventional treatment for portal vein stenosis after liver transplantation

ObjectiveTo evaluate the effectiveness of the interventional treatment for portal vein stenosis in patients who had undergone liver transplantation.MethodsFromApr.2004 to Oct.2011,30 patients with portal vein stenosis after liver transplantation were referred for angiographic analysis and interventional treatment. All patients had typical clinical signs and symptoms or surveillance by imaging.After percutaneous transhepatic portography and balloon angioplasty,stents were deployed.Embolization was performed on patients with varices or portal vein flow changes.The therapeutic results were monitored by the follow-up on clinical symptoms,laboratory tests and imaging examinations.ResultsAngiography was performed successfully on all patients.Twenty-four patients received balloon dilation and 26 stents were deployed subsequently.The guide-wire cannot pass through the lesion of portal trunk in 1patient.Four patients received balloon angioplasty only.The technical success rate was 96.7% (29/30).Stainless steel coils were applied in 7 patients for varices embolization.The complication related to interventional treatment was bleeding in thoracic cavity which happened in 2 patients.Portal vein patency was maintained in all the patients who received interventional treatment for 1-72 months (mean 21.5 months).No re-stenosis was identified.ConclusionInterventional therapy is an effective method for the treatment of portal vein stenosis after liver transplantation and excellent patency can be achieved by this method.
5.Contrast-enhanced ultrasound in the assessment of renal cortical perfusion in rabbits in low temperature conditions

Objective To explore the value of contrast-enhanced ultrasound(CEUS) in the evaluation of the renal cortical perfusion in rabbits in low temperature conditions.Methods 20 New Zealand rabbits were randomly divided into five groups,4 rabbits in each group:1) with the first group as the control group (under the conditions of room temperature) ; 2)The second group was treated under-15℃--23℃ for 8hours; 3)the third group was treated under-15℃--23℃ for 4 hours; 4)the fourth group was treated under-6℃--14℃ for 8 hours; 5) the fifth group was treated under-6℃--14℃ for 4 hours.SonoVue was injected via the ear vein at a dose of 0.1 ml/kg,the renal perfusion imaging was made,observing the dynamic image of 0-3 min after the injection and analyzing the time-intensity curves(TIC)obtained from the following parameters:the slope rate of ascending curve(A),the slope rate of descending curve(α),area under the curve (AUC),time to peak (TTP),and derived peak intensity(DPI).Results Compared with the control group,the A,α of the second and the fourth group were significantly decreased (P < 0.05),while the AUC was significantly increased (P < 0.05) and the TTP was significantly prolonged(P <0.05),for the third and the fifth group,only the A was decreased (P <0.05),and the DPI in each group showed no significant difference.Compared with the third and the fourth group,the AUC of the second group was increased and the TTP was prolonged (P <0.05).Conclusions Contrast-enhanced ultrasound can reflect the earlier renal cortical hemodynamic changes in rabbits in the low-temperature conditions,and is of great value for the early diagnosis of renal damage caused by low temperature.
6.Expression of MAP4K4 in bladder cancer and its biological effect on human bladder cancer T24 cells

Objective To observe the effect of MAP4K4 targeted shRNA on biological characteristics such as proliferation,invasiveness,and apoptosis in human bladder cancer cell.Methods Differentially expressed genes was screened out through cDNA microarray analysis in 5 pairs of fresh-frozen muscle-invasive bladder cancer(MIBC) and adjacent normal tissue obtained from radical cystectomy.Combining the results of genechip and literature review,MAP4K4 was picked up for further analysis.To verify the result of microarray analysis,16 pairs of fresh muscle-invasive bladder cancer (MIBC) and adjacent tissues were assessed for the expression of MAP4K4 mRNA and protein through RT-PCR,qRT-PCR and Western-blot.T24 cell line was stably trasfected with MAP4K4 targeted shRNA and control shRNA,respectively.The effects of MAP4K4 silencing on proliferation,invasiveness and apoptosis of T24 cells transfected with MAP4K4 targeted shRNA and control shRNA were assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT),transwell and flowcytometry (FCM) assay.Results MAP4K4 was overexpressed in muscle invasive bladder cancer than in normal tissue.Down regulation of MAP4K4 expression decreased bladder cancer cell proliferation(MAP4K4-targeted versus control,P<0.001),invasiveness(MAP4K4-targeted versus control,P=0.004)and promoted cell apoptosis(MAP4K4-targeted versus control,P=0.023).Conclusions MAP4K4 is overexpressed in muscle invasive bladder cancer than in normal tissue.Down-regulation of MAP4K4 expression inhibits the invasive ability of bladder cancer.Therefore,MAP4K4 might be a potential therapeutic target for bladder cancer.
7.Expression levels and significance of semaphorin3A and matrix metalloproteinase 14 in NSCLC

Objective To investigate the correlation between the expression levels of semaphorin3A and MMP14,and their subsequent prognostic significance in nonsmall cell lung cancer(NSCLC).Methods The expression of semaphorin3A and MMP14 protein levels was analyzed in 94 cases of NSCLC tissues and in 80 cases of normal lung tissues,using immunohistochemistry(IHC).Correlation and survival analysis were used to further investigate their association and prognostic value.The correlation analysis using Pearson test,and log-rank test for survival analysis.Results The NSCLC tissues exhibited a lower expression of semaphorin3A and a higher expression of MMP14 than in the control lung tissues.The downregulation of semaphorin3A and upregulation of MMP14 may promote pleural invasion,lymph node metastasis,vascular invasion and proliferating cell nuclear antigen expression.The expression of semaphorin3A was correlated with the maximum diameter of tumor.There was a negative correlation between the protein expression levels of semaphorin3A and MMP14 in NSCLC tissues.Conclusion The data suggest that lower expression of semaphorin3A and a higher expression of MMP14 may promote occurrence and development in NSCLC and that the combined detection of semaphorin3A and MMP14 protein may be a helpful tool in predicting the prognosis of NSCLC.
8.Prediction of early bacterial infection after liver transplantation by donor complement factor component 7 polymorphism

Objective To investigate the association between donor complement factor component 7 (C7) rs6876739 gene polymorphisms and risk of early bacterial infection following orthotopic liver transplantation (OLT).Methods A total of 113 patients who had undergone OLT in Shanghai General Hospital between July 2007 and January 2011 were included.A single nucleotide polymorphism (SNP),donor C7 rs6876739 was genotyped and analyzed together with clinical data.Results We demortstrated that donor C7 rs6876739 CC genotype had higher risk of early bacterial infection than TT genotype following OLT (55.6% vs.26.5%,P =0.021).The multivariate logistic regression analysis revealed that gender (P =0.018),biliary complications (P =0.018),ICU stay after LT (P<0.001) and donor C7 rs6876739 genotype (P =0.001) were identified as independent factors of early bacterial infection.Conclusion Donor C7 rs6876739 genotype polymorphism is associated with early bacterial infection following OLT and may be a new marker of risk for the development of potentially serious bacterial infection after liver transplantation.
9.Clinicopathological significance of lymphovascular invasion in high-grade pT1 bladder cancer after transurethral resection of bladder tumor

Objective To explore the clinicopathological significance of the presence of lymphovascular invasion (LVI) in the high-grade stage pT1 bladder cancer (BC) after first transurethral resection of bladder tumor (TURBT).Methods The retrospective study was performed with 27 patients of high-grade stage pT1 BC after first TURBT from January 2006 to December 2011,and another 54 patients were matched as negative control in terms of gender,age,pathological pattern,grading and staging.All the data were calculated by using SPSS17.0 software.Qualitative variables were compared by using chi-square test.The Kaplan-Meier method was used to calculate total survival and cancer-specific survival and differences were assessed with the Log-rank statistic.Results Twenty-four patients (89%) experienced cancer recurrence in LVI group,while 19 cases (35%) experienced recurrence in control group.Progress developed in 56% (15/27) and 24% (13/54) in the 2 groups respectively.LVI was detected to be a prognostic factor for overall recurrence (x2 =20.845,P<0.001) and progress (x2 =7.887,P =0.005) in patients with high-grade T1 stage BC.Furthermore,overall survival and recurrence-free survival according to the presence or absence of LVI was 54±6 months and 84±3 months,17±5 months and 67±5 months,respectively.LVI was proved to be associated with worse overall survival (x2=13.443,P<0.001) and recurrence free survival (x2=33.094,P<0.001).Conclusion In patients with high-grade T1 stage BC,LVI in first TURBT specimens predicts disease recurrence,progression and overall recurrence and recurrence free survival.
10.Mdm2 links genotoxic stress and metabolism to p53.

Mouse double minute 2 (Mdm2) gene was isolated from a cDNA library derived from transformed mouse 3T3 cells, and was classified as an oncogene as it confers 3T3 and Rat2 cells tumorigenicity when overexpressed. It encodes a nucleocytoplasmic shuttling ubiquitin E3 ligase, with its main target being tumor suppressor p53, which is mutated in more than 50% of human primary tumors. Mdm2's oncogenic activity is mainly mediated by p53, which is activated by various stresses, especially genotoxic stress, via Atm (ataxia telangiectasia mutated) and Atr (Atm and Rad3-related). Activated p53 inhibits cell proliferation, induces apoptosis or senescence, and maintains genome integrity. Mdm2 is also a target gene of p53 transcription factor. Thus, Mdm2 and p53 form a feedback regulatory loop. External and internal cues, through multiple signaling pathways, can act on Mdm2 to regulate p53 levels and cell proliferation, death, and senescence. This review will focus on how Mdm2 is regulated under genotoxic stress, and by the Akt1-mTOR-S6K1 pathway that is activated by insulin, growth factors, amino acids, or energy status.
