1.Clinical study on intrathoracic sarcoidosis:a report of 60 cases

Objective To explore the clinical features and diagnosis of pulmonary sarcoidosis.Methods The data of 60 cases of pulmonary sarcoidosis confirmed by pathology was reviewed to determine the clinical features and the value in the diagnosis of sarcoidosis.Results The main clinical manifestations of sarcoidosis included bilateral hilar and mediastinal lymphadenopathy.The results of lung function tests showed that these patients had restrictive ventilating obstacles with decreased DLCO.The diagnosis was confirmed in 80% of the bronchial mucosa biopsies and 77.8% of the transbranchial lung biopsies through fiberotic bronchoscopy,87% of the biopsies of peripheral lymphnodes and 87.5% of the subskin nodes biopsies.Conclusion Nonspecific constitutional symptoms may occur in the cases of sarcoidosis.To the diagnosis of intrathoracic sarcoidosis,peripheral lymph nodes biopsies,the bronchical mucosa biopsies and TBLB are the convenient methods with highly definite diagnosis rate and microinvasion in clinical practice.
2.Investigation on Pollution Status of Centralized Air Conditioning-Ventilating Systems in Five-Star Grade Hotels in Beijing

Objective To investigate the actual pollution status of heating-ventilating-air conditioning(HVAC) systems in five-star grade hotels in Beijing. Methods Seven sets of centralized HVAC system from five five-star grade hotels in Beijing were selected for sampling monitoring and videotaping in situ from August 2003 to October 2003. Results There were six sets HVAC systems seriously polluted and one set was in moderately polluted according collected dust weight. The total counts of bacteria in the dust were in medium pollution grade in four sets of HVAC systems; The total quantity of fungus in the dust were in medium pollution grade in five sets of HVAC systems; The video materials showed that inside the air duct there were fungus, construction rubbish and deposit of large quantity of dust. Conclusion The HVAC systems of those five hotels were seriously polluted.
3.On-line detecting of transformer winding deformation based on parameter identification of leakage inductance

Transformers are required to demonstrate the ability to withstand short circuit currents. Over currents caused by short circuit can give rise to windings deformation. In this paper, a novel method is proposed to monitor the state of transformer windings, which is achieved through on-line detecting the leakage inductance of the windings. Specifically, the mathematical model is established for on-line identifying the leakage inductance of the windings by applying least square algorithm (LSA) to the equivalent circuit equations. The effect of measurement and model inaccuracy on the identification error is analyzed, and the corrected model is also given to decrease these adverse effect on the results. Finally, dynamic test is carried out to verify our method. The test results clearly show that our method is very accurate even under the fluctuation of load or power factor. Therefore, our method can be effectively used to on-line detect the windings deformation.

Objective To investigate the effect of calcitonin gene related peptide(CGRP) on the expression of cyclic AMP response element binding protein(CREB) mRNA in rat hippocampus and parietal cortex during focal cerebral ischemia and reperfusion(I/R). Methods Focal cerebral ischemia/reperfusion model was induced by occluding of the right middle cerebral artery using the intraluminal suture method.Hybridization in situ experiment was used to detect the expression of CREB mRNA in the ipsilateral hippocampal CA1 region and parietal cortex during different reperfusion periods.The positive product of CREB mRNA was analyzed by image analysis system. Results There was a distinct expression of CREB mRNA in right hippocampal CA1 region and parietal cortex in sham group.The absorbency of CREB mRNA positive product reduced in I/R group as compared to sham group,while it increased in CGRP group than I/R group(P
5.Study on Quality Standard of Huangdi Cataplasm

Objective To develop the quality standard for evaluating Huangdi cataplasm. Methods Thin layer chromatography (TLC) was used to qualitatively identify Astragalus membranaceus (Fisch.) Bunge,Rheum palmatum Linn,Rhizoma Chuanxiong,Angelica sinensis and Resina Draconis in Huangdi cataplasm.HPLC method was used to determine astragaloside A and loureirin B in Huangdi cataplasm. Results The Astragalus membranaceus (Fisch.) Bunge,Rheum palmatum Linn,Rhizoma Chuanxiong,Angelica sinensis and Resina Draconis were well separated by TLC without interference in the negative control.content of Astragaloside A and loureirin B showed good liner relationships with respective peak area within the range of 6.96-23.2 μg,and 0.072-0.648 μg,with r = 0.999 5,r = 0.999 9, respectively;and the average recovery was 97.18%,and 96.93%,RSD was 1.21%(n= 6),1.53% (n = 6 ), respectively. Conclusion The established qualitative and quantitative detection method is simple, specific, reproducible, accurate and reliable, which can be used for quality control of Huangdi cataplasm.
6.The Effects of Autophagy Inhibitor on Activation of Alcohol induced Hepatic Stellate Cells

Objective To observe the effect of autophagy inhibitor on the activation of alcohol induced hepatic stel-late cells, and the mechanisms thereof. Methods HSC-T6 cells were cultured in vitro and divided into four groups, includ-ing blank control group, alcohol group, 5 mmol/L 3-MA+alcohol group (low alcohol group) and 10 mmol/L 3-MA+alcohol group (high alcohol group). RT-PCR was used to detect the expression levels ofα-smooth muscle actin (α-SMA) and typeⅠcollagen. The levels of LC3Ⅱ,α-SMA and typeⅠcollagen were detected by Western blot assay. The cell viability of HSC-T6 was detected by MTT assay. Results The mRNA expressions ofα-SMA, typeⅠcollagen and the protein of expressionsα-SMA, typeⅠcollagen and LC3Ⅱwere significantly up-regulated in alcohol group compared with those of control group (P<0.05), while the expressions of those parameters were significantly down-regulated in 10 mmol/L 3-MA+alcohol group (P<0.01). The mRNA and protein levels ofα-SMA and typeⅠcollagen were significantly decreased in two 3-MA-treated groups compared with those in alcohol group (P<0.05). Meanwhile, compared with the 5 mmol/L 3-MA+alcohol group,the protein expressions ofα-SMA, typeⅠcollagen and LC3Ⅱwere significantly decreased in10 mmol/L 3-MA+alcohol group (P < 0.05 ). Compared with the alcohol group,there was significantly lower proliferation activity in all two 3-MA-treated groups (P<0.05). Conclusion 3-MA can inhibit the protein expression of LC3Ⅱ,α-SMA and typeⅠcollagen induced by alcohol in HSC-T6 cells, and inhibit the proliferation of HSC cells.
7.Berberine regulates gut microbiota and Th17/Treg balance in sleep deprived rats

Objective To investigate the effect of berberine on gut microbiota and T helper cell 17 (Th17), regulatory T cell (Treg) cell in sleep deprived rats.Methods SD rats were randomized into control group, model group, low-dose and high-dose group (BBR1 and BBR2, 100 mg/kg and 200 mg/kg, administrated orally).Sleep deprived rat model was established by the small-platforms-over-water method.The number of bacteria in rectum content of rat was detected.The ratio of Th17/Treg was evaluated by flow cytometry.The expression of interleukin 17 (IL-17), RAR-related orphan receptor (ROR) C, and Forkhead Box Protein P3 (Foxp3) mRNA was evaluated.Results The counting of Clostridium perfringens was elevated (P<0.05), the amount of other microbiota decreased (P<0.05), Th17/Treg ratio(P<0.05), IL-17 and RORC expression enhanced (P<0.05), Foxp3 expression decreased (P<0.05) in the gut of model rats.In contrast, treatment with berberine inhibited the proliferation of Clostridium perfringens(P<0.05), and promoted the growth of other microbiota (P<0.05), and dampened Th17/Treg ratio (P<0.05), regressed IL-17 and RORC expression, augmented Foxp3 expression.Conclusions Various doses of berberine can counteract gut microbiota imbalance and Th17/Treg imbalance induced by sleep deprivation.
8.Interventional therapy of deep venous thrombosis of lower limb after burn: initial experience in 13 cases

Objective To discuss the curative effect and specificity of interventional therapy for deep venous thrombosis (DVT) of lower limb in burned patients.Methods The clinical data of 13 patients with lower limb DVT after burn,including 7 males and 6 females with a median age of 46.1 years (37-67 years),were retrospectively analyzed.The causes of burn included flame burn (n=9),electric injury (n=2) and hydrothermal bum (n=2).The burned area was 1%-88% of the total body surface,with a mean of (37.08± 30.60) %.Lower limb DVT complicated by lower limb bum was observed in 11 patients,among them bum of both lower limbs was seen in 8 patients.Lower limb DVT associated with inhalation injury was found in 5 patients.Clinically,lower limb DVT was usually detected in 13-72 days after burn,with a mean of (38.69± 16.83) days.Interventional treatment was carried out in all 13 patients,and the curative effect was assessed.Results Technical success of interventional treatment was obtained in all 13 patients.Both inferior vena cava filter placement via right internal jugular vein approach (n=3) or via unaffected-side femoral vein approach (n=10) and anticoagulant therapy were conducted.Catheter-directed thrombolysis was employed in 7 patients,intravenous thrombolysis was adopted in 4 patients,and no thrombolysis therapy was used in 2 patients.No pulmonary embolism occurred.The curative effect rate of interventional treatment was 84.6%(11/ 13).Conclusion For the treatment of lower limb DVT after burn,interventional therapy is safe and reliable,but the selection of puncture site and the use or not use of indwelling catheter for thrombolysis should be carefully taken into consideration.
9.Effect of penehycUdine hydrochloride on iNOS expression in lung in septic mice

Objective To investigate the effect of penehyclidine hydrochloride (PHC) on iNOS expression in lung in septic mice.Methods Female KM mice were subjected to cecal ligation and puncture (CLP,a model of polymicrobial sepsis) or shame operation.Thirty-six KM mice weighing 20~25 g were randomly assigned into three groups:shame CLP group,CLP group and PHC group.Mice in PHC group were given PHC 0.45 mg/kg intraperitoneally at 1 h before CLP. Six hours after CLP,the lungs of each group were sampled for light and electronic microscopy.The expression of iNOS mRNA in lung tissue were detected by in situ hybridization.The survival rates were observed at 24 h after the operation. Results The CLP group was observed thickened alveolar septa,as well as mitochondrial cristae swelling and mitochondrial vacuolation under electronic microscope.Emptied lamellar bodies could be also found.Histology of lung in PHC group had little changed.Expression of iNOS in lung in PHC group was significantly lower than that of the CLP group.At 24 h after CLP challenge,70.0% of the PHC mice lived,remarkably increased compared with that of the CLP group (26.7%),P<0.05.Conclusion PHC had effective effect for increasing the survival rates of septic mice,inhibiting the expression of iNOS and reducing the severity of lung injury.
10.MORPHOMETRIC ANALYSIS ON THE MITOCHONDRIA OF MYOCARDIUM IN EXERCISE TRAINED RATS

Adult male rats were divided at random into two groups and trained on the treadmill running schedules, 60 min/day and 10min/day for group A and B separately. Four weeks later the ultrastructure of the left ventricular myocardium was examined and the specific surface of mitochondria wasmeasured. Comparing to the corresponding control group, the specific surface of mitochondria in the two groups of trained rats was decreased significantly. (5.883+0.12 um and 6.050?0.12 um to 6.564?0.17 um). This means myocardium mitochondria in trained rats were swelling. The reason might be that the myocardium injury was induced by exercise training.
