1.The effects of erythrocyte CD35 on the immuno-response of lymphocyte activated by antigen

Objective To investigate the role of CD35 on erythrocytes played in antigen activating immunological reaction of lymphocytes. Methods Suspensions of lymphocytes(1?10~6/ml)and erythrocytes (1?10~8/ml) were respectively separated from anticoaguted whole blood of healthy adults with the lymphocyte separation medium. Using inactivated ascites carcinoma cell S180(1?10~6/ml) as activation antigen and autologous plasma as reactive medium, the role of erythrocytes in regulating the anti-neoplasm immunological reaction of lymphocytes was appraised. The expression of CD25 on lymphocytes was detected and compared using flow cytometry assay with CD35 on erythrocytes blocked by monoclonal antibody or complements in plasma were destroyed by EDTA. Objective To study the changes in the expressions of CXCR4 of leucocytes as modulated by erythrocytes from cord blood. Methods 0.2ml suspension of whole blood cells (or 0.2ml suspension of leucocytes) was added to 0.3ml of auto plasma, and 0.2ml suspension of S180 (5?10~6/ml) was added as stimuli. The suspension was incubated at 37 ℃ for 1 hour. Normal saline instead of S180 was used as control The expression changes in CD35 on erythrocytes and CXCR4 on leucocytes were determined with flow cytometry assay. The data could be grouped under four heads: (1) cancer cells 0.2ml were added to whole blood cells 0.2ml and plasma 0.3ml; (2) NS 0.2ml were added to whole blood cells 0.2ml and plasma 0.3ml; (3) cancer cells 0.2ml were added to white blood cells 0.2ml and plasma 0.3ml; (4) NS 0.2ml were added to white blood cells and plasma 0.3ml. Results When activated by S180, the expression of CD35 on cord erythrocytes was decline not so low as that of adult′; while the expression of CXCR4 on cord leucocytes was increased not so high as that of adults′. When activated by S180, the expression of CD35 on adult erythrocytes was significantly decreased than that of group of not activating (40.21%?11.52% and 28.65%?8.08% respectively), while the expression of CXCR4 on adult erythrocytes was significantly increased (40.62%?17.89% and 64.18%?11.69% respectively, P
3.Expanded and prefabricated forehead skin flap with grafted mucosa liner for eyelids reconstruction

Objective To explore a new method for reconstruction o f full-thickness defect of eyelid. Methods The composed skin flap which was lined the expanded forehead skin flap with oral mucosa were transferr ed to the defect of eyelid and then sutured anatomically to the eyelid skin. Fou r months later, the composed flap was divided to reconstruct upper and lower eye lids and put an artificial eye into it. Results The appearance and function of the eyelid was partly recovered. Conclusion The reconstruction of full-thickness eyelid defect with expanded and prefabricated skin flap with grafted mucosal liner is better and reliable.

5.Clinical observation of collagen sponge for reducing volume of drainage after surgery for lumbar spinal stenosis

ObjectiveTo observe the effectiveness of collagen sponge for reducing volume of drainage after surgery for lumbar spinal stenosis. MethodsOne hundred and eighty-six patients who suffered from lumbar spinal stenosis were divided into two groups by random digits table method. The test group(96 cases) used collagen sponge to cover dura mater before placing drainage tube,the control group (90 cases) was treated without collagen sponge. The volume of drainage at 1,12,24 h after surgery were observed, the blood routine test was carried out at before and 48 h after surgery and the volume and ratio of blood transfusion after surgery was also measured and compared between the two groups. ResultsThe volume of drainage were significantly decreased in the test group compared with the control group at 1,12,24h after surgery [( 106.11 ± 20.02 ) ml vs. ( 127.02 ± 25.09) ml, (236.12 ± 34.06) ml vs. (327.31 ± 51.21 )ml, (355.16 ± 49.03 ) ml vs.( 506.36 ± 85.29 ) ml](P < 0.05 ). The volume and the ratio of blood transfusion in the test group were ( 176.27 ± 21.37) ml and 10.42%(10/96) ,which were greatly lower than those in the control group[(445.94 ±24.56) ml and 32.22% (29/90)](P <0.05). The number of RBC and the concentration of Hb were (2.96 ± 0.45 ) × 1012/L and ( 106.75 ± 7.30) g/L, differently in the test group at 48 h after surgery,which were increased significantly compared with the control group[(2.35 + 0.57) × 1012/Land (90.45 ± 5.10) g/L](P < 0.05 ). ConclusionsCollagen sponge provides rapid ,effective and durable hemostasis and decreases the leak of cerebrospinal fluid after surgery for lumbar spinal stenosis. It can be used as an effective and economic method to reduce the volume of drainage after surgery.

7.Correlation between Leukoencephalopathy and Atherosclerosis

Objective To investigate the correlation between leukoencephalopathy and atherosclerosis. Methods 238 patients were enrolled. All the patients were underwent brain MRI and carotid and femoral artery duplex ultrasonography after hospitalized. The atherosclerotic plaque, intima-media thickness (IMT), peak systolic velocity (Vp) were measured with ultrasonography. The data was analyzed with SPSS 13.0 software. Results The leukoencephalopathy of the patients positively correlated with age, hypertension, serious of atherosclerosis and carotid IMT, negatively correlated with carotid Vp, but did not correlated with vertebrarterial Vp. Conclusion The leukoencephalopathy positively correlate with some atherosclerotic risk factors, the serious of atherosclerosis, and anterior circulation disorder, but do not correlate with posterior circulation.

This paper discussed anti blood group antibody and red blood cell innate immunological function changes in gravidas with various blood groups. The 1∶64 positive rate of anti blood group antibody and innate immunological adhesive ratio of CR1 on red blood cell of gravida were assayed. 10.Effects of FOLFOX regimen chemotherapy on immunity of patients with colorectal cancer

Background and purpose: Anti-tumor chemotherapy compromises normal immune function of the patients. There were many reports that chemotherapy for advanced colorectal cancer often inhibit the cellular immune function. The effect of FOLFOX regimen chemotherapy on immunity of the patients with colorectal cancer before and after therapy was studied, and healthy people were used as a control. Methods: Eighty colorectal cancer patients were treated by FOLFOX regimen, which consisted of 2-hour infusion of oxaliplatin(85 mg/m~2) and 2-hour infusion of leucovorin (CF)(200 mg/m~2) on Day 1, followed by 5-fluorouracil(5-FU) bolus (400 mg/m~2) on Day 1 and 46-hour infusion (2 400 mg/m~2). FOLFOX regimen was repeated at 2-week intervals. Two treatments of the above regimen were defined as one cycle. Flow cytometry was used to detect T lymphocyte subsets and NK cells in blood samples from patients with colorectal cancer before and after therapy. Data obtained fi'om healthy people was used as control. Results: CD3~+, CD4~+ T cells, NK cells and CD4~+/CD8~+ ratio in blood samples were not significant before and after chemotherapy in first day, second week and fourth week(P0.05). Lower CD3~+, CD4~+ T cells, NK cells and CD4~+/CD8~+ ratio were detected in blood samples from cancer group than that from the healthy control(P<0.05). CD8~+ T lymphocyte were reverse. This change was related to the TNM pathological stage. Conclusion: FOLFOX regimen was effective for patients with coloreetal cancer, which can improve patients' life quality and did not impact on the immune function of the patients. The immune function of lymphocytes in peripheral blood of the patients with colorectal cancer was low, and even worse in the patients with late TNM stage. It is valuable for estimating the function of cell immune of the patients, patients prognosis and the role of immune therapy in the treatment of the patients by detecting T lymphocyte subset and NK cell.
