1.Analysis of basic research in endocrine and metabolic projects granted by National Natural Science Foundation of China

ObjectiveTo investigate the effect of National Natural Science Foundation ( NSFC ) on the progress of the discipline of endocrine and metabolic research from 1987 to 2010.MethodsThe data regarding the NSFC allocated to endocrine and metabolic research from 1987 to 2010 were collected.Total expenses and numbers of the majority of programs,unit distribution,times of funding and the situation of completed program finished in recent two years were provided.ResultsFrom 1987 to 2010,a total of 731 projects and 178 398 thousands Yuan expenses of NSFC were allocated to endocrine and metabolic research.The detailed allocations are as follows:general program ( n =462 ),Young Scientists Fund ( n =187 ),regional fund ( n =28 ),Key Program ( n =9 ),National Science Fund for Distinguished Young Scholars ( n =5 ),Joint Research Fund for Overseas Chinese Young Scholars( n =2 ),Fund for Creative Research Groups ( n =1 ),International ( regional )joint research program ( n =11 ),and the others ( n =26 ).Taking the projects ( n =102 ) completed in 2009 and 2010 as an example,279 papers were published in Science Citation Index ( SCI ) included journals and 236 papers were published in Chinese journals.During the time of the projects completed,8 post-doctoral students,169 students for PhD degree,and 227 students for Master degree have been graduated.ConclusionOver the past 25 years,the continuously increased funding of NSFC on endowrine and metabolic research has led to substantial achievement.The grants of talent training and research program have increased dramatically,and the units of funding increased yearly.Talent training and subject-specific development have increased greatly.

3.Effect of miR-27a on cell proliferation and apoptosis in human melanoma cell line WM239

Objective To investigate the effect of miR-27a mimic and inhibitor on proliferation and apoptosis in melanoma cell line WM239. Methods The miR-27a mimic,inhibitor and its negative control were transfected into WM239 cells. The transfection efficiency was evaluated by fluorescence microscope. The expres-sion of miR-27a was detected by real-time fluorescent quantitative PCR. The proliferation of cells was detected by MTT. The cell apoptosis and cell cycle were analyzed by flow cytometry. Results The transfection efficiency in WM239 cells was 80% to 90%. The expression of miR-27a was markedly up-regulated in miR-27a mimic group (2-△△CT value is 26. 98 ± 0. 01),with statistically significant difference(t= -1 123. 67,P=0. 00);and the miR-27a inhibitor group showed lower expression of miR-27a(2-△△CT value is 0. 96 ± 0. 02),there was no statisti-cally significant difference compared with normal control group(t=0. 04,P=0. 06). Conclusion MiR-27a can suppress melanoma cell proliferation and act as a tumor suppressor gene,which is rel-evant to induce cell apoptosis and block cell cycle in G0-G1 phase.
4.The Interventional Sclerotherapy of Renal Cysts Guided by CT

Objective To compare the results of single and repeated percutaneous sclerotherapy in patients with simple renal cysts .Methods 96 patients with simple renal cysts underwent needle aspiration and sclerotherapy under CT guidance. 45 patients (group A) underwent one session of sclerotherapy with 99.7% ethanol immediately after aspiration and 51 patients (group B) underwent repeated sclerotherapy at least twice. The patients were followed up using ultrasonography or CT at 3~6 months intervals. The complete or almost disappearance of the renal cyst was considered a successful treatment.Results The efficacy of treatment was significantly different in two group . The overall effective rate were 91.1% and 98.8% in group A and group B , respectively. The rate of complete regression was significantly better in group B (80.4%) than in group A (62.2%,?
5.Chemokine receptor 4 and tumors

Chemokine receptor 4 (CXCR4) belongs to G protein-coupled receptor superfamily.It can induce immune cells-directed chemotaxis,thus keeping their homeostasis.CXCR4 expresses on a variety of tissues and cells.In different tumors and at different stages of tumors,CXCR4 expression is significantly higher than that in normal tissues.CXCR4 plays an important role in tumor progression since it is involved in tumor cell proliferation,adhesion,invasion and metasta.
6.The study of distinguishing solid solitary pulmonary nodule with diffusion weighted imaging

Objective To evaluate the value of diffusion weighted imaging (DWI)in distinguishing the solid solitary pulmonary nodule (SPN).Methods 42 patients with SPN (malignant in 25 and benign in 1 7)who were confirmed by operation,biopsy or follow up after treatment underwent routine chest T1 WI,T2 WI and DWI.The b values were chosen as 300,500,800 and 1 000 s/mm2 ,and the corresponding apparent diffusion coefficient (ADC)values and the signal intensity (SI)were respectively measured.Results The ADC values and SI of benign and malignant SPNs were gradually reduced with increasing b value.The ADC value between benign and malignant SPNs was statistically significant with b value of 500 s/mm2 (P 500 =0.03 <0.05 ),meanwhile the SI was statistically significant with b values from 300 to 1000 s/mm2 (P 300 <0.001,P 500 =0.03 <0.05,P 800 =0.01 <0.05, P 1 000 =0.02<0.05).Conclusion Both SI and ADC value of DWI play important role in distinguishing benign and malignant SPNs, and the diagnostic efficiency of SI is superior to ADC value.
7.Salmonella virulence gene analysis in poisoning food,slaughtering and commercial samples

We investigated the carrying status of the virulence genes of Salmonella from different sources in Shijiazhuang City,China,to provide the basic data for the further risk assessment of Salmonella.A total of 186 isolates of Salmonella from different sources were collected and identified serotypes in the area of Shijiazhuang from 2011 to 2016.PCR was performed for eight virulence genes (invA,sopE,agfA,spvR,hilA,stn,pefA,shdA).These Salmonella bacteria were detected in 13 kinds of serotypes.Enteritidis is a significant advantage of the group.The above 8 virulence genes were analyzed,and the virulence genes hilA,stn and invA were the most frequently carried,their respective carrying rate were 90.3% (168/186),86.6% (161/186) and 82.8% (154/186) respectively.We found the virulence genes of Salmonella from different sources were different.It is necessary to take measures to strengthen the food hygiene supervision and prevention and control of the storage and sale of raw poultry stalls in the morning market in Shijiazhuang area.
8.Analysis of the characteristics of Salmonella enteritidis virulence genes from different origins

Objective To investigate the differences and characteristics of virulence genes carried by Salmonella enteritidis from different sources in Shijiazhuang City.Methods One hundred and twenty-four strains of Salmonella enteritidis isolated from morning markets of raw and poultry stalls,slaughterhouses and food poisoning specimens in Shijiazhuang area were collected.Eight virulence genes (invA,sopE,agfA,spvR,hilA,stn,pefA,shdA) were detected by polymerase chain reaction (PCR).Results Salmonella enteritidis might have different virulence gene profiles.The above eight virulence genes were detected in different strains.The carrying rate of virulence genes invA,sopE,stn,hilA,spvR and pefA in the food poisoning strains was higher than 94%.There was no difference in the carrying rate of 8 virulence gene between the morning raw poultry stalls isolates and the patient strains,but was different with the slaughterhouse strains.Conclusion There were more risks of food poisoning caused by Salmonella enteritidis from morning markets,and the hygiene supervision should be strengthened to prevent and control foodborne disease.
9.Application of randomized blind sample test in the external quality assessment schemes for clinical hematologic examination laboratories

Objective To improve the quality of the clinical hematologic examination laboratories in national free preconception health examination project by using randomized blind sample test in the external quality assessment (EQA ) schemes .Methods Blind samples for clinical hematologic examination were prepared as higher ,middle ,lower three levels .Samples were dispensed in u‐nified way which included 4 times conventional EQA and in random way which included 1 time blind sample test .Samples will be tested by Clinical hematologic examination laboratories in national free preconception health examination project .The feedback re‐sults were summarized and analyzed by EQA organizer .Results In 4 times of conventional EQA ,the rates of accepted score of 134 laboratories were 72 .4% ,97 .8% ,97 .0% and 98 .5% respectively .The rates of accepted score in last three times were statistically significant higher than that in the first time(P<0 .05) .However ,the rates of accepted score (84 .3% ) in randomized blind sample test were significant lower than that(97 .0% ) in conventional EQA which was conducted at the same time(P<0 .05) .Conclusion The use of randomized blind sample test may help the EQA organizer to find the problems in laboratories participated EQA and find effective way to improve the quality of the laboratories .
