1.THE EXPRESSION IN SITU OF TELOMERASE RNA AND THE PROGNOSIS OF BLADDER TRANSITIONAL CELL CARCINOMA

To study the expression of human telomerase RNA(hTR) in different grades of bladder transitional cell carcinoma and its relation to its prognosis. With in situ hybridization, the expression of hTR was observed in 67 cases of bladder transitional cell carcinoma specimens, to be compared with specimens of 10 normal bladder tissue and 10 benign lesions. The RNA probe used in hybridization was telomerase reverse transcript (TERT), and the marker was digoxin. There was a significent correlation between the expression of hTR and grading and prognosis. hTR is considered as an important and independent prognostic factor in this carcinoma.
2.Percutaneous balloon catheter occlusion technique for tumor thrombectomy in renal or adrenal neoplasm extending into the inferior vena cava

Objective To assess the value of percutaneous balloon catheter occlusion technique for tumor thrombectomy in renal or adrenal neoplasm extending into the inferior vena cava(IVC). Methods Twelve patients with renal or adrenal neoplasm extending into retrohepatic or subhepatic IVC were diagnosed by the investigations of CT, MR1, and Doppler ultrasound. There were 7 men and 5 women with a mean age of 51 years (range, 20 to 76 years). Of these patients, 11 had renal ne-oplasm and 1 had adrenal tumor; 11 in the right and 1 in the left; 11 with retrohepatic caval thrombus (level 2a) extension and 1 with extension into the subhepatic vena cava (level 1). On the operation day, all patients had a pereutaneous preset of the balloon catheter into the IVC, at the position be-tween hepatic vein and the tip of tumor thrombus. During surgical resection, the balloon was filled via the catheter to occlude the IVC prior to vena cavotomy. Results Radical nephrectomy and resection of vena cava thrombus was successfully performed with this technique on 12 patients. There were no operative or perioperative pulmonary embolisms or deaths, no any complication. The mean postopera-tive hospital stay was 12 d (range, 9 to 15 d). Pathological investigation revealed clear cell carcinoma in 6 patients, papillary renal cell carcinoma (RCC) in 3, metastatic hepatocellular carcinoma in 1, an-gioleiomyolipoma in 1 and adrenal leiomyosarcoma in 1. The pTNM stage in 9 patients with RCC was T3b N0 M0 in 8 patients, T3b Nx M1 in I patient. The mean followup was 21±10 months (median follow-up 24 months). Four patients died of lung or liver metastasis, and the recurrence of liver cancer at 6, 9, 15, and 22 months postoperatively. Eight patients have survived for 6 to 35 months (mean 26 months). The postoperative 1-, and 3-year cancer-specific survival rates in 9 patients with RCC were 78% and 67%. Conclusions Percutaneous balloon catheter occlusion technique is a feasible, safe, and effective method for tumor thrombectomy of low retrohepatic (level 2a) or subhepatic (level 1) type in patients with renal or adrenal neoplasm extending into the IVC. Additional experience and con-tinued investigation are warranted.
3.Application of grey model (1, 1)-linear regression coupling model in the prediction of regional nursing human resource

Objective Taking the prediction of the number of nurses per thousand cases as an example, to discuss the application of grey model GM(1, 1)-linear regression Coupling Model in the prediction of regional nursing human resource, so as to provide methodology reference for health human resource prediction.Methods The grey model GM (1, 1) was built up by EXCEL formula to predict the influencing factors of the number of nurses per thousand people. The predictive value of the influencing factors were substituted into the regression equation for the annual prediction of regional nursing human resource. Results The fitting error between predicted value and actual value was small when using the grey model GM (1, 1)-linear regression coupling model in the prediction of regional nursing human resource. The accuracy level of this coupling model prediction is excellent, and the prediction result can be used as a reference for target year. Conclusions The coupling model not only makes up for the lack of 1inear factors in the grey system model, but also improves the defect that exponential growth cannot be expressed in the linear regression prediction model, so the construction of the coupling model is reasonable and feasible.
4.mA Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2.

N-methyladenosine (mA), catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14, is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However, the roles and precise mechanisms of mA modification in regulating neuronal development and adult neurogenesis remain unclear. Here, we examined the function of Mettl3, the key component of the complex, in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced mA levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neuronal development and skewed the differentiation of aNSCs more toward glial lineage, but also affected the morphological maturation of newborn neurons in the adult brain. mA immunoprecipitation combined with deep sequencing (MeRIP-seq) revealed that mA was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically, mA was present on the transcripts of histone methyltransferase Ezh2, and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. Collectively, our results uncover a crosstalk between RNA and histone modifications and indicate that Mettl3-mediated mA modification plays an important role in regulating neurogenesis and neuronal development through modulating Ezh2.
