1.Role of bone marrow cells in renewal of renal tubular epithelial cells under physiological condition
Academic Journal of Second Military Medical University 1985;0(06):-
Objective:To observe whether bone marrow cells (BMCs) can differentiate into the renal tubular epithelial cells under physiological condition. Methods: Transgenic C57BL/6 mice labeled with green flouresent protein(GFP) served as bone marrow donors and C57BL/6 mice without fluorescence labeling were divided into 3 groups: normal group(N,n=15),total body irradiation group(TBI,n=15),and bone marrow transplantation(BMT,n=15) group. The mice in Group N received no treatment, those in Group TBI were irradiated with lethal dose of ? rays but received no transplant(sacrificed within 1 week after irradiation),and those in Group BMT were transplanted with BMCs after the irradiation(sacrificed at 58 and 84 days after transplantation).Blood samples were obtained from all 3 groups for routine analysis and analysis of urea nitrogen and plasma creatinine.Meanwhile, mice kidneys were harvested for H-E staining. The amount of GFP positive BMCs in mice of BMT group was determined by flow cytometry;the distribution and amount of GFP positive cells in renal tissues were observed by immunohistochemistry. Results: There was no difference in the levels of urea nitrogen and plasma creatinine between the 3 groups.Lethal-dose total body irradiation resulted in no significant change in renal histological structure and function, but RBC,WBC and PLT of the mice in TBI group were markedly reduced compared with those in the other 2 groups(P0.05).Conclusion: Bone marrow cells may participate in renewal of tubular epithelial cells even without the renal tubular damage.

The arteries of the spinal cord on surface have been observed in 62 Chinese adults and 39 children.Anteriorly, the spinal cord was divided into 3 areas, according to the arterial supply. The superior cervical area(segments of C_(1~3) or C_(1~4))was mainly supplied by the anterior spinal arteries; the cervical enlargement area (segments of C_4 or C_5 to T_5 or T_6) was mainly supplied by the arteria intumescentia cervicalis and the lumbar enlargement area (below the segments of T_6 or T_7) was mainly supplied by the arteria intumescentia lumbalis. In the intermediate places between these areas (segments of C_(3~4) and T_(5~6)), the anterior median artery was often narrow or interrupted. Usually the anterior spinal artery united with its fellow of the opposite side to form a single trunk (46.53%). The anterior radicular arteries averaged 5.2 (2~10) in number. The arteria intumescentia cervicalis frequently existed at C_7 and C_8 (C_5~T_3). The arteria intumescentia lumbalis mostly located at T_9 (T_6~L_2), and 72.28% of them being on the left side.Posteriorly, the arteries developed well on the two enlargements. The posterior spinal arteries arised frequently from the vertebral artery (76.41%). The posterior radicular arteries averaged 9.3 (5~21) in number. The authors have raised an objection to the view that the segments of T_4 and L_1 of spinal cord are poor supplied with blood.

ObjectiveTo explore the cause and treatment of multi-drug resistant pulmonary tuberculosis (MDR-TB) in patients with human immunodeficiency virus (HIV) infection. Methods24 cases with HIV and MDR-TB who were found in some AIDS-high areas in Henan were analyzed.ResultsAll cases were later-required drug-resistant ones. The main reasons were unreasonable plan for chemotherathy, improper management to the patients, side-effect of drugs, immunosuppression and low drug-tolerance. After anti-virus and 9~12 months long anti-tuberculosis chemotherathy,7 cases died and other 17 survival patients improved in clinical symptoms and signs, including no tubercle bacillus were found in sputum of 15 cases, the focal of infection were absorbed in 16 cases, the pulmonary cavity closure rate was 66.7%(4/6)and the count of CD4+ lymphocyte was higher than before.ConclusionThe patients with HIV and MDR-TB are difficult to cure and the mortality rate is high;the short-term effect of the anti-virus and multi-drug antituberculosis chemotherathy is better.

Objective To investigate the expression of microRNA (miRNA) in placenta tissue, explore the function of miRNA in pathogenesis of gestational diabetes mellitus (GDM). Methods Placenta tissue from pregnant women with GDM and normal controls were selected from January 2013 to January 2014. Expression of miRNAs were detected by sequencing technique and quantity real time PCR(qRT-PCR). Target genes of miRNA were analyzed by KEGG. Results 52 insulin signaling pathway related miRNAs including 47 up-regulated and 5 down-regulated were differentially expressed in GDM compared with normal control. Expression of placental hsa-miR-548am-5p and hsa-miR-95-5p were significantly increased , but hsa-miR-1246 was significantly lower (P <0.05) in GDM group than normal control; all the results were consistent with sequencing results. Key points of insulin signaling pathway could regulate expression of targets such as PI3K. Conclusion miRNA expression in GDM placenta was abnormal.A variety of exp ressions of miRNAs were involved in the pathogenesis of GDM by regulating insulin signaling pathway.

The fabrication system for the electrochemical microfluidic device was set up based on the pulse driving and controlling of microfluids technology. The nano silver ink and glycerol solution were jetted on the glass substrates to form the microelectrode pattern and the liquid mold pattern for the microchannel. Then the microelectrode and microchannel were obtained through a sintering process and a molding process, respectively. The electrochemical mircrofluidic device was Finally prepared through a bonding process with the microelectrode and the microchannel. The influences of the system parameters on the formation of the droplet were studied, as well as the influences of the droplets diameter and the overlap on the formation of the liquid lines. The minimal width, the thickness and the resistance of the prepared microelectrode were 45 μm, 2.2 μm and 5.2 μΩ cm, respectively. The minimal width of the microelectrode was 35 μm and the surface was smooth. The electrochemical flow detection of glucose concentration was carried out with the device, and the results showed that the glucose concentration had a high linear correlation with the response current, which could be used in the quantitative detection of glucose concentration. The fabrication of the electrochemical microfluidic device based on the pulse driving and controlling of micro fluids technology has many advantages such as simple system structure, lower cost and higher accuracy of the micro droplet and can be used in the preparation of the devices in the biochemical analysis and biosensor areas.

Objective To study the relationship between cardiovascular diseases (CVD)and 24-h peritoneal protein losses (PPL) in continuous ambulatory peritoneal dialysis (CAPD)patients. Methods One hundred and seventy-eight CAPD patients in our department were enrolled in this study. Their 24-h PPL was measured and other clinical data were recorded at the beginning. Meanwhile, Doppler ultrasound examination was performed. They were then followed-up prospectively for the development of CVD. Results The average of 24-h PPL was (5.0±1.8) g.Patients with diabetic status or preexisting CVD or carotid arteries arteriosclerosis had higher 24-h PPL than those without (t=2.082, P=0.039; t=2.601, P=0.010; t=2.217, P=0.029). 24-h PPL was positively correlated with left ventricular end-diastolic diameter (LVDd), interventricular septal thickness (IVSTd), posterior wall diameter of left ventricle at end-diastolic (LVPWd) and left ventricular mass index (LVMI) (r=0.222, P=0.040; r=0.217, P=0.043; r=0.339, P=0.002; r=0.305, P=0.007). It was negatively correlated with ejection fraction of left ventricle (r=0.221, P=0.040). One hundred and fourteen CAPD patients were prospectively followed-up for at least twelve months. Patients developing CVD were 40.4% and 19.3% for high and low PPL groups respectively (x2=6.035, P=0.014). In the multivariable logistic regression analysis, the 24-h PPL was one of the independent factors for developing CVD. Conclusions There is a significant and independent relationship between 24-h PPL and new cardiovascular events. 24-h PPL may be an important predictor of cardiovascular disease.
