OBJECTIVE To investigate the role of brain-derived neurotrophic factor(BDNF) and its receptor TrkB in the development of allergic rhinitis(AR). METHODS SD rats were randomly divided into allergic rhinitis group(AR group, n=20) and control group (Con group, n=10) . A model of allergic rhinitis in health SD rat was established by using ovalbumin intraperitoneal immunization and nasal antigen challenge. The nasal mucosa obtained from 18 out of 20 AR models as well as 10 normal controls were studied with immunohistochemistry (Envision System) method to observe the expression and distribution of BDNF and TrkB. RESULTS Immunohistochemical staining showed that the distribution of BDNF and TrkB in nasal mucosa from normal rats was in accordance with that in the nasal mucosa from rats with allergic rhinitis on the whole. The study showed that immunoreactivity to BDNF (or to TrkB) was present both on the surface and in the cytoplasm of epithelial cells, glandular cells and their duct cells. The statistical analysis of imumunohistochemical staining showed that the IOD of BDNF in rat nasal mucosa with allergic rhinitis (77344.17?8567.02) was obviously higher than that in the normal rat nasal mucosa (54124.82? 18515). There was a statistical significance between the two groups (t=3.053, P

OBJECTIVE:
To evaluate the role of brain-derived neurotrophic factor (BDNF) in allergic rhinitis of rat.
METHOD:
Thirty SD rats free of disease were randomly divided into two groups. A model of allergic rhinitis rat was established by using ovalbumin intraperitoneal immunization and intranasal antigen challenge. The nasal mucosa from 18 out of 20 AR models as well as 10 normal controls were studied for expression of BDNF mRNA by reverse transcription-polymerase chain reaction (RT-PCR).
RESULT:
BDNF/beta-actin ratio in AR models was significantly higher than control (0.49+/-0.07 vs 0.28+/-0.01, P<0.05).
CONCLUSION
BDNF played an important role in the development of allergic rhinitis of rat.

Objective:To investigate the value of serum miR-145 and matrixmetallo proteinase-2 (MMP-2) levels in predicting pathologic complete response (pCR) after neoadjuvant chemotherapy in triple-negative breast cancer.Methods:125 patients with triple-negative breast cancer who received neoadjuvant chemotherapy in the Hospital from Jan. 2022 to Dec. 2023 were prospectively included as the study objects, and 130 healthy people matching the age of the case group who underwent physical examination in our hospital during the same period were included as the healthy control group. Real-time fluorescence quantitative polymerase chain reaction was used to detect the serum miR-145 level of all subjects. Serum MMP-2 levels were determined by enzyme-linked immunosorbent assay (ELISA). After neoadjuvant chemotherapy, patients were evaluated according to Miller-Payne (MP) grading criteria and divided into pCR group and non-PCR group. Results:The serum miR-145 level in patients with tertiary breast cancer was 1.49±0.27, which was significantly lower than that in healthy control group (2.79±0.49), with statistical significance ( t=20.33, P<0.001). The serum MMP-2 level in triple negative breast cancer patients was (153.07±38.36) ng/mL, which was significantly higher than that in healthy control group (84.38±12.63) ng/mL, and the difference was statistically significant ( t=26.13, P<0.001). After neoadjuvant chemotherapy, the serum miR-145 level in non-PCR patients before treatment was 1.36±0.21, which was significantly lower than that in pCR group (1.74±0.20), with statistical significance ( t=9.93, P<0.001). After neoadjuvant chemotherapy, the serum MMP-2 level in non-PCR patients before treatment was (169.57±30.45) ng/mL, which was significantly higher than that in pCR group (121.61±31.79) ng/mL, and the difference was statistically significant ( t=8.24, P<0.001). Pearson correlation analysis showed that there was a significant negative correlation between serum miR-145 and MMP-2 levels in patients with triple-negative breast cancer before treatment ( r=-0.47, P<0.001). ROC curve analysis results showed that serum miR-145 and MMP-2 levels before treatment predicted the sensitivity and specificity of pCR after neoadjuvant chemotherapy for triple-negative breast cancer were 82.9% and 74.4%, 45.1% and 99.8%, respectively. The sensitivity and specificity of pCR after neoadjuvant chemotherapy for triple-negative breast cancer were 84.1% and 88.4%, respectively. Conclusion:Serum miR-145 and MMP-2 levels are related to the efficacy of neoadjuvant chemotherapy in patients with triple-negative breast cancer, and the combined application of the two has certain predictive value.

Objective:To explore the application effect of 3D printing technology in comprehensive reconstruction of thumb and finger. Methods:From January, 2018 to January, 2020, 67 patients with 84 thumbs and fingers defects were selected, which were 37 thumbs and 47 fingers, and 45 of I-III degree and 39 of IV-VI degree. The method of operation was comprehensive reconstruction of thumbs and fingers assisted by 3D printing technology. The patient's hands and feet were scanned with CT and 3D modeling, and the 1∶1 model was derived. The skin model and bone model of thumb and finger defect were printed. Put the skin model on the great toes, and design the shape of the nail flap to be cut. Then the flap was spread in the groin area to design the perforating branch flap of the superficial circumflex iliac artery. The interphalangeal joint or metatarsophalangeal joint of the second toe was cut off according to the skeleton model (when the length of the bone joint was not enough, the iliac bone strip was taken to be connected in series). The effect of operation was observed in outpatient. Results:All of the 84 thumbs and fingers survived and were followed-up for 3 to 24 months. The appearance of the reconstructed thumbs and fingers was similar to that of the normal ones, with good texture and elasticity. The nail was smooth and glossy. The sensory recovery was S 3+, and the TPD was 4-6 mm. According to the Evaluation Standard of Finger Replantation and Reconstruction of Hand Surgery Society of Chinese Medical Association, 70 thumbs and fingers were excellent and 14 thumbs and fingers were good. In 64 thumbs and fingers, all toes were preserved in the donor area, and the color and texture of the flap were satisfactory, which did not affect walking, running and jumping, and had no walking pain. Conclusion:With the help of 3D printing technology, the tissues can be cut more accurately during the thumb and finger comprehensive reconstruction, which not only improves the beauty of reconstructed thumbs and fingers, but also avoids unnecessary trauma and improves the satisfaction of patients.
