2.Effects of sodium ferulate on A?_(25-35)-induced cognitive deficits and expression of IL-1? and p38MAPK in rats

Aim To study the effects of sodium ferulate on A?induced cognitive deficits and expressions of IL-1? and phospho-p38MAPK proteins.Methods Alzheimers disease model of rats was produced by intracerebroventricular injection of A?_(25-35)(10 ?g,once).Morris water maze was used to measure spatial memory performance.Nissl staining and immunohistochemical technique for glial fibrillary acidic protein(GFAP) were employed to determine the morphology of pyramidal neurons and astrocyte infiltration in hippocmpal CA1 regions.The levels of phospho-p38MAPK and IL-1? were determined by Western blot and ELISA method.Reverse transcription-PCR analysis showed changes in FasL mRNA.Results Intracerebroventricular injection of A?_(25-35)in rats resulted in spatial memory impairments shown by longer escape latency and decreased percentage of time spent in the target quadrant.These behavioral dysfunctions were accompanied by astrocyte activation and infiltration,increased IL-1? production and elevated FasL mRNA level,the loss of pyramidal neurons in hippocampal CA1,and the increase of phosphorylated p38MAPK.Oral administration of sodium ferulate(50,100,250 mg?kg~(-1)daily) and ibuprofen 15 mg?kg~(-1)daily markedly improved the memory impairment,attenuated pyramidal neuronal damage,and reversed the A?-induced increases in IL-1? and p38MAPK activation.Conclusion sodium ferulate prevents A?-induced neurotoxicity through suppressions of inflammatory response and the activation of p38MAPK.
3.Isolation and Purification of the Antifungal Antibiotic from the Fermentation Broth of Streptomyces luteogriseus H-103

The antifungal antibiotic produced by Streptomyces luteogriseus H-103 was purified by means of macroporous adsorbent resin, and the crystal of the antibiotic with high purity was got. In this paper, the methods of purification by adsorbing of microporous adsorbent resin and detection by reversed high performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD) were established. The result appeared that resin X-5 is the best adsorbent, the eluant is 50% ethanol. The antibiotic was successfully separated on Agilent~(TM)20RBA?310SB C_(18 )column (150mm?4.6mm i.d,5?m) , using a mixture of acetonitrile (A)-H_(2)O (B) as a mobile phase under gradient elution at a flow of 0.8mL/min at 30℃.0~4.0 min, V(A)∶V(B)=20∶80, 4.0~9.5min, V(A)∶V(B)=45∶55, then V(A)∶V(B)=80∶20. The drift tube temperature and the air carrier gas flow rate of the ELSD were set at 115℃ and 2.3L/min.
5.MAP kinase superfamily in amyloid β-protein fragment 25-35-induced inflammation andapoptosis in rat hippocampus in vivo

AIM To explore the mechanism of amyloid β-protein fragment 25-35(Aβ25-35)-induced inflammation and apoptosis in rat hippocampus in vivo by studying mitogen-activated protein kinase (MAPK) signaling pathway and the protective effect of anti-inflammatory drug ibuprofen. METHODS Rats were given ibuprofen (7.5 mg·kg-1 daily, ig) for 3 weeks prior to and 1 week after icv single dose of Aβ25-35 (10 μL, 1 mmol·L-1). Seven days after injection, Nissl staining and immunocytochemical technique were employed to determine the morphology of pyramidal neurons and astrocyte infiltration in hippocampal CA1. The expressions of IL-1β, extracellular signal-regulated kinase (ERK), p38 MAPK, PKC, and caspase-3 were determined by Western blot. Reverse transcription-PCR analysis showed changes in IL-1β mRNA level. RESULTS Intracerebroventricular injection of Aβ25-35 elicited astrocyte activation and infiltration and caused a strong inflammatory reaction characterized by increased IL-1β production and elevated IL-1β mRNA level. The inflammatory reaction was accompanied by the loss of pyramidal neurons in hippocampal CA1. The phosphorylation of p38 MAPK was significantly increased, on the other hand, the phosphorylation of ERK was significantly reduced and these were coupled with the increase of caspase-3 expression in hippocampal CA1. Ibuprofen (7.5 mg·kg-1 daily, 4 weeks) significantly reduced Aβ-induced IL-1β expression, caspase-3 expression and p38 MAPK activation. The loss of pyramidal neurons was also significantly attenuated by treatment with ibuprofen. CONCLUSION The activation of p38 MAPK and the down-regulation of ERK play a pivotal role in the inflam-matory response and apoptosis evoked by Aβ25-35 in vivo, which can be prevented by ibuprofen.
6.Early application of non-invasive ventilation in treating elderly patients with acute left ventricular failure and respiratory failure

Objective To study effects of early non-invasive ventilation (NIV) application in treating elderly patients with acute left ventricular failure induced respiratory failure. Method Totally 32 elderly patients with acute left ventrieular failure induced respiratory failure, admitted from August 1997 to February 2007, received NIV treatment, and were retrospectively studied. There were 22 male and 10 female, aged (81.5?8.6) yearsdd. The changes of rahs, respiration rate, heart rate, arterial blood gas, cardiac function before and after NIV application were compared. According to the application time of NIV, 32 patients were divided into two groups: group A (early NIV application group, n=17) and group B (non-early NIV application group, n= 15). The time to improve the symptoms, the application time of NIV, cure rates, tracheal intubation rates and mortality were compared between the two groups. Results Thirty of the 32 patients survived, cardiac function was improved from New York classⅣtoⅠ~Ⅱ, respiratory rate, heart rate and blood pressure significantly decreased, PaO_2 and SaO_2 significantly increased and PaCO_2 significantly decreased. 7.Analysis of epidemiological and clinical features of adenovirus infection in 80 children with acute respiratory tract infection.

By analyzing the epidemiological and clinical features of adenovirus in children with acute respiratory tract infection (ARTI), we provide a theoretical basis for early clinical diagnosis and treatment. Nasopharyngeal secretions were collected from 3480 children with ARTI, who were hospitalized at the No. 2 Hospital of Changzhou from January 2011 to December 2012. Adenovirus were detected using direct immunofluorescence assays. A total of 80 samples were positive for adenovirus (2.30%). The rate of adenovirus infection during 2011 was significantly higher than that in 2012, and the infection rate was higher in summer and autumn than in winter and spring. The infection rate was 1.14% among children aged < 1-year-old and the rates were higher among children in other age ranges. Adenovirus was found to be an important ARTI pathogen in children in Changzhou, mainly affecting children older than 1 year. ADV infections have various clinical presentations, but affected children tend to be severely ill with poor outcomes.

8.Investigation of Tilt Angle of Head-Up Tilt Test on Children

0.05).The different tilt angles impacted patients' fear psychology(?2=8.038,P=0.018).The order of the extent of children's fear psychology represented from low to high at the angle of 60?,70? and 80?,respectively.The extent of the children's fear psychology had positive correlation with the tilt angle(r=0.669,P=0.002).Conclusions The tilt test angle does not affect the positive rate of HUTT and the hemodynamics in children undergoing HUTT with angles from 60? to 80?.The children at the tilt angle 60? had less fear than at the angle of 70? or 80?.The 60? tilt angle of HUTT in children is recommended in the clinical practice.

10.Therapeutic observation of grain-sized moxibustion for chemotherapy-induced myelosuppression for non-small cell lungcancer

Objective: To observe the clinical efficacy of grain-sized moxibustion in treating chemotherapy-induced myelosuppression for non-small cell lung cancer (NSCLC) and its effect on quality of life (QOL). Methods: Eighty NSCLC patients admitted to the Inpatient Department of Zhejiang Cancer Hospital between September 2016 and March 2018 were recruited and divided into an observation group and a control group by random number method, with 40 cases in each group. The two groups both received chemotherapy with paclitaxel plus cisplatin (TP regimen). The control group received oral administration of leucogen tablets starting from the first day of chemotherapy, 20 mg each time, three times a day, for consecutive 14 d; the observation group was additionally given grain-sized moxibustion, once a day, five days per week at a two-day interval, until the fourteenth day. The myelosuppression severity was observed and compared between the two groups prior to chemotherapy, at the 3rd, 7th and 14th days of chemotherapy; the QOL in the two groups was evaluated before chemotherapy, at the 14th and 21st days of chemotherapy. Results: Regarding myelosuppression, the peripheral blood indicators increased significantly at the 3rd day of chemotherapy in both groups (P<0.05 or P<0.01); at the 7th and 14th days of chemotherapy, the peripheral blood indicators presented a decreasing tendency in the two groups, but the level in the observation group was still significantly higher than that before chemotherapy (P<0.01); at the 3rd, 7th and 14th days of chemotherapy, the peripheral blood indicators in the observation group were higher than those in the control group (P<0.05 or P<0.01); the occurrence rate of myelosuppression in the observation group was significantly lower than that in the control group (P<0.01). The QOL score in the observation group was markedly higher than that in the control group at the 14th and 21st days of chemotherapy (both P<0.05). Conclusion: Grain-sized moxibustion can effectively improve myelosuppression after chemotherapy for NSCLC, reducing its occurrence and enhancing the patient's QOL.
