1.Effect of ICAM-1 on the rejection of corneal transplantation in rabbits

· AIM: To investigate the role of intercellular adhesion molecule-1 (ICAM-1) in the graft rejection of rabbit penetrating keratoplasty (PKP).· METHODS: Sutures were used to produce corneal neovascularization (CNV) on New Zealand white rabbits to make PKP model. Graft mean survival time and rejection index were determined. The levels of sICAM-1 in serum and aqueous humor were dynamically determined by enzyme-linked immunosorbent assay (ELISA).Immunohistochemical technique was used on cornea to get the evidence of expression and distribution of ICAM-1.· RESULTS: Graft mean survival time is (12.4±1.3) days.The mean sICAM-1 levels in aqueous humor and serum in the control animals that did not undergo surgery were (16.6±3.6)ng/L and (95.2±6.3) ng/L respectively. Levels in aqueous humor and serum for the graft rejection group increased postoperatively, continued to increase to (53.9±19.2)ng/Land (378.8±30.6)ng/L, compared to those in the other group (P <0.01). ICAM-1 positive cells were found in the graft rejection group.· CONCLUSION: ICAM-1 plays a critical role in initiating and maintaining corneal graft rejection. Sequential determination of serum sICAM-1 after operation may be of value in the prediction and diagnosis of acute rejection.
2.The sensibility of pattern visual evoked potential on diagnosis of primary glaucoma in different temporal frenquency

AIM: To investigate the sensibility of pattern visual evoked potential (P-VEP) on the diagnosis of glaucoma in different temporal frequency.METHODS: The P-VEP were recorded in 51 eyes of 30 primary glaucoma (PG) patients, 16 eyes of 13 ocular hypertension (OHT) patients and 46 eyes of 23 age-equivalent normal people using an array of different temporal frequency and then the patients were compared respectively with the normal group.RESULTS: The P100 wave amplitude in OHT group was significantly lowered at 8Hz, and was lowered at all temporal frequency in PG group, especially when the temporal frequency was higher.CONCLUSION: The P-VEP can be abnormal when the PG is still in its early stage, so it can be an index of early diagnosis and this is more obvious when the temporal frequency is higher, especially at 8Hz.
3.Establishment of mice Kawasaki disease model by Lactobacillus casei cell wall extract

Objective To establish mice models of Kawasaki disease by Lcatobacillus casei cell wall extract(LCWE) ,in order to provide experimental materials for follow-up study .Methods LCWE group was given LCWE 0 .1 mL per mouse(containing 200 mg LCWE) via abdominal intramuscular injection .PBS group was given 0 .1 mL PBS per mouse .Orbital blood samples were collected respectively at the 1st ,3rd and 28th day after administration .The heart ,lung ,liver ,kidney ,spleen and other organs tissue samples were collected for pathological section ,HE staining .The histopathological changes were investigated ,and the routine blood test was proceeded .Results The WBC of LCWE group showed a trend of rising at the 1st ,3rd and 28th day after administration .PLT and MPV of LCWE group increased at the 3rd day after administration ,which returned to normal levels at the 28th day after adminis-tration .The pathological section showed the blood vessel walls of heart tissue enlargement ,surrounded and infiltrated by inflamma-tory cells infiltration ,the atheromatous plaque in blood vessels occasionally .Conclusion The study established mice model of Ka-wasaki disease successfully .

6.The comparison of mfVEP in normal eyes and glaucomatous eyes

AIM: To test the ability of multifocal visual evoked potential (mfVEP) in the detecting of glaucoma by comparing the mfVEP recorded from normal subjects and glaucoma patients.METHODS: The mfVEP of 32 normal eyes (n =21) and of 58 eyes (n =37) with primary glaucoma were recorded with the Vision Monitor electrophysical apparatus by the second kernel analysis and to determine the correlation of the topographic location between them.RESULTS: There were significant variability (the coefficient of variation was 43.05%) in mfVEP RMS amplitude in the normal subjects; The RMS amplitude of eyes with glaucoma were smaller than that of the normal eyes and significantly statistical difference were found in the relatively center (namely the 0° -10° ring zone) and in superior nasal quadrant (P<0.05) while there were no significantly statistical differences of the latency time between them.CONCLUSION: The normal subjects have large individual variability of mfVEP responses. The RMS amplitude of the mfVEP of glaucomatous eyes descends, especially in center zone and superior nasal quadrant.
7.Study on real-time probe technique detecting mtDNA 11778 point mutation in Leber hereditary optic neuropathy patients

Background Although Leber hereditary optic neuropathy (LHON) and optic neuritis have different causes and managements,their clinical manifestations are difficult to be distinguished.Real-time fluorescence quantitative polymerase chain reaction(RTFQ-PCR) is a high flux,simple,rapid and specific detecting technology,so establishing a specific diagnosis method of LHON with RTFQ-PCR has a practical significance.Objective Purpose of the present study was to establish a real-time Taqman probe for mitochondrial DNA (mtDNA)11778G>A mutation in LHON patients.Methods Primers and Taqman probe for mtDNA 11778G>A mutation were designed based on mtDNA complete geneme.Eighty-four patients with LHON were selected from the LHON DNA bank of Molecular Biology Laboratory,Henan Eye Institute,and 40 normal physical examinees aged 18-20 years were from Henan People's Hospital.2 ml of periphery blood was collected from each individual.Based on the double-blindness principle,mtDNA 11778G>A mutation was tested by both Taqman probe and sequencing to check the reliability of real-time Taqman probe.Results The mtDNA 11778G>A mutation was found in 23 out of 84 patients,and 61 showed a negative result by the technique of real-time Taqman probe.The Ct values of 23 patients with mtDNA 11778G>A mutation were 22.993 ±0.708,but those of 5 normal controls were 0.These findings showed a consistent rate of 100% with the sequencing results.In addition,both the false positive rate and the false negative rate were zero.Conclusions Real-time Taqman probe technique is an accurate,convenient,sensitive,specific and intuitionistic method for the diagnosis of mtDNA 11778G>A mutation in LHON patients.It is feasible and suitable to screen the LHON patients with mtDNA 11778G>A mutation in a large scale.
8.Effect of sulfur dioxide on vascular collagen remodeling in spontaneously hypertensive rats.

OBJECTIVETo study the modulatory effect of sulfur dioxide (SO(2)) on the accumulation of collagen type I and type III in the wall of aorta during spontaneously hypertensive rat (SHR) vascular remodeling.
METHODSEight male Wistar Kyoto rats at the age of 4 weeks with normal blood pressure were used as a control group. And sixteen male SHRs at the age of 4 weeks were randomly divided into an SHR control group and SHR + Na(2)SO(3)/NaHSO(3) (SO(2) donor) group. Na(2)SO(3)/NaHSO(3) solution was injected intraperitoneally everyday to the rats in the SHR + Na(2)SO(3)/NaHSO(3) group. After 5 weeks, the systemic blood pressure was measured. The weight ratio of left ventricle to the whole heart was also measured. The rat aorta was dyed with Hart's method. The morphometric parameters including outer radius, lumen radius and the wall thickness were calculated by Leica workstation. The plasma level of SO(2) was determined by HPLC method. The expressions of collagen type I and type III in aorta were detected by immunohistochemistry.
RESULTS(1) Compared with the WKY rat control group, the systolic blood pressure increased by 53%, the weight ratio of left ventricle to the whole heart increased by 6% but the plasma level of SO(2) decreased by 44% for rats in the SHR group (P < 0.01 or P < 0.05). Compared with the SHR control group, the systolic blood pressure decreased by 26%, but the plasma level of SO(2) increased by 28% (all P < 0.01) for rats in the SHR + Na(2)SO(3)/NaHSO(3) group. (2) Compared with the WKY rats, the ratio of media to lumen radius increased by 28% for SHR. Compared with the SHR group, the ratio of media to lumen radius decreased by 10% (P < 0.01) in rats of the SHR + Na(2)SO(3)/NaHSO(3) group. (3) Compared with rats in the WKY control group, collagen type I expression increased by 10% for rats in the SHR group (P < 0.01). Compared with the SHR group, however, the expression decreased by 58% for rats in the SHR + Na(2)SO(3)/NaHSO(3) group (P < 0.01). (4) Compared with rats in the WKY control group, the expression increased by 13% for rats in the hypoxia SHR group (P < 0.01); however, compared with rats in the SHR group, the expression decreased by 8% in the rats of the SHR +Na(2)SO(3)/NaHSO(3) group (P < 0.01).
CONCLUSIONSIn the process of SHR vascular collagen remodeling in the rats, SO(2) could inhibit the abnormal accumulation of collagen type I and type III in the wall of aorta. This effect may be one of the mechanisms by which SO(2) ameliorates SHR vascular remodeling.

9.Expression of SARS spike gene in Shizomycete pombe.

The viral spike protein is the main surface antigen of the coronavirus, and it could be useful in the research of clinical diagnosis, SARS vaccine and the structure biology.According to the analysis of the main antigen of the SARS spike protein, 5 fragments of the whole spike gene were cloned, and ligated to the vector pNMT1. Through electroporation transformantion to TCP1, the recombinant S. pombe strains capable of expressing the 5 fragments were constructed. SDS-PAGE or Western blot analysis of the induced expression products demonstrated that the 5 recombinant proteins were expressed in the fission yeast respectively.

10.Changes of color pattern reversal visual evoked potential of primary glaucoma.

OBJECTIVE:
To investigate the changes of color pattern reversal visual evoked potential (CPR-VEP)of primary glaucoma using different temporal frequencies, and to search for the best temporal frequency parameters and color parameters.
METHODS:
Vision Monitor visual electrophysiograph (Métrovision, France) was used to record CPR-VEP at different temporal frequencies (1, 2, 4, 8, 16, and 32 Hz) and different color stimulations (black/white, red/green, blue/yellow) in 41 cases (70 eyes) with primary glaucoma (glaucoma group) and 13 normal subjects (26 eyes) (normal control group). P100 wave amplitudes were compared.
RESULTS:
(1) In the normal control group, P100 amplitudes declined while the temporal frequency of black/white stimulation was increasing, but they had peaks at 2 Hz and 8 Hz red/green stimulation and blue/yellow stimulation. (2) In the glaucoma group, CPR-VEP P100 declined while temporal frequency was increasing under 3 color stimulations, but had a peak at 8 Hz. At 2 Hz-16 Hz, P100 amplitudes were related with the mean defect of Humphrey visual field, especially with all 3 color stimulations at 8 Hz and with blue/yellow stimulation at 2 Hz and 16 Hz. (3) P100 amplitude was most different under the 3 color stimulations between the 2 groups at 8 Hz.
CONCLUSION
The changes of CPR-VEP P(100) amplitude can objectively reflect the glaucoma visual function damage. CPR-VEP P100 amplitude has certain value in studying glaucoma under different color stimulations (black/white, red/green, blue/yellow) at 8 Hz, and blue/yellow stimulation at 2 Hz and 16 Hz.
