1. Influence of cultivation medium on growth of tissue cultured seedlings of Polygonatum cyrtonema

Objective: To find a solution to the problems in the growth of tissue cultured seedlings of Polygonatum cyrtonema, such as low seedling survival rate, poor growth, and leaf disease. Method: In this study, tissue cultured seedlings of P. cyrtonema were used as testing plants. Two types of commercially-available cultivation medium commonly used in Fujian Province, as well as six types of cultivation medium mixed using peat soil, pearlite, roseite, sand, fungi residues and plant ash were used as the raw materials; Among them, a customized nutrient solution was added into the T5 and T7 cultivation media. Besides measuring the physicochemical properties of the cultivation media mentioned above, the study focused on the evaluation of their influence on the rate of grown-up seedlings, biological characteristics, growth biomass, strength index and incidence of leaf disease of P. cyrtonema. It also attempted to find the correlation between the physicochemical properties of the cultivation media and the biological characteristics, growth biomass, as well as the strength index of tissue cultured seedlings of P. cyrtonema. Results: The difference in the cultivation media had a great influence on the tissue cultured seedlings of P. cyrtonema in the rate of grown-up seedlings, biological characteristics, growth biomass, strength index and incidence of leaf disease. The difference in bulk density of the cultivation media had a notable negative correlation with the plant height of the tissue cultured seedlings of P. cyrtonema. The difference in the organic content of the cultivation media had a notable positive correlation with the fresh weight and dry weight of the tissue cultured seedlings of P. cyrtonema. Treated with the T5 cultivation medium we prepared, the tissue cultured seedlings of P. cyrtonema were in better growing condition in the rate of grown-up seedlings, morphological index, growth bomass, strength index, and no disease was found on the leaf. Conclusion: The prepared T5 was worth being widely used as the cultivation medium for growth of tissue cultured seedlings of P. cyrtonema.
2.Effect of carbenoxolone on expression of Fos, NMDAR2 and GFAP in the hippocampus of pentylenetetrazo-kindled epileptic rats.

OBJECTIVEGap junctions, the clusters of intercellular channels, play an important role in synchronizing electrical activity. This study investigated the effect of gap junction blocker carbenoxolone (CBX) on epileptic activity in pentylenetetrazo (PTZ)-kindled rats.
METHODSThirty adult male SD rats were randomly divided into three groups: control, PTZ-kindled and CBX-treated groups (n=10 each). The rats from the PTZ-kindled and the CBX-treated groups were intraperitoneally injected with PTZ (35 mg/kg x d) to induce epilepsy. After epilepsy kindling, they were intraperitoneally injected for 3 days with CBX (10 mg/kg) (CBX-treated group) or with normal saline (PTZ-kindled group). The control group received intraperitoneal injections of normal saline. Anti-GFAP, anti-Fos, and anti-NMDARZ immunohistochemical ABC methods were used to detect the expression of GFAP-Li, Fos-Li and NMDAR2-Li in the hippocampus respectively.
RESULTSSpontaneous seizures occurred in PTZ-kindled epileptic rats. CBX administration reduced spontaneous seizures. The NMDAR2-Li and Fos-Li neurons as well as GFAP-Li astrocytes in hippocampi increased in PTZ-kindled epileptic rats compared with controls. The numbers of Fos-Li (93.75 +/-7.94 vs 165.25 +/-15.87, P < 0.05) and NMDAR2-Li neurons (61.47 +/-3.62 vs 148.72 +/-14.53, P < 0.01) in the CBX-treated group were significantly less than in the PTZ-kindled group. There were no significant differences in the GFAP-Li expression between the CBX-treated and the PTZ-kindled groups.
CONCLUSIONSCBX may inhibit spontaneous seizures and decrease the numbers of Fos-Li and NMDARZ-Li neurons, thus providing anti-epileptic effects.

3.Effects of Z-9-octadecenyl-2-propanesulfonamide on the learning and memory function of diabetes-associated cognitive decline

This study was designed to investigate the therapeutic effect of novel compound Z-9-octadecenyl-2-propanesulfonamide (N15) on diabetes-associated cognitive decline (DACD). Type 2 diabetes (T2DM) mice models were established with multiple injection of low doses of streptozotocin (STZ) in mice on high fat diet (HFD). Vehicle and different concentrations of N15 (50 and 100 mg·kg-1·d-1) were administrated orally for 6 weeks. The step-down test, dark avoidance task and Morris water maze were conducted at the 6th week. The level of glucose and lactic acid in hippocampus were determined and mRNA of growth associated protein-43 (GAP-43), synaptophysin (SYN), brain derived neurotrophic factor (BDNF) and neurotrophins-3 (NT-3) in hippocampus were analyzed by real time PCR. The beneficial effects of N15 on learning and memory were found in the test of step-down, dark avoidance and Morris water maze. N15 reduced the level of glucose and lactic acid in hippocampus of HFD+STZ-induced diabetic encephalopathy model mice. Additionally, the mRNA expression of GAP-43, SYN, BDNF and NT-3 in hippocampus of HFD+STZ-induced diabetic encephalopathy mice were significantly increased by N15 (P<0.01). These results suggest that the novel compound N15 can ameliorate diabetes-associated cognitive decline and the potential mechanism may be associated with the expressions of increased synaptic-related factors and neurotrophic factor in the hippocampus of diabetesassociated cognitive decline in mice.
