ObjectiveTo investigate the distribution of human immunodeficiency virus-1 (HIV-1)subtypes and the relationship with transmission routes in Shenzhen city. MethodsThe epidemiological survey was conducted in 429 subjects with HIV-1 infection.Vein blood was collected and prepared with anticoagulation.Thereafter, the viral nucleic acid was extracted and subsequently amplified for HIV-1 env,gag genes by reverse transcription nested polymerase chain reaction. The viral sequences were analyzed using Mega software and compared with those in HIV database. The HIV-1 subtypes were confirmed and evolutionary tree was established. The gene discrete rate of different subtypes was calculated.ResultsFour hundred qualified samples were identified as CRF01_AE (n=207),CRF07_BC (n=115),CRF08 _BC (n=14),CRF02_AG (n=1),B (n=49) and C(n=14),respectively.CRF01_AE was the predominant subtype that was detected in the subjects with different transmission routes, such as homosexual contact, heterosexual contact, intravenous drug use and hospital-acquired infection. Among these, sexual contact was the most common route. There were nine cases infected with subtype B HIV-1 in 10 case who were blood borne. The gene discrete rate was more significant in subtype B HIV-1 compared to the other subtypes.According to the evolutionary tree analysis, the viruses under the same subtype formed the relatively independent clusters by different transmission routes. But some overlaps were observed as well. Conclusions Tbere are many HIV-1subtypes in HIV-1 infected subjects in Shenzhen, while the recombinant viruses present as the predominant form.Transmission routes are various with sexual contact as the most common route.Every subset distributes unevenly and forms its own characteristic.Overlap can be observed among the gene distribution of different HIV-1 subtypes.

Objective:To study the mechanisms that acupuncture therapy of "Shuanggu Yitong" in treating postmenopausal osteoporosis(PMO).Methods:60female SD rats aged 6 months were resected bilateral ovaries and feeded for 90 days in order to make the model of experimental osteoporosis. Then, they were divided into 5 groups such as control group, regular acupoints treated group,"Shuanggu" group,"Yitong" group,"Shuanggu Yitong" group, with 12 rats each group, and the control group without any treatment;Pishu(BL 20),Weishu(BL 21),Shenshu(BL 23),Qihaishu(BL 24) was selected for the regular acupoints treated group;Guanyuan(RN 4),Zusanli(ST 36) for the "Shuanggu" group;and Shenshu(BL 23),Geshu(BL 17),Dazhu(BL 11) for the "Yitong" group;Guanyuan(RN 4),Zusanli(ST 36),Shenshu(BL 23),Geshu(BL 17),Dazhu(BL 11)for the "Shuanggu Yitong" group.Electroacupuncture therapy was given once a day,15 minutes each time, the needles were retained for 10 minutes. Then these rats rested for 1 day after consecutive therapy for 6 days. The therapy lasted for 12 weeks.RT-PCR was used to examine the expression level of interleukin-6(IL-6)mRNA in bone tissue. Results:Compared with control group, the expression level of IL-6mRNA in all acupuncture therapy groups decreased(P0.05).Conclusion:Acupuncture can reduce the expression level of IL-6mRNA in osteoporosis model rats without ovaries and "Shuanggu Yitong" acupuncture therapy is superior to other regular acupuncture therapy.

Macrophages,as an important member of innate immune system, constitute a major component of the microenvironment of tumours, and play an important role in the occurrence and development of tumor.According to particular phenotypes and functions, macrophages can be divided into M1 macrophages and M2 macrophages. In the research of breast cancer, lung cancer, gynecological malignancies, researchers have found that tumor-associated macrophage has the similar phenotype and function of M2 macrophages.Tumor-associated macrophage can promote the formation of new blood vessels and lymphatic vessels in tumor tissue, thus contributing to the occurrence and development of tumor. The relation between tumor-associated macrophage and poor prognosis was also be found. The tumor microenvironment can affect the phenotype and function of macrophages, such as TGF,IL-10 and PGE-2.This paper reviewed the role and mechanism of macrophages in the tumorigenesis.

Objective To improve the key steps of direct volume rendering(DVR) method to obtain ideal semitransparent 3D visualization of medical images from 2D slices.Method An arbitrary data set was classified into a simple one and a complex one according to the boundaries existing within the data set, thereby a piecewise linear function was used to construct the transfer function(TF).Low-grayed voxels were regarded as a kind of material which did not radiate but do induce attenuation of sampling lights. The TF was adjusted according to the opacity distribution when one resampling process ends, and the rendering procedure was completed.Result Interior structures of tissues in different levels were rendered. Conclusion Multi-level visualization of 3D medical images for the semitransparent effect can be obtained by considering all the tissues in the data set as semitransparent materials.

Background Excessive production, deposition and contraction of extracellular matrix (ECM) of human lens endothelial cells (LECs) is one of main causes to posterior capsular opacification (PCO).Researches indicated that Wnt3a protein was involved in production of ECM and fibrosis in epithelial cells, but its effect on LECs is still unclear. Objective This study aimed to elucidate the roles of Wnt3a in production of ECM and gel contraction of LECs.Methods Lipofectamine-mediated transient transfection technique was used to introduce cDNA of Wnt3a gene and pcDNA3-HA vector into the LEC cell line SRA01/04 to act as Wnt3a transfection group and control group respectively. After 48 h of transfection,Western blot was used to detect the expression of Wnt3a,main components of ECM Col-Ⅰ,Col-Ⅳ and integrin β1.Immunofluorescence was used to detect the expression and distrubition of α-SMA and F-actin;collagen contraction was observed by mingling SRA01/04 cells with Col-Ⅰ.Results After 48 hours of transfection using lipofectamine 2000,the expressions of wnt3a protein in SRA01/04 cells was 0.703 ±0.105 in the wnt3a transfected group, and that in the control group was 0.290 ± 0.066,showing a significant difference between the two groups (t =5.782,P<0.01).Western blot assay showed that the expression levels of Col-Ⅰ and Integrin β1 was 0.697±0.021 and 0.875±0.055 in the Wnt3a transfected group, and which was significantly higher than 0.370±0.020 and 0.580±0.030 in the control group (t =19.600,8.156,both P<0.01).The expression level of Col Ⅳ in the Wnt3a transfected group was higher than that of the control group (0.430±0.020 vs 0.383 ±0.031),but the difference was not significant (t =2.514,P>0.05).Immunofluorescence assay revealed that F-actin and α-SMA were weakly expressed in the cell membrane primarily in the control group, while they were strongly expressed in the cell membrane, cytoplasma in the Wnt3a transfected group.Tewnty-four hours after addtion of Col-Ⅰ,the gel contraction area ratio appeared to be more obvious in comparison with the 8 hours (64.1% ±2.3% vs 98.9% ± 1.0%),and gel contraction area ratio was lower in the Wnt3a transfected group than that in the control group (64.1% ± 2.3% vs 93.9% ± 3.1%).Conclusions The overexpression of Wnt3a activates the production of ECM,and the remodeling of celluar skeleton and cellular contraction.
