Objective: To evaluate the effect of core∶dentin thickness ratio on the flexure strength, fracture mode and origin of bilayered IPS Empress Ⅱceramic composite specimens. Methods: IPS EmpressⅡ core ceramic, dentin porcelain and bilayered composite specimens with core∶dentin thickness ratio of 2∶1 and 1∶1 were tested in three-point flexure strength. Mean strengths and standard deviations were determined. The optical microscopy was employed for identification of the fracture mode and origin. Results: The flexure strength of dentin porcelain was the smallest (62.7 MPa), and the strength of bilayered composite specimens was smaller than single-layered core ceramic(190.2 MPa). The core∶dentin ratio did not influence the strength of bilayered composite specimens. The frequency of occurrence of bilayered specimen delaminations was higher in the group of core∶dentin thickness ratio of 1∶1 than in the group of 2∶1. Conclusion: IPS Empress Ⅱcore ceramic was significantly stronger than veneering dentin porcelain. Core∶dentin thickness ratio could significantly influence the fracture mode and origin, and bilayered IPS Empress Ⅱceramic composite specimens showed little influence in the fracture strength.

OBJECTIVETo establish a method for rapid determination of airborne acrylonitrile using a portable gas chromatograph.
METHODSA single standard sample of acrylonitrile was prepared in a laboratory and sampled by the built-in constant flow pump of the portable gas chromatograph. The sample was then preconcentrated by the preconcentrator, thermally desorbed, separated by capillary columns, and detected by a micro argon ionization detector to determine the retention time. Retention time was then used to perform qualitative analysis. Under the set condition of gas chromatography, the external standard method was used to create a standard curve for quantitative analysis of acrylonitrile.
RESULTSThe linear range of acrylonitrile on the portable gas chromatograph was 0.25 to 3.00 mg/m(3). The regression equation was y = 10(-5) x-0.0275, r = 0.9977. The limit of detection was 0.005 mg/m(3), and the lower limit of quantification was 0.25 mg/m(3). The relative standard deviation was lower than 7.09%, and the degree of accuracy was 91.09-105.54%.
CONCLUSIONPortable gas chromatography is a simple, repeatable, and accurate method for rapid determination of airborne acrylonitrile.

Objective To explore application of the bolus-triggered technique and optimal match between collimation and pitch of MSCTA in intracranial and cervical arteries.Methods(1)The small-dose tests were performed in 19 healthy volunteers and the theoretical threshold was obtained by the time-density curve.(2)forty healthy volunteers were divided randomly into two groups and the scanning parameters were as follows.Group A:collimation 1.0 mm,pitch 1.750. Group B:collimation 2.5 mm,pitch 0.625. Statistical significance was determined with the X~2 test and t test(?=0.05).Results(1)In 19 volunteers,CT value 4 seconds before the peak was 75 HU,the CT value at the beginning segment of the carotid artery and the C_1 segment of internal carotid artery was in accordance with the standard.(2)Image quality ofⅠ,Ⅱgrades artery structures of group B was superior to that of group A.Image quality ofⅢ,Ⅴ grades artery structures of group A was superior to that of group B.Conclusions(1)MSCTA in intracranial and cervical arteries can display systematically the cerebral and carotid arteries.(2)The bolus-triggered technique can improve the image quality of the target vessels. The image quality of the MSCTA of intracranial and cervical arteries is better with the threshold of 75 HU on the basis of 3.5 ml/s injection rate.(3)On the basis of the same other parameters, the optimal scanning parameters are a collimation of 1.0 mm with a pitch of 1.750.

OBJECTIVE To investigate the anti-pyroptotic effects of melatonin in atherosclerotic endothelium and to elucidate the potential mechanisms.METHODS ApoE-/-mice were randomly divid-ed into four groups (n=8): the normal-diet group (ND), the normal-diet group treated with melatonin (10 mg·kg-1)(ND+MLT),the high-fat-diet group(HFD),and the high-fat-diet group treated with melatonin (HFD+MLT).After 12 weeks, the expression levels of pyroptosis related genes including NLRP3,ASC, cleaved-caspase 1,GSDMD-N,IL-1β and IL-18 were examined in aortic endothelium by Western blotting, qRT-PCR and immunofluorescent staining.Besides, levels of MEG3 and miR-223 were also tested by qRT-PCR.The interaction between MEG3 and miR-223 was detected by luciferase assay. For in vitro study, human aortic endothelial cells(HAECs)were transiently transfected with miR-223 mimic,miR-223 inhibitor (AMO-223), MEG3-overexpressing plasmid or negative controls. After 6 h of transfection, the medium was replaced by fresh medium with or without ox-LDL(25 μg·mL-1)for 24 h and then treated with or without melatonin (10 μmol·L-1) for 48 h. Cell pyroptosis was evaluated by Hoechst 33342/PI staining and differentially expressed pyroptosis related genes. RESULTS Melatonin markedly reduced the atherosclerotic plaque in aorta of ApoE-/- mice. Meanwhile, melatonin also attenuated the expression NLRP3, ASC, cleaved-caspase1, NF-κB/GSDMD, GSDMD-N termini, IL-1β, and IL-18 in aortic endo-thelium.Consistent anti-pyroptotic effects were also observed in ox-LDL-treated HAECs.We found that lncRNAMEG3 enhanced pyroptosis in HAECs. Moreover, MEG3 acted as an endogenous sponge by sequence complementarity to suppress the function of miR-223 and to increase NLRP3expression and enhance endothelial cell pyroptosis. Furthermore, knockdown of miR-223 blocked the anti-pyroptotic actions of melatonin in ox-LDL-treated HAECs. CONCLUSION Melatonin prevents endothelial cell pyroptosis via MEG3/miR-223/NLRP3 axis in atherosclerosis and therefore melatonin replacement might be considered a new strategy for protecting endothelium against pyroptosis thereby for the treat-ment of atherosclerosis associated with pyroptosis.
