2.THE EFFECTS OF CARVEDILOL ON NEUROHOMONE AND HEART FUNCTION IN CONGESTIVE HEART FAILURE

To investigate the effects of Carvedilol on neurohormone and heart function in patients with congestive heart failure(CHF) by double blinded, randomized clinical trial. 35 congestive heart failure patients were double blinded and randomly divided into 2 groups, Carvedilol group and placebo group. 15 healthy persons constitute the normal control group. We measured the plasma concentration of norepinephrine(NE), angiotensin (Ang) and plasma renin activity (PRA) , meanwhile left ventricular ejection fraction (LVEF) , left ventricular dimension and volume were observed before and after treatment. 3.Perioperative nutritional support in patients with obstruction caused by colon cancer

Objective:To investigate the importance and appropriate application of perioperative nutritional support in patients with obstruction caused by colon cancer.Methods:19 patients received parenteral or enteral nutritional support in the period of perioperation.When nutrition was applied,the nutritional indices,liver function and renal function were observed.Results:Hb,blood K~(+) and Na~(+) were significantly increased after nutritional support.The nutritional indices did not change and were kept within normal range during nutrition.Conclusion:The nutritional support is one of the important perioperative treatments in patients with obstruction caused by colon cancer.
4.Value of non phosphorylated insulin -like growth factor binding protein -1 in cervical vaginal excretion and residual amniotic fluid index monitoring in the diagnosis of premature rupture of membranes

Objective To explore the method of rapid diagnosis preterm premature rupture of membrane (PPROM)and prognosis factors of residual amniotic fluid in mother and infant through monitoring nonphosphorylated insulin -like growth factor binding protein -1 (IGFBP -1)and the residual amniotic fluid index (AFI)of the cervi-covaginal secretions from PPROM women.Methods 96 cases of PPROM were tested IGFBP -1 of cervicovaginal secretions,AFI were tested by abdominal ultrasound through fast chromatography methods,and the cause was ana-lyzed.The pregnancy outcome and neonatal prognosis were compared.Results 92 cases positive of IGFBP -1 were diagnosed with PPROM.The risk factors of PPROMwere mainly reproductive tract infections,abortion,induced labor history and unknown causes (including no evidence of infection,fetal membrane dysplasia unclear or other factors), the twins,multiplets,macrosomia,breech or cephalopelvic disproportion,gestational hypertension disease,uterine fibroids or deformity,malnutrition and polyhydramnios.In the 92 patients with PPROM pregnant women,the duration from premature rupture of membrane to delivery was (105.7 ±13.9)h,cesarean section rate was 51.1%,fever rate of pregnant women was 7.6%,the rate of placental abruption was 4.3%,the rate of fetal distress was14.1%,perinatal infection rate was 21.7%,and neonatal asphyxia rate was 7.6%,the neonatal mortality was 7.6%.To compare the AFI≤40mm group with AFI >40 mm group,the duration from premature rupture of membrane to delivery was short-ened remarkably (t =10.904,P <0.05),the neonatal mortality rate was significantly increased (χ2 =4.359,P <0.05).Conclusion Measurement of IGFBP -1 and AFI in cervicovaginal secretions can rapid diagnosis PPROMand evaluate the results of pregnancy and neonatal prognosis,and can guide clinical improvement of pregnancy safety and reduce neonatal mortality.

6. Risk factor analysis of emergence agitation during anesthesia recovery period in 90 patients with hepatobiliary diseases

Objective: To analyze the risk factors of emergence agitation in patients with hepatobiliary diseases. Methods: A total of 110 patients with hepatobiliary disease were enrolled in the present study. The level of emergence agitation was scored by sedation-agitation scale (SAS) 30 min after the surgery. Patients with SAS < 4, were excluded. Related medical data of the rest 90 patients were subjected to univariate and multivariate Logistic regression for analysis of risk factors of emergence agitation. Results: Univariate analysis revealed that the airway management method, anesthesia maintenance method, operation time and the use of anesthetic gas absorber were significantly correlated with the incidence of agitation (P values being 0.014, 0.011, 0.042 and 0.019, respectively). Multivariate analysis revealed that induction with midazolam, urinary catheterization after anesthesia and total inhalational anesthesia were significantly correlated with the incidence of agitation (P values being 0.026, 0.049 and 0.004, and the OR values being 5.481, 10.867, and 11.604, respectively). Conclusion: In patients with hepatobiliary diseases, induction with midazolam, total inhalational anaesthesia and urinary catheterization after anesthesia are associated with high incidence of emergence agitation following general anaesthesia.
7. Advances in the study of arrhythmogenic mechanisms

Arrhythmia is a common complication of cardiovascular diseases and a risk factor for human health. Especially, ventricular tachycardia and ventricular fibrillation may not only exacerbate original heart diseases, but also cause cardiac sudden death which has been an mportant death reason in China. However, anti-arrhythmic drugs nowadays cannot effectively treat these arrhythmias, with an efficiency of only 30%-60%, which indicates that our knowledge about arrhythmias is limited. Hence, to explore the potential mechanism, look for novel targets, and develop drugs with multiple-channel action are the focus of the research direction. Recent studies displayed that the atrial-specific potassium channels such as IKur and IKAch were involved in atrial fibrillation, which provided a prospective target for atrial fibrillation treatment. Calcium leak, gap junction protein and autoantibody against ICaL channel were shown to participate in arrhythmogenesis. These findings provided a theoretical basis for the development of more effective anti-arrhythmic drugs. Remarkably, as a kind of mportant RNA regulating gene expression, microRNA (miRNA) was shown to possess anti-arrhythmic activities which may prevent cardiac sudden death. miR-1, miR-133 and miR-590 regulated the arrhythmia in various types of animal models. Because of the multiple-gene regulation actions of miRNA, it has the potential to be developed as novel anti-arrhythmic target.
8. Investigation of anticancer drug 5-Fu loaded porous metal-organic frameworks of MIL-53

OBJECTIVE: To study the drug release and cytotoxicity in vitro of 5-fluorouracil (5-Fu)-loaded porous metal-organic frameworks of MIL-53. METHODS: MIL-53 was hydrothermally synthesized, and the synthetic MIL-53 was characterized by TEM, XRD, IR and TG. 5-Fu was loaded into MIL-53, and the release pattern and cytotoxicity were investigated. RESULTS: The structure of MIL-53 was confirmed. The diameter of MIL-53 was at nanometer level and MIL-53 displayed better thermo-stabilization. The highest drug loading rate was 0.431 g · g-1 MIL-53 for 5-Fu. The drug release profile of 5-Fu-loaded MIL-53 displayed a clear biphasic release pattern. Cytotoxicity test showed that MIL-53 had good biocompatility at lower concentrations. CONCLUSION: MIL-53 is a promising platform for drug delivery due to its large pore sizes for drug encapsulation and good biocompatility at lower concentrations.
9.Determination of c-fos mRNA in U937 cell line with in situ hybridization

The levels of c-fos mRNA in U937 cell line were determined with in situ hybridization.Theresults showed that TPA induced the expression of c-fos gene with a relationship of dosage-ef-fect.It was also proved that GM-CSF,TNF,IL-9,IL-3 and IL-6 increased the expression of c-fos gene.
