1.Establishment and application of fingerprints of Shuanghuanglian Powder-Injection by HPLC

AIM: To establish the fingerprints of Shuanghuanglian Pwder-Injection(Flos Lonicerae Japonicae,Radix Scutellariae,Fructus Forsythiae) by high performance liquid chromatography(HPLC) so as to control quality of the injection. METHODS: The experimental conditions of the HPLC method were as follows: YMC-Pack ODSA C_(18) column(4.6 mm?250 mm,5 ?m,mobile phase consisting of methanol and 0.012 5 N ammonium acetate/acetic acid buffer eluted with a linear gradient,flow rate at 1.0 mL/min,the detectors of phtodiode array detector(PDA) and evaporative light scattering detector(ELSD) connected in series. RESULTS: The method provided two kinds of fingerprints with excellent spearation,which were HPLC-PDA(254 nm) fingerprint and HPLC-ELSD fingerprint.They had a good correlation and complementation each other.Baicalin was used as a standard substance.Thirteen common peaks were the picture of fingerprints features.The results showed the coincidence of the quality in the total ten batches of the injections. CONCLUSION: The method is stable,accurate and creditable,meanwhile it provides much useful information for the quality control of Shuanghuanglian Powder-Injection.
3.Quality Analysis of 51 Batches of Diclofenac Sodium Sustained-release Capsules from 7 Manufacturers

OBJECTIVE:To investigate the quality of Diclofenac sodium sustained-release capsules testing in Shandong prov-ince in 2014,to provide the reference for improving the homogeneity and stability of product quality. METHODS:51 batches of Diclofenac sodium sustained-release capsules from 7 manufacturers(A,B,C,D,E,F,G)were tested by statutory inspection test in respects of property,identification,content uniformity,release rate,related substance,content determination,etc. The inspection results were analyzed statistically. The content of chromium in capsule shell and release curve of diclofenac sodium in dissolution mediums with different pH were explored and tested by literature method (compared by similarity factor f2 with sample from A). RESULTS:All items of samples were in line with the quality standard,but there was difference in the quality of samples among different manufacturers,such as the content ranged 92.4%-102.1%. In exploration test,the content of chromium was lower than the limit requirement,but the content of diclofenac sodium had great difference among different manufacturers and different batch-es. Compared with the sample from A manufacture,the f2 from B,C,D,E,F,G manufactures ranged 37-86. CONCLUSIONS:Di-clofenac sodium sustained-release capsules are superior in quality. But it is still suggested to strengthen quality study and control, and raise the standard for release rate test.
4.Clinical effect of salmon calcitonin in the treatment of senile osteoporosis and its effect on BMD and VAS scores

Objective To investigate the clinical effect of salmon calcitonin in the treatment of senile osteoporosis , and to observe the effect of salmon calcitonin on bone mineral density ( BMD ) and pain.Methods 180 cases of patients diagnosed as osteoporosis from September 2015 to September 2016 in our hospital were retrospectively analyzed, the patients were divided into control group and treatment group according to the random number table method.The clinical efficacy and side effects of the two groups were evaluated.The bone mineral density (BMD) of the lumbar vertebrae, femur and radius were measured by dual energy X-ray absorptiometry.The VAS was used to evaluate the two groups before and after the treatment.( ODI) were used to evaluate the functional status of the patients before and after treatment.The self-made questionnaire was used to evaluate the quality of life of the patients before and after treatment.Results The total effective rate of treatment group was 94.44%, significantly higher than the control group 75.56%(P<0.05).After treatment, the treatment group of lumbar spine bone mineral density, bone mineral density and radial bone mineral density was significantly higher than the control group (P<0.05).After treatment, spontaneous low back pain, flexion extension pain, turn over pain and weight pain were significantly lower in the treatment group than in the control group (P<0.05).The ODI index of the treatment group was significantly lower than that of the control group (P<0.05).After treatment, the quality of life scores in the two groups were significantly higher than those before treatment, and the treatment group was higher than the control group(P<0.05).No significant adverse reactions occurred in two groups during the treatment.Conclusion Salmon calcitonin treatment of osteoporosis in elderly patients with a significant effect, help to improve bone mineral density, reduce pain and improve its dysfunction, and has high security.
5.Effects of silencing and overexpression of CBX8 on proliferation and apoptosis of human glioma cells

AIM: To explore the effects of chromodomain protein 8 (CBX8) on the proliferation and apoptosis of human glioma cells.METHODS: The expression of CBX8 in the tissues and cells was detected by Western blot and RT-qPCR.The overexpression (Flag-CBX8) and silencing (sh-CBX8) vectors of CBX8 were constructed and transfected into glioma T98G cells and U87MG cells.The cell proliferation was detected by MTT assay and BrdU staining.The cell apoptosis was analyzed by flow cytometry.The protein expression of Rb/E2F1 was detected by Western blot.RESULTS: Compared with normal brain tissues and astrocytes, the expression of CBX8 was increased in the glioma tissues and glioma cells.Overexpression of CBX8 promoted the cell proliferation, inhibited the cell apoptosis, and upregulated the protein levels of Rb/E2F1.On the contrary, silencing of CBX8 inhibited the cell proliferation, promoted the cell apoptosis, and decreased the protein levels of Rb/E2F1 in the T98G cells and U87MG cells.Moreover, the expression of cyclin D1 and Bcl-2/Bax ratio were reduced after transfection with sh-E2F1 in the T98G cells and U87MG cells.CONCLUSION: CBX8 may regulate the proliferation and apoptosis of glioma cells through Rb/E2F1 pathway.
6.Animal models of brain metastasis:preparation methods and research progress

Brain metastasis ( BM) is a common brain tumor in a-dults, originating from extracranial location in the body. There has been a growing interest in producing suitable animal models for studying BM in vivo. Current BM models take peripheral or brain route for tumor cell inoculation, including intra-cardiac or intra-carotid artery injection or orthotopic injection. Direct im-plantation of patient tumor biopsies into rodent brain bears ad-vantages of clinical relevance. This review presents a compre-hensive introduction to key elements for establishing animal mod-els of BM, with highlights on selections of suitable model ani-mals, brain-seeking tumor cell lines, reasonable inoculation routes, as well as succedent phenotyping methods. In the end, perspectives and future directions in this field are discussed.
7.Determination of monosaccaride and disaccaride in condensed Siwu Decoction by HPLC-ELSD

Object To establish a method by high performance liquid chromatography/evaporative light scattering detector (HPLC-ELSD) for the determination of contents of D-fructose, D-glucose, and sucrose in Siwu Decoction (SWD). Methods Condensed SWD was analyzed by HPLC-ELSD after dilution, precipitation by ethanol. Results Condensed SWD, 1 mL, contained (33.4?1.5) mg of D-fructose, (24.2?0.9) mg of D-glucose and (112.7?6.1) mg of sucrose. The standard curves were linear within the range of 0.15—3.75 mg/mL for D-fructose and 0.15—5 mg/mL for D-glucose and sucrose. The recovery rates were 128.5% for D-fructose, 114.7% for D-glucose, and 124.7% for sucrose. The relative standard deviations (RSD) within-days were 3.0% for D-fructose, 3.2% for D-glucose, and 4.4% for sucrose. Conclusion SWD contains abundant monosaccharide and disaccharide. HPLC-ELSD can be used to analyse the monosaccharide and disaccharide in SWD.
8.Fixation by S_1 pedicle screws combined with iliac screws to treat sacroiliac joint dislocation and fracture

Objective To evaluate the clinical effectiveness of the combined use of S1 pedicle screws and the iliac screws in treatment of sacroiliac dislocation and fracture. Methods 11 patients with sacroiliac dislocation and fracture were treated with S1 pedicle screw and iliac screw fixation technique. In this group, the dislocation was associated with vertical displacement of the sacroiliac joint and rotational deformity of the pelvic ring. They were classified as type B or type C pelvic disruption according to the Tile's classification. 7 patients with disruption of the symphysis pubis or pubic branch fracture underwent additional fixation of the pelvic ring using a reconstruction plate. The remaining 4 patients were treated by the posterior procedure alone. Results The vertical displacement was completely reduced in 7 patients, and the rotational deformity completely corrected in 4 patients. The reduction was maintained at the time of final follow up evaluation. No perioperative complications were found. The combined use of S1 pedicle screws and the iliac screws provided immediate stability and sufficient reduction for sacroiliac dislocation in the 11 patients in this study. Conclusion This hybrid internal fixation procedure is useful for reduction and fixation of sacroiliac dislocation associated with the vertical and rotational instability of the pelvic ring.
