1.Identification of the reference genes' accuracy for lung tissue specimens by real-time quantitative PCR

Objective To analyze the expression stability of the three widely used reference genes β-glueuronidase (GUSB), glycera]dehydes-3-phosphate dehydrogenase (GAPDH), β2-microglobulin (β2-M) in Chinese lung cancer tissue specimens and normal lung tissue specimens. Methods Gene expression wasmeasured by quantitative real time PCR and expression stability was analyzed with two widely used softwares genorm and normfinder. Results The intra-and inter-group difference of GAPDH is maximum (The intra and inter-group s is 1.07 and 0.93 respectively, |△ Ct|=2.01±1.06; P =0.000). The mean of these three genes' Ct value is the most stable one analyzed by the two softwares. But the t test showed that the mean of Ct value of GUSB and β2-M is the unique combination that had the minimum intra-and inter-group difference, with no statistically significant differences between normal and malignant samples (The intra-and inter-group s is 0.53 and 0.79 respective]y, |△Ct|= 0.73±0.53; P =0.053). Conclusion It is inappropriate to normalize data derived from lung tissue specimens using one of these three housekeeping genes alone. Among the different combinations of these three genes, the mean of the Ct values of GUSB and β2-M is the best choice as the internal control of lung tissue specimens.

4.Prediction and analysis of T-cell and B-cell epitopes of Mycobacterium tuberculosis Rv2657c protein

Objective To predict the epitopes of Mycobacterium tuberculosis Rv2657c protein, to understand its immunogenicity Methods The T?cell and B?cell epitopes of Mycobacterium tuberculosis Rv2657c protein were predicted by DNAStar software package. The homology of Rv2657c amino acid sequence with the human protein sequences was prepared using Blast method, then the CTL epitopes were predicted using SYFPEITHI supermotif method, BIMAS quantitative motif method and NetCTL prediction method, and the Th epitopes were predicted by RANKPEP and SYFPEITHI supermotif prediction method. Results The prediction using DNAStar software package showed that Rv2657c protein had 5 B?cell epitopes and 6 T?cell epitopes. The protein had 6 CTL epitopes and there were 38 Th epitopes. Conclusion Rv2657c protein has both B?cell epitopes and T?cell epitopes. It may be a candidate target antigen for the studies of vaccine and diagnosis of tuberculosis.

6.Determination of residual EDTA-2Na in Japanese encephalitis attenuated live vaccine by capillary electrophoresis with electrophoretically mediated microanalysis

Objective: To establish a capillary electrophoresis (CE) with electrophoretically mediated microanalysis (EMMA) method for the determination of EDTA-2Na in Japanese encephalitis attenuated live vaccine.
Methods: The test was performed in disodium hydrogen phosphate buffer with pH 2.5, the online metal ions complexation of 1.5 mg·mL-1 Fe3+ and incubation time of 3 min. The separation voltage was 25 kV, the detection wavelength was 257 nm, and. the column temperature was 25.0 ℃.
Results: The established method had a good linear relationship in the concentration range of 0.01-0.5 mg·mL-1 (r=0.999 9), the detection limit was 5 μg·mL-1, and the relative standard deviation (RSD) of the measured samples was less than 2.87%. The recoveries of spiked samples were between 96.49%-101.02%.
Conclusion:The optimized method was applied to the determination of EDTA-2Na in Japanese encephalitis attenuated live vaccine. The satisfactory experimental results were obtained.
7.Research Review of the Action Mechanism of Acupuncture Based on Cell Apoptosis

Apoptosis refers to the programmed death process of cells modulated by genes. It plays an important role in promoting the evolution of organisms, regulating the development of multiple systems, and maintaining the stability of internal environment. The major apoptosis pathways include mitochondrial pathways, death receptor pathways, and endoplasmic reticulum pathways. With the rapid development of the scientific research on acupuncture, it’s revealed that cell apoptosis should be closely related to the action of acupuncture. So far, the major apoptosis factors involved in relevant researches are P53, Bcl-2 family, Cyt-C, Caspase, Fas/Fasl, TNFR1/TNF-?, and FADD-Caspase8, and PERK, etc. Via influencing these apoptosis factors or genes, acupuncture can produce various effects in improving tissue ischemia, protecting neural cells, reducing inflammation, and correcting endocrine disorders. This article summarized and reviewed the apoptosis pathways and key factors related to the action of acupuncture, to provide some beneficial references for future studies.
8.The relationship between calcaneal bone mineral density and bone mineral, fat and muscle content in adolescents

Objective:
To investigate the correlation between calcaneal bone mineral density and bone mineral, fat and muscle content in adolescents.
Methods:
A total of 368 adolescents who received health examination in Hainan People’s Hospital were selected as the study subject. Calcaneal bone mineral density, bone mineral content, fat content and muscle content of adolescents were measured. Bone mineral density and body composition of adolescents of different ages and sexes were compared. The correlation between calcaneal bone mineral density and bone mineral content, fat and muscle content were analyzed.
Results:
The BUA, muscle mass and bone mineral content of boys were significantly higher than those of girls, and the fat content of boys was significantly lower than that of girls, the differences were of statistical significance(t=13.51, 10.65, 4.52, -7.55, P<0.05). Calcaneal bone mineral density in adolescents was positively correlated with BMI, bone mineral content and muscle mass(r=0.39, 0.42, 0.69, P<0.05). There were significant differences in BUA, BMI, muscle mass and bone mineral content among boys of different ages(F=7.95, 8.63, 6.96, 5.01, P<0.05). There were significant differences in BUA, fat, muscle and bone mineral contents among girls of different age groups(F=8.65, 10.33, 7.96, 4.87, P<0.05). There was no significant correlation between BUA value of calcaneus and BMI and muscle mass in adolescents aged 12, 13-year old(P>0.05), while BUA value of calcaneus in adolescents aged 14-year old, 15-year old and 16-year old was positively correlated with BMI and muscle mass(P<0.05).
Conclusion
Calcaneal bone mineral density in adolescents is closely related to bone mineral and muscle content, but not to fat content.
9.Dosimetric study of influence of intensity modulated radiotherapy for nasopharyngeal carcinoma on hippocampal formation

Objective To evaluate the dosimetric characteristics of hippocampal formation (HF) in the intensity-modulated radiotherapy (IMRT) for nasopharyngeal carcinoma (NPC).Methods Fifty-nine NPC patients underwent IMRT.Simultaneous integrated boost technology was used to determine the doses for the target areas.The dose ranges of the HF were collected by dose-volume histogram.The influence of T stage on the exposure doses ( Dmax,Dmean,D20,V10,V20,V30,and V40 ) were compared.Results The maximum dose for the HF (Dmax) ranged from 11.1 to 78.2 Gy(F =24.2,P <0.05) and the Dmean ranged from 3.2 to 44.6 Gy ( F =16.3,P < 0.05 ).The Dmax and Dmean of the T1-2 stage patients were (40.8 ±9.4) Gy and ( 12.5 ±5.1 ) Gy,respectively,both significantly lower than those of the T3-4 stage patients [ (58.6± 14.8) Gy,(20.9± 9.3 ) Gy].The mean exposed volume of the T4 stage patients was significantly larger than that of the T1 and T2 stages patients.Conclusions In the IMRT of NPC,the HF receives rather high irradiation dose.T stage is the main factor influencing the dose,especially T3 and T4 stages deserve serious attention.
10.Primary leiomyosarcoma of the vagina: a clinical analysis of 9 cases

Objective To analyze the clinical characteristics,diagnoses,treatments and prognoses of vaginal leiomyosarcoma (LMS).Methods The clinical and pathological recordings of 9 patients suffering from vaginal leiomyosarcoma from January 1973 to May 2011 were analyzed retrospectively.Results The study group constituted 2.7% (9/330) of all the malignant vaginal tumor admitted to our hospital in the same period.The major clinical manifestations were asymptomatic vaginal mass (3/9),bellyache and vaginal bleeding (2/9),vaginal pain (2/9),and difficulty in defecation (1/9) and micturition (1/9).Preoperative diagnosis was highly unreliable.Based on the sample chose,the diagnosis were made in 3 cases by preliminary biopsy before undertaking surgery,while 6 cases were confirmed shortly after simple tumor excision or unexpected recurrence.All the patients were pathologically confirmed diagnosis by tumor resection specimen or biopsy,5 cases of them were also confirmed by virtue of immunohistochemistry staining.All 9 patients,8 cases received initial surgery,four of them patients received postoperative adjuvant chemotherapy or radiation therapy,the remaining 1 patients received initial chemotherapy and radiotherapy.Follow-up was performed for all the 9 patients,of which 3 cases were lost,the median follow-up time was 50 months (range 7-134).Four patients underwent local recurrence and adjacent organ metastases within two years,with 5 years survival rate 4/9.Conclusions Primary vaginal leiomyosarcoma is a rare and difficult to diagnose preoperatively tumor.In this regard,It is strongly recommend preoperative biopsy for definitive diagnosis and perform surgery as the principal approach,in conjunction with radiotherapy or chemotherapy when needed to improve survival.
