1.Monitoring results and epidemic analysis of gerbil plague in northern Ningxia in 2009

Objective To analyze gerbil plague in northern Ningxia and the monitoring results,to master the plague epidemic dynamics,and to provide the basis for developmenting countermeasures.Methods Monitoring data of gerbil plague focus in northern Ningxia were collect in 2009,counted main host density,rate of dye fleas,flea body index and bacteriology,serology detect and analyzed the epidemic situation.Results An average density of main host was 1.74/hm2,the average rate of infected fleas was 28.60%,and the average rat body flea index was 0.76.Monitoring found 4 plagues from rat plague epidemic,plague bacteria were found in 16 strains,of which gerbils inspection bacteria 10 strains,Meriones unguiculatus 2 strains,the same type cheopis 2 strains,and bald disease fleas 2 strains.Indirect hemagglutination (IHA) was used to test 529 copies of samples,2 copies were found positive,and hemagglutination-positive rate was 0.38%; eight copies were examined by reverse indirect hemagglutination(RIHA),7 material were found positive,and hemagglutination-positive rate was 87.50%.Conclusions In recent years,the disease is active among animals in gerbil plague focus Ningxia.The density of rodents is higher,and local plague epidemic is found in rats.Monitoring efforts should be strengthened and scope of monitoring should be expanded.We should pay close attention to the epidemic dynamics,control the prevalence and spread of animal disease,and prevent the occurrence of human plague.
2.Correlation of 5-HTTLPR genetype and behavioral problems in school-age twins

Objective To analyze the correlation between 5-hydroxytryptamine transporter gene linked polymorphic region(5-HTTLPR) and twin children's behavioral problems and its interaction with environmental factors.Methods A total of 147 pairs of school-age twins aged 6-12 years were enrolled.The behavioral problems were assessed with the Child Behavior Checklist (CBCL).The genotype of 5-HTTLPR was detected by the method of PCR.The correlation of 5-HTTLPR and children's behavioral problems was performed using generalized estimating equation.Results (1) The overall incidence rate of behavioral problems in school-age twins was 24.15% (27.94% in the boys;20.89% in the girls),in which the rate of thinking problems (15.31%) was the highest,and the rates of other problems were lower (3.40%-8.16%).(2) The results of correlation analysis showed that 5-HTTLPR was only related to the social problems (P< 0.05).The S allele of 5-HTTLPR maybe associated with the social problems.(3) The results of interaction of 5-HTTLPR with twin factors of themselves and family environmental factors showed that 5-HTTLPR was the main effect of the anxious/depressed problems,and S allele carriers were easier to occur anxious/depressed problems.Gender was the main effect of the social problems,and boys were easier to occur social problems.There was no significant interaction between 5-HTTLPR and twin factors of themselves and family environmental factors.Conclusion 5-HTTLPR genetype may have some relevance to the social and anxious/depressed problems,and S allele may be a susceptibility gene of social and anxious/depressed problems.
3.Inhibitory effects and mechanisms of snake venom tripeptide pENW on platelet adhesion.

This study was designed to investigate inhibitory effects and possible mechanisms of snake venom tripeptide (pENW) on platelet adhesion in order to promote the development of a novel anti-platelet therapy. To study the inhibitory effects of pENW on platelet adhesion, washed platelets pre-incubated with pENW (116.5-466.2 μmol x L(-1)) were used to test the ability of platelet adhesion to fibrinogen. Effect of pENW on fibrin clot retraction was also tested. Effect of pENW on platelets viability was tested by MTT assay. Effect of pENW on reactive-oxygen species (ROS) levels of platelet was studied by flow cytometry assay. Calcium mobilization in Fura-2/AM-loaded platelets was monitored with a spectrofluorimeter. Cyclic guanosine monophosphate (cGMP) and cyclic adenosine monophosphate (cAMP), thromboxane A2 (determined as its metabolite thromboxane B2) were measured using enzyme immunoassay kits. Akt, ERK and p38 phosphorylation were tested by Western blot. The results showed that pENW inhibited platelet adhesion and fibrin clot retraction in a concentration-dependent manner without cytotoxicity. Intracellular cGMP and cAMP in both resting and thrombin-activated platelets were increased by pENW. In addition, pENW attenuated intracellular Ca2+ mobilization and TXA2 production in platelets stimulated by thrombin. As shown by Western blot assay, Akt, ERK and p38 phosphorylation in thrombin-induced platelet were attenuated by pENW. However, inhibitory effects of pENW had nothing to do with ROS. Thus, pENW exhibited a significant inhibition on platelet adhesion to fibrinogen, which means pENW could block the first step of thrombosis as while as retard the more stable clot formation. The mechanisms of pENW on inhibition platelet adhesion might be related to instant regulations, such as protein kinases.

4.Factors influencing functional recovery in patients with spinal cord injury at discharge

Objective To explore the factors influencing the functional recovery of patients with spinal cord injury (SCI) at discharge. Methods A total of 105 patients with SCI admitted to the rehabilitation medicine department at Huashan Hospital between December 2004 and October 2009 were studied. Data on eleven variables were collected including the patients' medical history, physical examination results and American Spinal Injury Association (ASIA) scores at admission. Functional status was registered according to the modified Barthel index (MBI) assessed at admission and before discharge. Linear regression analysis was used to assess the influence of the variables.Results After rehabilitation, average MBI and ASIA scores were significantly higher. The multiple regression analysis revealed that injury grade, motor and pinprick sensation scores at admission were related to MBI before discharge.The duration of rehabilitation, the rehabilitation treatment course and motor scores at admission were related to MBI increases during hospitalization. Conclusions Patients with different ages, injury levels and severity can improve their functional abilities through rehabilitation treatment, especially patients with better initial motor ability, longer treatment and earlier intervention.
5.Study on correlation of genetic polymorphism of PRKAA2 with metformin response

Aim To study the single nucleotide poly-morphism ( SNPs ) of AMP-activated protein kinase ( AMPK) ?2 subunit gene PRKAA2 ( rs2051040 and rs17848595) and its relationship with metformin response. Methods A total of 32 patients with type 2 diabetes( T2DM) were enrolled. All patients were required to take metformin for 1 week. The serum levels of FPG,TG,TC,LDL-C,HDL-C were assayed before and after therapy. The gene polymorphism of PRKAA2 was analyzed by PCR-DGGE,the effects of metformin were compared between patients with different phenotypes. Results FPG,TG,TC,LDL-C were significantly improved after therapy in wild genotype carriers( P 0. 05) . Conclusion The results of this study suggest that PRKAA2 polymorphism may be associated with metformin treatment effects in T2DM patients.

7.Optimum process for Kangfuxin Drop Pill by orthogonal design

AIM: To selecte the best process for Kangfuxin Drop Pills. METHODS: With appearance,weight coefficient of variation and dissolution time for index sign,orthogonal design was applied. RESULTS: The results indicated that the excellent factors and levels were as follows: the scale between PEG6000 and drug extraction was four to one,the temperature of drug fluid was at 85 ?C,drip rate was 40 drops/min. CONCLUSION: The method adopted is steady and feasible.
8.Perioperative nursing of patients undergoing resection of rectal neoplasms with transanal endoscopic microsurgery

Objective To introduce the application and the surgical cooperation of the transanal endoscopic microsurgery in resection of rectal neoplasms and summarize the nursing points.Method The clinical histories of 135 patients undergoing resection of rectal neoplasms with transanal endoscopic microsurgery were reviewed and analyzed.Results All the surgeries were successful. Four patients developed with wound infections,three of them getting better after treatment and one of them diagnosed with poorly differentiated adenocarcinoma and selected another operation.Conclusion Well-prepared、pre-operation,skillful operation and instrument procedure and postoperative nursing were critical to the success of operations.

10.Changes of Akt mRNA expression in rat pulmonary arterial smooth muscle cells induced by hypoxia

Objective To observe the changes of Akt1,Akt2,Akt3 mRNA expression level in rat pulmonary arterial smooth muscle cells(PASMC) induced by hypoxia and to investigate the value of Akt signaling pathway in vascular reconstruction during phyoxia pulmonary hypertension(HPH).Methods The pure PASMCs were established by tissue-sticking methods.Semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) were used to examine the Akt1,Akt2 and Akt3 mRNA expression after PASMCs were exposed to normoxia(21%O_2) or hypoxia(3%O_2) for 2,8,12 and 24 h respectively.Results The pure PASMCs were established.The mRNA of Akt1,Akt2,Akt3 could be detected in all groups.Exposed to hypoxia,the mRNA expression levels gradually increased,reached the peak at 8 h,then decreased,and at 24 h down to the level under normoxia.As compared with that exposed to normoxia,the mRNA expression levels of Akt1 at 2,8,12,24 h,Akt2 at 8 h,Akt3 at 8 h in PASMCs exposed to hypoxia showed significant difference(P
